1.Some Visible Properties of T Lymphocyte Activation in Human Umbilical Cord Blood

Objective To explore visible properties of T lymphocyte activation in human umbilical cord blood.Methods T lymphocytes of human umbilical cord blood were cultured in RPMI 1640 complete medium and divided into two groups: the experimental group activated with CD3 McAb(10mg/L)plus PHA and the control group cultured without these reagents. Afcer the cell viability being measured, T cell activation was observed with inverted microscope. And then ultrathin section of the cells sample was prepared tobe observed with transmission electron microscope(TEM).Results Morphologically, activated T cells were obviously bigger, their nuclei clear, and most of divided cells had irregular outline;there were cell colonies to be seen in growing regions. Under TEM, there were more vacuoles (visible property of structure) in typically activated T lymphocytes , chromatic agglutination and nucleoli to be observed.Conclusion Main properties of T cell activation of human umbilical cord blood were chromatic agglutination, more vacuoles in cytoplasm and cell colonies formation.

4.EFFECT OF LYCOPENE ON PREVENTION OF LIPID METABOLIC DISTURBANCE IN RATS AND ITS MECHANISM

Objective To study the protective mechanism of lycopene by modulating lipid metabolic disturbance in rats. Method The hyperlipidemic rats were induced by high fat diet and were divided into groups given different doses of lycopene. The serum total cholesterd(TC), triglyceride(TG), low density lipoprotein cholesterol(LDL-C), high density lipoprotein cholesterol(HDL-C), the hepatic lipase(HL) and lipoprotein lipase(LPL)activity in liver were tested by biochemical method, and the expression of low density lipoprotein receptor(LDL-R)in liver was detected by Western blot analyses. Results Lycopene decreased serum TC, TG, LDL-C, and the activity of LPL and HL in three lycopene groups were higher than those in model group, and the high dose group was the highest. Conclusion Lycopene may increase the lipases’activity and the expression of LDL-R, and decrease the deposit of TG and LDL in liver to prevent lipid metabolic disturbance.

6.The CT findings and its pathological basis of gastrointestinal neuroendocrine tumors

Objective To investigate the CT findings and its pathological basis of gastrointestinal neuroendocrine tumors (GEP-NETs ). Methods The CT findings of 23 patients with GEP-NETs confirmed by pathology were analyzed retrospectively and compared with the pathological results.Results The GEP-NETs were found on CT in 20 patients,including focal gastrointestinal wall thickening in 4,mass formation in 6,and both in 10.The tumor diameter ranged from 0.9 cm to 5.2 cm with a mean value of (2.6±0.6)cm. Plain CT showed homogenous isodensity in 15 lesions,little necrosis with low density in 4,and hemorrhage with high density in 1. The dynamic contrast-enhanced CT showed the tumors with obvious enhancement in 16 cases and mild enhancement in 4 in arterial phase.In addition,serosa invasion was found in 7,enlargement of mesentery lymph node in 7,and liver metastases in 2.The pathol-ogy showed the location of submucosa and invasion of the tumors.Most small tumors had intact gastrointestinal mucosa,and some large ones had surface or infiltrated ulcer.Conclusion Some specific CT findings of GEP-NETs depend on its pathological character-istics.CT plays an important role in assessment of invasion extent and metastasis of the tumor.
7.Causes and Countermeasures of Violent Trespass against Medical Right

8.Relationship between the triglycerides to high-density lipoprotein ratio and coronary heart disease

Objective To investigate the relationship of triglycerides/high-density lipoprotein (TG/HDL C) ratio with coronary heart disease (CHD) and its severity.Methods 158 patients with CHD were enrolled in this study.Patients were divided into 4 groups according to the selective coronary arteriography:the control group,single-vessel CHD group,double-vessel CHD group,multiple-vessel CHD group.The relationship between multiple risk factors and CHD was analyzed by stepwise multiple regression.The severity of coronary artery stenosis was analyzed by Gensini scoring system.Results TG/HDL-C ratio was elevated in patients with coronary artery stenosis as compared with the control group [(1.8±1.0) vs.(1.7±1.4),t=2.68,P<0.05].TG/HDL-C ratio was elevated along with the severity of CHD.TG/HDL-C ratio was elevated in multiple-vessel C HD group as compared with the control group by ANOVA analysis (P<0.05).Stepwise multiple regression showed that multiple-vessel CHD was positive related with total cholesterol (TC),LDL-C and TG/HDL-C ratio (β value:0.326,0.339,0.158,respectively,all P<0.05).Conclusions TG/HDL C ratio is related with multiple vessel CHD,but it does not have more clinical significances than other indicators.
9.Study on the extent of lymphadenectomy in thoracic esophageal carcinoma

Objective To study the rule of lower-cervical lymphatic metastasis in thoracic esophageal carcinoma,and make evaluation about the reasonable extent of lymphadenectomy. Methods One hundred and eight cases of thoracic esophageal carcinoma through chromatic ultrasound and CT before operation were divided into different groups selectively,while three fields lymphadenectomy (3-FL) was adopted in 31 cases,and two fields lymphadenctomy (2-FL) was adopted in 77 cases. Results The rate of lower-cervical lymphatic metastasis was 87.1%(27/31) through chromatic ultrasound and CT,and that was 25.0%(27/108) before the two up-mentioned examinations (P<0.05). In all cases, the rate of lower-cervical lymphatic metastasis in the upper pectoral esophageal carcinoma was 47.6% (10/21),that in the middle pectoral esophageal carcinoma was 21.3%(13161),and that in the middle and lower pectoral esophageal carcinoma was 19.5%(17187) ,P<0.05. Through 3-FL, the rate of lower--cervical lymphatic metastasis in the upper pectoral esophageal eareinoma was 57.1%(12/21), that in the middle pectoral esophageal carcinoma was 23.0%(14/61), and that in the middle and lower pectoral esophageal carcinoma was 21.8%(19/87), P<0.05. Conclusions The regionality metastasis is the main fashion in thoracic esophageal carcinoma with lower-cervical lymphatic metastasis. It is feasible to judge lower-cervical lymphatic metastasis through chromatic ultrasound and CT before operation.The 3-FL of the upper pectoral esophageal carcinoma is recommended. The 3-FL is selectable in the middle and lower pectoral esophageal carcinoma according to the result of chromatic ultrasound and CT.
10.Effects of zinc-finger proteins and artificial zinc-finger proteins on microbial metabolisms--a review.

Zinc-finger proteins have been widely studied due to their highly conserved structures and DNA-binding specificity of zinc-finger domains. However, researches on the zinc-finger proteins from microorganisms, especially those from prokaryotes, are still very limited. This review focuses on the latest progress on microbial zinc-finger proteins, especially those from prokaryotes and the application of artificial zinc-finger proteins in the breeding of robust strains. Artificial zinc-finger proteins with transcriptional activation or repression domain can regulate the global gene transcription of microbial cells to acquire improved phenotypes, such as stress tolerance to heat, ethanol, butanol, and osmotic pressure. Using the zinc-finger domain as DNA scaffold in the construction of enzymatic system can enhance the catalytic efficiency and subsequently the production of specific metabolites. Currently, zinc-finger domains used in the construction of artificial transcription factor are usually isolated from mammalian cells. In the near future, novel transcription factors can be designed for strain development based on the natural zinc-finger domains from different microbes, which may be used to regulate the global gene expression of microbial cells more efficiently.
