1.Development of scoliosis surgery

Scoliosis surgery includes fusion and non-fusion techniques.Fusion technique includes posterior fusion technique and video-assisted anterior thoracoscopic fusion.Non-fusion technique is suitable for children patients with growing potential of the spine.It includes growing rod,vertical expandable prosthetic titanium rib(VEPTR),U-shape screw,and multi-vertebral wedged osteotomy(for severely rigid scoliosis).Short term outcome from these techniques has been encouraging,but medium and long term outcomes are still need investigation.
2.The Expression and Significance of LGR5 in Endometrial Cancer

Objective:To investigate the expression of Leucine-rich repeat-containing G protein-coupled receptor 5(LGR5) in endometrial cancer(EC) and their relationship with clinicopathological characteristics.Methods:Immunohistochemistry were performed to measure the LGR5 expression in EC(n =90) and normal endometrium tissue(n =30).The expression of LGR5 and its relationship with clinicopathological characteristics were analyzed.Results:The expression of LGR5 was significantly higher in EC than that in normal tissue (63.3% vs 23.3%,P<0.001).The expression of LGR5 in < 1/2 myometrium infiltration group was higher than in ≥1/2 myometrium infiltration group(72.5% vs 33.3%,P =0.001).There was no significant difference between the expression of LGR5 in different group of age,histological type,histological differentiation,cervical stroma invasion,lymph node metastasis,FIGO stage(P > 0.05).Multivariate analysis showed that LGR5 was an independent influential factor of myometrium infiltration (OR =0.163,95% CI 0.034 ~ 0.772,P =0.022).Conclusions:LGR5 is up-regulated in EC,and is correlated to myometrium infiltration.LGR5 may play an important role in EC tumorigenesis.

4.Cryopreservation of bone marrow mesenchymal stem cells from Sprague-Dawley rats at-80℃

BACKGROUND:We attempt to explore a low-cost, simple and effective way to cryopreserve bone marrow mesenchymal stem cels at-80℃.
OBJECTIVE:To screen the optimal cryopreservation fluid for bone marrow mesenchymal stem cels and to verify the biological features of bone marrow mesenchymal stem cels after long-term cryopreservation.
METHODS: Bone marrow mesenchymal stem cels were cultured using adherent method and the biological features and purity of cels were detected using immunofluorescence method. Bone marrow mesenchymal stem cels were cryopreserved in the cryoprotectant medium containing low-sugar DMEM, fetal bovine serum and dimethyl sulfoxide at different proportions at-80℃ for a short term. Then, the optimal cryoprotectant was selected to storage the bone marrow mesenchymal stem cels. After 1, 3, 6 months of cryopreservation, the cels were resuscitated, cultured and passaged. Passage cels were identified immunofluorescence method to determine the biological features of bone marrow mesenchymal stem cels cryopreserved at-80℃.
RESULTS AND CONCLUSION:Cryoprotectant medium of 80% DMEM+10% fetal bovine serum+10% dimethyl sulfoxide was suitable for cryopreserving MSCs at -80℃, and resuscitated cels were able to proliferate in vitro, and passage normaly, indicating the cryopreserved bone marrow mesenchymal stem cels stil maintain the original biological activity.

The presence of D-amino acid in proteins is thought to be a result of racemization of L-amino acid,which may influence the space structure and the functions of proteins and the connection to the occurence of some diseases has been demonstrated.Recently,tremendous advancement in instrumental techniques has brought the rapid development to analytical methods and detecting techniques in the research on racemization of protein amino acid.The advances in the relationship between racemizadon of protein amino acid and diseases,the influence factors on racemization of protein amino acid and the analytical techniques are reviewed in this article,and the research directions of racemization of protein amino acid are also given.
6.Research progress in regulation roles of G protein-coupled receptors in hepatocellular carcinoma

G protein- coupled receptors (GPCRs), also known as seven- transmembrane domain receptors, constitute the largest superfamily of cell surface receptors. By coupling to heterotrimeric G proteins, arrestins and other signaling molecules, GPCRs modulate diverse signal transduction pathways under physiological and pathological conditions. Recent studies have revealed crucial roles of GPCRs in tumorigenesis and development of cancer metastasis. This review summarizes roles of GPCRs, particularly the roles of those coupled to chemokines, prostaglandin, lysophosphatidic acid, endothelin, catecholamine and angiotensin in proliferation, invasion, metastasis and angiogenesis of hepatoma cells and development of hepatocellular carcinoma. The potential of GPCRs- based therapeutics being used for hepatocellular carcinoma is also highlighted.
7.Type Ⅲ TGF-βreceptor mediated signaling pathway and its role in fibrotic diseases

Transforming growth factor β(TGF-β)superfamily ligands play an important role in regulating cellular homeostasis including proliferation,differentiation,apoptosis,immune sur-veillance and angiogenesis.Type Ⅲ TGF-βreceptor (TβRⅢ) is considered to be the coreceptor of TGF-βsuperfamily.TβRⅢnot only has an effect on classical Smad signaling pathway,but also on non-Smad signaling pathway.TβRⅢplays a crucial role in fibrosis,tumor,cardiovascular diseases via mediating kinds of signaling pathways.This paper reviews TβRⅢ mediated sig-naling pathway and its role in fibrotic diseases.
8.Mechanism of CCCP-induced non-canonical autophagy

Objective To identify the factors those regulate CCCP-induced non-canonical autophagy.Methods Different cells expressing GFP-LC3 were treated with or without CCCP (30μM) for 6 h.Fluorescent images were taken and cell lysates were analyzed by western blot assay.Real-time PCR was used to measure the mRNA levels of LC3B.FIP200-KO MEF cells were cultured and treated by 30 μM CCCP with or without water channel inhibitors, for 6 h.Cell lysates were analyzed by Western blot assay.Results CCCP could not induced autophagy in Atg5-KO MEF cells.CCCP could induce non-canonical autophagy in ULK1-KO MEF, FIP200-KO MEF, and Beclin1-KD U251.CCCP treatment in FIP200-KO MEF cells had no effect on the expression level of LC3B mRNA.We also found two distinct aquaporin water channel inhibitors could inhibit the generation of LC3 which was induced by CCCP.Conclusion CCCP induced non-canonical autophagy was Atg5-dependent, but Beclin1-, ULK1-and FIP200-independent.Osmotic imbalance could regulate CCCP-induce non-canonical autophagy.

10.Modern Literature-based Analysis of Factors Influencing the Effect of Acupoint Combination

Acupoint compatibility is a basic element for acupuncture prescription and an important link for clinical acupuncture workers to obtain a therapeutic effect. Studying factors influencing the effect of acupoint combination and ascertaining its concept and connotation are of important significance for studying the mechanism of acupuncture action. By sorting out, summing up and analyzing relevant modern literature, it is found that factors influencing the effect of acupoint combination are those affecting the therapeutic effects of combination of two or more than two acupoints. Its contents include five aspects: combination mode, combination effect, stimulation method, time factor and body status. Analysis and induction show that acupoint selection is the primary task in studying factors influencing the effect of acupoint combination.
