1.Expression and clinical significance of EphA8 in colon cancer

Objective To investigate the expression of EphA8 in colon cancer and its clinical significance.Methods Real-time quantitative polymerase chain reaction (RT-PCR) and Western blot were respectively used to assess the expressions of EphA8 mRNA and protein in normal colon mucosa cell line and colon cancer cell lines.Immunohistochemistry for EphA8 and vascular endothelial growth factor (VEGF) were performed in 98 cases of colon cancer and 12 matched normal mucosa tissues.CD31 immunohistochemical staining was used for microvascular density (MVD) counting.The relationships between the expression of EphA8 and the expression of VEGF and MVD,the clinical pathological significance,and the prognosis of patients were analyzed by statistical methods.Results EphA8 mRNA expressions were significantly higher in colon cancer cell lines than those in normal mucosa cell line (t =11.98,13.54,P <0.001).EphA8 protein expressions were significantly higher in colon cancer cell lines than those in normal mucosa cell line (t =4.63,P =0.006;t =4.92,P =0.004).The high expression of EphA8 was closely related to tumor size (x2 =22.97,P<0.001),TNM stage (P<0.001),differentiated degree (P =0.007)and lymph node metastasis (P<0.001),distant metastasis (x2 =6.97,P =0.008) and poor survival rote (x2 =17.3,P <0.001).But it was not associated with the gender and age (x2 =1.36,P =0.30; x2 =0.83,P=0.44).Besides,EphA8 had positive correlation with the VEGF and MVD (r =0.434,P <0.001; r =0.584,P <0.001).Conclusion EphA8 may play important roles in the development and metastasis of colon cancer,which has potential clinical values in the therapeutic intervention and prognosis of colon cancer.
2.Effects of different frequencies of glucocorticoids on the soft tissues and the immune system in a model of osteoporosis

Objective To investigate different frequencies of glucocorticoids (GCs) on the tissue in a model of osteoporosis. Methods Thirtytwo three-month-old SD female rats were randomly divided into four groups: (1) the control group (group C); (2) the low-frequency group (group L); (3) the middle-frequency group (group M); (4)the high-frequency group (H). The rats in the group C were given intramuscular injection (im) of 0.9% saline. Im of dexamethasone (Dex) was 1 mg/(kg·time). Rats were given two times im a week in the group C, four times im a week in the group M, and six times im a week in the group H. Each rat was sacrificed on thirty days post-administration. Results (1)The body weight of rats gradually increased in the Ctrl group , however , the body weight of rats declined gradually during the experiment in the group L, M and H. The size of immune organs (spleen and thymus) significantly decreased in rats of the group L, M and H. (2)Compared with the group C, cell edema was changed in the heart, renal and lung morphological fatty degeneration in liver , atrophy in spleen , atrophy in lymphoid nodules , and cell edema in kidney tubular were observed. Conclusion GCs cause serious degradation in the thymus and atrophy of the spleen. Administration has different inhibitory effect on immune function; the high frequency will lead to strong inhibition.

On this basis, combined with the self-character of TCM, the potential path for the academic cultivation of young scholars is refined: focusing on clinical practice, confirming clinical efficacy, deepening mechanism exploration, improving clinical therapeutic effect, optimizing clinical program, extending clinical disease spectrum, inheriting, seeking, breaking through, innovating, expecting and topping over. It is believed that during the academic cultivation, the importance is to form the stable research direction and continuity, organize research team, absorb actively new knowledge and thoughts, and adopt multidisciplinary cross and coordination so as to achieve the greater success ultimately.

4.Analysis of relevant factors in patients with diabetic retinopathy

Objective To explore the relevant factors of diabetic retinopathy(DR).Methods 400 patients with type 2 diabetes were selected,and the relevant factors with DR of type 2 diabetes were analyzed.Results The incidence of DR in type 2 diabetes was 49.0%.In type 2 diabetes when diabetes duration>15 years,the incidence of DR was 84.7%,duration<5-year incidence rate was 30.0% respectively,there were significant differences(P<0.05).Incidence rate in lower blood glucose group and poor glycemic control group were 38.1% and 68.5% respectively,and there was statistically difference(P<0.05),and in normal blood pressure and well control group,the incidence rates were 44.1% and 41.2%,which were significantly better than 80.0% of poorly controlled hypertension group.Conclusion Duration of diabetes,glycemic control level,blood pressure,exercise,and regular schedule were related factors of DR.The most important thing of DR treatment and prevention was to control blood glucose.
5.Clinical Observation of Adiponectin in Inhibiting Fatty Toxicity by Acupuncture

Objective:To investigate if acupuncture has an inhibitory effect on fatty toxicity and its possible mechanism. Methods: Thirty-four patients with simple obesity were clinically treated with acupuncture for 3 courses. Before and after treatment, body mass index(BMI) and fat percentage(F%) were determined, and serum insulin and adiponectin were measured by enzyme linked immunoassay, blood lipid and sugar by biochemical colorimetry and eight kinds of free fatty acids(FFAs) by high performance liquid chromatography. Twenty normal persons were selected as a control. Results:The total efficacy rate was 88.2%. Acupuncture could increase the decreased insulin sensitivity index(ISI) in the patients(t=-5.103, P=0.000). The correlation of a decrease in F% with an increase in unsaturated fatty acid after acupuncture was of significance(r=0.402, P=0.019) and its correlation with an increase in the ratio between unsaturated fatty acid and saturated fatty acid was also of significance(r=0.348, P=0.044). The correlation between increases in high density lipoprotein and in eicosapentaenoic acid was of significance(r=0.352, P=0.041). The correlation between a decrease in BMI and an increase in high density lipoprotein was of significance(r=0.357, P=0.038). Adiponectin level significantly rose after acupuncture (compared with pre-treatment, P=0.000). The correlation between a decrease in FFAs and an increase in adiponectin was of significance(r=-0.349, P=0.043). Conclusion: Acupuncture can lower the levels of free fatty acids in the patients and increase their sensitivity to insulin to inhibit the fatty toxicity. The inhibitory effect of acupuncture on fatty toxicity is somewhat related to a rise in adiponectin level.
6.Effects of electroacupuncture versus sibutramine on adipocyte products in obesity rats

BACKGROUND:Both acupuncture and moxibustion and drug ale effective methods to treat simple obesity.Whether electroacupuncture is different from sibutramine to affect adipocyte products in obese organisms?OBJECTIVE:To observe the different effects of electroacupuncture and sibutramine on adipocyte produces in obesity rats and to investigate the mechanism of weight reduction by acupuncture and moxibustion.DESIGN:Layering-randomized controlled study.SETTING:Laboratory of Acupuncture and Pharmacology.Nanjing University of Chinese Medicine.MATERIALS:12045-day-old male SD rats were provided by Shanghai Experimental Animal Center.The experiment was in accordance with animal ethics standards.Sibutramine hyrdrochloride capsule was made in Fuling Pharmaceutical Factory of Taiji Group(batch number:X20010279).Electro-acupuncture apparatus(LH402A acupoint neutoelectrical stimulation equipment)was made in Beijing Huawei Chanye Developing Company.METHODS:This study was performed at the Laboratory of Acupuncture and Pharmacology,and Expefimental Animal Center,Nanjing University of Chinese Medicine from October 2004 to November 2006.①100 male SD rats underwent high-fat diet.Fifty-four alimentary obesity rats were randomly divided into electroacupuncture group,sibutramine group and obesity group.wim 18 rats in each group.Another 18 rats were regarded as a normal group.②Rats in the electroacupuncture group were given electrical stimulation(2/15 Hz,4 mA)at lateral Housanli and Neiting acupoints,contralateral alternation every other day.Rats were fixed with self-made binding instrument,and rats maintained a consciousness but not an intense struggle.The electricai stimulation lasted for 15 minutes every day,one day was an interval every 6 days.and the electroacupuncture was given for 49 days and 42 times in total.Rats in the sibutramine group were ingragastrically administrated with 0.2 g/L sibutramine hydrochloride(2.0 mg/kg/d).Rats in the control group and obesity groups were fixed the same as electroacupuncture group but not given electrical stimulation.MAIN OUTCoME MEASURES:①Obese marker:To detect body mass,body length,weight reduction rate and body mass/body fat mass,and to measure total cholesterol(TC),high density lipoprotein cholesterol(HDL-C)and triacylglycerol (TG)with biochemical chromatometry;②insulin sensitivity index:To measure fasting blood glucose,insulin and insulin sensitivity index with enzyme-linked immunosorbent assay;③adipocyte secreting regulatory substance level:To measure serum resistin,leptin,adiponectin,tumor necrosis factor-α(TNF-α),interleukin-6(IL-6)and its soluble receptor level with enzyme-linked immunosorbent assay.RESULTS:Among 120 rats,54 alimentary obesity rat models and 18 normal rats were included in the final analysis.①Obesemarker:Bodymass,bodyfatmass,cholesterol and TG levels in the obesity group were significantly higher than in the normal group(P<0.01).Body mass,body fat mass,body mass/body fat mass,cholesterol and TG levels in the electroacupuncture group were significantly lower than in the obesity group(P<0.01).TG level in the sibutramine group was significantly lower than in the obesity group(P<0.01).Weight reduction rate in the electroacupuncture group was significantly higher than in the sibutramine group(P<0.01).Body mass/body fat mass in the electroacupuncture group was significantly lower than in the sibutramine group(P<0.01).②Insulin sensitivity:Blood glucose and serum insulin levels in the obesity group were significantly higher than in the normal group,and insulin sensitivity index was significantly lower than in the normal group(P<0.01).Blood glucose and insulin levels in the electroacupuncture and sibutramine groups were significantly lower than in the obesity group,but insulin sensitivity index was significantly higher than in the obesity group(P<0.01).③Adipocyte secreting regulatory substance level:Adiponectin in the electroacupuncture group was significantly higher than in the obesity group (P<0.01).Resistin and TNF-αin the electroacupuncture group was significantly lower than in the obesity grollp(P<0.01).Resistin level in the sibutramine group was significantly lower than in the obesity group(P<0.01).CONCLUSION:The effect of electroacupuncture on abnormally secretary products of adipose tissues in obesity rats is superior to the effect of sibutramine.This may be one of important mechanisms of weight reduction by acupuncture and moxibustion,and prevention and cure of obesity and its related diseases.
7.Level of plasma D-dimer in the patients of type 2 diabetes mellitus with hypertension

Objective To evaluate the plasma D-dimer level in the patients of type 2 diabetes mellitus with hypertension and investigate their correlation.Methods Eighty-five subjects were divided into three groups according to clinical manifestation:control group:20 subjects ; type 2 diabetes mellitus group:21 subjects; type 2 diabetes mellitus combined with hypertension group:44 subjects.The level of plasma D-dimer was measured and the difference was compared between groups.The results were showed as mean ± sd,and the difference was compared using ANOVA Test ( SPSS13.0).Results The plasma D-dimer concentrations in normal control group was ( 102.15 ± 32.48 ) μg/L,in single type 2 diabetes mellitus was ( 148.62 ± 80.99 ) μg/L,while plasma concentrations in type 2 diabetes mellitus combined with hypertension was ( 206.28 ± 92.99 ) μg/L.plasma D-dimer concentration was higher in single type 2 diabetes mellitus than that in normal control cases( P <0.05) ;And plasma D-dimer concentration was also found increased in type 2 diabetes mellitus combined with hypertension when compared with control group (P < 0.01 ) ;And there was also significant difference on plasma D-dimer concentration between single type 2 diabetes mellitus and type 2 diabetes mellitus combined with hypertension cases ( P < 0.05 ).Conclusion The plasma levels of D-dimer was increased obviously in single type 2 diabetes mellitus and type 2 diabetes mellitus combined with hypertension,it may be related to the imbalance of coagulation and fibrinolytic system.Monitoring of plasma D-dimer concentration in type 2 diabetes and patients with hypertension may have important clinical implications for the prevention of thrombotic diseases.
8.CT and MRImaging of gynecological acute abdomen

Objective To explore the manifestations and diagnostic value of CT and MRI in gynecological acute abdomen.Methods CT and MRI features of 45 patients with gynecological acute abdomen proved by surgery were reviewed,including 17 ectopic pregnancy,10 ovarian cyst rupture,6 pedicle torsion of ovarian tumors,9 tubo-ovarian abscess and pelvic inflammation and 3 uterine perforation.31 cases underwent CT examination,14 cases underwent MRI examination and 39 cases underwent non-enhancement and enhancement scanning.Results In the 17 ectopic pregnancy cases,cystic structure surrounded by a thick wall or heterogeneous mass located in adnexal area was found,with 10 cases showing increased number of peripheral vessels penetrating into tumors.In the 10 cases with rupture of ovarian cyst, thick capsule wall was incomplete and cystic cavities collapsed in 7 lesions.In the 6 cases with pedicle torsion of ovarian tumors,irregular thickening of tumor pedicle were found in 4 cases,ovarian enlargement with surrounding follicles arrayed as fruit platter were found in 2 cases.In the 9 cases with tubo-ovarian abscess and pelvic inflammation,honeycomb-shaped lesions and multilocular changes were found in 6 cases and fallopian tube expanded like sausages in 5 cases.3 cases with uterine perforation exhibited intrauterine gas and fluid.Uterus with overflowed gas was found in 2 cases.One case had the thin and incomplete uterine wall.Conclusion CT and MRI can be used to confirm the causes of gynecological acute abdomen,and to comprehensively display the anatomical structures and pathological changes of pelvic tissues and organs.Therefore,CT and MRI are effective supplement means for the clinical diagnosis of gynecological acute abdomen.

10.Clinical analysis of 14 cases of niacin deficiency

The clinical data of 14 patients with niacin deficiency diagnosed and treated in Department of Dermatology, Affiliated Hospital of Jining Medical College from 2012 to 2021 were retrospectively analyzed. There were 11 males and 3 females aged 26-65 years. The etiological factors were alcoholism in 8 cases, gastrointestinal disease in 3 cases, medication history in 1 case, and unknown etiology in 2 cases.Patients had typical skin lesions, 1 case also had both digestive system and nervous system symptoms, and 3 cases had combined digestive system symptoms and 2 cases had neurological symptoms. All patients were systematically treated with oral nicotinamide and vitamin B complex, and also with topical drugs; and they all improved after 14-52 days of treatment. During regular follow-up, 2 cases of alcoholics and 1 case with diarrhea had recurrence. It is suggested that the typical clinical triad of niacin deficiency is uncommon, and the diagnosis is based on the medical history, clinical manifestations and relevant laboratory test, and the treatment with nicotinamide and vitamin B complex is usually effective; alcoholism is the main cause in male patients and is prone to recurrence.
