1.CXCR4 gene transfection enhancing the distribution of dMSCs to the wounded skin of rats with combined wound and irradiation injury

Objective To observe whether the transplanted dermal multipotent stem cells(dMSCs)transfected by adenovirus vector of CXCR4(Adv-CXCR4)can distribute more frequently to the wound of rats with combined wound and irradiation injury.Methods dMSCs transfected by Adv-CXCR4(group A),or transfected by adenovirus vector of green fluorescent protein(group B),and non-transfected dMSCs were labeled with 3H-TdR and then transplanted into combine-injured rats.The amount of dMSCs in wound were determined by liquid scintillation,and wounds healing process was observed by measuring the remaining wound area.Results From the 5th day after transplantation,the amount of dMSCs in the wound of group A accounted for 1.95%-3.85% of the total transplanted dMSCs,significantly greater than those in group B and group C,which accounted for 1.07%-1.86% of the total transplanted dMSCs.The remaining wound area in group A was smaller than those in group B and group C from day 12 after injury,and the healing time of group A was 1.5 day ahead than group B and group C.Conclusions dMSCs transfected by Adv-CXCR4 distributes more frequently to the wound of combine-injured rats and could accelerate wound healing.
2.Morphological study on the growth of human amniotic membrane loaded with porcine bone marrow-derived mesenchymal stem cells

BACKGROUND: Human amniotic membrane (HAM) contains various ingredents such as collagen, glycoprotein,proteoglycan, integrin and laminated body, and so on, and expresses many kinds of growth factors and mRNA-associated proteins. And these ingredents can supply abundant nutriments for cellular proliferation and differentiation, and benefit cells to grow and propagate. Whether or not HAM can load porcine bone marrow-derived mesenchymal stem cells (BMSCs) to well grow on it deserves to be further investigated.OBJECTIVE: To set up a method of tissue engineering of human amniotic membrane loading porcine BMSCs and observe the morphological characteristics of growth and proliferation of BMSCs seeded on HAM.DESIGN: Randomized controlled observation.SETTING: State Key Laboratory of Trauma, Burn and Combined Injury, General Institute of Combined Injuries, Academy of Preventive Medicine, Third Military Medical University of Chinese PLA.MATERIALS: This experiment was carried out in the State Key Laboratory of Trauma, Burn and Combined Injury,General Institute of Combined Injuries, Academy of Preventive Medicine, Third Military Medical University of Chinese PLA between January and November 2003. Three Guizhou minipigs of either gender, aged 2 to 3 months, weighing from 6 to 8 kg, were provided by the Experimental Animal Center, Third Military Medical University of Chinese PLA. Main reagent:ISCOVE'S modified DULBECCO'S medium (IMDM) culture medium (Hyclone, USA); high-quality fetal bovine serum PAA (Germany); haematoxylin (China); Eosin B (Sigma, USA) and OCT embedding medium (USA). Main instruments: BX51 stereoscopic fluorescence microscope (Olympus, JaPan); IX70 inverted fluorescence microscope (Olympus, Japan);cryostat (2700-Frigcut, Germany); myeloid puncture needle (Jiangsu); superclean bench (Sujing Bloc Antai Company);CO2 constant-temperature incubator (QUEUE, USA).METHODS: HAM was prepared as previously described. The BMSCs of Guizhou minipigs isolated and cultured according to method described previously were primarily cultured and passaged, then they were inoculated to the stromal surface of HAM at different densities (0.84×105 cells/cm2,1.54×105 cells/cm2,2.75×105 cells/cm2); The growth and proliferation of BMSCs of different densities were observed under an inverted microscope and scanning electron microscope; BMSCs of the second or the third passages were inoculated on HAM held with tissue-holding device at a density of 1.54×105 cells/cm2, and they were cultured for 18 days at most. The HAM was daily rolled, sliced and stained by HE for observing the growth of BMSCs loaded on HAM under the light, scanning and transmission electron microscopes.MAIN OUTCOME MEASURES: The growth of BMSCs on HAM was examined at different densities and different time points.RESULTS: ① Comparison of growths of BMSCs promoted by different densities of HAM: BMSCs,which were planted on HAM at the density of 0.84×105 cells/cm2 were irregular and scattered under an invert microscope. Distances between BMSCs were biggish. BMSCs seeded on HAM at the density of 1.54×105 cells/cm2 were regular in arrangement and moderate in density, with clear cell outline and good cell activity before 24 hours, and seeded at the density of 2.75×105 cells/cm2 were congested with many nonattached cells and the longer the growing time of the cells was, the more the cellular debris were observed. BMSCs,which were planted on HAM at the density of 0.84×105 cells/cm2 under the scanning electron microscope, scatted on HAM presented in shapes of irregular, long, thin and flat polygon. Their membrane protuberances presented in shapes of thick and thin, and the distances between cells were biggish. BMSCs,which were planted on HAM at the density of 1.54×105 cells/cm2 have similar appearance of their bodies and membrane protuberances, and the membrane protuberances were more compared with the BMSCs planted at the density of 0.84×105 cells/cm2. Their membrane protuberances intercrossed each other, and the margin of some BMSCs overlapped each other. BMSCs planted at the density of 2.75×105 cells/cm2, arraved on HAM crowdedly and overlappedly with many debris. Their membrane protuberances were not obviously. The margin of some BMSCs was overlapped.② Comparisonof growths of BMSCs promoted by HAM at different time points: Under the inverted microscope, the BMSCs adhered quickly to HAM after being incubated for about 30 minutes. All of BMSCs adhered to HAM within 24 hours, and formed monolayer on it within 48 hours, and grew densely on HAM after being cultured for 4 to18 days. Under the light and electron microscopes, HE results revealed that BMSCs adhered tightly and grew on HAM in different arrays, such as emitting, whirlpool or parallel,and their nuclei located in middle, dense in staining, were big and clear. The shapes of BMSCs were comparatively consistent on HAM. HAM loaded with BMSCs grew 4 days, and BMSCs covered HAM completely. The densities of BMSCs on HAM were suitable, and their bodies were large, and presented irregular, long,thin and flat polygon under the scanning electron microscope. The margin of some BMSCs overlapped each other. The protuberances of cellular membrane of BMSCs were abundant in the shapes of thick and thin. Some protuberances intercrossed each other in the shape of net. BMSCs adhered tightly to HAM through these protuberances. HAM loading BMSCs grow 4 days; most of BMSCs grew on HAM in double layers with the shapes of cambiform under the transmission electron microscope, Their nucleoli were clear. The protuberances of cellular membrane of BMSCs, which situated at two sides of nuclei and overlapped each other, were long. Most of chromatins of BMSCs were autosome.Abundant organell such as rough endoplasmic reticulum (RER),mitochondria could be observed in BMSCs.CONCLUSION:HAM is able to promote the proliferation of BMSCs significantly. BMSCs may be cultured on HAM ex vivo.HAM is a good carrier of BMSCs.
3.Expression of recombinant human ?-defensin 2 in dermal multipotent stem cells and its antiseptic activity

Objective To examine the expression of human ?-defensin 2 (hBD_ 2 ) recombinant adenovirus expression vector in rat dermal multipotent stem cells (dMSCs) and to observe the antiseptic activity of recombinant hBD_ 2 . Methods The expression of hBD_ 2 in dMSCs was examined by RT-PR, fluorescent immunochemistry and Western blotting, and the concentration of recombinant hBD_ 2 in supernate was measured by ELISA. The antiseptic activity of recombinant hBD_ 2 was assessed by K-B disc agar diffusion test. Results hBD_ 2 could be effectively expressed in dMSCs, and the concentration of recombinant hBD_ 2 in supernate was about 743.6 ng/ml . Recombinant hBD_ 2 in supernate showed antiseptic activity. Conclusion Recombinant adenovirus expression vector of hBD_ 2 could be effectively expressed in dMSCs, and the recombinant hBD_ 2 in supernate showed obvious antiseptic effects toward some standard bacteria lines.
4.Repair effect of hBD_2-modified dermal multipotent stem cells transplantation on infected wound

Objective To observe the repair effect of human ?-defensin 2 (hBD 2)-modified rat dermal multipotent stem cells (dMSCs) transplantation on infected wound. Methods Thirty Wistar rats were excised a piece of whole-layer back skin, 3 mm in diameter, then infected the wound with 1?10 8/ml pseudomonas aeruginosa 1 ml, then the rats were injected on the wounded back respectively with dMSCs modified by hBD 2 (n=10), or pure dMSCs (n=10) or none as control (n=10). Objective To explore the activation of PI3k/Akt pathway of serum deprived IEC-6 cells by the serum of rats exposed to single radiation,burn or combined injury.Methods The IEC-6 cells were cultured in serum deprived media for 24 h,and stimulated by the serum of rats exposed to single radiation(~(60)Co ? ray at dose of 9 Gy),single burn(exposure to 5 kW tungsten-halogen light till whole body Ⅲ degree burn) or combined injury(burn first and radiation),and the cells stimulated by the serum from the normal rats and serum starved cells served as the control group.The total proteins of different group cells were extracted and the levels of phosphorylation of Akt were tested by Western blotting.The differentially expressed low mass proteins in the serums were detected by SELDI proteinchip technology,and primarily analyzed by related software as well as bioinformatic methods.Results The level of phosphorylation of Akt in the IEC-6 cells stimulated by serum from rats exposed to single radiation,single burn or combined injury was higher than in the cells stimulated by the serum from normal rats,in which the burn serum caused the highest level.As compared to burn rat serum,the serum of radiation and combined injury had 13 and 6 differentially expressed protein peaks respectively.Conclusion All the serums from rats exposed to different kinds of damage agents could activate the PI3K/Akt pathway of IEC-6 cells efficiently.The special components of burnt rat serum may contribute to the highest effect on the phosphorylation of Akt.
6.Relationship of hypertension with interactions among ABCA1 gene, AGT gene, and occupational stress

Background Hypertension is influenced by both genes and environment. At present, most studies on the relationship among occupational stress, polymorphisms of ATP binding cassette subfamily A member 1 (ABCA1) or angiotensinogen (AGT) genes, and hypertension focus on single gene or single environmental effects. Objective To investigate the relationship of potential interactions between ABCA1 and AGT gene polymorphisms and occupational stress with the prevalence of hypertension. Methods A total of 198 hypertensive patients were selected as the case group from the 1200 oilfield workers in Karamay Oilfield in 2018 with random cluster sampling method, and the control group was selected as 1∶1 matched subjects for sex, age (±3 years), and ethnicity, after excluding blood samples, questionnaires, or DNA purity (concentration) that did not meet the inclusion criteria. Finally, 153 workers in the hypertension case group and 153 workers in the control group were determined. A questionnaire was used to collect general information of the oilfield workers, and the Occupational Stress Inventory Revised Edition (OSI-R) was used to evaluate occupational stress. Polymerase chain reaction-restriction fragment length polymorphism technology was used to detect the genotypes of V825I and R219K loci of ABCA1 as well as M235T and T174M loci of AGT. The gene-gene interaction of ABCA1 and AGT and the relationship between the interaction of gene-occupational stress and the prevalence of hypertension were analyzed by generalized multi-factor dimensionality reduction method. Results The difference of reported occupational stress between the hypertension case group and the control group was statistically significant (P=0.001), and the reporting rate of high occupational stress in the case group (65.4%) was higher than that in the control group (47.7%). The genotype and allele distributions of ABCA1 V825I, ABCA1 R219K, and AGT M235T between the hypertension case group and the control group were significantly different (P<0.05). The results of conditional logistic regression analysis showed that VI and II genotypes at V825I locus of ABCA1 (ORVI=1.682, 95%CI: 1.099-2.573; ORII=1.708, 95%CI: 1.045-2.790), TT genotype at M235T locus of AGT (OR=1.645, 95%CI: 1.022-2.647), and high occupational stress (OR=2.642, 95%CI: 1.228-5.686) increased the risks for hypertension (P<0.05). There was no significant correlation between ABCA1 R219K or AGT T174M polymorphisms and the prevalence of hypertension (P>0.05). The gene-gene interactions between ABCA1 V825I and R219K loci and AGT M235T locus were associated with hypertension (accuracy on training and test sets was 0.68 and 0.63, respectively, with a cross-validation coefficient of 10/10, P<0.05), and ABCA1 V825I locus positively interacted with AGT M235T locus. The gene-environment interactions among ABCA1 V825I and R219K loci, AGT M235T locus, and occupational stress were associated with hypertension (accuracy on training and test sets was 0.74 and 0.63, respectively, with a cross-validation coefficient of 10/10, P<0.05), and AGT M235T locus negatively interacted with occupational stress. Conclusion Genotype VI and II of V825I locus at ABCA1, genotype TT of M235T locus at AGT, and high occupational stress may be risk factors for oilfield workers’ hypertension in Karamay, and the interactions of gene-gene and gene-environment among ABCA1 and AGT gene polymorphisms and occupational stress may be associated with hypertension.
7.Prescription Technology Optimization of RPV Modified Paclitaxel and Schisandrin B Liposomes and Preliminary Evaluation of Antitumor Activity in vitro

OBJECTIVE:To prepare paclit axel and schisandrin B liposomes modified by cell penetrating peptide RPV ,and to preliminarily evaluate its anti-tumor activity in vitro . METHODS :RPV modified paclitaxel and schisandrin B liposomes were prepared by film dispersion method. Box-Benhken design-response surface methodology was used to optimize the prescription technology of RPV modified paclitaxel and schisandrin B liposomes using the amount of cholesterol and paclitaxel ,the time interval of ultrasound probe as factors ,average entrapment efficiency of paclitaxel and schisandrin B was used as the index. The liposomes prepared by the optimal technology were characterized. Sulfonylrhodamine B staining method was used to investigate in vitro toxicity of RPV modified blank liposomes ,paclitaxel and schisandrin B liposomes ,RPV modified paclitaxel and schisandrin B liposomes to human ovarian cancer cell SK-OV- 3. The effects of 3 kinds of liposomes on the migration and invasion ability of SK-OV-3 cells were investigated by cell scratch test and Transwell chamber invasion test. RESULTS :The optimal prescription technology was phospholipid 44 mg,cholesterol 8 mg,paclitaxel 0.64 mg,schisandrin B 1.5 mg,ultrasonic probe time interval 5 s,prescription dosage 5 mL. According to the optimal prescription technology ,the liposomes were spherical in shape ,and the particle size was (126.49±1.19)nm,Zeta-potential was (-4.83±0.61)mV,average entrapment efficiency of liposomes was (93.88±1.67)%. Compared with RPV modified blank liposomes ,after treated with paclitaxel and schisandrin B liposomes and RPV modified paclitaxel and schisandrin B liposomes ,the survival rate ,migration inhibition rate and invasion rate of SK-OV- 3 cells were significantly decreased (P<0.05). The effects of RPV modified paclitaxel and schisandrin B liposomes was better than those of paclitaxel and schisandrin B liposomes (P<0.05). CONCLUSIONS :RPV modified paclitaxel and schisandra B liposome are successfu lly prepared ,and they have certain antitumor activity in vitro .
8.Infraoccipital needle-knife for cervical vertigo.

OBJECTIVETo observe the clinical effect differences between infraoccipital needle-knife and massage for cervical vertigo.
METHODSA total of 366 patients with cervical vertigo were randomly assigned into a needle-knife group (186 cases) and a massage group (180 cases). With cases dropping excluded, 183 cases in the needle-knife group and 176 cases in the massage group were included. Needle-knife was used at Fengchi (GB 20), infraoccipitalpoint, etc. in the needle-knife group. The treatment was given for one course, once three days, 5 times as one course. The traditional massage was applied in the massage group for one course, including systematic stroking, kneading, and the application of pressure and plucking, etc., once every two days and 7 times as one course. The dizziness handicap inventory (DHI) score was observed before and after treatment, as well as 3, 6, and 12 months after treatment. The effects were also evaluated.
RESULTSThe total effective rate was 92.3% (169/183) in the needle-knife group, which was better than 85.2% (150/176) in the massage group (<0.05). Compared with those before treatment, the DHI scores at all the observation time points after treatment were improved in the two groups (all<0.05), with better improvements after treatment as well as 3 and 6 months after treatment in the needle-knife group (all<0.05). There was no significant difference in the improvement of DHI scores between the two groups 12 months after treatment (>0.05). The recurrence rate was 10.3% (12/117) in the needle-knife group, and it was 10.7% (11/103) in the massage group 12 months after treatment (>0.05).
CONCLUSIONSInfraoccipital needle-knife achieves apparent effect for cervical vertigo, which is superior to massage in short period.
9.Health related life quality and its influencing factors in elderly community residents in Shanghai

ObjectiveTo understand the health-related life quality and its influencing factors in elderly community residents in Shanghai, and to provide reference for improving their life quality. MethodsDuring April to June 2021, a cross-sectional survey was conducted in five selected community health care centers, from Baoshan District, Minhang District, Songjiang District and Pudong New District of Shanghai, respectively. The elderly residents aged 65 and over who received physical examination were face-to-face interviewed using EQ-5D-5L scale. SPSS 26.0 and Stata 16 were used for statistical analysis. Binary logistic regression, multiple linear regression and Tobit regression were used to analyze influencing factors of each dimension in EQ-5D scale, EQ-VAS score and health utility value. ResultsA total of 1302 elderly residents were included in the study, and the proportions of problems in the five dimensions of EQ-5D-5L scale were pain/discomfort (13.9%), mobility (9.1%), daily activity (5.8%), self-care ability (4.8%) and anxiety/depression (3.6%), while age was a factor influencing problems in each dimension. The average score of EQ-VAS was 84.28±10.32, and the average health utility value based on Chinese population was 0.97±0.08, which was higher than the rest of the country. Tobit regression analysis showed that female (t=-3.96, P<0.001), age group over 75 (t=-5.28, P<0.001), high school education or above (t=-2.53, P=0.012), chronic disease (t=-5.12, P<0.001) had poor quality of life (P<0.05). ConclusionThe overall quality of life of the elderly in Shanghai community is good, but the quality of life in the elderly women and in the group suffering from chronic diseases is relatively poor. It is necessary to pay more attention to the key populations to improve their health status.
