AbstractObjective: To explore the effect and mechanism of curcumin on human T-cell acute lymphoblastic leukemia (T-ALL) cell apoptosis induced by Mcl-1 small molecule inhibitors UMI-77.
METHODS:
T-ALL cell line Molt-4 was cultured, and the cells were treated with different concentrations of curcumin and Mcl-1 small molecule inhibitor UMI-77 for 24 h. The MTT method was used to detect the cell survival rate after different treatment; According to the results of curcumin and UMI-77, the experimental settings were divided into control group, curcumin group (20 μmol/L curcumin treated cells), UMI-77 group (15 μmol/L Mcl-1 small molecule inhibitor UMI-77 treated cells) and curcumin+ UMI-77 group (20 μmol/L curcumin and 15 μmol/L Mcl-1 small molecule inhibitor UMI-77 treated cells), MTT method was used to detect cell proliferation inhibition rate, Annexin V-FITC/PI double staining method and TUNEL staining were used to detect cell apoptosis, DCFH-DA probe was used to detect cell reactive oxygen species, JC-1 fluorescent probe was used to detect mitochondrial membrane potential, Western blot was used to detect the expression levels of apoptosis-related proteins and Notch1 signaling pathway-related proteins.
RESULTS:
After the treatment of Molt-4 cells with different concentrations of curcumin and Mcl-1 small molecule inhibitor UMI-77, the cell survival rate was decreased (P<0.05); Compared with the control group, the cell proliferation inhibition rate of the curcumin group and the UMI-77 group were increased, the apoptosis rate of cell was increased, the level of ROS was increased, the protein expression of Bax, Caspase-3 and Caspase-9 in the cells were all increased, and the protein expression of Bcl-2 was reduced (P<0.05); Compared with the curcumin group or UMI-77 group, the cell proliferation inhibition rate and apoptosis rate of the curcumin+UMI-77 group were further increased, and the level of ROS was increased. At the same time, the protein expression of Bax, Caspase-3 and Caspase-9 in the cells were all increased, the protein expression of Bcl-2 was reduced (P<0.05); In addition, the mitochondrial membrane potential of the cells after curcumin treatment was decreased, and the proteins expression of Notch1 and HES1 were reduced (P<0.05).
CONCLUSION
Curcumin can enhance the apoptosis of T-ALL cells induced by Mcl-1 small molecule inhibitor UMI-77 by reducing the mitochondrial membrane potential, the mechanism may be related to the inhibition of Notch1 signaling pathway.

Objective:To investigate the value of ADC derived from DWI combined with texture analysis derived from T 2WI fat suppressed images in distinguishing benign and malignant soft tissue tumors. Methods:The MRI and DWI images of 94 patients with soft tissue tumors (44 cases with malignant and 50 cases with benign) confirmed by pathology were analyzed retrospectively in the First Affiliated Hospital of USTC West District. ADC values of solid components were measured at GE ADW4.6 workstation. The texture features were extracted by manually drawing the ROI on the maximum level of the T 2WI fat suppressed images; the ADC values and texture parameters between the two groups were statistically analyzed by SPSS17.0, and the multivariate logistic regression model were conducted to analyze and calculate the diagnostic performance. Results:ADC value of benign and malignant soft tissue tumors was (1.6±0.3)×10 -3 mm 2/s, (1.2±0.5)×10 -3 mm 2/s, respectively, and the difference was statistically significant( t=-5.382, P<0.05). Taking 1.28×10 -3 mm 2/s as the critical value, the area under curve (AUC) for the diagnosis of benign and malignant soft tissue tumors was 0.783, the sensitivity was 92.00%, and the specificity was 65.91%. Among the texture features, the AUC of frequency size, skewness, Inertia All Direction_offset7, Inverse Difference Moment angle0_offset1, Inverse Difference Moment angle0_offset7 and Haralick Correlation All Direction_offset4_SD distinguishing benign and malignant soft tissue tumors were 0.825, 0.739, 0.826, 0.816, 0.820 and 0.783, respectively. The AUC, sensitivity and specificity of the best predictive model distinguishing benign and malignant soft tissue tumors were 0.930, 88.00% and 86.36% respectively using multivariate logistic regression analysis. Conclusion:ADC combined with texture analysis is of great value in preoperative differentiation of benign and malignant soft tissue tumors.
8.Situation and suggestions for the service trade of traditional Chinese medicine in Singapore
International Journal of Traditional Chinese Medicine 2018;40(10):897-899
With the establishment of the singapore college of traditional Chinese medicine (TCM) and the landing of the TCM Practitioners Act in 2000, TCM is booming in Singapore. At present, TCM service trade consumes less outside Singapore, but cross-border consumption, commercial existence and natural person flow have greater advantages. At the same time, the treatment of damp-heat disease, geriatric diseases and tourism of TCM in Singapore made the trade of TCM services show broad prospects for its development. This paper suggests that we should attach importance to the location of service agencies, integrate resources to treat special diseases, increase peer exchanges, take mobile payment as an entry point, and adopt a culture-first approach to develop trade in TCM services.
9.Analysis on the development status and routes of traditional Chinese medicine service trade in the Netherlands
International Journal of Traditional Chinese Medicine 2020;42(8):724-727
There are close economic and trade exchanges between China and the Netherlands. At present, the development of Chinese medicine service trade in Holland is centered on the association of traditional Chinese medicine. The drug access policy is relatively loose, but there is no clear legislation, the relevant laws and regulations are not perfect, the drug access route needs to be widened, the construction of medical institutions still needs to be developed, and there is a gap in the service trade of traditional Chinese medicine. Based on this, we should pay attention to the legislation of traditional Chinese medicine in the Netherlands, promote the establishment of overseas centers and other large-scale institutions, pay attention to Chinese medicine insurance and pharmaceutical investment, and comprehensively promote the sustainable development of Chinese medicine service trade in the Netherlands.

International Journal of Traditional Chinese Medicine 2020;42(9):835-838
In recent years, the cooperation between China and the Republic of Lebanon (hereinafter referred to as "Lebanon" ) in the field of Traditional Chinese Medicine (TCM) service trade has become increasingly urgent. At present, TCM in Lebanon is facing challenges such as the westernized medical environment, the imperfect system of TCM doctors’ recognition, the strict drug access conditions, the imperfect industrial chain of Chinese medical products, and the significant impact of cultural and regional differences on the market demand for Chinese medicine. It is suggested that health care products should be taken as the breakthrough point to develop service trade, enterprises should be attracted to settle in Lebanon from the national level to produce Chinese medicine preparations on the spot, and pay attention to high-level contact, establish TCM hospitals, and deepen cultural exchanges, so as to promote the development of Chinese medicine in the Lebanon.
