1.microRNA-27a regulates maturation process and cytokine in LPS-stimulated dendritic cells

Objective:To explore the effects of miR-27a on the phenotype and cytokine secretion in LPS-stimulated dendritic cells.Methods:Murine bone marrow-derived dendritic cells were transfected with miR-27a mimics and negative control mimics,and then stimulated by LPS for 24 hours.Dendritic cells exposed to LPS were collected for analysis of the DC immunophenotype by flow cy-tometry and supernatants were collected to determine cytokine lever.Moreover,the capability of stimulating allogeneic CD4 T+cell prolif -eration was measured by MLR ( mixed lymphocyte reaction) .Results: The levels of MHCⅡ, CD80, and CD86 were significantly increased in LPS-stimulated dendritic cells when compared with imDC ( P<0.001).Transfection with miR-27a mimics resulted in sig-nificantly lower expression levels in levels of MHCⅡ,CD80,and CD86 (P<0.001).For cytokine secretion,transfection with miR-27a mimics enhanced IL-10 production (P<0.01) and reduced the production of IL-12 (P<0.05).For MLR,transfection with miR-27a mimics suppressed allogeneic CD4+T cell proliferation.Conclusion: MiR-27a may play critical roles in regulating the maturation process and cytokine secretion in LPS-stimulated dendritic cells.
2.Analysis on risk factors of hospital-acquired pneumonia based on expert questionnaire

Objective To explore the risk factors of hospital-acquired pneumonia from the point of view of experts′experiences with the method of expert questionnaire .Methods The questionnaire on the risk factors of hospital-acquired pneumonia , which was made based on the literature consultation and group discussion , were distributed to 210 experts selected from the whole city .The degree of mean , the coefficient of variation ( CV) and the cumulative percent of the risk factors were surveyed .Results The survey distributed 210 cases of questionnaires in all .The effective retrieving rate was 96.2%.The mean value of degree of mean , the cumulative percent and CV were 3.80, 86.7% and 0.30 respectively.Male, aerosol inhalation, low hemoglobin production and cognitive disorder were excluded , according to the discussed criteria .The resting 27 items were considered as the risk factors of hospital acquired pneumonia .Conclusions This study determines the risk factors of hospital-acquired pneumonia by conducting expert questionnaire , which can be a complementary solution to previous retrospective case control study and afford the foundation to further prevention of hospital -acquired pneumonia .
3.Evaluation analysis of nurse′s satisfaction based on quality of nursing

Objective To investigate job satisfaction of clinical nurses in Shanghai, and explore the influencing factors so as to provide evidence for improving nursing quality and nursing science development. Methods A total of 1 149 nurses had been selected to take part in the nurse′s satisfaction measurement in the level two or beyond hospital, and we explored the impacted factors in order to provide evidence for the improvement of nursing quality by stratified sampling method in 2013 in 71 level two and beyond hospitals Shanghai. Results The total score of nurse′s satisfaction was over 4, and nurses were satisfied the colleague cooperation highest but the career development was lowest. The top three dimension scores of satisfaction were colleague cooperation (4. 94 ± 0. 24), supervisor support (4. 93 ± 0. 26), and leader communication (4. 92 ± 1. 75). The reward satisfaction of nursing job and satisfaction of nursing management had positive correlation with satisfaction of nurse experience (r=0. 85, 0. 82;P<0. 05). Conclusions Multiple influencing factors impacts on satisfaction of nurses, but how adjust rigid system, arrange human resources well and achieve the value. The recognition of nursing job are a significant topic and are worthy to study further for the improvement of nursing quality.
4.Comparison of inpatient satisfaction between general hospital and special hospital of grade-III in Shanghai

Objective To explore the satisfaction of inpatients in general hospital and special hospital of grade-Ⅲ in Shanghai, thus to provide the basis for policy making to improve nursing service quality. Methods Inpatients of 13 grade-Ⅲ general hospitals and 5 grade-Ⅲspecial hospitals in Shanghai were drawn-off by random sampling method and underwent questionnaires by recruited and the third party group. The satisfaction of inpatients was evaluated by relevant data analysis. Results A total of 1 377 calls were dialled in this investigation, but just 689 questionnaires were returned,so the return-rate was 50. 04%, among which 551 questionnaires were valid (valid return rate was 40. 01%). There was statistically significant difference between general hospital and special hospital of grade-Ⅲ in terms of daily care, awareness of patient′s needs during nursing round, nursing skill, communication before nursing care, privacy and health education before discharge by χ2 test (P<0. 05). The mean score of overall satisfaction in general hospital was 99. 42, and 99. 75 for special hospitals, which was of statistically significant difference (P<0. 05). Conclusions Survey of inpatient satisfaction between different level hospitals is beneficial for detecting the difference between general hospital and special hospital of grade-Ⅲ and finds out the shortcomings of different types hospitals, so as to provide the evidence to improve the quality of care targeted.
