1.Effect of SIRT6 on migratory and proliferative activity of skin fibroblasts in the elderly and their mechanisms

Objective To assess the effect of silent mating type information regulation 2 homolog 6 (SIRT6) on migratory and proliferative activity of skin fibroblasts in the elderly,and to explore their mechanisms.Methods Circumcised foreskins were obtained from patients of different ages in Department of Urologic Surgery,Second Hospital of Shanxi Medical University,including 8 elderly patients and 8 young patients.Human skin fibroblasts were isolated from the foreskin tissues by using a collagenase digestion method.Western blot analysis was performed to determine the expression of SIRT6 and phosphorylated p65 (p-p65) in human skin fibroblasts in different age groups,scratch assay to evaluate cell migratory activity,and cell counting kit-8 (CCK8) assay to assess cellular proliferative activity.Skin fibroblasts in the elderly group were divided into 2 groups:SIRT6 group transfected with a lentiviral vector overexpressing SIRT6,and control group transfected with an empty lentiviral vector.Then,the cell migratory and proliferative activity as well as p-p65 expression were measured by the above methods,and the mRNA expression of type Ⅰ and Ⅲ collagens,and integrin subunits α3,α5 and β1 was determined by real-time PCR in the SIRT6 group and control group.Statistical analysis was carried out with GraphPad Prism 5 software by using t test for comparison between 2 groups.Results Compared with the young group,the elderly group showed significantly decreased SIRT6 expression in skin fibroblasts (0.434 ±0.179 vs.1.000 ± 0.067,t =3.040,P =0.012),migration rate (43.81% ± 18.84% vs.94.63% ± 12.32%,t =5.903,P =0.003)and cellular proliferative activity at 24 and 48 hours (both P < 0.05),but significantly increased p-p65 expression (1.694 ± 0.148 vs.1.000 ± 0.093,t =2.949,P =0.015).Compared with the control group,the SIRT6 group showed significantly decreased p-p65 expression (P < 0.05),but significantly increased migratory and proliferative activity (both P < 0.05),and elevated mRNA expression of type Ⅲ collagen and integrin subunits oα3,α5 and β1 (all P < 0.05).Conclusion SIRT6 can improve the migratory and proliferative activity of human fibroblasts in the elderly,possibly by inhibiting the nuclear factor-κB pathway.
2.Treatment with 3-Bromo-4,5-Dihydroxybenzaldehyde Improves Cardiac Function by Inhibiting Macrophage Infiltration in Mice

BACKGROUND AND OBJECTIVES: Appropriate inflammatory response is necessary for cardiac repairing after acute myocardial infarction (MI). Three-Bromo-4,5-dihydroxybenzaldehyde (BDB) is a potent antioxidant and natural bromophenol compound derived from red algae. Although BDB has been shown to have an anti-inflammatory effect, it remains unclear whether BDB affects cardiac remolding after MI. The aim of this study was to investigate the potential role of BDB on cardiac function recovery after MI in mice. METHODS: Mice were intraperitoneally injected with BDB (100 mg/kg) or vehicle control respectively 1 hour before MI and then treated every other day. Cardiac function was monitored by transthoracic echocardiography at day 7 after MI. The survival of mice was observed for 2 weeks and hematoxylin and eosin (H&E) staining was used to determine the infarct size. Macrophages infiltration was examined by immunofluorescence staining. Enzyme-linked immunosorbent assay (ELISA) was used to test the production of cytokines associated with macrophages. The phosphorylation status of nuclear factor (NF)-κB was determined by western blot. RESULTS: BDB administration dramatically improved cardiac function recovery, and decreased mortality and infarcted size after MI. Treatment with BDB reduced CD68+ macrophages, M1 and M2 macrophages infiltration post-MI, and suppressed the secretion of pro-inflammatory cytokines, such as tumor necrosis factor (TNF)-α, interleukin (IL)-1β, monocyte chemoattractant protein (MCP)-1, and IL-6 in the injured hearts. Furthermore, BDB inhibited the phosphorylation of NF-κB in the infarcted hearts. CONCLUSIONS: These data demonstrate, for the first time, that BDB treatment facilitated cardiac healing by suppressing pro-inflammatory cytokine secretion, and indicate that BDB may serve as a therapeutic agent for acute MI.

4.Analysis of influencing factors and impact path of self-management in tumor patients with PICC of the intermission of chemotherapy

Objective:To analysis the effect of health-promoting lifestyle and patient activation on self-management among tumor patients with PICC of the intermission of chemotherapy, so as to provide a basis for the targeted caring intervention programs.Methods:Using cross-sectional survey methods, from January 2021 to December 2022, the tumor patients with PICC of the intermission of chemotherapy of Tianjin Third Central Hosptial were investigated, using the General Demographic Questionnaire, Cancer Patients PICC Self-management Scale, Health Promotion Lifestyle Profile Ⅱ and Patient Activation Measure. Pearson correlation analysis and multiple linear stepwise regression analysis were used to analyze the data, and the structural equation model between related factors were established.Results:A total of 251 patients were collected in the this study, the scores of self-management were (153.12 ± 20.33) points, health-promoting lifestyle were (98.58 ± 16.05) points, and patient activation were (50.52 ± 6.51) points. The dimensions scores and total scores of self-management were positively correlated with the health-promoting lifestyle and patient activation in tumor patients with PICC ( r values were 0.503-0.910, all P<0.01). Multiple linear regression analysis showed that the health-promoting lifestyle and the patient activation were the influencing factors of the patient's self-management ability ( t=2.67, 1365, both P<0.05). Pathway analysis results showed that patient activation played a partially mediating role between health-promoting lifestyle and self-management, the effect value was 0.33 (95% CI 0.301-0.557), accounting for 46.5% of the total effect. Conclusions:Health-promoting lifestyle and the patient activation were the influencing factors of the self-management ability of tumor patients with PICC, patient activation played a partially mediating role between health-promoting lifestyle and self-management.
5.Study on the Effects of Different Primary Processing Methods on the Quality of Polygonatum sibiricum

OBJECTIVE: To explore the effects of different primary processing methods on quality change of Polygonatum sibiricum, and to optimize primary processing method of P. sibiricum with different grades and shapes. METHODS: After processed with different drying methods, boiling method and steaming method, sulfuric acid-phenol method was used to determine the contents of indicator component P. sibiricum polysaccharide in samples. The contents of ethanol-soluble extract, water-soluble extract and total ash were investigated. The optimal primary processing methods were screened. RESULTS: After processed with different methods, the contents of polysaccharide (≥7%), ethanol-soluble extract (≥45%) and total ash (≤4%) of samples at all levels conformed to the requirements of the 2015 edition of Chinese Pharmacopoeia (part Ⅰ); from aspect of drying method, microwave drying method was better than oven drying method;for P. sibiricum with different morphologies (individual and lamellar), the quality of lamellar samples of P. sibiricum was better than that of individual P. sibiricum. As primary processing method, steaming was better than boiling. The contents of P. sibiricum polysaccharide were in relatively high level, when the steaming and boiling time of gradeless and uniformly-priced goods of P. sibiricum were 20 and 15 minutes respectively. CONCLUSIONS: By discussing the different primary processing methods, high-efficient primary processing method for the individual and lamellar samples of P. sibiricum as well as gradeless and uniformly-priced goods of P. sibiricum are preliminarily obtained, which can provide a scientific basis for the effective utilization of P. sibiricum resources.
