1.Evaluation of left ventricular function in children with dilated cardiomyopathy using quantitative tissue velocity imaging(QTVI)

Objective:To evaluate the application of Quantitative tissue velocity imaging(QTVI) in assessing the left ventricular (LV) systolic function in Children with dilated cardiomyopathy(DCM).Methods:Fifteen consecutive children with DCM were included in the present study,who were divided into group A(with light heart failure) and group B(with moderate and severe heart failure).Twelve children without any cardiac diseases were as controls.The dynamic tri-plane QTVI images of apical four-chamber,LV two-chamber and LV long-axis views were acquired simultaneously and the myocardial velocity curves in basal and middle segments were analyzed.Results:(1)The peak systolic velocity (Vs)redueed generally in group A and B compared with that in controls,especially in group B(P0.05).(2)There were forty-two segments with the velocity of postsystolie shortening(PSS) more than that of the systole in DCM group,among which thirty segments(71.43%)were from group B,and twelve segrnents(28.57%)from group A. Analyzed with strain,twenty-one segments (50.00%)were actively shortened and eleven segments (26.19%)were paradoxical motion.Conclusion:QTVI can assess LV systolic function accurately in Children with DCM.
2.Analysis of bacteriology of drug sensitivity in the inactive stage of chronic suppurative otitis media.

OBJECTIVE:
Detecting the sensitivities of bacterium in the inactive stage of chronic suppurative otitis media, to guide a clinical rational drug use after the type I tympanoplasty.
METHOD:
Collecting 148 hospitalized patients from Guangdong General Hospital diagnosed as inactive stage of chronic suppurative otitis media, who had been dry ear for more than 1 months. Analyse the results of drug sensitivity on the mucosa surfaces of the middle ear.
RESULT:
There are 16 species of bacterium, including Staphylococcus aureus, Pseudomonas aeruginosa, Enterobacter cloacae etc. Total positive rate of bacteria was 33.1%, the detectable rate of gram-positive bacteria was 69.4%, gram-negative bacteria was 30.6%. Drug sensitive test demonstrated that vancomycin, linezolid, teicoplanin, rifampicin etc were effective in gram positive bacteria, piperacillin/tazobactam, cefoperazone/sulbactam etc were effective in gram-negative bacteria.
CONCLUSION
Patients with dry ears,who were going to take type I tympanoplasty, still had potential pathogens. Bacterial cultivation and drug sensitive test before operation can guide the clinical medication and prevent infection.

3.Effect of Osthole on Mast Cells and Expression of STAT5 Gene and Protein in Mice with Eczema

Objective To explore the effect of osthole on mast cells and expression of STAT5 gene and protein in them. Methods Passive cutaneous anaphylaxis was made in mice to copy eczema model, then the allergic mast cells were separated, and the ovalbumin was used to induce allergy of mast cells. Different concentrations of osthole were used to intervene the sensitized mast cells.Then the sensitized mast cells were divided into blank control group, osthole high-dose group and low-dose group.At the end of the experiment, morphology of the mast cells was detected by immunofluorescence technology.MTT assay was used to detect the influence of drugs on mast cells proliferation. RT-PCR and Western blotting were used to detect the expression of STAT5 gene and protein. Results As compared with blank control group, the number of mast cells in the osthole groups was significantly reduced, cells and nuclei obviously shrank, even apoptosis of some cells could be observed; the inhibition rate of mast cells in osthole groups was significantly increased in concentration-dependent manner ( P<0. 01 ) . As compared with blank control group (2.16±0.57), gene expression of STAT5 was significantly decreased in osthole high-dose group (0.59±0.12) and low-dose group (0.82±0.13) (P<0.01).The protein expression of STAT5 was also significantly decreased in osthole high-dose group and low-dose group as compared with blank control group (P<0.01). Conclusion Osthole can inhibit the proliferation of sensitized mast cells, and reduce the expression of STAT5 gene and protein.
4.Expression of KAI1 gene in cervical cancer and its clinical significance

Protein transduction is an emerging technique in biological fields in recent years. Through the medium of protein transduction domain(PTD) composed by a small number of alkaline peptides, the functional proteins linked covalently with DNA, peptides, proteins or other macromolecules can across the membranes of mammalian cells, even the blood-brain barrier and transfer unconventionally that isn't like classical pathway. The distinct property of PTD that can take along fused protein entering into cells unconventionally offers a new vehicle to gene therapy for some diseases and has dulcet applied perspective.
6.Effect of decorin on proliferation and cell cycle of rabbit tendon cells in vitro

Objective To investigate effect of decorin on proliferation and cell cycle of rabbit tendon cells in vitro so as to explore the role of decorin in tendon wound healing.Methods Tendon cells derived from the tissue of rabbits flexor tendon were harvested and cultured in vitro with decorin of 0.25,1.25,2.5,5 ?g/ml.After culture of 12,24 or 48 h,the cell proliferation rate was measured by MTT colorimetric determination.After 24-hour culture with 0.25?g/ml decorin,the morphology of tendon cells was obtained and the cell cycle distribution was detected by flow cytometer.Results The proliferation of tendon cells was inhibited after 12-hour culture and significantly increased after 24-hour culture with 0.25,1.25,2.5 ?g/ml decorin.However,5 ?g/ml decorin could increase the proliferation after 12-hour culture,increase after 24-hour culture,with no significant difference between 24 h and 48 h.decorin at 0.25 ?g/ml could significantly increase the cells at S phase and PI after 24-hour culture(P
7.Cloning and sequencing of the CTX-M-12 encoding gene of ?-lactamase

Objective To investigate the ESBL gene in a Klebsiella pneumoniae strain isolated from a patient in Huashan Hospital, Shanghai. Methods ESBL gene in the transconjugant from Klebsiella pneumoniae isolate was amplified by PCR. The PCR products were cloned into pHSG398 vector. The antibiotic susceptiblity and ?-lactamases activity of the clone strain were tested; The PCR product was sequenced and analyzed. Results The ESBL gene was identified as CTX-M-12 gene and CTX-M-12 is much more active against cefotaxime than against ceftazidine.Conclusions A clinical isolate of Klebsiella pneumoniae k99-442 isolated from a patient in Huashan Hospital, Shanghai produced ESBL type CTX-M-12 which caused this isolate resistant to most ?-lactams.
8.Laparoscopic and open treatment for choledocholithiasis:a prospective clinical comparative study

Objective To compare the therapeutic results of laparoscopic and open treatment for choledocholithiasis. Methods A prospective comparison was conducted between 29 cases of laparoscopic choledocholithotomy and 35 cases of open choledocholithotomy in terms of stone removal,operative time,hospitalization day,hospitalization cost and postoperative complications. Results Stone removal was achieved in 28/29 patients by a laparoscopic approach and in 32/35 patients by a open approach( ? 2=0 71, P =0 399);average operative time was (178 3?32 4) min in laparoscopic approach and (145 8?27 1)min in open approach( t =4 27, P =0 01);the patients in laparoscopic procedures had flatus in (20 8?2 9) hour and those in open procedures did in (58 2?4 9) hours( t =37 8, P 0 05);no major complication was found in laparoscopic approach. Conclusions Laparoscopic choledocholithotomy is safe and effective and it is superier to open treatment.
9.Application of temporary cardiac pacemaker in perioperative period of laparoscopic cholecystectomy

Objective To evaluate the safety of temporary cardiac pacemaker during perioperative period of laparoscopic cholecystectomy (LC) in patients with bradyarrhythmia. Methods Temporary cardiac pacemakers were installed preoperatively in 34 elderly patients with bradyarrhythmia for LC. On operation the ultrasonic harmonic scalpel, instead of electrotome, was used to perform dissection and coagulation. Results LC was completed successfully in all the 34 patients without surgery-related complications or perioperative serious cardiovascular diseases. Conclusions Utilization of temporary cardiac pacemaker and ultrasonic harmonic scalpel may considerably enhance the perioperative safety and reduce the incidence of cardiovascular complications in elderly patients with bradyarrhythmia.
10.P300 Wave Extraction by Wavelet Transform Combined with Independent Component Analysis

Objective To study P300 wave extraction by a new method. Methods According to the respective characteristics of the brain electrical signal and the signal analysis method, the algorithm by Wavelet Transformation (WT) combined with Independent Component Analysis (ICA), abbreviated to WICA, was applied to P300 wave extraction. Results With WT, the target component of the signal could be strengthened, while the non-target component and the noise were weakened which became weak in multi -channel ICA input signal, and then WICA algorithm could effectively separate strong target component. Conclusion WICA algorithm behaves well in P300 wave extraction.
