1.The curative effect of 61 patients with procedure for prolapse and hemorrhoids

Objective To evaluate the curative effect of procedure for prolapse and hemorrhoids.Methods To retrospectively anabpe the clinical data of 61 patients with procedure for prolapse and hemorrhoids of Inner Mongolia medical school affiliated hospitals form 2002 to 2009.Results The average time of operation was 20~30 minutes.After operation,2 cases appeared urine retention,2 cases appeared bleeding and 1 case appeared prolapse lightly.The average time of being hospitalized was 5 days.The curative effect of allpatients was well after 1~6 years follow-up.Conclusion It was a safe and effective method to use procedure for prolapse and hemorrhoids,but the longterm curative effect of it still need further observation.

3.Expression and significance of multidrug resistance-associated protein 3 in pancreatic cancer and pancreatic cancer cell lines

Objective To investigate the expression and significance of MRP3 in pancreatic cancer and different tumor cells. Methods MRP3 expression was detected in 30 cases of pancreatic cancer tissues and the adjacent tissues through immunohistochemical method. RT-PCR was used to detect MRP3 mRNA in 7 tumor cell lines and human embryo kidney cell lines 293T. Monoclonal antibody against MRP3 was employed to detect MRP3 protein expression through flow cytometry. Results There was significant difference for MRP3 expression in nucleus between the pancreatic cancer and normal tissues beside the cancer ( P= 0.003 ) while no significant difference was found in cytoplasm (P = 0. 472). MRP3 mRNA expression was found in 3 pancreatic carcinoma cell lines. The positive expression rate of MRP3 protein was relatively high in BxPC-3 and AsPC-1 cells (68. 5% and 33.6% respectively) while it was only 5.4% in PNAC-1. Conclusions The difference of MRP3 expression in pancreatic cancer and normal tissues lies mainly in nucleus. BxPC-3 may serve as cellular models for in vitro studies on multidrug resistance of pancreatic carcinoma.
4.Expression of PXMP4 in breast cancer tissues and its effects on biological behavior of breast cancer cells

Purpose To detect the expression of peroxiso-mal membrane protein 4(PXMP4)in breast cancer tissues and to explore the effect of PXMP4 on the proliferation,invasion,and epithelial-mesenchymal transition(EMT)of breast cancer cells.Methods Bioinformatics and immunohistochemistry(IHC)were used to detect the expression of PXMP4 in breast cancer tissues.In breast cancer cells,Western blot was used to detect the expression of Cyclin D1,E-cadherin,vimentin and N-cadherin after knockdown and overexpression of PMXP4.The proliferation ability of breast cancer cells was analyzed by CCK-8 and plate cloning assay.Scratch healing and Transwell assay an-alyzed the migration and invasion ability of breast cancer cells.Lentivirus was used to construct a PXMP4-silenced MCF-7 cell line,and the PXMP4-silenced MCF-7 cells were injected into the subcutaneous or tail vein of mice to observe lung metastasis and the number of subcutaneous tumors.Results Bioinformat-ics and IHC showed that the expression of PXMP4 in breast cancer tissues was significantly increased(P<0.05),and the prognosis of breast cancer patients with high expression of PXMP4 was poor(P<0.05).The clinicopathological analysis showed that the expression of PXMP4 was correlated with tumor grade and lymph node metastasis(P<0.05).In vitro knock-down of PMXP4 inhibited the proliferation,invasion and EMT process of breast cancer cells(P<0.05).Conversely,overex-pression of PXMP4 promoted the proliferation,invasion and EMT process of breast cancer cells(P<0.05).In vivo,the number of lung metastases,the size of subcutaneous tumor,and the expression of Ki67 in tumor tissue were significantly de-creased after silenced PXMP4(P<0.05).Conclusion PXMP4 is related to tumor grading and lymph node metastasis.PXMP4 promotes proliferation,invasion and EMT process of breast cancer cells.
