Acta Academiae Medicinae Sinicae 2022;44(2):208-212
Objective To study the influence of recombinant bovine basic fibroblast growth factor as an adjuvant therapy on scar alleviation and inflammatory cytokines in patients with atrophic acne scar. Methods The random number table was employed to randomly assign 120 patients with atrophic acne scar into a test group and a control group. Both groups of patients were treated with CO2 lattice laser. After the operation, the control group was routinely smeared with erythromycin ointment and the test group was coated with recombinant bovine basic fibroblast growth factor gel.The clinical efficacy, clinical indicators, scar alleviation, and inflammatory cytokine levels before and after treatment were compared, and adverse reactions were counted. Results The test group had higher total effective rate(P=0.040) and lower total incidence of adverse reactions(P=0.028) than the control group.Compared with the control group, the test group showcased short erythema duration after treatment(P=0.025),early scab forming(P=0.002),and early edema regression(P<0.001).After treatment, the proportion of grade 1 scars graded by Goodman and Baron's acne scar grading system in the test group and control group increased(P=0.001,P=0.027),and the proportion of grade 4 scars decreased(P<0.001,P=0.034).Moreover,the proportion of grade 1 scars in the test group was higher than that in the control group(P=0.031) after treatment, and the proportion of grade 4 scars presented an opposite trend(P=0.031).After treatment, the levels of tumor necrosis factor-α(TNF-α) and interleukin-1β(IL-1β) in both groups declined(all P<0.001),and the test group had lower TNF-α and IL-1β levels than the control group(all P<0.001). Conclusion The recombinant bovine basic fibroblast growth factor gel as an adjuvant therapy of CO2 lattice laser can effectively alleviate the atrophic acne scar, relieve local inflammatory reaction, and has good curative effect and less adverse reactions.

China Pharmacy 2019;30(2):244-249
OBJECTIVE: To investigate the characteristics and regularity of hepatotoxicity induced by quinolones, and to provide reference for clinical use of drug safely. METHODS: Using “quinolone” “floxacin” “hepatotoxicity” “hepatic injury”as retrieval words, relevant literatures about hepatotoxicity induced by quinolones were retrieved from domestic and foreign databases as CNKI, Wanfang, VIP, PubMed (during database establishment to 31th, Dec. 2017). Those literatures were summarized and analyzed. RESULTS: A total of 59 valid literatures were collected, including 61 cases of hepatotoxicity induced by quinolones, 8 types of drugs as ciprofloxacin, moxifloxacin, ofloxacin, lomefloxacin, norfloxacin, levofloxacin, gatifloxacin and enoxacin. Ciprofloxacin, levofloxacin, moxifloxacin and ofloxacin were the most common drugs that caused hepatotoxicity, involving 19, 13, 11, 7 cases, respectively; accumulative constitute ratio was 81.97%. The ratio of male to female was 1.54 ∶ 1, and hepatotoxicity always happened at the age of 61 to 80 (30 cases, 49.18%). Primary diseases of 46 cases were single disease (75.41%), and mainly were infection of respiratory system and urogenital system. There were 15 cases of combined disease (24.59%). Thirty-one cases used quinolones alone, most of which was ciprofloxacin. There were 30 cases of drug combination. Thirty-four cases were given drug intravenously and mainly were domestic cases. The hepatotoxicity first occurred within 10 minutes after administration and at the latest 8 weeks after administration. Forty-nine patients suffered from hepatotoxicity within 10 days after medication, accounting for 80.33%. Besides general fatigue, nausea and vomiting, clinical symptoms also included abnormal elevation of alanine aminotransferase, aspartate aminotransferase and total bilirubin, etc. Fifty-four patients were improved after withdrawal or symptomatic treatment, while 7 patients died. The results of causality evaluation of drug-induced hepatic injury showed that there were 4 probably association cases, 45 likely association cases and 12 possible association cases. CONCLUSIONS: The hepatotoxicity caused by quinolones is related to drug variety, patient’s age, primary disease, drug combination and route of administration, and mostly occurs within 10 days after administration. Great importance should be attached to patient’s liver function indexes, strengthen medication monitoring, and carefully combined use of drugs.

Objective:To investigate the effects of ultra-high dose rate radiation (FLASH-RT) and conventional radiation (CONV-RT) on plasma metabolites in glioma mice. Methods:Tocally 21 male C57BL/6J mice bearing intracranial glioma xenograft were randomly divided into healthy control group ( n=3), CONV-RT group ( n=9) and FLASH-RT group ( n=9). The CONV-RT group was administered a single dose of 24 Gy radiation on the head of mice at a dose rate of 0.4 Gy/s, and the FLASH-RT group was administered a single dose of 24 Gy radiation on the head of mice at a dose rate of 60 Gy/s, and the healthy control group was given 0 Gy pseudoradiation under the same condition. Mice blood was collected through the inner canthus vein for plasma separation at 1, 3 and 7 d after radiation in the two radiation groups, and the blood plasma of healthy control group was collected at 7 days after sham radiation. The changes in plasma metabolites were detected by the non-targeted metabolomics based on liquid chromatography mass spectrometry tandem platform. Results:After irradiation, the metabolites in plasma of two irradiation groups had significant difference. Compared with the healthy control group, 12 and 5 differential metabolites were screened out in the FLASH-RT group and CONV-RT group at three time points, respectively. The difference of plasma metabolites had the largest value at 1 day and decreased at 3 and 7 d after radiation. The arachidonic acid and isovaleric acid, involving arachidonic acid metabolism, biosynthesis of unsaturated fatty acids, and tyrosine metabolism pathways were screened in both FLASH-RT group and CONV-RT group, and the 10 differential metabolites, mainly involving energy metabolism and redox reactions, only existed in the FLASH-RT group. Conclusions:Arachidonic acid and isovaleric acid may be the common sensitive biomarkers to different radiation patterns, which provides ideas for further exploring the molecular mechanism of FLASH-RT in the treatment of glioma.

Journal of Zhejiang University. Medical sciences 2021;50(1):32-40
The metabolic reprogramming of tumor cells is characterized by increased uptake of various nutrients including glutamine. Glutamine metabolism provides the required substances for glycolysis and oxidative phosphorylation and affects the homeostasis of carbohydrate, fat and protein metabolism to induce the chemoresistance of tumor cells. Combination of chemotherapeutic agents with inhibitors specific to different components of glutamine metabolic pathway has obtained favorable clinical results on various tumors. Glutamine metabolic pathway plays a role in drug resistance of tumor cells in various ways. Firstly, the dynamic change of glutamine transporters can directly affect intracellular glutamine content thereby causing drug resistance; secondly, tumor stromal cells including adipocyte, fibroblast and metabolite from tumor microenvironment would give rise to immune-mediated drug resistance; thirdly, the expression and activity of key enzymes in glutamine metabolism also has a critical role in drug resistance of tumors. This article reviews the effects of glutamine metabolic pathway in the development of tumor chemoresistance,in terms of transporters, tumor microenvironment and metabolic enzymes,to provide insight for improving the therapeutic efficacy for drug-resistant tumors.
