Objective To investigate the growth inhibition and apoptosis induction effect of vitamin E succinate on MCF-7 human breast cancer cells and to analyze the modulation of Fas expression in this process. (Methods) Estrogen receptor positive MCF-7 human breast cancer cells were treated with VES for 12h, 24h and 48h. The concentrations of VES were 5?g/mL, 10?g/mL and 20?g/mL. The inhibitory effect was measured with MTT method and the cell cycle and cell surface Fas expression were analyzed with flow (cytometry). Fas protein level was detected by Western blotting assay. Results VES had significant inhibitory effect on the growth of MCF-7 human breast cancer cells and the effect was dependently related to time and dosage. The apoptotic rate rose from 1.2% to 11.2%,16.4%, 41.2%,after treated with VES for 48h at the concentrations of 5?g/mL,10?g/mL,20?g/mL respectively. Fas protein level and cell (surface) Fas expression in cancer cells increased after the administration of VES. Conclusions VES had (significant) growth inhibition and apoptosis induction effect on MCF-7 estrogen receptor positive breast cancer cells. The mechanism was related to Fas upregulation on the surface of cancer cells.

Objective: To investigate the growth inhibition and apoptosis induction effect of vitamin E succinate (VES) on human colon cancer cells and to analyze the modulation of apoptosis-mediator Fas expression in this process. Methods: Human colon cancer cell line LS174T was treated with VES for 12 h, 24 h and 48 h at the concentrations of 5 mg/L, 10 mg/L and 20mg/L. 1-(4,5-dimethylthiazo-2-yl)-3,5-diphenylformazan (MTT) assay was employed to detect the inhibitory effect of VES on the growth of colon cancer cells. Flow cytometry was then used to analyze the cell cycle of the colon cancer cells after being treated with VES and the apoptotic rate was calculated at the same time. To find out whether the Fas protein expression was modulated in this process, Western blotting assay and flow cytometry were used to detect the Fas protein level in whole cell lystates and on cell surface. Results: VES exhibited a significant inhibitory effect on the growth of human colon cancer cells in a doseand time-dependent manner. After being treated with VES at 5 mg/L, 10 mg/L and 20 mg/L for 48 h, the apoptotic rate of LS174T cells rose from 0.90% to 15.9%, 46.7% and 64.5%, respectively. Fas neutralizing antibody can significantly block VES-induced apoptosis. After the administration of VES, total Fas protein in whole-cell extracts increased in a dose-dependent manner. The flow cytometry showed that the mean fluorescence intensity rose from 5.43 to 9.88, 13.21 and 18.0 after being treated with VES. Conclusion: VES can induce significant growth inhibition and apoptosis in human colon cancer cells. The modulation of Fas expression is one of the mechanisms involved in this process and may be related to the upregulation of Fas molecule on the cancer cell surface.

Objective To evaluate the effect of selective angiography and embolization for the pa-tients with haemodynamically unstable pelvic fracture. Methods Sixty-two patients with haemodynami-cally unstable pelvic fracture were treated in our hospital from 2002 to 2008. Their pelvie fractures were classified on the basis of their X-ray and CT data. By AO classification, there were 2 cases of type A2, 6 B1, 9 B2, 8 B3, 16 C1, 13 C2, and 8 C. There were 6 cases of open injury (by Gustilo classification, 1 type Ⅱ and 5 type Ⅲ). Of the 46 eases of multiple injuries, 12 had shock. While active fluid infusion and vascular medication were conducted, the angiograph was done for all patients, 59 of whom also received embolization. Results Seventy-nine angiographs were performed for the 62 cases, and 12 of them received more than twice angiographa. Exams revealed that 32 patients had suffered injury to more than 2 arteries. The injured arteries were sequentially superior gluteal artery, iliolumbar artery, lateral sacra] artery, obturator artery, inferior gluteal artery, internal pudendal artery, internal iliae artery and external lilac artery. Of the 59 cases of embolization, 56 were successful. Seven patients died due to severe abdominal organ injury. Forty-five patients got an average follow-up of 14 months (range, 6 to 52 months) . No severe complications were no-ticed. Conclusions Selective angiography and embolization is an effective intervention for patients with haemodynamically unstable pelvic fracture. It is mandatory to conduct recurrent angiographic embolization for patients with multiple artery injuries or delayed artery injuries.

Methods Bcap-37 human breast cancer cells were treated with VES combined with chemotherapeutic drugs for 24h and 36h. The ~concentrations of VES were 10?g/mL and 20?g/mL and those of 5-florouracil, mitomycin and ~cyclophosphamide were 16.9?g/mL and 33.8?g/mL, 1?g/mL and 3.3?g/mL and 100?g/mL and 300?g/mL respectively. The inhibitory effect was measured with MTT method and the cell cycle and cell ~surface Fas expression were analyzed with flow cytometry assay. Results The combination of VES with ~chemotherapeutic drugs had a significant inhibitory effect on the growth of Bcap-37 human breast cancer cells. Flow cytometry assay of cell cycle showed that the natural apoptptic rate of Bcap-37 cells was 0.7%;after treatment with VES 20?g/mL,the apoptotic rate was 19.2%;after treatment with 5-Fu,mitomycin and ~cyclophosphamide the apoptotic rates were 16.2%,16.7% and 12.3%,respectively;after the combined use of VES and the 3 chemotherapeutic drugs, the apoptotic rates were 40.3%,44.8%,39.6%,~respectively .Fas expression in cancer cells increased after the co-administration of VES and chemotherapy drugs. Conclusions VES combined with chemotherapeutic drugs had significant inhibitory effect on the growth of Bcap-37 human breast cancer cells. The mechanism may be related to Fas upregulation on the surface of cancer cells.

Objective To investigate the effect of Pericarpium Citri Reticulatae on changes of the intestinal absorption of ginsenoside components in Yi Gong San based on the rat model of spleen deficiency and whether its effect is related to the exocytosis of P-gp protein. Methods The group design included the P-gp protein agonist rifampicin and the inhibitor verapamil, and 36 SD rats were randomly divided into a blank control group(6 rats)and a model group(30 rats),and then subcutaneously injected with rifampicin to establish a model of splenic deficiency, and then the model group was divided into the Yigong San fullparty(Y)group, the lack of Pericarpium Citri Reticulatae(Y-C)group,the YiGong San combined with rifampin(Y+R)group, the lack of Pericarpium Citri Reticulatae combined with verapamil(Y-C+V)group and the lack of Pericarpium Citri Reticulatae combined with rifampin(Y-C+R)group,Ginsenoside Rb1,ginsenoside Re and ginsenoside Rg1 were used as the index components in an in situ unidirectional intestinal perfusion test, and HPLC-MS was used to determine the content of the index components and to calculate the kinetic parameters of intestinal absorption, the effective permeability coefficient(Peff)and the absorption rate constant(Ka).Results Compared with group Y,Peff of ginsenoside Rb1 and Re were significantly lower in Y-C at multiple perfusion sessions, and Peff and Ka of ginsenoside Rb1,ginsenoside Re,and ginsenoside Rg1 were lower in Y+R at multiple perfusion sessions, suggesting that Pericarpium Citri Reticulatae promotes the intestinal absorption of ginsenoside Rb1 and ginsenoside Re and that this effect is related to the activity of the P-gp protein, but failed to change the intestinal absorption of ginsenoside Rg1.The P-gp protein inhibitor verapamil significantly increased the Peff of ginsenoside Rb1 and Re in the Y-C+V compared with the Y-C,while the P-gp protein agonist rifampicin significantly decreased the Peff and Ka of ginsenoside Rb1,ginsenoside Re,and ginsenoside Rg1 in the Y-C+R in multiple perfusion sessions. The significant decrease in Peff and Ka at multiple perfusion times suggests that ginsenoside Rb1,ginsenoside Re,and ginsenoside Rg1 may be substrates for P-gp proteins in the absence of Pericarpium Citri Reticulatae intervention. Conclusion Pericarpium Citri Reticulatae promotes the intestinal absorption of ginsenoside Rb1 and ginsenoside Re by exerting a verapamil-like effect that inhibits the activity of P-gp proteins in Yi Gong San.

Journal of Southern Medical University 2012;32(2):270-273
OBJECTIVE[corrected] To understand the role of miRNA-146a in the proliferation and migration of primarily cultured rat vascular smooth muscle cells (VSMCs) and investigate the mechanisms.
METHODSPrimarily cultured rat VSMCs were transfected with a synthesized miRNA-146 inhibitor, a scramble sequence or PBS via Lipofectamine2000. Cell counting kit 8 (CCK8) and transwell assay were employed to assess the proliferation and migration of the transfected cells, and the expressions of nuclear factor-κB p65 (NF-κBp65) and proliferation cell nuclear antigen (PCNA) were detected using Western blotting.
RESULTSA 48-h transfection of the VSMCs with miRNA-146 inhibitor caused significantly lowered miRNA-146a expression as compared with that in VSMCs transfected with the scramble sequence or PBS (P<0.01), resulting also in lowered proliferative and migration ability of the cells (P<0.01). The expression levels of NF-κBp65 and PCNA were remarkably lower in cells transfected with miRNA-146 inhibitor than in the cells in the other two groups (P<0.05).
CONCLUSIONmiRNA-146a is capable of promoting the proliferation and migration of rat VMSCs probably by enhancing the expression of NF-κBp65.

Objective To explore the localization and guidance value of infraorbital/maxillary nerve in endoscopic transmaxillary approach, and to provide anatomical data for clinical operation. Methods The eight adult cadaver heads were dissected by means of the endoscopic transmaxillary approach to expose the infraorbital/maxillary nerve, and relevant data were collected. Results In 8 cases, 16 maxillary nerves arose from the trigeminal ganglion of the trigeminal nerve and emerged from the foramen rotundum, transited to the infraorbital nerve at the infraorbital fissure, then traveled in the infraorbital canal and out of the infraorbital foramen. The infraorbital/maxillary nerve could be divided into four segments according to the foramen rotundum, infraorbital groove and infraorbital foramen: the terminal segment, the infraorbital nerve and its terminal branches to the face, distal to the infraorbital foramen; the orbitomaxillary segment and the orbitomaxillary segment of the infraorbital nerve within the infraorbital canal from the infraorbital foramen along the infraorbital groove (length 11.7 ±2.5 mm ), which was readily identified in the roof of the maxillary sinus in all specimens. The pterygopalatine segment, the pterygopalatine segment within the pterygopalatine fossa, which started at the infraorbital groove to the foramen rotundum (length 13.4±2.1 mm); The intracranial segment, the intracranial segment from the foramen rotundum to the trigeminal ganglion(length 15.2±3.9 mm). Conclusion The infraorbital nerve can serve as a anatomical landmark for endoscopic transmaxillary approach to get access to infratemporal fossa, pterygopalatine fossa, trigeminal ganglion and lateral wall of the cavernous sinus.

Objective:
To investigate the effect of online and offline mindfulness training on improving anxiety and depression and sleep quality of college students, and to provide a reference for mental health promotion among college students.
Methods:
From October 2020, a total of 1 203 university students from North China University of Technology were screened with the Self rating Anxiety Scale (SAS), Self rating Depression Scale (SDS) and Pittsburg Sleep Quality Index (PSQI) using the whole group radom cluster sampling method. Totally 103 students who met the inclusion criteria were randomly divided into 64 online and 39 offline groups. The degree of improvement in anxiety, depression and sleep quality was assessed after the intervention.
Results:
The SAS, SDS and PSQI scores of college students after the online and offline the mindfulness training intervention significantly decreased compared with score before the intervention( t =5.57, 5.31, 3.99; 4.88,5.02, 5.88, P <0.01). The difference in the degree of improvement in sleep quality between the two interventions, online and offline, was statistically significant ( t =-2.55, P <0.05). The less the three symptoms of anxiety, depression and sleep were combined in university students, the higher the symptom remission rate of the positive mindfulness training (25% remission rate for all three symptoms together, 40% remission rate for two symptoms together and 100% remission rate for only one symptom).
Conclusion
Both online and mindfulness training can be used as an effective intervention for sleep, anxiety and depression; offline mindfulness training is more effective than online in improving sleep quality in university students; mindfulness training is more effective in relieving single symptoms.
