1.A Research Progress on Matrine and Its Preparations

Matrine is an effective natural product isolated from the traditional herb,Sophora flavescens,with wide range of pharmacological and clinical effects and treating as the treatment for chronic viral hepatitis in clinic.Currently,the mainstream clinical preparations of matrine involved injections,tablets,capsules and suppositories,while preparations under development included targeting preparations,sustained-release preparations and transdermal drug delivery preparations.Admittedly,It can significantly improve the biological utilization of matrine through the optimization of its clinical preparations for with scientific researches and the development of new dosage forms.While the clinical applications will be expanded and the clinical efficacy of the preparations of matrine enhanced with the development of researches for new dosage forms and drugs and the application of new materials.It may be also benefical to the study of monomer compositions of Chinese material medica,including the development of the preparation of matrine.Rested on the recent studies,pharmacological activities,preparation methods and researches for the preparations of matrine was summarized in this text for providing a reference for developing its preparations in the future.
2.Safety evaluation of Ad-shRNA-NgR in brain tissue of EAE rats

Objective To observe the survival of EAE rats after recombinant adenovirus NgR specific RNA interference(Ad-shRNA-NgR) transfected the brain tissue of the experimental autoimmune encephalomyelitis(EAE),and provide the basis for EAE intervention.Methods EAE rats were randomly divided into high,medium,low and control groups(20 rats in each group).The lateral ventricle of EAE rats were injected with a titer of 1×1011 pfu/mL,1×1010 pfu/mL and 1×109 pfu/mL Ad-shRNA-NgR.The survival of EAE rats at third and seventh days after injection was observed.Results The survival rate of EAE rats of the high titer group was significantly lower than those of the middle titer group and low titer group at third and seventh days after Ad-shRNA-NgR transfected into EAE brain tissue.There was no significant difference in survival rate in middle titer group,low titer group and control group.Conclusion The titer of Ad-shRNA-NgR is safe in the experiment of EAE rats from 1×1010 pfu/mL to the range of 1×109 pfu/mL.
3.Genetic analysis of thalassemia among Tujia and Miao people of reproductive age in Chongqing

Objective:The aim of this study was to investigate the carrier ratio and the genotype of thalassemia among Tujia and Miao people of reproductive age in Chongqing.Methods:According to forward-looking design and multi-stage stratified cluster sampling method, fasting venous blood samples of Tujia and Miao people of reproductive age were collected from 11 survey sites in Chongqing from March to July 2019. Gap-PCR and high-throughput sequencing were used to screen thalassemia genes.Results:A total of 516 Tujia people (258 males, 258 females) and 270 Miao people (139 males, 131 females) were included in this study, and their age were (28.63 ± 5.26) and (28.62 ± 5.35) years, respectively. About 5.04% (26/516) Tujia people carried thalassemia gene, with 1.94% (10/516) and 2.52% (13/516) for α and β thalassemia, respectively. Three kinds of new variants (1 case of each variant), HBA 2: c.46G>A (Gly>Ser), HBB: c.*+129T>A and HBB: c.-39T>G with unclear pathogenicity, were identified in Tujia people. About 7.78% (21/270) Miao people carried thalassemia gene, among these, α and β thalassemia were 3.33% (9/270) and 4.44%(12/270), respectively. The most common mutation type of α-globin gene was -α 3.7/in the two ethnic groups. Three kinds of β-globin gene mutation types, Codons 41/42 (-TTCT) beta 0, Codon 17 (A>T) beta 0 and IVS-Ⅱ-654 (C>T) beta +, were the most common in Tujia people. Meanwhile, the chief β-globin gene mutation type was Codons 41/42 (-TTCT) beta 0 in Miao people. Conclusions:The carrying rate of thalassemia gene is higher in Tujia and Miao people in Chongqing, and the genotypes of thalassemia gene are different between Tujia and Miao people. The clinical significance of three kinds of new variants with unclear pathogenicity should be focused on.
4.A Study on the Recombinant 26 kDa Glutathione-S-Transferase as a Vaccine Candidate:Dynamics of Antibodies in Immunized Buffaloes and Protection against Schistosoma japonicum Infections

ObjectiveTo observe the dynamics of antibodies and protection against Schistosoma japonicum infections in buffaloes after immunized with recombinant 26 kDa glutathione S transferase (reSjc26GST). Methods Buffaloes in 2 villages endemic for schistosomiasis japonica were selected as test and control groups, respectively.In test group initially 96 buffaloes were vaccinated with reSjc26GST, and 90 buffaloes in the control group did not experience vaccination. The indicators included levels of antibodies to reSjc26GST in buffaloes before and after infection with S japonicum and changes in infection rate. Results Specific antibodies, which showed a trend of trapezoid increase, were induced in buffaloes after immunized with reSjc26GST. Twenty months after immunization, the infection rate of the test group was decreased by 62 2% when compared with that before vaccination,and by 67 7% when compared with that of the control in the corresponding period.Conclusion Specific antibodies and a certain extent of protection were induced in buffaloes after immunized with reSjc26GST, which played an significant role in ameliorating morbidity.
5.Reconstruction of hair follicles in three-dimensional alginate scaffolds with mixed cells of hair follicles

Objective To induce the formation of hair follicles in vivo and in vitro with primary human hair follicle outer root sheath cells (ORSCs),human hair dermal papilla cells (DPCs) and fibroblasts seeded on three-dimensional alginate scaffolds at different sequences.Methods Second-passage ORSCs,DPCs and mitomycin-c-treated human fibroblasts were seeded into three-dimensional alginate scaffolds at different sequences and a certain ratio to reconstruct hair follicles in vitro.After 8 weeks of culture,some of the substitutes were subjected to hematoxylin and eosin (HE) staining,and some were transplanted into the subcutaneous tissue of bal/bcl nude mice followed by additional culture for 8 weeks.Then,the mice were sarcrificed and transplants were harvested and subjected to HE staining,immunohistochemical staining and transmission electron microscopy for the observation of hair follicle formation.Results After 8 weeks of culture in vitro,hair follicle cells were evenly scattered in three-dimensional scaffolds,with no keratinized material or hair follicle formation.When the substitutes were cultured in vivo for 8 weeks,DPCs and ORSCs gathered together to form cell aggregates,with the visualization of hair follicle-like structure in circular arrangement,which was positive for cytokeratin (CK) 14,CK15,β1-integrin and vimentin.Electron microscopy showed hair follicle cells and erythrocytes adhering to the scaffolds.One-week preculture of mitomycin-c-trcatcd human fibroblasts in the scaffolds followed by the seeding of the suspension of ORSCs and DPCs at a ratio of 1∶5 resulted in increased follicular like structures with more typical appearance in mice compared with the other strategies for cell inoculation and culture.Conclusions In the reconstructed hair follicle models,DPCs keep the capability to induce the formation of hair follicles,and ORSCs keep the characteristics of hair follicle stem cells.The optimal cell density,ratio and process were determined in the experiment,which may lay a methodological and theoretical basis for hair follicle reconstruction.
6.Relationship of proliferation and activation of T lymphocyte subsets and disease progression in human immunodeficiency virus-I-infected individuals

Objective To study the relationship of proliferation and activation of T lymphocyte subsets and disease progression in antiretroviral-naive human immunodeficiency virus(HIV)-1-infected individuals.Methods Forty-nine antiretroviral-naive,chronically HIV-1 infected patients and 16 healthy,HIV-1 negative controls were enrolled in this study.The patients were divided into 3 groups according to their CD4+T cell counts:<200×106/L,(200-350)×106/L and>350×106/L.Peripheral blood mononuclear cells(PBMC)were isolated.T cell proliferation index was measured by Ki-67 staining.T cell activation was detected by CD38 staining.The samples were analyzed by flow cytometry.The data were compared by one-way ANOVA.Results The percentage of Ki-67+cells in CIM+T ceils was 7.92%±4.37%in CD4+T cell<200×106/L group,which was significantly higher than those 0.39%d:0.24%in control group,2.61%±2.12%in(200-350)×106/k group and 2.65%±2.13%in>350 X106/L group(F=21.961,P<0.01).The percentage of Ki-67+cells in CD8+T ceils in CD4+T cells<200×106/L group was 2.87%±1.13%,which was also much higher than those in other 3 groups(0.15%±0.90%,1.40%±1.17%,1.22%±0.80%,respectively F=19.203,P<0.01).The Ki-67'CD4'T cells and Ki-67+CD8+T cells were inversely correlated with CD4+T cell counts(r=-0.654,r=-0.539,respectively;P
7.The efficacy and safety of antiviral treatment in inactive hepatitis B surface antigen carriers

Objective To evaluate the feasibility and safety profile of pegylated-interferonα-2a (Peg IFNα-2a) combined with adefovir dipivoxil (ADV) in inactive hepatitis B surface antigen (HBsAg) carriers (IHC).Methods This was a single center, prospective and open-label study.IHC were divided into therapeutic group (T, 112 subjects) and control group (C, 72 subjects) according to personal willingness.Patients with hepatitis B virus (HBV) DNA<20 IU/mL were treated with Peg IFNα-2a monotherapy, and those with HBV DNA ≥20-<2 000 IU/mL were treated with Peg IFNα-2a combined with ADV.Total therapy duration was 96 weeks.For patients who achieved HBsAg seroconversion and continued consolidation treatment for 24 weeks, the treatment duration could be less than 96 weeks.t test was used for continuous variable comparison between the two groups, while chi-square test or Fisher′s exact probability method was used for counting data analysis.The related factors affecting HBsAg clearance was analyzed by univariate or multivariate logistic regression analysis.Results A total of 194 patients were enrolled with 112 in therapeutic group and 72 in control group.The HBsAg clearance rate and seroconversion rate at week 48 in therapeutic group were 30.8% (32/104) and 26.0% (27/104), respectively.The rates at week 96 increased to 45.2% (47/104) and 38.5% (40/104), respectively.The HBsAg clearance rates at weeks 48 and 96 in control group were both 1.5% (1/68).HBsAg seroconversion was not achieved in control group.The HBsAg clearance rate in treatment group was significantly higher than that in control group (χ2=39.066, P<0.01).The quantitative HBsAg levels at baseline (OR=2.313, 95%CI: 1.258-4.251, P=0.007), week 12 (OR=3.159, 95%CI: 1.826-5.466, P<0.01) and week 24 (OR=3.347, 95%CI: 2.050-5.465, P<0.01), the decline of HBsAg at week 12 (OR=5.343, 95%CI: 2.085-13.689, P<0.01), and week 24 (OR=4.855, 95%CI: 2.380-9.902, P<0.01), and alanine transaminase (ALT) elevation at week 12 (OR=3.520, 95%CI: 1.369-9.052, P=0.009) were independent predictors for HBsAg clearance.Conclusions Peg IFNα-2a-based treatment for IHC could achieve higher HBsAg clearance rate and seroconversion rate, and has a safety profile.Decline of HBsAg at week 12 and week 24 with ALT elevation at week 12 could predict a higher HBsAg clearance rate.
8.Analysis of high- risk HPV infection status among rural women in Chongqing City

Objective To analyze the situation of high-risk human papillomavirus(HPV) infection and its relationship with cervical lesions among rural women in Chongqing City. Methods A total of 20000 rural women were selected from the districts of Wanzhou, Yongchuan,Banan and Tongnan of Chongqing City from January to April 2016. The cervical cancer preliminary screening was performed by detecting HPV gynotyping. The cases of HPV 16 and HPV 18 positive or other 12 high risk HPV types positive with abnormal colposcopy examination conducted the colposcopy fixed point biopsy. Results The positive rate of high-risk HPV infection was 9.66% among rural women in Chongqing City. The positive rates of high-risk HPV infection and different types of high-risk HPV infection had difference among different age groups. Among women of high-risk HPV infection positive, the occurrence rate of CIN1 was 2.33 %,which of CIN2,CIN3 and AIS was 1.35 %,and which of cervical cancer was 0.31%, the incidence rate of different grades of cervical lesions had no statistical difference among different age groups. Among women with different types of high-risk HPV infection,the incidence rate of different grades of cervical lesions in women of HPV 16 positive was highest, those with the other 12 types of high risk HPV positive had the lowest occurrence rate of different grades of cervical lesions, the differences were statistically significant. Conclusion HPV genotyping detection can increase the efficiency of cervical cancer screen ing and is more suitable for the screening of large population.

10. Low-levels of HBsAg quantification at 48-week in HBeAg-negative chronic hepatitis B patients are the advantageous population for HBsAg clearance

Objective:
To analyze the therapeutic effect on HBeAg-negative chronic hepatitis B patients treated with Peg-IFNα-2a combined with NAs to obtain the influencing factors for predicting HBsAg clearance.
Methods:
A retrospective study was conducted to investigate the effect of pegylated interferon alpha-2a combined with nucleoside analogues (lamivudine/adefovir dipivoxil) on HBeAg-negative chronic hepatitis B. The treatment course was 96 weeks. Patients were followed up 120 weeks after the treatment. HBsAg clearance at 120 weeks was taken as the objective of the study. Logistic regression and receiver operating characteristic curve analysis screened the related factors affecting HBsAg clearance.
