Objective To observe the local immue response and changes of angiogenic factors of tumor cells in Heps-bearing mice after mesenchymal stem cell (MSC) are administrated. And to explore the feasibility and safety of MSC for liver tumors therapy. Methods MSC were obtained through adherent culture method. Phe-notypes of MSC were analyzed by flow cytometry. MSC were labeled with DAPI in vitro. 54 Mice of 8 weeks of age with subcutaneously transplanted liver carcinomas were developed randomly. When the maximal diameters of the tumor reached 0.5 - 0.8cm, they were divided into three groups randomly: MSC group, DAPI group and NS control group. 2 × 10~6 MSC and MSC marked by DAPI were administrated into the mice right rear back tumor tissue. The survival time of the tumor-bearing mice was recorded and the mean survival time was calculated. Immunohistochemical staining was performed to count CD4~+ T cells and CD8~+ T cells in the local tumor,as well as to examine the expression of vascular endothelial growth factor ( VEGF) in tumor cells. Results In the MSC group,the mean survival time was 45 d (95%CI;33 ~56 d) ,in the NS control group, the mean survival time was 33 d ( 95%CI : 28 ~ 37 d). There was a statistical significance in the difference between them ( P < 0.05). Immunohistochemical staining results showed as follow: the number of CD4~+ T cells and CD8~+ T cells in the MSC group decreased significantly in comparison with the NS control group at early stage. The expression of VEGF also decreased obviously in comparison with the NS control group and induced tumor cells necrosis at late stage. The survival time of MSC group was prolonged. Conclusion MSC can engraft in Heps-bearing tumor tissue, and inhibit T lymphocyte cellular immunity at early stage. It can reduce the number of CD4~+ T cells and CD8~+ T cells and promote tumor growth. MSC can down regulate VEGF expression and induce tumor cells necrosis at late stage. By this way,it can prolong the survival time of Heps-bearing mice.
3.Anatomic study on lumbar cortical bone trajectory of adults

Objective To determine anatomic parameters related to the surgery of lumbar cortical bone trajectory of normal adults and the feasibility of screw application in cortical bone and its specification and dimension.Methods Lumbar 3D image data of 80 adults between 18 and 40 years old (40 for each gender) were randomly selected as subjects of our research.With the help of the imaging software for 3D interactive viewing,the ideal starting point was supposed to be the junction of the center of the superior articular process and 1 mm below the inferior border of the transverse process.The diameter,length,lateral angle to the vertebral horizontal plane,cephalad angle to the vertebral sagittal plane of the trajectory and the horizontal distance from the insertion point to the lateral edge of the vertebral plate were measured.Differences of anatomic parameters for each gender,side and segment were analyzed.Results Differences of anatomic parameters on both sides of each segment had no statistical significance.At the ideal trajectory,the mean screw length was 37.56±2.41 mm at L1,38.72±2.36 mm at L2,39.51 ±2.51 mm at L3,39.78± 2.87 mm at L4 and 38.83±2.74 mm at L5.The mean screw diameters from L1 to L5 were 6.04±1.23 mm,6.17±1.24 mm,7.15±1.22 mm,8.02± 1.41 mm and 8.68± 1.42 mm respectively.However,differences of ideal entry angle of L1 to L5 had no statistical significance.The mean lateral angle from L1 to L5 were 8.46°±2.11°,9.37°±2.84°,9.62°±2.16°,9.53°± 1.98°,9.04°± 1.97°,while the mean cephalad angle to the vertebral sagittal plane from L1 to L5 were 26.49°±4.97°,25.94°±4.56°,26.42°±4.42°,26.29°±3.48°,26.89°±3.69°.The mean distance from the insertion point to the lateral edge of the vertebral plate gradually increased from L1 to L5,which were 1.19±0.75 mm,1.54±1.08 mm,2.01±1.45 mm,3.49±1.52 mm,4.47±1.32 mm respectively.The screw diameters of each segment for men were greater than those for women.Conclusion The common length of screw for lumbar cortical bone trajectory of normal adults might be from 35 mm to 40 mm.The safe upper limits of the screw diameter were 5.5 mm at L1,5.5-6.0 mm at L2,6.5-7.0 mm at L3,7.5 mm at L4 and 8 mm at L5.The average lateral angle of all 5 lumbar segments was 9.20°± 2.11° and the average cephalad angle was about 26.41°±4.22°.
4.Differentiation of Non-puerperal Mastitis from Breast Carcinoma with Non-mass-like Enhancement by Texture Analysis of Contrast-enhanced Magnetic Resonance Imaging

Purpose To investigate the feasibility of texture analysis of breast contrastenhanced magnetic resonance imaging in differentiating non-puerperal mastitis and breast carcinoma with non-mass-like enhancement in order to prevent misdiagnosis of nonpuerperal mastitis.Materials and Methods In this retrospective study,the contrastenhanced MRI images of 42 female patients of invasive ductal carcinoma with non-masslike enhancement and 30 female patients of non-puerperal mastitis were analyzed.3234 texture features were generated from manually selected region of interest (ROI) of normal breast tissue and breast lesions.By means of genetic algorithm and linear discriminative analysis,10 texture features were selected based on their stability and accuracy in breast tissue classification.Results With these 10 features,the linear discriminative analysis classifiers had sensitivity of 92.9% and specificity of 90.0% in classifying two lesions,and accuracy of 89.6% in classifying all three types of tissue.The result showed that texture analysis successfully differentiate non-puerperal mastitis and breast carcinoma with nonmass-like enhancement.Conclusion Texture analysis demonstrates the ability of differentiating invasive ductal carcinoma with non-mass-like enhancement,non-puerperal mastitis and normal breast tissue,and provides reliable results for clinical diagnosis.
5.Pharmacophore identification of novel dual-target compounds targeting AChE and PARP-1.

Multi-target drugs attract increasing attentions for the therapy of complicated neurodegenerative diseases. In this study, a computer-assisted strategy was applied to search for multi-target compounds by the pharmacophore matching. This strategy has been successfully used to design dual-target inhibitor models against both the acetylcholinesterase (AChE) and poly (ADP-ribose) polymerase-1 (PARP-1). Based on two pharmacophore models matching and physicochemical properties filtering, one hit was identified which could inhibit AChE with IC50 value of (0.337 +/- 0.052) micromol x L(-1) and PARP-1 by 24.6% at 1 micromol x L(-1).
6.Electrophysiological characteristics of Hirayama disease, amyotrophic lateral sclerosis and distal cervical spondylotic amyotrophy

Objective To explore the electrophysiological charaterstics of upper extremities nerves on the patients with Hirayama disease (HD),amyotrophic lateral sclerosis (ALS),and distal cervical spondylotic amyotrophy (DCSA).Methods The data of electrophysiological examination of the upper limbs of 87 patients with HD,83 with ALS and 28 with DCSA were reviewed retrospectively.Seventy-two patients with HD among 87 had unilateral upper limb's amyotrophy and the other 15 ones had bilateral amyotrophy.There were 30 patients had unilater upper limb's amyotrophy and 53 ones had bilateral amyotrophy from the group of patients with ALS; 20 patients with DSCA were affected unilaterally and 8 ones were bilaterally affected.Results Compound muscle action potential (CMAP) evoked by ulnar stimulation had a lower ampititude compared with that evoked by median stimulation in HD patients.In ALS cases that was just the opposite.However,the CMAPs were similar in DCSA cases.The mean ratio of CMAP amplitude by ulnar stimulation to by median stimulation was 0.58±0.40 in HD group; 2.28±1.25 in ALS and 1.31±0.63 in DCSA.The differences in the three groups were statistical significance.The U/M CMAP ratio was less than 0.6in 62 patients with HD,3 with ALS and 1 with DCSA,and more than 1.7 in 73 cases (57 ALS,12 HD and 4 DCSA).Conduction velocities (CV) of the sensory and motor nerves,the amplitude of the sensory nerve action potential in bilateral limbs,and the CMAP amplitude of the unaffected limb were normal in all cases.Conclusion This study could concluded that the severity of amyotropy in hypothenar mucles were higher than that in thenal muscles in patients with HD; there was just opposite in ALS cases and similar in DSCA.
7.Effect of hydrogen on mitochondrial function after cerebral ischemia/reperfusion injury in rats

Objective To study the effect of hydrogen on mitochondrial function in cerebral cortex after cerebral ischemia/reperfusion(I/R)injury in rats. Methods 48 male SD rats were randomly divided into sham operation group(sham group),brain I/R injury group(MOD group),hydrogen treatment group(H2group). 24 hours after reperfusion,the neurological deficit scoring was performed. The changes of mitochondrial membrane potential(△ψm),permeability openness(MPTP),ROS production rate and mitochondrial swelling were detected. Results Compared with the Sham group,neurological deficit score,MPTP openness,mitochondrial swelling degree and ROS production rate were increased in the MOD group(P<0.01),△ψm levels were reduced(P<0.01). Compared with the MOD group,the neurological deficit score,MPTP openness,mitochondrial swelling degree and ROS production rate were decreased in H2group(P<0.01),△ψm levels increased(P<0.01). Conclusions Simul-taneous intraperitoneal injection of pure hydrogen can reduce the generation of reactive oxygen species,protect the mitochondrial function of neuronal cells in the ischemic region after brain I/R,improvement the rat brain I/R after the neurological scoring.
8.Effect of hydrogen on mitochondrial membrane potential during focal cerebral ischemia-reperfusion in rats

Objective To investigate the effect of hydrogen on mitochondrial membrane potential during focal cerebral ischemia-reperfusion (I/R) in rats.Methods Eighty-four healthy adult male SPF Sprague-Dawley rats,weighing 220-240 g,were divided into 3 groups (n=28 each) using a random number table:sham operation group (Sham group),I/R group and hydrogen group (H2 group).Focal cerebral I/R was produced by mid-cerebral artery occlusion in I/R and H2 groups.Hydrogen 10 ml/kg was intraperitoneally injected immediately after onset of reperfusion and at 12 h of reperfusion in group H2.Neurological deficit was scored at 24 h of reperfusion.The rats were then sacrificed and brain tissues in cortex were removed for microscopic examination of the pathological changes and for determination of cerebral infarct size (by TFC staining),nerve cell apoptosis (by TUNEL),mitochondrial membrane potential (by JC-1 staining) and expression of Bcl-2 and Bax (by Western blot).Apoptotic index (AI) was calculated.Results Compared with Sham group,the neurological deficit score,percentage of cerebral infarct size and AI were significantly increased,the mitochondrial membrane potential was decreased,Bax expression in brain tissues was up-regulated,and Bcl-2 expression in brain tissues was down-regulated in I/R and H2 groups (P<0.05).Compared with I/R group,the neurological deficit score,percentage of cerebral infarct size and AI were significantly decreased,the mitochondrial membrane potential was increased,Bax expression in brain tissues was down-regulated,and Bcl-2 expression in brain tissues was up-regulated (P<0.05),and the pathological changes of brain tissues were significantly attenuated in H2 group.Conclusion The mechanism by which hydrogen reduces focal cerebral I/R injury is related to decreasing dissipation of mitochondrial membrane potential and inhibiting nerve cell apoptosis in rats.
9.Effect of discharge preparation service strategy in hospital-community chronic disease management of patients with liver cirrhosis

Objective:To explore the effect of discharge preparation service strategy in hospital-community chronic disease management of patients with liver cirrhosis.Methods:From January 2018 to May 2019, the convenience sampling was used to select 162 patients with compensatory cirrhosis hospitalized in the Department of Infectious Diseases, Puyang People's Hospital of Henan Province. Patients were randomly divided into observation group and control group, with 81 cases in each group. Control group used routine management, and observation group carried out the discharge preparation service strategy. We compared the differences in re-admission rate and laboratory indicators between two groups after discharge.Results:Within 6 months after discharge, the re-admission rates of patients in observation group and control group were 4.94% (4/81) and 20.99% (17/81) respectively, and the difference was statistically significant (χ 2=9.246, P<0.05) . Six months after discharge, prealbumin of observation group was (226.48±27.52) mg/L higher than that of control group, and alanine aminotransferase of observation group was (90.26±6.72) U/L lower than that of control group, and differences were statistically significant ( t=5.482, -6.834; P<0.05) . Conclusions:Discharge preparation service strategy can reduce the re-admission rate of patients with liver cirrhosis and delay the progression of the disease, which is worthy of clinical application.

To investigate the effect of hSCGF-alpha on human Umbilical Cord Mesenchymal Stem Cells (hUCMSCs), we obtained hSCGF-alpha using genetic engineering, hSCGF-alpha gene was amplified from hUCMSCs cDNA using two-step PCR and was inserted into pET-28a(+) plasmid vector. Induced by IPTG at 20 degrees Celsius for 24 h, the fusion protein expressed in E. coli BL21 (DE3) was mainly existing in soluble form. The recombinant hSCGF-a was purified using NI-NTA affinity chromatography and the purity was up to 90%. The colony forming test revealed that combined use hSCGF-alpha and rmGM-CSF (recombinant murine GM-colony stimulating factor, rmGM-CSF) had granulocyte/macrophage (GM) promoting effects on murine bone marrow GM progenitor. In addition, the results indicated that hSCGF-alpha and rhGM-CSF had stimulatory effect on hUCMSCs and their synergetic effect was the strongest.
