1.Effects of Arsenic Trioxide on Mouse Cultured Melanoma Cloudman S91 Cells

Objectives To observe the effects of arsenic trioxide (As2O3) on the cell cycle, differentiation and proliferation of cultured mouse melanoma Cloudman S91 cells. Methods The cell viability was measured by MTT reduction assay. The morphological changes of the cells were examined by Wright-Giemsa staining. Clone forming was carried out to test the proliferation ability. Apoptosis rate and cell cycle of S91 cells were measured by flow cytometry. Results As2O3 in the concentrations of 0.031~0.25 ?mol/L promoted cell growth markedly and blocked the transformation of cells from G0 to G1. As2O3 in the concentrations of 0.5~8.0 ?mol/L inhibited the cell growth and induced apoptosis markedly. Conclusions As2O3 has marked effects on mouse melanoma S91 cells, which include inducing apoptosis and promoting differentiation and proliferation. These effects are dose- and time-dependent.
2.Effect of ginsenoside Rg1 on multiplication of pig's bone marrow stromal cells in vitro

BACKGROUND:It is shown in past researches that total saponin of ginseng has the effect of promoting multiplication of bone marrow hematopoietic stem cells and progenitor cells in vitro, and promoting secretion of bone marrow hemotopoietic factors, however there has been little researches on the effect of multiplication of bone marrow stromal cells.OBJECTIVE: To investigate the effect of ginsenoside Rg1 on pig's bone marrow stromal cells multiplication in vitro.DESIGN: A controlled observation.SETTING:Department of Cardiology and Department of Dermotology,First Affiliated Hospital, Nanjing Medical University; Department of Pharmacology, Nanjing Medical University.MATERIALS: The experiment was conducted at the Cardiovascular Pharmaceutical Laboratory of Nanjing Medical University from January 2002 to February 2003. The experimental pigs ahd the powder of ginsenoside Rg1were chosen METHODS :The bone marrow stromal cells cultured in vitro were divided as control and experimental groups. For experimental group insenoside Rg1 in different concentrations (10-7,5×10-7,10-6,5×104 mol/L)were added for induction culture, and for control group the media of the same volume was added. The multiplication conditions of bone marrow stromal cells were respectively detected by fibroblast colony forming in vitro, tritiated thymidine {[3H]TdR} incorporation and MTT.MAIN OUTCOME MEASURES: ① The effect of ginsenoside Rgl on the fibroblast colony forming units of bone marrow stromal cells in vitro. ②The effect of ginsenoside Rg1 on [3H]TdR incorporation of bone marrow stromal cells. ③ The effect of ginsenoside Rg1 on bone marrow stromal cells multiplication detected by MTT.RESULTS:① The effect of ginsenoside Rg1 on the fibroblast colony forming units of bone marrow stromal cells in vitro: The numbers of units in experimental group were all higher than that in control group (P < 0.05-P < 0.01),especially obvious in (5-50)×10-7 mol/L experimental group, reaching peak in the concentration being 10-6 mol/L. ② The effect of ginseno side Rg1 on [3H]TdR incorporation of bone marrow stromalcells: As the concentration of ginsenoside Rg1 was 10-6 mol/L, the [3H]TdR incorporation efficiency of bone marrow stromal cells was obviously higher that that in control group (P < 0.05-P < 0.01), and the effect was enhanced as the dosage increased. ③ The effect of ginsenoside Rgl on bone marrow stromal cells multiplication detected by MTT: The value of absorbance in experimental group was obviously higher than that in control group (P < 0.05-P < 0.01), and it was dose-dependent. CONCLUSION: Ginsenoside Rg1 had an effect of multiplication on pig's bone marrow stromal cells in vitro, and it was dose-dependent.
3.Effects of thyroxine on β-adrenoceptors expression of peripheral lymphocyte in elderly patients with congestive heart failure

Objective To determine whether the supplementation of thyroid hormone(TH)improves β-adrenoceptors(β-AR)density and expression of peripheral blood lymphocyte in elderly patients with chronic heart failure(CHF). Methods A total of 42 elderly patients with advanced CHF(NYHA classⅢorⅣ)due to dilated cardiomyopathy(DCM)or ischemic cardiomyopathy (ICM)were randomly divided into a treatment group and a control group.Low-dose L-thyroxine(LT50)was added to the treatment group,both groups had routine anti-heart-failure treatment.Before and after 1 month of treatment,125I-Pindolol radioligand binding assay was used to measure β-AR in peripheral lymphocytes.Reverse-transcriptase polymerase chain reaction(RT-PCR)was used to measure β1-AR expression in peripheral lymphocytes. Results The density and expression of β-AR of lymphocytes in patients with CHF due to ICM or DCM were lower than those in normal health people.and the serum TH level in patients with CHF was lower than that in normal health people.The L-T4 administration induced an up-regulation of β-AR density and expression in peripheral lymphocytes. Conclusions Thyroxine administration induces an up-regulation of β-AR density and expression in peripharal lymphocytes.
5.Recurrent swelling of lower limbs (lymphedema) as manifestations of idiopathic hypereosinophilic syndrome: a case report

A 21-year-old male was hospitalized for recurrent swelling of lower limbs (lymphedema) for 1 year and worsening of it for 1 week.Physical examination revealed several smooth,firm enlarged lymph nodes of the neck,groin without apparent tenderness measuring about 1 cm in diameter.Cardiac and pulmonary auscultation showed no obvious abnormality.The abdomen was soft on palpation without tenderness or rebound tenderness.Skin examination revealed swelling of both lower limbs,especially the left lower limb,as well as scattered irregularly sized,dark erythematous patches with a wood-like consistency on the swollen lower limbs,with high temperature but no tenderness.Elevated peripheral eosinophil count was observed before and after admission,with the eosinophil percentage higher than 70%.Vascular ultrasonography showed thrombosis in the right anterior tibial artery,left dorsal artery of foot and lower portion of the left great saphenous vein.Multiple enlarged lymph nodes were found by computed tomography in the mediastinal,bilateral axillary,retroperitoneal regions and around the abdominal aorta.Lymph node and bone marrow biopsies showed eosinophilia.Histopathology of lesions on the extensor aspect of the left medial thigh and left lateral malleolus showed massive eosinophilic infiltration and lymphatic dilation in the dermis,as well as eosinophil emboli in some lumens.The FIP1L1-PDGFRA fusion gene was undetected.A diagnosis of idiopathic hypereosinophilic syndrome was finally made.The symptoms rapidly regressed after glucocorticoid treatment.
6.Effect of Co-treatment Method of Stagnation of Phlegm and Blood Stasis (Danlou Tablet) on Vascular Endothelial Function in Patients with Atherosclerosis

ObjectiveTo clarify the protective effect of Danlou tablet, a representative traditional Chinese medicine of the stagnation of phlegm and blood stasis co-treatment method, on vascular endothelial function in patients with atherosclerosis (AS). MethodsA randomized controlled trial was conducted. From September 2023 to November 2023, a total of 72 patients who were diagnosed at Guang'anmen Hospital, China Academy of Chinese Medical Sciences and met the inclusion and exclusion criteria for carotid atherosclerosis (CAS) combined with coronary atherosclerotic heart disease (CHD) and stable angina pectoris (SAP) were enrolled. The patients were randomly divided into a control group (receiving conventional Western medicine treatment) and an observation group (receiving Danlou tablet combined with conventional Western medicine treatment), with 36 cases in each group. The intervention lasted for 12 weeks. The frequency of angina pectoris attacks was recorded to evaluate the clinical efficacy of Danlou tablet. Peripheral blood samples were collected from patients, and the expression levels of serum endothelial injury markers before and after treatment were detected by enzyme-linked immunosorbent assay (ELISA). The nitrate reductase method was employed to evaluate the protective effect of Danlou tablet on vascular function. The expression levels of serum inflammatory factors and lipoproteins were determined by ELISA and an automatic biochemical analyzer (dynamic timed scatter turbidimetry and enzymatic method) to assess the anti-inflammatory and lipid-regulating effects of Danlou tablet. ResultsIn terms of angina pectoris attacks, compared with that in the control group, the frequency of attacks in the observation group was reduced (P<0.05). In terms of endothelial injury markers, compared with the levels before treatment within the same group, the levels of endothelin-1 (ET-1), intercellular adhesion molecule-1 (ICAM-1), and vascular cell adhesion molecule-1 (VCAM-1) in the peripheral blood of the observation group were decreased (P<0.05), while the levels of nitric oxide (NO) and vascular endothelial growth factor (VEGF) were increased (P<0.05). Compared with the control group after treatment, the differences in ET-1, NO, ICAM-1, and VCAM-1 were significant (P<0.05). In terms of serum inflammatory factors, after treatment, the interleukin-6 (IL-6) level in the observation group was decreased significantly (P<0.05). Compared with the control group after treatment, the IL-6 level in the observation group was decreased significantly (P<0.01). In terms of serum lipoproteins, after treatment, the level of low-density lipoprotein cholesterol (LDL-C) in the observation group was decreased (P<0.05). After treatment, the level of high-density lipoprotein cholesterol (HDL-C) in the observation group was significantly higher than that in the control group (P<0.05). In terms of safety evaluation, no serious adverse events occurred in either group during the intervention period. ConclusionDanlou tablet applied to patients with CAS combined with CHD can improve endothelial function, reduce inflammatory indicators, alleviate symptoms, improve the quality of life of patients, and demonstrate good safety.

8.Logistic regression analysis for factors affecting the successful rate of nano-carbon in sentinel lymph node biopsy.

OBJECTIVE:
To explore the factors affecting the successful rate of nano-carbon in sentinel lymph node biopsy.
METHODS:
A total of 270 patients with breast cancer, who were treated in First Affilitated Hospital of Henan University of Science and Technology from January 2013 to March 2015, were chosen and given sentinel lymph node biopsy (SLN) with nano-carbon, and the influencial factors were examined by logistic analysis.
RESULTS:
Successful rate of biopsy, accuracy, sensitivity and false negative rate was 92.2%, 97.6%, 93.1% and 6.8%, respectively. Age, primary tumor lesions, body mass index, axillary lymph node status, number of SLN and pathological grade were the factors affetcing successful biopsy (all P<0.05), and body mass index, age, and number of SLN were three independent factors affecting the successful rate of biopsy (all P<0.05). The history of biopsy, tumor location, affected sides, injection sites and chemotherapy showed little effect on the successful rate of biopsy (all P> 0.05).
CONCLUSION
Nano-carbon tracer method is a reliable method in sentinel lymph node biopsy. The body mass index, age, and number of lymph node metastasis greatly impact the successful rate of biopsy.
