1.In vitro susceptibility testing of Candida albicans to berberine hydrochdoride

ObjectiveTo test the antifungal activity of berberine (BBR) against Candida albicans in vitro. MethodsAccording to the National Committee for Clinical Laboratory Standards (NCCLS) M27-A protocol, broth microdilution test was performed to assess the susceptibility of 30 clinical isolates of Candida albicans to BBR and fluconazole alone or in combination. ResultsThe geometric mean minimum inhibitory concentration (MIC) was 248.18 mg/L and 2.64 mg/L for BBR and fluconazole alone, respectively. When combined with BBR of 320, 160 and 80 mg/L, the geometric mean MIC decreased to 0.03, 0.35 and 1.35 mg/L respectively for fluconazole. Fluconazole and BBR showed synergistic effects in 26.7% of these isolates including 3 fluconazoleresistant strains, additive effects in 56.7%, indifferent effects in 16.7%, antagonistic effects in none. Conclu sionIn vitro, BBR has antifungal activity against Candida albicans and can enhance the activity of fluconazole against Candida albicans.
2.Evaluation of four approaches in screening of cervical lesions

Exfoliative cell samples were collected from 545 patients in outpatient clinic of Kelamayi Centre Hospital from Sep 2009 to Dec 2010. Liquid-based cytology test ( LCT ) and high-risk human papillomavirus (HR-HPV) test were performed in exfoliative cells; and colposcopic biopsy was undertaken in all patients.With the pathological results as gold standard the diagnostic values of HR-HPV test,LCT test,HPV-LCT serial test and HPV-LCT parallel test were evaluated for cervical cancer screening.The sensitivity of HPV-LCT parallel test was 99.17% with a Youden's index of 0.81.Our results indicate that HPV-LCT parallel test can improve significantly the detection rate of cervical cancer screening.

4.Expression and significance of Ezrin in skeletal muscle of patients with myopathy

Objective To investigate the expression and significance of Ezrin in skeletal muscle of myopathy patients. Methods The skeletal muscle specimens with obviously regenerating muscle fibers were obtained from 9 patients with pseudohypertrophic muscular dystrophy (DMD) and 5 patients with polymyositis (PM). Then frozen serial sections were made and the sections were treated with HE stain, immunohistochemistry stains including anti-Ezrin and anti-Neural cell adhesion molecule (NCAM) monoclonal antibodies. The pathological changes and the expression of Ezrin were observed. Results The small regenerating fibers, central location of nucelus and basophilia endochylema were observed under HE stain both in the patients with DMD and PM. Anachromasis of anti-NCAM monoclonal immunohistochemistry stain was found in the regenerating muscle fibers. Positive Ezrin expression was also detected in the regenerating fibers. However, this expression tapered gradually as the mature process of skeletal muscle. The expression of Ezrin was negative in the mature fibers. Ezrin expressed in the myoblasts. Conclusions Ezrin may be closely related to the regenerating skeletal muscle fibers of DMD and PM patients.
5.In-vitro Antibacterial Effect and Sterility Test of Danshen Injection

Objective To investigate the in-vitro antibacterial effect and sterility test of Danshen injection(DI). Methods Three kinds of in-vitro antibacterial and bacteriostatic methods were used to investigate the in-vitro antibacterial effect of DI and to establish its sterility test according to the results of the antibacterial effects. By validation test, the validity of its sterility test was evaluated. Results The results of doubling dilution test showed that the minimum bacteriostatic concentration of DI is 0.098 mg/mL, and its minimum bactericidal concentration is 1. 563 mg/mL in ethanolsulfate medium and 0.195 mg/mL in nutrient meat soup medium. Rate of recovered bacterium was much more than 80% by membrane filtration method. Conclusion DI has strong in-vitro bacteriostatie effects for staphylococcus aureus and the effectual method of its sterility test is membrane filtration method.

7.Evaluation of clinical application of approaches in cervical cancer screening

Objective To evaluate the function of Hybrid capture 2 (HC2) human papillomavirus (HPV) test,liquid-based cytology test (LCT),HPV serial test,LCT serial test and HPV-LCT parallel test for cervical high-grade lesions screening and to guide in selecting the scheme of cervical cancer screening.Methods HPV test,LCT,and colpescopic biopsy were performed in 545 women treated in Centrale Hospital of Klamayi from Sep.2009 and Dec.2011.The pathology results were the final diagnosis to evaluate the value of each approach for screening cervical cancer.Results Of 545 patients, 10 had cervical cancer.41 had cervical intraepithelial neoplasia CIN3,69 had CIN2,and 120 had CIN 1.Positive rate of high risk HPV was 64.22 %.The sensitivity,specificity,accuracy,positive pre-value,and negative pre-value of HPV test for detecting high-grade cervical lesions were 95.83 %,84.71%,88.48 %,30.68 %,99.12 %,respectively.The values of LCT were 84.20 %,89.53 %,85.53 %,36.69 %,91.76 %,respectively.The values of LCT serial test were 81.03 %,95.96 %,78.33 %,40.69 %,92.92 %,respectively.The values of HPV serial test were 81.03 %,95.26 %,78.33 %,42.15 %,99.52 %,respectively.The values of HPV-LCT parallel test were 99.20 %,81.53 %,90.03 %,32.09 %,99.33 %,respectively.Conclusion HPV-LCT parallel test can significantly improve the detection rate of cervical cancer screening.
8.EFFECTS OF ELECTROACUPUNCTURE ON CELL-MEDIATED IMMUNITY OF THE HUMAN BODY

70 patients including 60 cases under acupuncture anesthesia and 10 cases under drug anesthesia were observed. Before and 20 minutes after acupuncture the blood samples were taken from the patient ear lobes respectively, and in some patients taken once again 24 hrs after acupuncture. The electroacupuncture point Hegu or Zusanli was mainly adopted. As the method for detection of cell-mediated immunity(CMI) in vitro the improved microtechnique of whole blood for E-rosette (active and nonactive) and lymphocyte transformation tests was used. In performance of active rosetting the total leucocyte count and the differential lymphocyte count were done for calculation of absolute number of active rosette forming cells (RFC). The mean value of increase of active RFC was 12.7?1.43, the decrease was 6.8?1.77 after acupuncture. The increment of the absolute number of active RFC was 175?63.59. However no marked effect on the drug anesthesia group was found. In the lymphocyte transformation assay the increase was 12.7?1.49, the decrease was 7.0?2.19, and the enhancement effect still exhibited 24 hrs after acupuncture. In these tests an increase was mostly found in those with a lower or a usual CMI level; a decrease often found in those with a higher CMI level prior to acupuncture. The increase or decrease level in the results of three kinds of test (active, nonactive RFC and lymphocyte transformation) was similar, the increase range was 12~13%, the decrease range 6~7%. As the former compared with the latter, the promotion was prominent by all means.
9.Glycometabolic state in hypertensive and normotensive patients: identifying candidates for oral glucose tolerance tests.

OBJECTIVETo characterize unknown glycometabolic state in patients with essential hypertension (EHT) and normotensive patients and determine which EHT patients are candidates for oral glucose tolerance tests (OGTTs).
METHODSThis cross-sectional study consecutively recruited 895 EHT patients and 486 normotensive patients. The data including blood glucose, blood pressure, blood lipids, angiography profiles, and left ventricular parameters were collected.
RESULTSOGTTs performed in all patients revealed that the prevalence of abnormal glucose metabolism (AGM) was significantly higher in EHT patients than in normotensive patients at both baseline (P<0.001) and post-OGTT analysis (P<0.001). In total, 76.4% of the individuals with impaired glucose tolerance and 78.2% of individuals with newly diagnosed diabetes would have remained undetected if OGTTs had not been performed. Newly diagnosed AGM was significantly correlated with the presence and severity of coronary stenosis and left ventricular structure abnormalities and dysfunction. EHT patients with fasting glucose ≥5.6 mmol/L, hypertension duration exceeding 10 years, coronary artery disease, high-sensitivity C-reactive protein >3 mg/L, or high levels of apoB/apoA-1 ratio were at high risk of AGM.
CONCLUSIONSAGM is more common in patients with EHT than in normotensive patients, and OGTTs is a cost-effective strategy to detect AGM in EHT patients.

10.Analysis on present situation of post-hospital care needs scale in patients with colostomy and validity of the research

Objective To build demand scale for Post-hospital intestinal colostomy patients and test the reliability and validity. Methods Through the investigation of relevant literature at home and abroad,Based on the Maslow's Need Hierarchy Theory, in combination with intestinal colostomy patients psychological scale, using qualitative research, a semi-structured interview was conducted in 12 patients with colostomy A thorough understanding of colostomy patients Post-hospital care needs content (interview time as follows:September-October 2015), to analysis the interview recording data, the item pool of the scale for post-hospital care needs in patients with colostomy was constructed. Choosing 200 patients from the department of Proctology of third-grade class-A hospitals of Jilin province, and carries on the scale of clinical testing, after collecting the relative data , and make use of Critical Ration、the correlation coefficient method and the internal consistency coefficient method、further concludes that the scale items should be added or not, and picking up the internal consistency reliability 1/2 reliability retest reliability content validity and structure validity to analysis each item. Results The demand scale for Post-hospital intestinal colostomy patients included 6 dimensions and 24 items. The total content validity index (S-CVI) of the scale was 0.968, and the content validity index (I-CVI) of each item was 0.78-1.00. By exploratory factor analysis, 6 principal components were extracted, and the cumulative explanation variation of 6 principal components was 91.753%. This indicated that the scale had good validity. The total Cronbach a coefficient of the scale was 0.751, and the Cronbach a coefficient of each dimension was 0.968-0.974, all above 0.7, which indicated that the internal consistency was good. The half-fold reliability and retest reliability of the scale were 0.794 and 0.827 respectively. Conclusion The scale designed in this study has good reliability and validity, and can be used as an effective assessment tool for nursing needs of patients after discharge from enterotomy.
