1.Effect of Xiangdan Glucose Injection on blood hemorrheology

Objective:The effect of Xiangdan Glucose Injection (XDGI) (Radix Salviae Mitiorrhizae, Lignum Dalbergiae Odoriferae) on blood hemorrheology was studied. Methods: We compared the changes of blood viscosity, haematocrit, platelet aggregation and fibrinolytic activity after the treatment with XDGI. Results: Results showed that XDGI can significantly reduce blood visosity, haematocrit, platelet aggregation, and enhance red blood cell MAXDI. Conclusion: XDGI can markedly improve hemorrheology.
2.Changes in the level of cytokine in rats with chronic obstructive pulmonary disease of phlegm heat cumber lung type after treatment of Maxing Shigan decoction

BACKGROUND: It is generally thought that the pathogenesis of the chronic blocking pulmonary disease (COPD) is characteristics in chronic inflammations of air passage, lung essence and the lung blood vessels. As the classic Chinese medicine, Maxing Shigan decoction has the pharmaco logical effects on fever alleviation, anti-inflammation, anti-virus, cough relieving and phlegm resolving, which is suitable for the treatment of phlegm heat cumber lung COPD, and is remarkable in the curative effect.OBJECTIVE: To probe into the mechanism of Maxing Shigan decoction ini the treatment of COPD by studying the changes in interleukin-8 (IL-8),tumor necrosis factor-α (TNF-α), interferon-γ (IFN-γ), interleukin-4 (IL-4)and the ratio between IFN-γ and IL-4 in rats with COPD.DESIGN: Randomized control animal experiment.SETTING: Affiliated Hospital of Shandong University of Traditional Chinese Medicine.MATERIALS: Seventy-two healthy male Wistar rats of SPF level with the body mass of (200±20) g were selected. Maxing Shigan decoction, composed of 5 g of ephedra, 9 g of almond, 6 g of liquorices, 18 g of gypsum.The water-decocted enrichment was 0.475 g/mL, and the medical herb pieces were supplied by the TCM Dispensary of Affiliated Hospital of Shandong Medical University of Traditional Chinese Medicine.METHODS: The experiment was conducted in the Central Laboratory,Affiliated Hospital of Shandong University of Traditional Chinese Medicine from April to October 2005. ① 72 healthy male Wistar rats were randomly divided into 3 groups: blank control group (n=24), retention of phlegm-heat modeling group (modeling group, n=24) and retention of phlegm-heat treatment group (treatment group, n=24). Improved smoking and trachea dropping lipopolysaccharide was adopted to copy COPD model, and model of phlegm heat cumber lung was remodeled for copy.② Rats in the treatment group were lavaged with 2 mL of Maxing Shigan decoction per day (the medicine composition: Chinese ephedra 5 g, almond 9 g, licorice 6 g, gypsum 18 g, the water fries the concentration for the density 475 g/L concentrated solution) from the 30th day, while rats in the modeling group were lavaged with 2 mL of normal saline each day.Rats in the blank control group received no interventions, which ate and drank freely under the same environment. ③ The contents of IL-8, IL-4 and TNF-α in the homogenate of lung tissues were tested by radio-immune method. ④ The content of IFN-γ in the homogenate of lung tissues was detected by the enzyme immunoassay method. ⑤ The ratio of IFN-γ and IL-4 was calculated. MAIN OUTCOME MEASURES: The contents of IL-8, TNF-α, IFN-γ,IL-4 as well as the ratio between IFN-γ and IL-4 in the homogenate of lung tissues in rats of each group.RESULTS: ① The level of IL-8 in the modeling group was markedly higher than that in the blank control group [(581.41±74.72), (318.63±40.01 ) ng/L,P < 0.01], which was significantly decreased after the treatment [(533.95±68.67) ng/L,P < 0.01], while there was difference in comparison with the blank control group (P < 0.05). ② The level of TNF-α in the modeling group was significantly higher than that in the blank control group [(160.78±20.90), (100.29±24.2) ng/L, P < 0.01], which was remarkably decreased after the treatment [(147.08±19.43) ng/L,P < 0.05],whereas it was different in comparison with that in the blank control group(P < 0.01). ③ Compared with the blank control group, the level of IFN-γ in the modeling group was markedly decreased [(230.32±72.33), (328.08±50.57) ng/L ,P< 0.01], which was obviously increased after the treatment [(267.24±55.14) ng/L ,P < 0.05], and there were differences between the two groups (P < 0.01). ④ The level of IL-4 was significantly increased in the modeling group than that in the blank control group[(135.08±29.17), (82.41±14.66) ng/L,P < 0.01], which was significantly decreased after the treatment [(107.99±23.76) ng/L,P < 0.01], and there was difference between the two groups (P < 0.01 ). ⑤ The ratio between IFN-γ and IL-4 declined markedly in the modeling group than that in the blank control group (1.77±0.73,4.09±0.91 ,P < 0.01), and the ratio increased markedly after the treatment(2.59±0.79, P < 0.01 ). There was difference between the two groups(P < 0.01 ).CONCLUSION: Maxing Shigan decoction can regulate the changes in level of multiple cytokines in rats with COPD, which has the effects of anti-inflammation and enhancing the immunity of organism immunization with clear curative effect on COPD (syndrome of retention of phlegmheat).
3.Study on the effects of microRNA-375 on gastric cancer cell function

Objective To investigate the effects of microRNA(miRNA)-375 on gastric cancer cell function and the possible mechanism.Methods miRNA-375 eukaryotic expression vector was stably transfected into human gastric cancer cell line HGC-27.A miRNA-375 highly expressed gastric cancer cell line PC3,1-375 was screened and established.Blank plasmid transfection group (PC3.1 group) and blank control group were also established.The cell proliferation ability and apoptosis rate in three groups were detected by cell counting kit-8 (CCK-8) method and flow cytometry.The protein expression of astrocyte elevated gene 1 (AEG-1) in three groups was determined by Western blotting.The nude mouse subcutaneous xenografts model was also established.After 30 days,the nude mice were executed and the xenografts were dissociated and measured.The expression ofAEG-1 in the tumors at protein level was tested by immunohistochemistry method.The statistical inference includes t-test and analysis of variance.Results After stable transfection and cell screening,compared with that of blank control group,the expression of miRNA-375 in PC3.1-375 group significantly increased (t=2.82,2.01,bothP<0.05).The cell proliferation of PC3.1-375 group was significantly lower than that of PC3.1 group and blank control group and the difference reached a maximum in day 3 (t3d=3.56,2.79,both P<0.05).But with the apoptosis rate of PC3.1-375 group,the opposite was true (t=2.05,2.96,both P<0.05).The results of Western blot shows that the expression of AEG-1 at protein level of PC3.1-375 group decreased compared with that of blank control group and PC3.1 group.The xenografts growth rate,average weight and tumor volume of PC3.1-375 group were significantly lower than those of blank control group and PC3.1 group (t=3.78,2.26,both P<0.05).By the end of experiment,the volume of nude mouse subcutaneous xenografts in blank control group ((3.12 ± 0.77) cm3) and PC3.1 group ((3.76 ± 0.65) cm3) was significant bigger than that inPC3.1-375 group (0.72±0.21) cm3,t=5.67,6.92,both P<0.05).The average weight of xenografts in PC3.1-375 group ((0.85 ± 0.15)g) was obviously lower than that in PC3.1 group ((4.01±0.52) g) and blank control group ((3.86±0.56) g,t=4.56,5.01,both P<0.05).The results of immunohistochemistry indicated that the expression of AEG-1 protein in xenografts in PC3.1-375 group was remarkably decreased compared with that in blank control group and PC3.1 group (F=35.000,P<0.05).Conclusions miRNA-375 can effectively inhibit the cell proliferation of gastric cancer,promote cells apoptosis and it's a tumor suppression factor in gastric cancer.AEG-1 may be a target gene of miRNA-375.
4.Effects of high and low audio-motor stimulation mode on attention and memory in rural elderly with mild cognitive impairment

Objective:To explore the intervention effects of high and low audio-motor stimulation mode on attention and memory of elderly with mild cognitive impairment in rural areas.Methods:From September 2019 to May 2020, 80 subjects with mild cognitive impairment meeting the inclusion and exclusion criteria were selected and randomly divided into experimental group and control group according to the drawing lots, with 40 participants in each group.The experimental group was given high and low audio-motor stimulation mode training, and the control group was given finger exercises training.Before and after the intervention, sustained and selective attention were assessed using number cancellation test(NCT), E-Prime paradigm based on Stroop effect design, and memory was assessed using the Rivermead behavioral memory test, version 2 (RBMT-Ⅱ). SPSS 17.0 software was used to perform data statistics. The independent sample t test, paired sample t test and Mann Whitney U test were used for statistical analysis. Results:After intervention, the sustained attention index of the experimental group (0.86±0.48) was significantly higher than that of the control group (0.63±0.47, t=4.336, P<0.05). After intervention, the unanimous response time of the experimental group ((2 263.38±437.87) ms) and the reverse reaction time ((2 335.75±599.86) ms) were significantly lower than those of the control group ((2 703.95±506.34) ms, (2 667.43±585.38) ms) ( t=-4.068, -2.447, P<0.05). The consistent correct rate ((0.84±0.14)) and the reverse correct rate ((0.87±0.10))were significantly higher than those of the control group ((0.71±0.19), (0.73±0.15)) ( t= 3.571, 4.931, both P<0.05). RBMT-Ⅱ dimensions and total standard scores of the experimental group (19.00 (18.00, 20.00)) were higher than those of the control group (11.00 (10.00, 12.00))( Z=7.299, P<0.05). Conclusion:High and low audio-motor stimulation improves attention and memory in rural elderly with mild cognitive impairment.
5.Plasma cytokines in the patients with acute graft-versus-host disease and significance

Objective To study the relationship between type 1 and type 2 cytokines and human acute graft-versus-host disease (aGVHD).Methods In 20 patients undergoing allogeneic peripheral blood stem cell transplantation (allo-PBSCT), plasma concentrations of interleukin-18 (IL-18), interleukin-2R (IL-2R), tumor necrosis factor-a (TNF-?) and interleukin-10 (IL-10) were measured by using enzyme-linked immunosorbent assay (ELISA).Results All the patients achieved engraftment. Eight cases deve- loped grade Ⅰ-Ⅱ aGVHD and 3 cases developed Ⅲ-Ⅳ aGVHD. The concentrations of IL-18 and IL-2R in the patients with aGVHD were significantly higher than those without aGVHD. The levels of IL-18 were correlated with the severity of aGVHD; The levels of TNF-? showed no difference between the patients with or without aGVHD; The concentrations of IL-10 in the patients with aGVHD were significantly declined but in those without aGVHD were significantly increased.Conclusion Type 1 and type 2 cytokines play an important opposite role in development of aGVHD.
6.Clinical research on the correlation of liver function CTP classification and MELD score of post-hepatitis B cirrhosis between different TCM syndromes

Objective To explore and identify the correlation of CTP classification,MELD score of patients with cirrhosis after hepatitis between different TCM syndromes.Methods For 135 patients comply with hepatitis B cirrhosis standard,standardized TCM syndrome differentiation and laboratory tests is executed so as to analyze the correlation of CTP classification,MELD score of patients with cirrhosis after hepatitis between different TCM syndromes.Results The liver function CTP classification in liver Qi stagnation syndrome and dump-heat accumulation syndrome patients mainly scores to Class A,and the MELD score less than 10 points-based.The liver function CTP classification in Yin deficiency of Liver and kidney,Yang deficiency of spleen and kidney and blood stasis syndromes mainly scores to Class B-C,and the MELD scores more than 10 points basically.Yin deficiency of Liver and kidney,Yang deficiency of spleen and kidney and blood stasis syndromes take majority in advanced cirrhosis patients compared with liver Qi stagnation syndrome and dump-heat accumulation syndrome take majority in early stage cirrhosis patients.Conclusion Matches the traditional view in TCM that form Qi stagnation,to damp heat-water stopped,to blood stasis-Yang and Yin deficiency is the regular pathogenesis evolution pattern of post-hepatitis cirrhosis.
7.Study of immunological function in severe acute respiratory syndrome

Objective To discuss the relationship between disease progress with the change of cell im munological function as well as the effect of immunological function in severe a cute respiratory syndrome. Methods Retrospective study was designed to analyze the relationship of disease progres s with the change of immunological function. According to the disease outcomes, the patients are divided into two groups: died group and survival group. The dif feren ce of immunological function in groups was performed statistics analysis. 8.Maxillofacial and periodontal tissue-engineered bone:vascularization and osteogenesis strategies

BACKGROUND:Maxilofacial bone and periodontal tissue defect is one of the important diseases that affect human functionality and aesthetic appearance, and bone tissue engineering becomes the main means to repair maxilofacial and periodontal tissue defects. Currently, the basic mode is constructed by the combination of co-culture of seed cels and cels, scaffolds and micro-environment. Pre-vascularization and rapid osteogenesis of tissue-engineered bone can reduce implant necrosis and absorption, and improve repair success rate. OBJECTIVE:To summarize the new progress of bone tissue engineering used in the oral and maxilofacial and periodontal tissue in the past 5 years. METHODS:CNKI database and PubMed database from 2010 to 2015 were searched using the keywords of “oral and maxilofacial, bone tissue engineering, bone regeneration, vascularization, genetic modification, seed cels, support material, microenvironment” in Chinese and English, respectively. After elimination of independent and repetitive studies, 68 articles were included in result analysis. RESULTS AND CONCLUSION: The tissue-engineered bone has achieved tremendous progress in the repair of oral and maxilofacial and periodontal tissue defects. The three-dimensional scaffold with gene-modified seed cels can effectively promote the vascularization, improve the osteogenic effect and increased the probability of success in mandibular defect repair. In addition, tissue-engineered bone implantation into the alveolar ridge defects or fresh extraction fossa can effectively restore and preserve alveolar ridge height and width, to ensure a good bone condition for subsequent restorative treatment. After the implantation of tissue-engineered bone, different external environmental stimuli could be loaded at defect sites, and the extracelular matrix components or signal pathway could be adjusted to change the process of vascularization. Vascularization is a premise condition for the establishment of an effective blood circulation to ensure the success of scaffold implantation.
9.RP-HPLC gradient elution analysis for the determination of pantoprazole soldium and its related substances

Aim: To establish a method for the determination of pantoprazole sodium and its related substances. Methods: A column packed with octadecylsiance bonded silica gel (250 mm × 4. 6 mm, 5 pjn) was used. The 0. 01 mol/L monopotassium phosphate solution( adjusted with phosphoric acid to pH 7. 0) -acetonitrile were adopted as the mobile phase, a gradient elution was programmed as follows: 0→30 min(90:10-60:40), 30→45 min(60: 40→15: 85); the detection wavelength was 289 nm; the column temperature was 40 ℃; the flow rate was 1. 0 mL/min. Results: Pantoprazole sodium, the intermediates and its related substances could be well separated. A good linear relationship was obtained over the range of 6. 96-48. 72 μg/mL( r =0. 999 9). The limit detection and quantisation of pantoprazole sodium were 8.51 ng and 17.0 ng, respectively. Conclusion: This method can be applied to control the related substances of pantoprazole sodium and determine pantoprazole sodi-um.
10.Expression of repulsive guidance molecule A in the hippocampus of rat models of cerebral ischemia-reperfusion injury during treadmill exercise

BACKGROUND:Exercises play an important role in the recovery of neurological function after stroke. Few studies concerned the amount of exercise in rats after stroke. Hippocampus is strongly associated with cognitive function, but no reports addressed the expression of repulsive guidance molecule A in the rat hippocampus after ischemia and reperfusion. OBJECTIVE:To explore the effects of exercise on repulsive guidance molecule A expression in the hippocampus on the ischemic side in rats after cerebral ischemia-reperfusion injury. METHODS:120 Sprague-Dawley rats were equal y and randomly divided into normal group, sham-operation group, and 7-, 14-, 28-day model groups. The model of right cerebral ischemia-reperfusion injury was induced by ligation with nylon monofilament in rats of 7-, 14-, 28-day model groups. Low exercise group received treadmil training of 5 m/min, 5 minutes;7 m/min, 5 minutes;9 m/min, 20 minutes. Moderate exercise group received treadmil training of 8 m/min, 5 minutes;10 m/min, 5 minutes;13 m/min, 20 minutes. High exercise group received treadmil training of 8 m/min, 5 minutes;11 m/min, 5 minutes;20 m/min, 20 minutes. RESULTS AND CONCLUSION:Repulsive guidance molecule A mRNA and protein expression was highest in the ischemic side of the hippocampus in the 7-day model group without excercise. Moreover, repulsive guidance molecule A relative expression gradual y reduced over time. Compared with non-exercise, repulsive guidance molecule A mRNA and protein expression significantly decreased in the 14-and 28-day model groups during moderate exercise (P<0.05), but repulsive guidance molecule A mRNA and protein expression increased during high exercise. Above data confirmed that moderate exercises could decrease repulsive guidance molecule A expression in the affected side of the hippocampus of rats with cerebral ischemia-reperfusion injury.
