1.Observation of the Efficacy of Astragalus Injection in Skin of Aging Mice Induced by D-galactose

Objective To investigate the efficacy of acupoint injection of Astragalus on contents of SOD,HYP and MDA in skin of aging mice in-duced by D-galactose. Methods The mice were randomly divided into normal group,model group,Astragalus group,and saline group(n=15). Except for the normal control group,the mice of other groups were treated with continuous subcutaneous injection of D-galactose to establish mice ag-ing model. The Astragalus group mice was injected with 0.01mL Astragalus in Sanzuli acupoint per day,The saline group mice received an injection of same amount of saline. SOD,HYP and MDA changes in the skin tissue and serum were evaluated after 21 days. Results All the index in the skin tissue and serum showed significant difference(P<0.01)between model group mice and normal control group. Compared with model group,SOD activity and MDA content in the skin tissue and serum of the Astragalus group mice were significantly raised,HYP content was significantly reduced (P<0.01). There also were significant differences of these index between the saline group mice and the model group(P<0.05),but the effect was not as good as the Astragalus group mice. Conclusion Acupoint Injection of Astragalus can prevent and protect against skin aging induced by D-galactose.
2.The expression of serum squamous cell carcinoma antigen in patients with common skin diseases

Objective There is a significant increase of squamous cell carcinoma antigen ( SCCA) in patients with common skin diseases .The aim of this study was to investigate the difference expression of serum SCCA in patients with common skin diseases . Methods A total of 497 patients with 14 kinds of common skin diseases from the Department of Dermatology were recruited , and the serum SCCA was measured . Results The positive rates of SCCA in 116 patients with psoriasis and 37 patients with erythroderma were 90 .5% and 100%, respectively .The common skin diseases with positive rate of SCCA greater than 60% includes eczema (61.7%), generalized dermatitis (65.1%),pemphigus (61.1%), bullous pemphigoid (62.2%).The SCCA values in pustulosa (1.76 ±1.22 ng/mL) and arthropathica psoriasis (1.69 ±1.05 ng/mL) were decreased compared with pustular (2.86 ±1.46 ng/mL) and arthropathica psoriasis (3.56 ±1.84 ng/mL) (P<0.05). Conclusion The SCCA value may have a relationship with the increase of the performance of keratinization and desquamation in the skin diseases , and it could be used as a valuable indicator for the diagnosis and treatment in patients with cutaneous diseases .
3.Changes of drug resistance of Acinetobacter baumannii isolated from our hospital in recent three years

Objective To analyze the changes of drug resistance of Acinetobacter baumannii in our hospital during 2010-2012 to provide reference for clinical rational drug use and the effective control of Acinetobacter baumannii infection .Methods The drug resistance of Acinetobacter baumannii clinically isolated from 2010 to 2012 and its infection situation were retrospectively performed the statistical analysis .Results The clinical isolation rate of Acinetobacter baumannii in our hospital was increased from 218 cases (12 .19% ) to 741 cases (18 .48% ) in 2012 .The resistance rates of 18 kinds of antibiotics were almost more than 70% ,and pan-drug-resistant Acinetobacter baumannii appeared in some clinical departments ,such as ICU and neurology .The selective pressure of antibiotic was one of the main motive causes of drug resistance change ,and age ,underlying diseases ,duration of hospitalization and invasive procedures were the high risk factors in patients infection .Conclusion Hospital should enhance a series of measures including reinforcing the disinfection of the medical environment ,medical equipments and medical staffs ,strengthening the monito-ring of hospital infection and using antibiotics rationally to reduce and control the prevalence of Acinetobacter baumannii .
4.Observation and Application of Oral Ambroxol in Improving Positive Rate of Tuberculosis Laboratory Test by Sputum Smear

Objective To research the application of oral ambroxol in improving positive rate of sputum smear laboratory testing of Tuberculosis.Methods After immediate microscopic phlegm examination on 1 536 tuberculosis outpatients,them were divided into two groups alternatively and randomly.The referential group took oral placebo while the test group took oral ambroxol.Recheck would be conducted onmorning phlegm after three days,then comprated the positive rate of each group.Results The positive rates of immediate microscopic phlegm examination of the referential and test groups were 4.82% and 4.56% (x2 =0.058,P>0.05) respectively.For the referential group,the positive rates before and after taking medicine were 4.82% and 5.60% respectively (x2 =0.475,P>0.05).The differences had no statistic meaning.The positive rates of morning microscopic phlegm examination after taking medicine of the referential and test groups were 5.60% and 8.59 % ()x2 =5.224,P<0.05) respectively.The positive rates of the test group before and after taking medicine were 4.56 % and 8.59% respectively (x2 =10.18,P<0.05).It could be deemed that the difference had statistic meaning.Conclusion O-ral ambroxol can help raise the positive rate of microscopic phlegm examination of the patient with suspected tuberculosis.
5.Observation on serumγ-glutamyl transpeptidase levels of patients with sub-clinical schistosomiasis before and after pathogen treatment

Objective To observe the changes of serumγ?glutamyl transpeptidase(GGT)levels before and after the patho?gen treatment in patients with subclinical schistosomiasis,and explore its clinical value in the diagnosis and treatment of sub?clinical schistosomiasis. Methods Totally 109 patients with subclinical schistosomiasis,who were found in the endemic inves?tigation of schistosomiasis in Ezhou City,were selected as the investigation subjects,and then they were treated with praziquan?tel. The serum GGT levels of the subjects before and after the treatment were detected and compared. Results Before the treat?ment,the average value of the GGT levels of the 109 patients was(48.1 ± 45.9)IU/L,among which,the GGT levels of 69 cases (63.3%)were normal,and the levels of 40 cases(36.7%)were increased. After the treatment,the average GGT level of the pa?tients was(32.1 ± 23.4)IU/L,which decreased by 33.3%comparing with that before the treatment,and the difference had a statistical significance(U=2.17,P=0.01). The GGT levels of 65 patients decreased in different degrees. Among the 40 pa?tients whose GGT levels had increased before the treatment,the GGT levels of 31 ones returned to the normal. Conclusion The GGT level detection can accurately reflect the liver function in the patients with subclinical schistosomiasis ,and also it has certain clinical application value to judge the liver function damage and recovery of the patients before and after the pathogen treatment.
6.Botulinum-A toxin injection into detrusor to treat detrusor hyperreflexia and neurogenic incontinence in patients with spinal cord injury

Objective To evaluate the effects of Botulinum-A toxin (BTX-A) injection into detrusor to treat detrusor hyperreflexia and neurogenic incontinence in the patients with spinal cord injury (SCI). Methods A total of 31 patients with SCI(mean age,30 years;male 20,female 11) were included into the study.All the patients underwent urodynamic examination and voiding diary was recorded.300 U of BTX-A was dissolved in 15 ml of saline,and the solution of BTX-A was injected into 30 different points in detrusor using a flexible cystoscopic needle.The evaluation for the effects and follow-up included voiding diary,urodynamic testing and observation of adverse and toxic effects. Results After the first injection,29 of the 31 patients had symptom relief within 3 to 14 days;4 cases received the second injection;4 cases were treated in combination with anticholinergic medication;and 2 cases had no improvement and,therefore,received other treatments.The mean follow-up was 8.5 months. After 3-week treatment of BTX-A,the mean frequency of incontinence decreased from 14.2 to 2.5 times per day.The mean volume of intermittent catheterization (IC) increased from 124 to 495 ml each time.Urodynamic data showed that mean cystometric bladder storage volume increased from 133 to 475 ml,mean maximum storage detrusor pressure decreased from 62.7 to 17.1 cmH 2O(1 cmH 2O=0.098 kPa).No adverse and toxic effect was observed. Conclusions Our initial experience suggests that Botulinum-A toxin injection into detrusor to treat detrusor hyperreflexia and neurogenic incontinence in patients with SCI may be an effective,safe,feasible and micro-invasive treatment choice.However,it is necessary to observe its long term outcome.
7.Role of transient receptor potential vanilloid 1 channel in pancreatitis

Transient receptor potential vanilloid 1 (TRPV1) is a non-selective cation channel,which can be activated by multiple pathways during the course of the diseases.Recent studies indicate that primary sensory neurons of the pancreas express TRPV1 receptor and the activation of TRPV1 receptor promotes pancreatic inflammation.Moreover,blockade of these transient receptor potential channels can greatly ameliorate the pain response in experimental pancreatitis.
8.Application of abdominal puncture in ventriculoperitoneal shunt

Objective To introduce a new method that abdominal puncture is used in ventriculoperitoneal shunt. Methods A retrospective review of the clinical materials of 49 patients who underwent improving ventriculoperitoneal shunt.Results After operations, all the patients had fellow-up survey that the rates of infection and obstruction are both 4.08%. They are lower than that of the lectures. Conclusion It has many advantages that abdominal puncture taken the place of incision of abdominal cavity. They are less wounded, less infection, less abdominal complication , not easy to be packed by the omentum and so on.
9.Observation on Relationship Between Different Serum Markers Model for Patients with Hepatitis B Virus Infection and HBV-DNA Quantitative Analysis

Objective To study the relationship between different serum markers model for patients with HBV infection and content of HBV-DNA,to provide evidence of diagnosis,treatment and provention of hepatitis B.Methods Determinations of HBV serum markers and HBV-DNA content by ELISA and real time fluorometric quantitative-PCR technique respectively in 814 sera samples.Results HBV-DNA positive rate was 87 28%(302 of 346 coses) in HBsAg(+),HBeAg(+),HBcAb(+) model.HBV copy quantity ≥10 8/ml accounted for 63 87%.HBV-DNA positive rate was 48 778%(40 of 82 cases) in HBcAb positive model HBV-DNA copy quantity in the partial samples was higher,it can occure in gene mutation of HBV precore regions,resulting in the experssion of HBeAg was failed,but virus replication was not influenced.The HBV-DNA positive rate and copy quantity in HBeAg(+) model was obviously high than those of other different serum markers model.Conclusion Serum marker model only applied to judg the degree of HBV replication and its infectivity were considerable diffecult,HBV-DNA quantitative determination could real reflect HBV replication condition,provede objective evidences of diagnosis,prevention and treatment of HBC hepatitis.
10.The Effect of overexpressed Daxx in Liver Tumor Cells on The apoptosis Induced by Oxidative Stress

In order to study the effects and the possible mechanisms of Daxx overexpressed in HepG2 to hydrogen peroxide treatment, and to search new targets for cancer chemotherapy, HepG2cells were transfected using lipofectamine 2000, and selected by treatment with G418. Stable cell lines were confirmed by reverse transeriptase polymerase chain reaction (RT-PCR) targeting vector gene. Experiments include the following groups: (1) control group (non-transfected cells); (2) transfected with empty vector (HepG2/GFP cells); and (3) transfected with pEGFP-C1-Daxx (HepG2/GFP-Daxx cells). After incubation with hydrogen peroxide (H2O2) for 24 h, cellular viability was analyzed by MTT, and cellular apoptosis was measured by flow cytometric analysis. Gene expression at protein level was detected by Western blot. The RT-PCR results showed that Daxx RNA in cells transfected with pEGFP-C1-Daxx was increased significantly compared with that in the HepG2/GFP cells. Fluorescence microscopy revealed that Daxx protein was localized in the nuclei. Hydrogen peroxide was used to induce apoptosis of HepG2 cells and observed that the hydrogen peroxide decreased the viability of HepG2 cells in concentration-dependent pattern. The IC50 values in three groups (Normal cells, HepG2/GFP cells and HepG2/GFP-Daxx cells) were 0.72, 0.76, and 0.49 mmol/L respectively. The apoptotic ratio was significantly higher in HepG2/GFP-Daxx cells as compared to the other two groups. HepG2/GFP-Daxx cell incubated with hydrogen peroxide, showed a significant increase in the activation of caspase-3 and JNK as compare with the other groups. Over-expression of Daxx facilitated HepG2 cells apoptosis induced by hydrogen peroxide. Furthermore, there may be a synergetic relation with apoptosis and increase of JNK activity.
