1.Effect of Electroacupuncture on Learning and Memory and Heme Oxygenase in Rats with Vascular Dementia

[Objective] To observe the effect of electroacupuncture (EA) on the disorder of learning and memory in rats with vascular dementia (VD) and to explore its molecular mechanism. [Methods] Among 60 male SD rats, 10 rats were in sham-operation group and the other 50 rats were used to establish models of global ischemia and reperfusion by four-vessel occlusion (4-VO) method. Then the survival model rats were randomized into EA group (n = 14) , model group (n = 13) and nimotop group (n = 13). EA group was treated with EA at points of Baihui (GV20) and Dazhui (GV14) , nimotop group with nimotop 12 mg/kg, and sham-operation group and model group did not receive any treatment. After a 20-day treatment, the changes of learning and memory behavior in rats were detected by Morris water maze, as well as the protein expression and mRNA expression of heme oxygenase (HO) . [Results] The escape latent period, HO-1 protein expression and HO-1 mRNA-expression were increased obviously in model rats ( P 0.05). [Conclusion] EA can improve the learning and memory behavior in rats with VD and this may be related to the decrease of HO-1 protein expression and HO-1 mRNA-expression in cortex and hippocampal neurons.
2.Influence of applied times of Batroxobin on neuroprotective effects in cerebral ischemic-reperfusion injury gerbils

Objective To study the influence of applied times of Batroxobin on neuroprotective effects in cerebral ischemic-reperfusion(IR) injury gerbils.Methods 45 gerbils were randomly divided into five groups:normal group;cerebral IR injury model group(IR group);three times group;five times group;seven times group.The later three groups were administered Batroxobin 8 U/kg by abdominal injection q.o.d with certain times respectively.The normal group and IR group were not given drug but isometric physiological saline.Flow cytometer was used to measure apoptotic neurons of the hippocampal CA1,meanwhile electronic microscope was used to detect the ultrastrctural change of the hippocampal CA1 region.Result The quantity of apoptotic neurons in the every Batroxobin groups were significantly less than those in the IR groups(all P0.05).The ultrastrctural changes in five times group were lighter than those in three times group,but no significant difference was found between five times group and seven times group.Conclusion Three times,five times and seven times of Batroxobin can reduce the number of apoptotic neurons after IR injury.However,applied five times and seven times of Batroxobin has stronger neuroprotective effect than three times.
3.Effect of parecoxib united with local anethsia analgesia system on analgesia after radical resection of liver cancer

Objective To observe the effect of parecoxib united with local anaesthesia analgesia system on analgesia after radical resection of liver cancer. Methods Ninety patients undergoing elective radical resection of liver cancer were randomly assigned to three groups, with 30 patients in each group. The patients in parecoxib group (P group) received parecoxib 40 mg through vein before the end of operation for 30 min, and were injected parecoxib 40 mg at 12 h, 24 h and 46 h after operation. The patients in local anethsia analgesia system group (L group) received embedding hypodermic pervasion canal in incision subcutaneously, then connected the pump with 0.25% ropivacaine 250 ml, with infusion speed of 4 ml/h.The patients in united group(U group)receiced the same methods with those in P group and L group. When the patients'visual analogue scale (VAS) scores were higher than 5 scores, they were given muscle injection of pethidine 50 mg. The VAS scores and Ramsay sedation scores at the time of tracheal extubation (T0), postoperative 2 h(T1), 6 h(T2), 12 h(T3), 24 h(T4) and 48 h(T5), the adverse reaction rate, dosage of pethidine, and analgesia satisfaction were compared. Results The resting and movement of VAS scores and Ramsay sedation scores at each time in P group and L group had no significant difference (P>0.05). The resting and movement of VAS scores and Ramsay sedation scores at each time in U group were significantly better than those in P group and L group (P<0.05). The rate of nausea and vomiting in three groups had no significant difference among three groups (P > 0.05). The rate of drowsiness in U group was significantly lower than that in P group and L group:6.67%(2/30) vs. 26.67%(8/30) and 23.33%(7/30), P < 0.05. The analgesia satisfaction in U group was significantly higher than that in P group and L group:93.33%(28/30) vs. 56.67%(17/30) and 53.33%(16/30), P<0.05. Conclusions The analgesia effect of the parecoxib united with local anaesthesia analgesia system for radical resection of liver cancer is perfect.
4.The clinical study of stent implantation and platelet activation in acute coronary syndrome

Objective To explore the change of platelet activation before and after stent implantation in acute coronary syndrome. Methods Detection of positive expression of platelet membrane glycoprotein CD62p,CD63 and thrombin-sensitive protein (TSP) were performed using flow cytometry (FCM) in 82 patients which included 45 unstable angina pectoris (UAP) and 37 acute myocardial infarction (AMI). Another 45 angiographically normal patients served as the control. Results Expression of CD62p, CD63 and TSP after stent implantation in ACS group. On the other hand, the expression of the above factors was higher in ACS group compared with the control group. Conclusion Acute thrombogenesis induced by platelet activation is a basic pathophysiologic process in ACS. Endothelial injury and plaque rupture produced by stent implantation may be a cause of platelet activation.
5.Effects of Nimodipine on the c-fos and c-jun mRNA expressions in dorsal root ganglia of rate following acute injury

6.Oxygen radical absorbance capacity assay and its application

Aim To summarize the principles, applications and future development in oxygen radical absorbance capacity (ORAC) assay. Methods The ORAC assay method use Sodium Fluorescein as fluorescent probe, AAPH as free radical initiator, and results are expressed as Trolox equivalents. Results This method has several advantages compared with other methods in linear responses with concentration, specificity, precision, accuracy and ruggedness. Conclusion The ORAC assay has been widely accepted and largely applied to the assessment of free radical scavenging capacity of pure compounds, biological samples, plant and food extracts.
7.The prognostic value of dynamic monitoring blood plasma colloid osmotic pressure of the danger patients

Objective To investigate clinical value of the level of arterial plasma colloid osmotic pressure (COP) in predicting the prognosis of severe patients.Methods According to the prognosis of patients,the patients were divided into survival group and death group,12 h arterial plasma COP change level in the two groups were compared.According to APACHE Ⅱ score,the patients were divided into≤25 and >25 group,the COP into the ICU,12 h plasma COP change were compared.The correlation of APACHE Ⅱ score and arterial lactate into ICU,12 h arterial plasma COP were analyzed by Spearman correlation analysis.Results The level of plasma COP into the ICU and 12 h levels of survival were significantly lower than those of the death group; in the APACHE Ⅱ score≤25 group,plasma COP into the ICU levels and 12 h plasma COP levels were significantly higher than those of the APACHE Ⅱ score > 25 group,and 12 h COP levels were significantly higher than those of behind group (t =8.34,P < 0.01),Further analysis showed that the COP level into the ICU and APACHE Ⅱ score was significantly negative correlated; 12 h COP levels and APACHE Ⅱ score was significantly negative correlated.Conclusions Dynamic monitoring of COP levels is a good indicator to determine the prognosis of patients with the danger patients,the lower plasma COP,the more severe disease and poor prognosis.
9.Effects of Salidroside on expression of substance P and neurokinin-1 receptor of bone marrow cells in bone marrow depressed anemia mice

Objective:To observe the effects of salidroside on the expression of substance P (SP)and neurokinin-1 receptor (NK-1R)of bone marrow cells(BMCs)in bone marrow(BM)depressed anemia mice,and to explore its roles for hematopoietic regulation.Methods:Automatic blood cell analysator was used to detected the changes of peripheral blood cells in each group ,immuno-histochemistry and reverse transcription-polymerase chain reaction ( RT-PCR) were used to analyze the expressions of SP and its receptor NK-1R of BMCs in each group respectively.Results: Peripheral blood testing results showed that the number of white blood cells (WBC),red blood cells(RBC)and hemoglobin(HB)of model group were significantly decreased when compared with control group.Compared with model group ,low-dose,middle-dose and high-dose salidroside obviously elevated the number of white blood cells ,and middle-dose salidroside obviously elevated the number of platelets.Immunohistochemistry studies showed that the expression of SP and its receptor NK-1R of BMCs was found in each group.Compared with control group , the expression of SP of BMCs was decreased obviously in model group(P<0.05),and the expression of NK-1R of BMCs was decreased slightly in model group (P>0.05),and the expression of SP and its receptor NK-1R of BMCs was increased significantly in low-dose,middle-dose and high-dose salidroside groups (P<0.05).RT-PCR data showed that the expression of SP mRNA of BMCs was increased obviously in model group ,low-dose,middle-dose and high-dose salidroside groups when compared with control group (P<0.05).The expression of NK-1R mRNA of BMCs was un-detected in control group and model group.The expression level of NK-1R mRNA of BMCs was elevated gradually with the increase of salidroside dosage in low-dose,middle-dose and high-dose salidroside groups.Conclusion: The mRNA and protein expressions of SP and its receptor NK-1R of BMCs were up-regulated significantly by salidroside in dose-dependent manner.These data suggest that salidroside could participate in the recovery of hematopoietic function of BM depressed anemia mice by increasing the expression of SP and its receptor NK-1R of BMCs.
10.Study of the protective effect of salvia miltiorrhiza ennoblement for enteral nutrition on intestinal mucosa in MODS model rats

Objective: To explore the protective effect of salvia miltiorrhiza for enteral nutrition in S-MODS model rats.Methods: 70 Wistar rats were randomly divided into four groups: control group (n=10), model group (n=20), nutrition fibre group (n=20) and salvia miltiorrhiza for enteral nutrition group (n=20) . The experiment period was 12 days. At the end of experiment, blood routine, biochemistry index (ALB, AST, ALT, BUN, Cr, CK and CK-MB), the concentrations of IL-6,TNF-α and IL-1β were measured. Results: The model was successful by clinical manifestation,blood routine and pathology. The clinical symptom was improved, blood routine and pathology damage were relieved in salvia miltiorrhiza group. The concentrations of IL-6, TNF-α and IL -1 in salvia miltiorrhiza group were lower than model group and nutrition fibre group(P<0.05).Conclusions: Salvia miltiorrhiza for enteral nutrition could prevent gut function damage,protect intestinal mucosa and decrease the concentrations of pro-inflammatory cytokines.
