1.Bioactive glass modified polyethylene terephthalate promotes bone healing

BACKGROUND:Polyethylene terephthalate (PEA) holds good mechanical properties, but its biocompatibility needs to be improved. OBJECTIVE:To investigate the effect of bioactive glass modified PEA on bone healing. METHODS:In vitro experiment:PEA (control group) and bioactive glass modified PEA (experimental group) were respectively co-cultured with MC3T3-E1 cells. The cell morphology was observed at 3 days of culture, and the cell proliferation and alkaline phosphatase activity were detected at 1, 3 and 5 days of culture. In vivo experiment:24 New Zealand white rabbits were enrolled, modeled into unilateral knee ligament rupture, and then randomly allotted to two groups. Bioactive glass modified PEA and PEA were implanted into experimental and control groups, respectively. Biomechanics and histological changes were compared between groups at 6 and 12 weeks after transplantation. RESULTS AND CONCLUSION:In vitro experiment:the cell density in the experimental group was significantly higher than that in the control group after 3-day culture (P<0.05);the cell proliferation and alkaline phosphatase activity were significantly higher than those in the control group at 3 and 5 days of culture (P<0.05). In vivo experiment:the maximum axial pull-out strength in the experimental group was significantly greater than that in the control group at 6 and 12 weeks after implantation (P<0.05). In the control group, the defect was obvious and there were a large number of inflammatory cells at 6 weeks after implantation;the defect region became fuzzy, abundant inflammatory cells existed and new bone formed on the surface at 12 weeks after implantation. In the experimental group, there were newly formed bones and trabeculae formed on the interface, the scar tissue became less, and the interface width was narrow at 6 weeks after implantation;numerous new tissues were observed, and only a few inflammatory cells observed in the defect region at 12 weeks after implantation. These results indicate that the modified PEA with bioactive glass possesses good biocompatibility and biomechanics, which can promote bone healing.
2.Reconstruction of the anterior cruciate ligament using Ligament Advanced Reinforcement System: its biocompatibility and bone healing

BACKGROUND: Ligament Advanced Reinforcement Sytstm (LARS) artificial ligament can avoid the interfibrillar friction compared wiht traditional ligaments, which is conductive for autogenous tissue growth. OBJECTIVE: To investigate the biocompatibility of LARS artificial ligament used for cruciate ligament reconstruction and bone healing. METHODS: Twenty goats were randomly divided into two groups: the anterior cruciate ligament insertion at the femur was cut as stump group; the anterior cruciate ligament insertions at the femur and tibia, and all the ligaments and synovial tissues between them were cut as stump resection group. LARS artificial ligament was then implanted into both two groups. At 20 months after implantation, the ligament healing was observed using CT and MRI examinations, the fibrous tissues were observed under scanning electron microscope, and the growth of newborn ligaments was observed by histological staining. RESULTS AND CONCLUSION: CT findings showed that in the stump group, the number of femoral tunnel enlargement was significantly more than that in the stump resection group (P < 0.05), while the number of tibial tunnel enlargement in the stump group was significantly less than that in the stump resection group (P < 0.05). LARS artificial ligament in the femoral and tibial tunnel and free part of the joint presented with hypointensity on T1- and T2-weighted images, which showed no significant difference between two groups. Under the scanning electron microscope there were obvious dense fibrous tissues between LARS artificial ligaments in both two groups; and the autologous fibrous tissues arranged closely and evenly under high magnification. Hematoxylin-eosin staining results showed that in the two groups, the LARS artificial ligaments in the free part of the articular cavity and bone tunnel were surrounded by a large number of autologous tissues in a fusiform arrangement; autologous fibrous tissues and artificial ligament were wrapped well but arranged irregularly; mature fibrous tissues and few capillary infiltration were found in the bone tunnel and free part of the joint cavity, but without necrotic tissues. Masson staining results showed that the inner and outer parts of the LARS in the bone tunnel were wrapped by blue collagen, the red-stained bones located outside the collagen, and no obvious transitional zone appeared. These results showed that the LARS artificial ligament holds good biocompatibility and fibrous tissue induction, which can promote bone healing.

Objective To observe the effects of stone needle on chronic obstructive pulmonary disease in acute exacerbation period (AECOPD). Methods One hundred and forty five cases of AECOPD were randomly divided into three groups. Patients in combined treatment group (n =49) received that stone needle hot compresses the back and Chinese herbal medicine besides routine treatment. Patients in Chinese herbal medicine group (n =48) received Chinese herbal medicine besides routine treatment. Patients in placebo group (n =48) received routine treatment. 4.Regulatory effect of protein tyrosine phosphatase non-receptor type on the cardiac Kv11.1 (HERG) potas-sium channel

Objective Cardiac HERG potassium channel currents can cause long QT syndrome .The function of the cardiac HERG potassium channel is regulated by tyrosine kinase and phosphatase , and protein tyrosine phosphatase non-receptor type 11 ( PTPN11) negatively regulates the HERG potassium channel .This study aimed to investigate the effect of PTPN 11 on the electrophysio-logical characteristics of the HERG channel . Methods The plasmids of pcDNA3.1-PTPN11-EGFP were constructed by PCR technique and transfected or cotransfected with the pcDNA 3.1-PTPN11-EGFP plasmid into HEK293 cells using Lipofectamine 2000.The patch clamp technique was employed to record the HERG channel currents in the control group ( HEK293 cells transfected with pcDNA3.0-HERG-EGFP), PTPN11 overexpression group (pcDNA3.0-HERG and pcDNA3.1-PTPN6-EGFP cotransfected HEK293 cells), and PTPN11 overexpression with PAO group . Results The pcDNA3.1-PTPN11-EGFP plasmid was successfully constructed .Green fluorescence was observed in the HEK293 cells transfected with pcDNA3.0-HERG-EG-FP or cotransfected with pcDNA3.0-HERG and pcDNA3.1-PTPN11.The maximum densities of pulse and tail currents were significantly decreased in the PTPN11 overexpression group as compared with the control ([31.85 ±5.54] vs [45.92 ±3.18] pApF, P<0.05;[73.82 ±11.31] vs [108.43 ±7.98] pApF, P<0.05) but markedly in-creased in the PTPN11 overexpression with PAO group ([48.08 ±4.32] pApF;[120.06 ±8.02] pApF) (P<0.05).The time con-stant of deactivation was significantly higher in the PTPN 11 overexpression group than in the control ([622.16 ±46.49] vs [440.70 ± 49.49] ms, P<0.05). Conclusion The overexpression of PTPN11 decreases HERG potassium channel currents , which can be re-versed by PAO.This finding provides a theoretical basis for the application of certain regulatory enzymes in the HERG channel as a treat -ment of long QT syndrome .
5.Preventive effect of alkaline drinking water on hyperuricemia in mice

Objective:
To observe the preventive effect of alkaline drinking water on hyperuricemia in mice.
Methods:
Sixty male SPF Kunming mice were randomly divided into six groups: pH 7.3, pH 8.0, pH 9.3 intervention groups, in which the mice were given water with pH values of 7.3±0.5, 8.0±0.5 and 9.3±0.6, respectively; the control group, model group and positive drug group ( with 2 g/L allopurinol ) were given double distilled water. Except for the control group, the mice in each group were given yeast by gavage (1.5 g/mL) for 13 days. On the 14th day, the mice were injected with 300 mg/kg potassium oxyzinate by intraperitoneal injection, and then fasted for 1 day. On the 16th day, serum uric acid, creatinine and urea nitrogen were detected, and renal tissues were stained to observe the morphology.The expression levels of neutrophil gelatinase-associated lipocalin ( NGAL ), tissue inhibitor of metalloproteinase 1( TIMP1 ), organic anion transporter 1 ( OAT1 ) and urate transporter 1 ( URAL-1 ) in renal tissues were determined bywestern blotting. The mRNA expression levels of URAL-1 and OAT1 were detected by real-time fluorescent quantita⁃tive polymerase chain reaction.
Results:
The level of serum uric acid was higher in the model group than in the control group and in the pH 9.3 intervention group (both P<0.05). The number and area of renal tubular lesions were less in the pH 9.3 intervention group than in the model group (all P<0.05). The relative expression levels of NGAL and
URAT-1 proteins were lower in the pH 9.3 intervention group than in the model group, and the relative expression level of OAT1 protein was higher in the pH 9.3 intervention group than in the model group ( all P<0.05). The relativeexpression level of URAT-1 mRNA was lower in the pH 9.3 intervention group than in the model group, and the rela⁃tive expression level of OAT1 mRNA was higher in the pH 9.3 intervention group than in the model group ( all P<0.05 ).
Conclusion
Alkaline drinking water with pH value of 9.3±0.6 can effectively prevent hyperuricemia and acute
kidney injury in mice.
6.Effects of FOLFOX neoadjuvant chemotherapy protocol on non-resectable advanced gastric Cancer

Objective To study the clinical efficacy and adverse reactions of the preoperative neo-adjuvant chemotherapy with oxaliplatin plus fluorouracil and calcium folinate (FOLFOX) in the treatment of advanced gastric carcinoma that was difficult to undergo radical operation. Methods We enrolled 16 patients with advanced gastric carcinomas that were unable to undergo radical operation admitted to our hospital from April 2008 to October 2009. The neo-adjuvant chemotherapy regimen was oxaliplatin 130 mg/m~2 for first one, fluorouracil 500 mg/m~2 and calcium folinate 200 mg/m~2 from first day to fifth day,and one treatment course was three weeks and the patients underwent two courses.The changes of the primary lesion and the adverse reactions of the patients were observed. Results Carcinomas of 13 patients were degraded after the treatment, of which, 11 patients underwent radical resections after 4-6 weeks after operation.There were two cases of complete remission (CR) , 10 cases of partial remission (PR), three cases of stable disease(SD) and one case of progress disease(PD) , and the total effective rate was 75.0% (12/16). Adverse reactions included bone marrow suppression, diarrhea, nausea and vomiting and paresthesia, which were relieved after appropriate symptomatic treatment. Conclusions Neoadjuvant chemotherapy with FOLFOX can improve the resection rate of the advanced gastric carcinoma that is unable to undergo radical resection, and the patients' tolerability to this regimen is favorable.Thus, the regimen is worthy of generalizing.
7.Study on the Applicability of NIR Combined with LS-SVM Used for the Quality Monitoring of Medicated Leaven Fermentation

This article investigated the applicability of the near infrared spectroscopy (NIR) technology, combined with the least squares support vector machines (LS SVM) used for the quality monitoring of medicated leaven fermentation. First, near infrared spectra of 67 medicated leaven samples were obtained by near infrared spectroscopy system in the wavelength range of 400 2 500 nm, and then the protease and amylase activity were measured by Folin phenol method and DNS method. Thereafter, the LS SVM was employed to calibrate models. The Rc and Rp of protease in near infrared model were 0.975 and 0.938, respectively; The RMSEC and RMSEP were 5.297 and 9.795, respectively. The Rc and Rp of amylase in near infrared model were 0.987 and 0.973, respectively; The RMSEC and RMSEP were 7.215 and 6.864, respectively. This model has good prediction ability and is suitable for quality monitoring in medicated leaven fermentation process. The research achievement could lay a certain foundation for the near infrared spectral analysis technology applied in the field of traditional fermentation processing.
8.Effect of ligustrazine on cardiac inflammation in rats with type 2 diabetes mellitus

AIM: To study the effect of ligustrazine on the cardiacmyocyte lesion in rats with type 2 diabetes mellitus.METHODS: Male Wistar rats were injected with STZ via tail vein under high-glucose and high-fat feeding for 4 weeks to establish the animal model of type 2 diabetes mellitus.Ligustrazine at different doses was used to treat the diabetic rats.The body weight, blood glucose and the morphology of heart tissues were observed.The myocardial levels of IL-1β, IL-6 and TNF-α were detected by ELISA, and the protein expression of IKKβ and NF-κB in the myocardium was determined by Westeren blotting.RESULTS: Ligustrazine at high dose alleviated the body weight reduction and blood glucose elevation cause by diabetes, and reduced pro-inflammatory factors IL-1β, TNF-α and IL-6.Moreover, the protein expression of IKKβ and NF-κB was significant decreased by ligustrazine.CONCLUSION: Ligustrazine inhibits the myocardial inflammation caused by diabetes through anti-inflammatory pathway.
9.Clinical observation of percutaneous vertebroplasty assisted by four-axis positioning 3D printing guide plate

Objective To compare the difference between four-axis positioning 3D printing guide assisted puncture and manual puncture percutaneous vertebroplasty,to clarify the clinical efficacy of 3D printing guide assisted percutaneous vertebroplasty,and to explore its clinical application value.Methods A total of 70 patients who underwent single-segment vertebroplasty for osteoporotic vertebral compression fractures in our hospital were randomly divided into a manual group(control group)and a guide plate group(observation group).Thirty-five patients in the control group underwent PVP under the traditional manual puncture,and 35 patients in the observa-tion group underwent PVP under the four-axis positioning 3D printing guide assisted puncture.The general data,perioperative data,imaging data,preoperative and postoperative functional scores of the two groups were collected and compared.Results In terms of intraoperative fluoroscopy times,operation time,and postoperative adverse reactions,the observation group was significantly lower than the control group,and the difference was statistically significant(P<0.05).However,there was no significant difference between the two groups in intraoperative blood loss,bone cement leakage,and whether bone cement perfusion included fracture sites.In terms of clinical efficacy,the ratio of anterior vertebral height,postoperative VAS score and postoperative JOA score of the two groups were significantly better than those before operation,and the difference was statistically significant(P<0.05).At the same time point before and after operation,there was no significant difference in the height ratio of the anterior edge of the injured vertebra,VAS score and JOA score between the two groups(P>0.05).Conclusion The clinical efficacy of four-axis positioning 3D printing guide plate-assisted puncture is the same as that of experienced clinicians with free hand puncture for PVP,which can quickly relieve pain and improve dysfunction.However,guide plate assistance can shorten the operation time,reduce the number of fluoroscopy and postoperative adverse reactions,and make vertebroplasty faster and safer.
10.Impact of sample data repeatability on NIR calibration model.

OBJECTIVETo investigate the impact of repeated data acquisition on the stability of NIR quantitative calibration model, and make a preliminary analysis on reasons for the impact.
METHODYinhuang decoction was used as the subject, and NIR spectrum samples were collected. By reference to HPLC's determination value, the baicalin quantitative calibration model was established by using recursive least square algorithm to detect cumulative-LVs curve of latent variables. The impact of calibration model caused by repetitive samples was explained in latent variance space.
RESULTAfter averaging the repetitive spectrum samples, quantitative prediction model, which was built by optimal method of spectrum pretreatment, showed the ideal prediction result (RMSECV = 1.824). The area under the cumulative-LVs curve of latent variables was obviously larger than other modeling methods, i. e., this model is more stable.
CONCLUSIONAveraging of multiple measurements can dramatically improve the predictive ability of the model and make the model more stable.
