Cerebral palsy (CP) is the most common activity limitition of children, their movement and posture impairments persist throughout whole life.In recent years, CP has been significantly increasing with the improved survival rate of newborns.This may lead to a great burden and costs to both the family and the society.A variety of risk factors have been proposed to be associated with CP.However, recent abroad researches indicate that genetic factors may predispose to CP of newborns and initial results of related researches infer that several susceptibility genes may contribute to CP's development, masqueraders have a great impact on CP's clinical symptoms.Now, the recent publications related to virulence genes and masqueraders of CP are reviewed.
3.Depression and oxidative damage in TNM stage Ⅲ patients with poorly differentiated gastric adenocarcinoma

Objective To investigate the association between psychological stress and oxidative damage in TNM stage Ⅲ patients with poorly differentiated gastric adenecarcinoma (GA). Methods One hundred and six patients with newly diagnosed poorly differentiated GA were assessed using the Hamilton Depression Rating Scale (HAMD), Zung Self-rating Depression Scale (SDS), Zung Self-rating Anxiety Scale (SAS), Symptom Checklist 90 (SCL-90), activities of daily living (ADL) and other multiple-item qnestionnaires. Oxidative-stress-related parameters in serum and the expression of DNA repair genes were monitored during a pretreatment period. Results The patients were divided into depression and nondepression groups (Groups A and B, respectively) based on a HAMD score cutoff of 20. The mean SDS, SAS, SCL-90, ADL and passive coping scores were higher in Group A, whereas social support and quality of life were lower. Serum total antioxidant capacity, eatalase, superoxide dismutuse concentrations and anti-superoxide anion capacity (A-ASC) were significantly decreased in Group A, whereas serum malondialdehyde (MDA) and 8-hydroxy-deoxyguanosine (8-OHdG) levels were significantly increased. Pearson correlation analysis revealed that depression was pesitively correlated with MDA, SAS, SCL-90 and ADL, but negatively correlated with A-ASC. Furthermore, real-time PCR revealed that the expression levels of hOGG1 and APEX1 were increased in Group A. Conclusion Psychological stress might be related to impaired antioxidant system in patients with GA, and it presents the first evidence of the involvement of oxidative DNA damage in the pathogenesis of depression.
4.Bioinformatics analysis of microRNAs differently expressed in major depression disorder

Objective To predict the target genes and function of has-miR-26b,has-miR-1972,has-miR4485,has-miR-4498,and has-miR-4743 by bioinformatics analysis,and provide the theoretical basis for the further research.Methods The targets of the five microRNAs were predicted by Target Scan,miRBD,and DIANA-microT-CDS,and the result were analyzed by gene ontology and pathway analysis using FunNet.Results 734 predicted targets were obtained by finding the intersected genes of Target Scan,miRBD,and DIANA-microT-CDS.GO analysis showed that biological processes regulated by the differentially expressed microRNAs included diverse terms,among which some terms (e.g.,central nervous system development,neuron differentiation,axonogenesis,synaptic transmission,learning,and memory,etc.) had direct relationship with the central nervous system and brain functions.The pathway analysis showed that a significant enrichment in several pathways related to neuronal brain function,such as axon guidance,glutamatergic synapse,Wnt signaling pathway,mTOR signaling pathway,VEGF signaling pathway,etc.Among the five microRNAs,has-miR-26b,has-miR-1972,has-miR-4498 might have more important regulatory functions.Conclusion Bioinformatic analysis indicates that has-miR-26b,has-miR-1972,has-miR-4485,has-miR-4498,and has-miR-4743 are closely related to the mechanism and pathogenesis of major depressive disorder.
5.Diagnostic value of microRNAs in peripheral venous blood for patients with depressive disorder

Objective To investigate the diagnostic value of microRNA (miRNA) in peripheral venous blood for depressive disorder.Methods Real-time fluorescence quantitative PCR (RT-qPCR) was used to verify expression level of miRNAs in peripheral blood of non-specific mental retardation children,which were aberrantly expressed in depressive disorder patients,and then Receiver Operating Characteristics (ROC)curve was employed to confirm the sensitivity and specificity of abnormal miRNA expression in depressive disorder and non-specific mental retardation.Results MiR-1972,miR-26b,miR-4485,miR-4498 and miR-4743 were upregulated significantly in case group of depressive disorder(P<0.05),meanwhile miR-4485 and miR-4743 in aforementioned 5 miRNAs also upregulated significandy in patients of non-specific mental retardation(P<0.05),but miR-26b showed no significant difference between case group of non-specific mental retardation and the control group (P>0.05).The ROC curve of miR-26b in depressive disorder patients and their control group showed that the sensitivity and specificity were 0.609,0.664 respectively,and the area square under the curve was 0.614(P=0.021).The ROC curve of miR-26b in patients of depressive disorder and non-specific mental retardation indicated that the sensitivity and specificity were 0.784 and 0.471,and area square under the curve was 0.643 (P=0.003).Conclusion miR-26b probably have diagnostic value for depressive disorder,which may comorbidity with non-specific mental retardation.But genetic and psychosocial mechanism of comorbidity still needs further exploration.

7.Literature Analysis of Hepatotoxicity Induced by Quinolones

OBJECTIVE: To investigate the characteristics and regularity of hepatotoxicity induced by quinolones, and to provide reference for clinical use of drug safely. METHODS: Using “quinolone” “floxacin” “hepatotoxicity” “hepatic injury”as retrieval words, relevant literatures about hepatotoxicity induced by quinolones were retrieved from domestic and foreign databases as CNKI, Wanfang, VIP, PubMed (during database establishment to 31th, Dec. 2017). Those literatures were summarized and analyzed. RESULTS: A total of 59 valid literatures were collected, including 61 cases of hepatotoxicity induced by quinolones, 8 types of drugs as ciprofloxacin, moxifloxacin, ofloxacin, lomefloxacin, norfloxacin, levofloxacin, gatifloxacin and enoxacin. Ciprofloxacin, levofloxacin, moxifloxacin and ofloxacin were the most common drugs that caused hepatotoxicity, involving 19, 13, 11, 7 cases, respectively; accumulative constitute ratio was 81.97%. The ratio of male to female was 1.54 ∶ 1, and hepatotoxicity always happened at the age of 61 to 80 (30 cases, 49.18%). Primary diseases of 46 cases were single disease (75.41%), and mainly were infection of respiratory system and urogenital system. There were 15 cases of combined disease (24.59%). Thirty-one cases used quinolones alone, most of which was ciprofloxacin. There were 30 cases of drug combination. Thirty-four cases were given drug intravenously and mainly were domestic cases. The hepatotoxicity first occurred within 10 minutes after administration and at the latest 8 weeks after administration. Forty-nine patients suffered from hepatotoxicity within 10 days after medication, accounting for 80.33%. Besides general fatigue, nausea and vomiting, clinical symptoms also included abnormal elevation of alanine aminotransferase, aspartate aminotransferase and total bilirubin,etc. Fifty-four patients were improved after withdrawal or symptomatic treatment, while 7 patients died. The results of causality evaluation of drug-induced hepatic injury showed that there were 4 probably association cases, 45 likely association cases and 12 possible association cases. CONCLUSIONS: The hepatotoxicity caused by quinolones is related to drug variety, patient’s age, primary disease, drug combination and route of administration, and mostly occurs within 10 days after administration. Great importance should be attached to patient’s liver function indexes, strengthen medication monitoring, and carefully combined use of drugs.

Objective:To construct the recombinant prokaryotic plasmid to express HCV HLA-A2 restricted multi-CTL epitopes and to purify the fused protein for antigenic analysis.Methods:The human ubiquitin gene and multi-CTL epitopes gene was synthesized respectively,and digested by restrict enzyme before being cloned into pRSET-A.Then it was transformed into E.coli DH5α and the positive recombinant plasmid named pRSET-Ub-Mep was sequenced.Target protein was distinctly expressed after transformed into E.coli BL21 and induced with IPTG.Thus the protein was scanned and purified on Ni~(2+)-NTA column as well as Western blot performed after solubility analysis.Results:The recombinant plasmid pRSET-Ub-Mep was successfully constructed and it could efficiently express the target gene.Protein production was mainly in inclusion body and could be purified through Ni~(2+)-NTA column.The purified protein kept the antigen activity.Conclusion:The gene encoding for HCV HLA-A2-restricted multi-CTL epitopes is efficiently expressed and the target protein is purified,which establishes a foundation of further research to evaluate the cellular immune response induced by the target gene.
9.Diagnostic performance of ADC value and texture features based on T 2WI fat suppressed image to distinguish benign and malignant soft tissue tumors

Objective:To investigate the value of ADC derived from DWI combined with texture analysis derived from T 2WI fat suppressed images in distinguishing benign and malignant soft tissue tumors. Methods:The MRI and DWI images of 94 patients with soft tissue tumors (44 cases with malignant and 50 cases with benign) confirmed by pathology were analyzed retrospectively in the First Affiliated Hospital of USTC West District. ADC values of solid components were measured at GE ADW4.6 workstation. The texture features were extracted by manually drawing the ROI on the maximum level of the T 2WI fat suppressed images; the ADC values and texture parameters between the two groups were statistically analyzed by SPSS17.0, and the multivariate logistic regression model were conducted to analyze and calculate the diagnostic performance. Results:ADC value of benign and malignant soft tissue tumors was (1.6±0.3)×10 -3 mm 2/s, (1.2±0.5)×10 -3 mm 2/s, respectively, and the difference was statistically significant( t=-5.382, P<0.05). Taking 1.28×10 -3 mm 2/s as the critical value, the area under curve (AUC) for the diagnosis of benign and malignant soft tissue tumors was 0.783, the sensitivity was 92.00%, and the specificity was 65.91%. Among the texture features, the AUC of frequency size, skewness, Inertia All Direction_offset7, Inverse Difference Moment angle0_offset1, Inverse Difference Moment angle0_offset7 and Haralick Correlation All Direction_offset4_SD distinguishing benign and malignant soft tissue tumors were 0.825, 0.739, 0.826, 0.816, 0.820 and 0.783, respectively. The AUC, sensitivity and specificity of the best predictive model distinguishing benign and malignant soft tissue tumors were 0.930, 88.00% and 86.36% respectively using multivariate logistic regression analysis. Conclusion:ADC combined with texture analysis is of great value in preoperative differentiation of benign and malignant soft tissue tumors.
