Objective To explore the inhibition effects of CD4+CD25+/highCD127low on the EBV immortalized cells(CD23+)in children with infectious mononucleosis (IM). Methods The expression of CD3+, CD3+ CD4+ , CD3+ CD8+ , CD25+/highCD127low and CD19+ , CD19+ CD23+ were analyzed by flow cytometry in 23 children at the acute stage of IM and 10 children recovering from IM in comparison with the ones of 20 healthy controls. Results Compared with the following results in controls, CD3+, CD3+ CD8+ were significantly increased and CD3+ CD4+, CD4+/CD8+ ratio, CD4+ CD25+/highCD127low, CD19+, CD19+ CD23+ were significantly reduced in the IM patients. Compared with the ones in the IM pa-tients recovering from IM, CD3+ , CD3+ CD8+ were significantly increased and CD3+ CD4+, CD4+/CD8+ ratio, CD4+ CD25+/highCD127low, CD19+ , CD19+ CD23+ were significantly reduced in the IM patients at the acute stage. There was no obvious difference in all the marks except CD4+/CD8+ratio, CD8+ CD28+ , CD19+ , CD19+ CD23+ ratio between the children recovering from IM and the normal controls. There is a positive correlation between CD3+ and CD19+, CD19+ CD23+ during acute phase. There is a negative correlation between CD4+ CD25+/highCD127low and CD19+ , CD19+ CD23+ during acute phase. Conclusion The results indicate that the decreasing of CD4+ CD25+/highCD127low may play an important role in elimina-ting EBV immortalized cells.

Objective To develop and validate a ultrasonographic (US)imaging agent with targeted microbubbles that attaches to chemokine receptor 2 (CCR2)and to compare the US single obtained from targeted microbubble with that from control microbubble in murine breast tumor model. Methods The microbubble which carried CCR2 antibody (MBCCR2 )and isotype-macthed immunoglobulin G-labled control microbubble (MBcontrol ) were prepared. The microbubble size and distribution were assessed by AccuSizer780.Binding specificities of targeted microbubble compared with control microbubble were tested with murine microvascular endothelial cells (bEnd.3 ).Orthotopic breast tumor model was estabished in BALB/c mice with mouse breast cancer 4T1 cell.In vivo imaging signals of contrast material-enhanced ultrasound by use these two different types of microbubble which were injected respectively into each mouse at random order and 30 min interval.Tumor tissue was stained for CCR2 and CD3 1 .Results Automatic Particle Sizer showed size uniform of two kinds of microbubbles, and narrow distribution of particle size (mean diameter of about 1 -2 μm),which were not significantly different (P >0.05).Adhension to bEnd. 3 endothelial cells was significantly higher (P < 0.001 )for MBCCR2 (mean,9.50 ± 1 .5 1 )than that for MBcontrol (mean,0.01 ±0.01).Imaging signal in the murine tumor model was significantly higher for MBCCR2 [mean,(6.76±0.26)dB]than that for MBcontrol [mean,(1 .06 ±0.62)dB,P <0.001 ].Immunofluorescence confirmed expression of CCR2 on tumor vasculature. Conclusions The targeted microbubbles with CCR2 monoclonal antibody had been successfully prepared, which precisely targeted to CCR2 of tumor angiogenesis in the murine breast cancer xenograft tumor models in vivo. These results suggest that the targeted microbubbles as a kind of ultrasound molecular imaging agent with a better specificity can be used for both evaluating tumor neovascularization and monitoring therapeutic effect of anti-angiogenesis.

Objective To explore the mechanisms of synergic action of commonly-used herb pair of Radix Sophorae Flavescentis (Kushen, KS) and Fructus Cnidii (Shechuangzi, SCZ) for the treatment of pruritus. Methods We predicted and analyzed the potential targets of KS and SCZ based on network pharmacology method, and then established Chinese herbs-compound skeletons-targets networks to reveal anti-pruritus targets, predicting targets, and the interaction of KS and SCZ, as well as the components of the herb pair. The RAW264.7 inflammatory cell model was established to observe the synergistic anti-inflammatory activity of KS and SCZ. Results KS and SCZ had possible synergic action on the pruritus-related targets such as histamine receptors, cannabinoid receptors and proteinase-activated receptor 2. Additionally, KS and SCZ probably had synergic regulation of inflammation-related pathway (Toll -like receptor signaling pathway , chemokine signaling and Fc epsilon RI signaling pathway) and nerve-related pathway(neurotrophin signaling pathway) during the treatment of pruritus. Flavones from KS and coumarin of SCZ had various synergistic anti-inflammatory activities(P<0.05), indicating that they played an important role in inhibiting pruritus induced by inflammation. Conclusion The method may reveal the molecular mechanism of KS and SCZ in inhibiting pruritus, which is important for the modernization of Chinese medicine and new drug development.
