1.A Study in Effect of Artemisinin and its Derivatives on Reversion of Multi-drug Resistance of Tumors

Objective Effect of artemisinin and its derivatives on multi-drug resistance of tumor cells. Method The inhibition of cell proliferation and RI (reversal index) were determined by MTT method. Results The inhibitory effect of dihydroartemisinin and artemisunate on the proliferation of tumor cell is stronger than that of artemisinin. Artemisinin could partly improve KBv200 cell sensitivity to VCR in different concentrations. Conclusion Artemisunate and dihydroartemisinin effectively inhibited the proliferation of KBv200. Artemisinin could partly improve KBv200 cells sensitivity to chemotherapy medicine.
2.Reliability of flexural strength and subcritical crack growth of two kinds of zirconia ceramics

Objective:To evaluate the reliability of flexural strength and subcritical crack growth(SCG)of WL and HT zirconia ce-ramics under cyclic loading.Methods:Bar-shaped speciments of WL(n =30)and HT(n =30)were fabricated and loaded in three-point bending and cyclically fatigued.The flexural strength and the number of cycles to fracture were obtained from three-point flexural test and cyclic fatigue test respectively.Data were examined using Weibull statistics.Pairs law was used to analysize the subcritical crack growth(SCG),the behavior of which was described in terms of crack velocity as a function of maximum stress intensity factor (KImax ).Results:The Weibull module(m)of WL and HT was 10.64 and 12.04,the stress corrosion cracking susceptive indexes(n) 23.71 and 32.87,the SCG 90% and 76.7%,the KImax 49.6% and 50.9% of its maximum stress intensity factor(KIC ),respectively. Conclusion:WL and HT produce pronounced SCG.The minimum fatigue crack growth rate of HT is lower than that of WL(νHT <νWL ).
3.Tissue distribution of PEGylated puerarin in acute myocardial ischemia mode rats.

The aim of this study is to explore the tissue distribution of PEGylated puerarin in acute myocardial ischemia model rats. Healthy male SD rats were randomly divided into two groups (30 each). Both were given PEGylated puerarin at a dose of 488 mg x kg(-1). After 5 min of medication, one group was normal rats, another group with acute myocardial ischemia was established by peritoneal injection of 50 mg x kg(-1) isoprenaline. After administration, the animals were executed at 30, 60, 90, 120, 150 and 180 min, then heart, liver, spleen, lung, kidney were extracted. The content of puerarin in organ tissue was determined by HPLC. The results showed that the AUC of tissue distribution of PEGylated puerarin in normal rats was liver > kidney > heart ≈ spleen > lung > brain. While the AUC of tissue distribution of PEGylated puerarin in acute myocardial ischemia model rats was liver ≈ heart > kidney > lung ≈ spleen > brain. AUC(heart) of PEGylated puerarin in acute myocardial ischemia model rats was 1.7 times than that of the normal rats, and there was significant difference (P < 0.05). Thus, PEGylated puerarin had a good heart-targeting property in early myocardial infarction area, drugs could accumulate in the ischemic myocardium. It provided important information for further study and clinic use of PEGylated puerarin.
4.Clinical application of the adjacent horn shaped perforator fasciocutaneous flap in the trunk area.

OBJECTIVETo study the anatomy basis for the clinical application of the adjacent horn shaped perforator fasciocutaneous flap for the reconstruction of small and medium-sized defects in the trunk area.
METHODS(1) Ten adult antiseptic cadavers (20 sides) were perfused with red latex. The skin blood supply, line of the blood vessels, branches in accordance with the distribution and crossing were observed. (2) Fifteen cases with defects in the trunk were treated with the adjacent horn shaped perforator fasciocutaneous flaps. The defects size ranged from 5 cm x 5 cm to 13 cm x 13 cm with the size of the flaps ranging from 10 cm x 6 cm to 35 cm x 15 cm.
RESULTSThe trunk skin is supplied by mainly 17 groups arteries such as thyrocervical trunk, internal thoracic artery, posterior intercostal arteries, superior epigastric artery, arteria epigastrica inferior, lumbar arteries, and so on. The perforators (diameter > 0.5 mm) numbers are about 20, 40, 24, 6, on the chest, abdomen and perineum, upper back, waist, respectively. All the flaps survived completely with primary healing both on donor and recipient sites. The flaps color, texture, function and appearance were satisfactory during the follow-up period of 1-24 months.
CONCLUSIONSThe adjacent horn shaped perforator fasciocutaneous flap should be designed flexibly. The defects in the donor sites could be closed directly without skin graft. It is an effective, easy and ideal method for the reconstruction of large defects in the trunk.

5.Relationship between homocysteine and sarcopenia in the elderly population

Objective:
To investigate the relationship between total homocysteine (tHcy) levels and sarcopenia among the elderly, so as to provide insights into the prevention and treatment of sarcopenia.
Methods:
The elderly aged 65 years and older who participated in the physical examination of Shibantan Township Health Center in Xindu District, Chengdu City from April to June 2021 was selected as the study subjects. The elderly with sarcopenia (diagnosed according to the diagnostic criteria of the Asian Sarcopenia Working Group in 2019) and non-sarcopenia were matched 1︰1 by gender and age (±2 years). Demographic information, skeletal muscle mass, skeletal muscle strength and tHcy were collected through questionnaire surveys, physical examination and laboratory testing. Multivariable conditional logistic regression model was used to explore the relationship between tHcy and sarcopenia.
Results:
A total of 320 individuals, including 160 sarcopenia patients and 160 non-sarcopenia individuals, were investigated. There were 138 males (43.13%) and 182 females (56.87%), with a median age of 71.00 (interquartile range, 6.00) years. There were 57 drinkers (17.81%), 78 smokers (24.37%), 173 cases of hypertension (54.06%) and 124 cases of hyperhomocysteinemia (38.80%). Multivariable conditional logistic regression analysis showed that elevated tHcy was associated with an increased risk of sarcopenia (OR=1.107, 95%CI: 1.024-1.197), after adjusting for smoking, alcohol consumption, hypertension, waist circumference, neck circumference, body mass index, platelet count and high density lipoprotein cholesterol.
Conclusion
Elevated tHcy is associated with sarcopenia, and intervention should be carried out for the elderly with higher tHcy.
6.Effectiveness of exercise intervention on sleep quality and negative emotion among female college students with anxiety

Objective:
To investigate the effect of exercise intervention on sleep quality of female college students with anxiety, and to further explore the mediating effect of negative emotions in this association.
Methods:
Seventy female college students were randomly divided into experimental group (34 patients) and control group(36 patients). The experimental group received eight-week aerobic exercise and yoga training. The control group received no training. Sleep Quality Scale, Negative Emotion Scale and Anxiety Self-rating Scale were used to assess the level of sleep and negative emotion before and after exercise intervention.
Results:
After exercise intervention, sleep quality (4.31±1.26), anxiety (36.41±7.32) and negative emotion (2.37±0.50) in the experimental group significantly improved(P<0.01), no similar changes were found in the control group(P<0.05). By examining the mediating effects, negative emotions played a partial mediating role in the association between exercise and sleep quality(t=6.77, P<0.01).
Conclusion
Exercise intervention significantly improved the quality of sleep and negative emotion among female college students with anxiety. Negative emotions play a partial role in the positive effect of exercise on sleep quality.
7.Inhibition of growth and proliferation of Hep-2 cells by targeting c-myc gene using small interference RNA technology.

OBJECTIVE:
To investigate the effect of small interfering RNA (siRNA) targeting c-myc gene in Hep-2 cells.
METHOD:
siRNA targeting c-myc mRNA was designed and synthesized. In vitro cultured Hep-2 cells were transfected with lipofectamine 2000 and the inhibitory effect was detected by MTT, morphology, real time PCR assay.
RESULT:
1) The MTT result showed the c-myc siRNA to be able effectively to suppress the Hep-2 cell multiplication; 2) The real time PCR result showed c-myc at mRNA level inhibition ratio at 94% in group S3; 3) The morphology result showed the c-myc siRNA to be able effectively to suppress the Hep-2 cell multiplication, the cell heteromorphism was diminished.
CONCLUSION
siRNA targeting c-myc can remarkably suppress the Hep-2 cell growth and multiplication.

8.Clinical efficacy of a new low notch plate system and Zero-P in the treatment of single segment cervical spondylotic radiculopathy

Objective To compare the clinical efficacy of a new low notch plate system(Carmen synchronous adaptive cervical fusion system)and Zero-p in the treatment of single level cervical spondylotic radiculopathy(CSR).Methods Retrospective analysis was made on 48 patients with single level cervical spondylosis of nerve root type admitted from January 2015 to January 2020,including 26 patients with new low notch steel plate system(observation group)and 22 patients with Zero-P(control group).The operation duration,intraoperative blood loss,incidence of postoperative dysphagia,preoperative and postoperative JOA scores,cervical dysfunction index(NDI)Visual analog score of pain(VAS)and Cobb angle and intervertebral space height of postoperative imaging.Results There was no significant difference between the two groups in terms of operation time,intraoperative blood loss and the incidence of postoperative dysphagia(P>0.05).All patients were followed up for 12-15 months,with an average of(12.6±0.7)months.There was no significant difference in JOA score,NDI index and VAS score of pain between the two groups before and 3 and 12 months after operation(P>0.05).Conclusion The clinical efficacy of the new low notch plate system in the treatment of single segment cervical spondylotic radiculopathy is equivalent to Zero-P,and it is a reliable new type of anterior cervical internal fixation system.
9.Association of greenness surrounding school with aggression among primary school students

Objective:
To explore the association between greenness surrounding school and aggression among primary school students, and to explore the potential mediating roles of social support, loneliness, particulate matter (PM2.5) and Nitrogen Dioxide (NO2) in this association, in order to provide a scientific reference for preventing and ameliorating aggressive behaviors of primary school students.
Methods:
The data was used from a survey of children and adolescents conducted in 2015. The Chinese version of the Buss-Warren Aggression Questionnaire was used to assess total and subtypes of aggression, and the mean values of normalized difference vegetation index (NDVI) of 100 m, 500 m, 1 000 m circular buffers surrounding school were used to indicate the participants greenness exposure. PM2.5 and NO2 datas were obtained from the China High Air Pollutants Dataset.Generalized Linear Mixed Models were used to assess the associations of greenness surrounding school with total and subtypes of aggression.
Results:
Per IQR increment of NDVI-500 m [OR(95%CI)=1.09(1.03-1.15)] and NDVI-1 000 m[OR(95%CI)=1.07(1.02-1.13)] were positively correlated with physical aggression among primary school children, and per IQR increment of NDVI-100 m [OR(95%CI)=0.94(0.90-0.99)], NDVI-500 m [OR(95%CI)=0.93(0.89-0.98)] and NDVI-1 000 m [OR(95%CI)=0.95(0.91-1.00)] were negatively associated with verbal aggression (P<0.05). Mediation analyses revealed that social support partially mediated the association between the NDVI-500 m and physical aggression (mediation ratio:18.0%) and verbal aggression (mediation ratio:-8.3%) among primary school students, and loneliness partially mediated the association between the NDVI-500 m and physical aggression and verbal aggression among elementary school students effects, with proportion mediated ratios of -10.0% and 21.0%, respectively (P<0.05).
Conclusions
Exposure to school surrounding greenness is likely to associated with physical aggression and verbal aggression in primary school students, and social support and loneliness may partially mediate these associations.
10.Effect and mechanism of ophthalmic acupuncture combined with tail moxibustion in post-stroke cognitive impairment rats

Objective To explore the effects and possible mechanisms of ophthalmic acupuncture combined with tail moxibustion on the behavior and hippocampus in rats with post-stroke cognitive impairment.Methods Seventy-five male SD rats were randomly divided into the sham operation group (15 rats) and the modeling group (60 rats) using the random number table method. A modified suture-occluded method was used to replicate the occlusion model of the middle cerebral artery in the modeling group,whereas only the right carotid artery was exposed in the sham operation group. After modeling,48 rats with post-stroke cognitive impairment were selected using the Morris water maze experiment,and were randomly divided into the model group,the ophthalmic acupuncture group,the tail moxibustion group and the ophthalmic acupuncture+tail moxibustion group using the random number table method,with 12 rats per group. The sham operation group and the model group were bound with no intervention;the ophthalmic acupuncture group was needled once a day in the bilateral "liver area","kidney area","heart area",and "spleen area",leaving the needle for 30 min;the tail moxibustion group was given mild-warm moxibustion on the area between "Changqiang" (GV1) and the tip of the tail for 20 min,once a day;the ophthalmic acupuncture+tail moxibustion group was given the above-mentioned ophthalmic acupuncture and tail moxibustion interventions simultaneously. After 7 days of intervention,the behavior of the rats was detected. Hematoxylin and eosin staining was used to observe the pathological changes in the hippocampus;the malondialdehyde (MDA) content and superoxide dismutase (SOD) activity in the hippocampus were detected by colorimetric method;Western blotting was used to detect the protein expressions of Kelch-like ECH associated protein 1 (KEAP1),phosphoglycerate mutase 5 (PGAM5),apoptosis-inducing factor mitochondria associated 1 (AIFM1),nuclear factor erythroid 2-related factor 2 (Nrf2),and heme oxygenase-1 (HO-1) in the hippocampus of rats;and real-time fluorogenic quantitative PCR was used to determine the mRNA expressions of KEAP1,PGAM5,AIFM1,Nrf2,and HO-1 in the hippocampus. Results Compared with the sham operation group,the escape latency in the model group was prolonged,and the crossing platform number was decreased (P<0.05);the number of neurons in the hippocampal CA1 area was significantly decreased,with a disordered arrangement and irregular morphology,and necrotic neurons were observed;the SOD activity in the hippocampus was decreased,while the MDA content was increased (P<0.05);the protein and mRNA expressions of KEAP1,PGAM5,and AIFM1 were increased,while the protein and mRNA expressions of Nrf2 and HO-1 were decreased (P<0.05). Compared with the model group,the escape latency of rats in the ophthalmic acupuncture+tail moxibustion group was shortened,and the crossing platform number was increased (P<0.05);the loss of neurons in the hippocampal CA1 area of the rat was significantly reduced,and the cell morphology was more plump;SOD activity in the hippocampus was increased,and MDA content was decreased (P<0.05);and the protein and mRNA expressions of KEAP1,PGAM5,and AIFM1 were decreased,while the protein and mRNA expressions of Nrf2 and HO-1 were increased (P<0.05).Conclusion The combination of ophthalmic acupuncture and tail moxibustion can be used to treat rats with post-stroke cognitive impairment,and its mechanism may be related to alleviating oxidative stress damage and oxeiptosis in the hippocampus,thereby improving the degree of hippocampal neuronal damage and enhancing the cognitive ability of rats after stroke.
