AIM:To explore the relationship and molecular mechanism between microRNA-21(miR-21) and Schwann cells ( SC) following peripheral nerve injury.METHODS: The mRNA expression of miR-21 and phosphatase and tensin homologue deleted on chromosome ten ( PTEN) in animal model were detected by real-time PCR.The over-ex-pression of miR-21 and inhibition of miR-21 expression in the Schwann cells according to transfection of lentiviral vectors were performed, the nonspecific miRNA was used as a negative control ( NC) .The cell apoptosis was measured by flow cy-tometry.The mRNA expression of miR-21 and PTEN in the cells was detected by real-time PCR.The protein expression of PTEN and cleaved caspase-3 was determined by Western blot.RESULTS: The level of miR-21 was significantly higher and the mRNA level of PTEN was significantly lower in the model of nerve injury than those in control group.miR-21 over-expression decreased the number of apoptotic Schwann cells compared with NC-SC.The mRNA expression of PTEN was down-regulated by over-expression of miR-21.The protein expression of PTEN and cleaved caspase-3 was down-regulated by over-expression of miR-21 (P<0.05).CONCLUSION: miR-21 may play an important role in the peripheral nerve injury through inhibiting apoptosis of Schwann cells by down-regulating the expression of PTEN.

Objective To study the synergistic effects of Mitomycin C in photodynamic therapy for colonic carcinoma. Methods The colon carcinoma cell line SW800 was assigned to be treated with 002 ?g/ml Mitomycin C, or 2 ?g/ml PSD-007 (photosensitizer), or Mitomycin C and PSD-007 (PSD+MC group). The cells without any special treatment served as control. Fluorescence spectrophotometer and MTT assay were used to detect the content of intracellular photosensitizer and survival rate of SW700 cells under photodynamic therapy. The obtained data was analyzed with SPLM statistical software. Results After PSD was added for 4, 8, 16 and 24 h, intracellular PSD level of PSD+MC group increased significantly as compared with that of PSD group. The intracellular PSD contents of PSD group, PSD+MC group had a strong and negative correlation with cell survival rate respectively after photodynamic therapy, but the cell survival rate curve shifted to the left obviously. The best killing effects on SW700 cells of photodynamic therapy could be obtained when the SW700 cells had been exposed to photosensitizer for 8-16 h. Conclusion There is insignificant influence of lowconcentration of Mitomycin C alone on cell survival rate. The better synergistic effect of Mitomycin C on killing the colon carcimoma cells can be achieved by enhancing the intracellular content of photosensitizer for 6-8 h before photodynamic therapy.

Objective To develop a measurement method for determination of optical parameters of a red blood cell ( RBC) suspension based on the measurement of spatial scattered light signals without using an integrating sphere. Methods Multiple independent photoelectric sensors and light intensity modulation were used to obtain the measured values of diffuse reflectance, diffuse transmittance and collimated transmittance. The measured data results were imported into a Monte Carlo simulation based RTE to inversely determine the absorption coefficient, scattering coefficient and anisotropy factor of the measured sample using a new perturbation method. Results The described measurement method was applied to determine the optical parameters of a polystyrene microsphere suspension with a mean diameter of 2.6μm,and the results were essen-tially consistent with the calculated optical parameters by Mie code. Then, the RBC suspension was used for testing optical parameters, and the results were basically consistent with the parameters in the literature. Conclusion The system based on the measurement of spatial scattered light signals without using an integrating sphere will provide a quick and accurate approach for quantitative analysis of free hemoglobins and RBC suspensions.

Objective To observe imaging characteristics and changes of vivo two-dimension multi-voxel proton magnetic resonance spectroscopy (~1H-MRS) in the rabbit models of VX2 hepatic carcinoma after high intensity focused ultrasound (HIFU) treatment. Methods VX2 hepatic carcinoma models were established in 20 New Zealand rabbits. Routine MR and 2-dimentional ~1H-MRS scanning were performed before and after HIFU treatment. The central regions of interest (ROI) of the VX2 tumor, tumor border and paratumor normal liver tissues were selected. The Cho/Cr and Lip/Cr of the same ROI before and after HIFU treatment were compared. Results Total 28 satisfied spectrogram diagram of ~1H-MRS were brought into statistical analysis. Of all the spectra, 6 metabolite peaks were detected as lipids (Lip), glutamine and glutamate complex (Glx), choline (Cho), lactate (Lac) and creatine (Cr). Cho and Lac peak in tumor center and tumor border regions after HIFU treatment were higher than those before HIFU treatment. Lip peak was lower than before, and major metabolites of paratumor normal liver tissues did not changed significantly. Statistical differences of Cho/Cr and Lipid/Cr of tumor center and border region were found between before and after HIFU treatment (P<0.05). Conclusion Two-dimension multi-voxel ~1H-MRS can reflect major changes in the level of metabolites of different ROI for hepatic VX2 carcinoma after HIFU treatment.
