AIM: To achieve an optimum manufacturing process for acetaminophen extended release tablets. METHODS: To evaluate and set the manufacturing process according to the compressibility of the granule and the dissolubility of the finished tablets. RESULTS & CONCLUSION: The results showed that the bi-layer tablet technology was the most preferable pro- cess. The in vitro dissolution behavior and the in vivo bioavailability of the tablets are comparable to those of the imported products from McNEIL Consumer Healtheare Co. USA.

On the basis of summarizing association rules algorithm and algorithm optimization,the paper explores the rules of prescription compatibility in the scheme for apoplexy diagnosis and treatment through Traditional Chinese Medicine (TCM),establishes the TCM Electronic Medical Records (EMR) database,processes and analyzes the data based on the classical association rule-Apriori algorithm,introduces the parameter optimization mining results of correlated measurement parameters Kulc and IR,eliminates pseudo strong association principle,gets the drug compatibility with strong correlation and theoretical significance,and provides theoretical guidance and reference for apoplexy medication through TCM.
3.The 1 H-Magnetic Resonance Spectroscopy Study of Auditory Cortex Metabol ism in Patients with Metabol ic Syndrome

Objective To investigate the characteristic changes of the metabolism products in the auditory cortex (transverse temporal gyrus)in patients with metabolic syndrome combined with sensorineural deafness using 1 H magnetic resonance spectroscopy (1 H-MRS),and to find out the early warning indicators of sensorineural deafness in patients with metabolic syndrome.Methods The pure tone audiometry (PTA)was performed in 142 pa-tients with metabolic syndrome (diagnosed by endocrinology department),and 15 healthy subjects as the control group.The patients were divided into four groups:the metabolic syndrome group;the metabolic syndrome group with unilateral,the bilateral deafness group,and the control group.Cerebral metabolism was studied by assessing the ratios of nitro-acetyl aspartate contrast to choline (NAA/Cho)as well as to creatine (NAA/Cr),myo-inosi-tol to creatine (mI/Cr)and choline to creatine (Cho/Cr)ratios in the auditory cortical separately in these groups. ROC curves were made for those metabolism markers to find the best diagnostic threshold.Results Significantly lower values of NAA/Cho ratio and higher values of Cho/Cr were observed in metabolic syndrome in the control group (P<0.05),Cho/Cr higher than those (P<0.05).NAA/Cho ratios in the injured and uninjured auditory cortex of MS with unilateral deafness were significantly lower than those in the control group (P<0.05).A self-comparison was made between the inj ured and uninj ured auditory cortex,the result showed that NAA/Cho ratio had significant differences.All of the metabolisms were tested by the curve of ROC.The area of NAA/Cho under the ROC curve was 81%,which had a higher accuracy.NAA/Cho equating to 1 .82 could be used as the boundary indi-cators between metabolic syndrome without deafness and with deafness groups.The areas of the other indicators un-der the ROC curve were <50%.Conclusion NAA/Cho may be the early warning marker of sensorineural deafness in patients with metabolic syndrome.
4.Study of pathogen aetiology and drugs resistance in neonatal septicemia

Objective To study pathogen aetiology and distribution of drug resistance in neonatal septice-mia. Methods A retrospective study of positive organisms of blood cultures and the results of antimicrobial suscep-tibility testing was conducted from January 2000 to December 2006 in 1293 cases of neonatal septicemia;The results were divided into three groups:the earlier group,the middle group and the later group. The strains were identified by VITEK-AMS and antibiotics susceptibility was tested by K-B method, results were interpreted according to NCCLS. inant in three groups(76.3% ,74.1% and 65.7% ) (P <0.05). Among Gram-pesitive coccus,the most prevalent strains was staphylococcus, Gram-negative bacilli increased gradually,accounting for 21.3% ,21.3% and 28.4% in cpidermids to erythromycin and sulfamethoxazole/trimethoprim were rather higher:93.3% ,73.7% vs 87.8% vs 100.0%, respectively, the resistance -rates to penicillin were less,but to susceptible to vancomyein,piperacill/tazo-batam,amikacinand ciprofloxacin;gram-negative bacilli were only sensitible to imipenem, cefepime, and cefopera zone/sulbactam;multiresistances of drug strains was commonly seen. Condusions It is important for using antibi-otics rationally according to drug sensitivity test, because multiresistant strains and opportunistic pathogen are com-monly seen.
5.Study on Doppler blood flow spectrums of umbilical artery and ductus venosus in fetus with arrhythmia by ultrasound

Objective To explore the clinical value of Doppler flow spectrums of umbilical artery and ductus venosus in fetus with arrhythmia by Doppler ultrasound in evaluating the fetal prognosis.Methods Umbilical artery and ductus venosus flow spectrums were obtained in a total of 796 pregnancies with gestation age between 16-41 weeks who were found abnormal heart rhythm by maternity auscultation,fetal electronic monitoring or ultrasound.All these fetuses were followed up by electrocardiography and echocardiography examination until the end of pregnancy and the pregnancy outcomes were recorded.Results Among 796 fetuses,136 cases of fetal arrhythmia were detected.There were 18 cases appeared with umbilical artery absent or reversed end-diastolic flow,and 9 cases with absent or reversed wave A.Among 18 fetuses with arrhythmia whose blood flow spectrums of umbilical artery were abnormal,twelve were compared with congenital heart disease.The proportion of fetal bradycardia associated with abnormal flow spectrums and congenital heart disease were higher than other types of arrhythmias.Conclusions Pregnancy outcome of the fetus with arrhythmias who appeared with normal umbilical artery and ductus venosus will be good,and conversely is poor.So Doppler flow spectrums of the fetal umbilical artery and ductus venosus is one of the important indicators in early intervention.
6.Synchronous Detection of Rat Neural Spike Firing and Neurochemical Signals Based on Dual-mode Recording Instrument

A dual-mode recording system used for synchronous detection of neuroeletrical and neurochemical signals was developed, and a dual-mode synchronous detection experiment was carried out using the instrument. The device comprised 64-channel neuroelectricity recording module with voltage resolution of 0. 3 μV and 4-channel neurochemistry recording module with current resolution of 1 pA. The software had many basic features, including Spike separation and sort, chronoamperometry, cyclic voltammetry, etc. In particular, the software could observe and analyze the dual-mode neural signals synchronously. The performance of the system was demonstrated in the single mode detection experiments. In neuroeletrical experiments, 64-channel simulate neural signals were detected and the signal to noise ratio ( S/N) of Spike recorded from cortex of Sprague-Dawley ( SD ) rat was 6. In the K3 [ Fe ( CN )6 ] and ascorbic acid measurement experiments, the current response of K3 [ Fe ( CN)6 ] in the range of 0. 1-10 mmol/L was obtained by cyclic voltammetry, with a correlation coefficient of 0. 9889, and the current response of ascorbic acid ( concentration:10-800 μmol/L) by chronoamperometry increased linearly with a correlation coefficient of 0. 9841. Based on the rat model of global cerebral ischemia, a dual-mode detection experiment was carried out. In the experiment, the neuroelectrical and neurochemical signals were synchronously recorded in the SD rat primary visual cortex. According to the experimental results, we got the conclusion that the concentration of ascorbic acid negatively related to the Spike firing in the SD rat primary visual cortex.
7.A Wearable Wireless Electrochemical Instrument Used for In_vivo Neurotransmitter Detection

A wireless electrochemical recording device was designed for in_vivo neurotransmitters real_time detection. Low_power microcontroller MSP430 was chosen as main control unit in hardware system. Other modules were current detection module, waveform generator module and data transceiver module. This device had the merits of small size (2. 3 cm×1. 8 cm×0. 6 cm) and low power consumption. Firmware program design was based on uC/OS operating system. Combined with the PC software, the device could achieve online display and analysis of the recording data. For neurotransmitter detecting needs, the device implemented fast_scan cyclic voltammetry ( FSCV) and fixed_potential amperometry. By using fast_scan cyclic voltammetry method, a linear relationship ( R=0. 99 ) between the concentration of dopamine and response current was acquired in the range of 5. 0×10-7-7. 0×10-5 mol/L. In the in_vivo experiments, the electrically evoked dopamine was recorded in the caudate_putamen area of brain in rats. Experimental results showed that the system had high detection accuracy, which could realize qualitative and quantitative analysis of the brain neurotransmitter. This work would have a broad application prospect in the field of neuroscience research.
8.Analysis of echocardiographic missed diagnosis and misdiagosis of infants with coarctation of the aorta

Objective To explore the clinical value of two dimensional and color Doppler echocardiography in diagnosis of infants with coarctation of the aorta(CoA),and to analyse the cause of missed diagnosis and misdiagnosis.Methods The echocardiographic characteristics of 87 infants with CoA were analyzed retrospectively,and the results were compared with surgical findings.Results Seventy-five patients were correctly diagnosed,while 9 patients were missed and 3 patients were misdiagnosed.Discrete coarctation was found in 67 cases,whereas tubular coarctation was detected in 8 cases of CoA.Conclusions Mastering the echocardiographic characteristics of infants with CoA and carefully scanning the sternum nest section,combined with two dimensional and color Doppler echocardiography,could avoid missed diagnosis and misdiagnosis.
9.Study of Microelectrode Array Probe for Simultaneous Detection of Glutamate and Local Field Potential during Brain Death

High extracellular potassium can induce spreading depression-like depolarizations, elevations of extracellular glutamate and even neuronal death in normal brain. To investigate the contribution of high potassium in vivo, a microelectrode arrays ( MEAs ) probe integrated with recording sites for glutamate concentration (50í150 μm) and local field potential ( LFP) ( diameter=15 μm) was fabricated by Micro-electro-mechanical-systems ( MEMS) technologies. We implanted the MEA probe acutely in the rat brain and exposed the brain to a high potassium solution. During these multi-modal recordings, it was observed that high potassium elevated extracellular glutamate while suppressing the LFP irreversibly. This is one of the first studies in which a dual mode MEA probes is applied in vivo for neuronal death, and it is concluded that our MEA probes are capable of examining specific spatiotemporal relationships between electrical and chemical signaling in the brain.
10.Detection of Human Chorionic Gonadotropin by Highly Sensitive Magnetic EnzymE-linked Chemiluminescent Immunoassay

A highly sensitive magnetic enzymE-linked chemiluminescent immunoassay method was developed for the detection of human chorionic gonadotropin(HCG). The monoclonal antibody was covalently coupled on the surface of carboxylated magnetic beads to generate magnetic-biotargeting; Alkaline phosphatase(ALP) was utilized as a labeled reagent of another monoclonal antibody, whereas 3-(2-spimadamantane) 4-methoxy-4-(3-phosphoryloxy)phenyl-1,2-dioxetane(AMPPD) was utilized as the chemiluminescent substrate. Based on this concept, a highly sensitive chemiluminescent immunoassay method was established to test HCG. Then, several modifications were made to optimize the method, and the detection sensitivity and procedure were improved accordingly. The detection of the assay could be fulfilled within 60 min and the test result of HCG concentration was linear over the range of 0.15 150 IU/L with good relativity(r=0.960). The relative standard deviation(RSD) were below 5% and the sensitivity of this method was 0.15 IU/L. The proposed method with wide linear range, simple operation and fast detection showed good prospect in practical application on-site.
