Objective To investigate the effects of ropivacaine on the whole cell sodium currents in rat sympathetic ganglion neurons in order to determine whether sympathetic ganglion is involved in the ropivacaine cardiotoxity. Methods The sympathetic neurons were enzymatically isolated from the superior cervical ganglion. The effects of ropivacaine on the whole cell sodium channel currents were assessed using patch clamp technique. Results Ropivacaine blocked the whole cell sodium channel currents dose dependently. When the holding potential (Vh) was -80mV and Vt Omv , 0.01?mol/L ropivacaine reduced peak currents by 30.02% with a mean IC50 of 2.68?mol/L. The blockade was also potential dependent. When Vh was -60mv, the mean IC50 was 1.55?mol/l. 1.0?mol/L ropivacaine reduced the peak value of I V curve by 30.66% but did not affect the shape of I V curve and caused 2.56mv shift of voltage dependence activation curve to depolarized potentials and 3.56mv shift of steady state inactivation curve to more hyperdepolarized potentials. The conditioning pulse potential at which half maximal channels were activated (V1/2), changed from -52.99mv to -56.44mv and the test potential at which half maximal channels were activated (V1/2), changed from -25.2mv to -22.64mv. Conclusions Subconvulsive concentration of ropivacaine significantly inhibits sympathetic ganglion neurons in a dose dependent and potential dependent way through the inactivation of sodium channel, indicating that sympathetic ganglion neurons may contribute to the cardiotoxity of ropivacaine.

Objective To compare the clinical outcomes of cervical vertebral reconstruction using titanium mesh and nano-hydroxyapatite and polyamide 66 (n-HA/PA66) mesh as two different kinds of artificial vertebrae. Methods From January 2008 to June 2009 had surgically treated 37 cases of cervical spondylosis with anterior corpectomy for decompression, and artificial vertebrae implanted combined platescrews fixation for cervical reconstruction and fusion. As a vertebral substitution, titanium mesh were implanted in 25 cases ( group A ), and the others (12 cases, group B) were implanted with n-HA/PA66 mesh. The height of fusion space immediately postoperatively and the implanted condition were observed. The fusion rate and the JOA score were recorded to compare the clinical outcomes. Results Two groups received 6-20 ( 15.4 ±4.2 )months follow-up. When follow-up to the end grafts were already fusion. The preoperative JOA score in group A,B [(8.40 ±0.96) scores vs. (8.33 ±1.07) scores(P > 0.05)] were increased to (14.36 ±0.86)scores and (14.83 ±0.71) scores after postoperatively (P> 0.05),but there were statistically significant differences between preoperative and postoperative each group (P<0.05). There were 6 cases artificial vertebrae in group A and 1 case in group B subsided asymptomaticly with 100% choiceness rate. Conclusion These two kinds of artificial vertebrae could show similar outcomes in cervical vertebral reconstruction, but the n-HA/PA66 mesh has a lower subsidence rate.

OBJECTIVETo study the effects of the ingredients from Chinese herbs with the nature of cold or hot on the expression of TRPV1 and TRPM8.
METHODThe effects of ingredients from herbs on primary culture DRG neurons are observed in vitro. The expression quantity of gene is detected by the method of real time PCR. the 2 (-deltadeltaCT) method is applied to analyze the data.
RESULTIngredients from herbs with the nature of cold up-regulate the expression level of TRPV1 and down-regulate that of TRPM8, especially under the temperature condition of 39 degrees C; while ingredients from herbs with the nature of hot up-regulate the expression level of TRPM8 and down-regulated that of TRPV1, which is more significant under the temperature condition of 19 degrees C.
CONCLUSIONThe regulatory changes of TRPV1 and TRPM8 mRNA expression induced by the chemical ingredients might be related to the cold and hot natures of the herbs from which the ingredients are extracted. And this could be one of the therapeutic mechanisms for the treatment of Chinese herbal medicines to cold- and heat-related diseases.
