1.Dietary survey and nutritional evaluation of one meal among the four-star hotel customers in Nanjing city

Objective To investigate the food intake of one meal of the hotel customers and evaluate it rationality ,then provide dietary suggestions .Methods Six hundreds hotel customers were investigated in 2 four‐star hotel and 2 five‐star hotel of Nanjing , who were selected with a stratified sampling method .Their 5 days diet were investigated and then the dietary intake ,the percentage of energy from protein ,fat and carbohydrate and the percentage of protein resource were calculated based on the food composition table .Results It was shown that the daily energy intake of one meal was appropriate among the surveyed hotel customers ,and the energy proportion from 3 major nutrients were not reasonable ,and the energy proportion from fat were higher than other 2 major nutrients ;the proportion from the source of good proteins was reasonable ,but the intake of protein was too much;the customers fell short on their daily serving of grains ,eggs ,beans and milk products and the intake of cooking oil and salt was excessive in diets . Their intake of fat ,protein ,retinol equivalent ,niacin ,vitamin E ,sodium and iron were too much .Instead ,they ate less ascorbic acid;only the intake of energy ,riboflavin and thiamine were appropriate .Conclusion The one meal dietary structure of hotel customers is not appropriate ,while nutrition collocation is not reasonable .It is necessary to promote nutrition and health education and some intervention measures must be taken .
2.Effects of RNA Interfering of MBP-1 on Proliferation of Saos-2 Cell Line

Objective To investigate the effects of c?myc promoter binding protein 1(MBP?1)gene on the proliferation of human Saos?2 osteo?sarcoma cells in vitro. Methods Saos?2 cells were divided into three groups:blank control group(untransfected cells),negative group(cells transfected with missense sequence)and experimental group(cells transfected with MBP?1 shRNA). Two MBP?1 shRNA sequences and one neg?ative control shRNA sequence were designed ,synthesized and cloned into pSIREN?retroQ plasma. Then the recombinant plasmids were construct?ed and transfected into human Saos?2 osteosarcoma cells by Lipofectamine 2000. The expressions of MBP?1 mRNA and protein in Saos?2 cells were detected by real?time PCR and Western blot ,respectively. The effects of altered expression of MBP?1 on cell proliferation were measured by CCK?8 cell proliferation assay. The expressions of cyclin D1 and cyclin E in Saos?2 were determined by Western blot. Results PCR and sequenc?ing results indicated that the recombinant plasmids pSIREN?retroQ was constructed. The relative expression level of MBP?1 mRNA in the MBP?1 siRNA transfection group was significantly decreased than that in blank control group(P<0.05). Compared with the blank control group,the ex?pression levels of MBP?1 protein in the experimental group also significantly decreased. The proliferation abilities of Saos?2 cells at 48,72,and 96 hours after MBP?1 siRNA transfection were significantly increased than those in the blank control group(P<0.05). Compared with the blank con?trol group,the expression levels of cyclin D1 and cyclin E protein in the experimental group also significantly increased(P<0.05). Conclusion Knockdown of the expression of MBP?1 gene promotes the proliferation of human Saos?2 osteosarcoma cells. MBP?1 gene may become the new tar?get of gene therapy for osteosarcoma.
3.Progression of early urine markers in diabetic nephropathy

Diabetic nephropathy (DN) is one of the major microvascular complications in diabetic patients, and it is also the main global cause of chronic kidney disease(CKD) and end-stage renal disease(ESRD). Albuminuria and estimated glomerular filtration rate (eGFR) are currently recognized as clinically diagnosed early DN, but their sensitivity and specificity are poor. Therefore, it is very important to find new biomarkers to reflect the potential diabetic nephropathy in the clinical silent period.
4.The analgesic mechanism of exosomes derived from umbilical cord mesenchymal stem cells in rats modelling osteoarthritis

Objective:To observe any effect of exosomes derived from umbilical cord mesenchymal stem cells on pain, cartilage repair and the expression of transcriptional activator 3 (ATF-3) and growth related protein 43 (GAP-43) in the dorsal root ganglia (DRG), as well as to explore the mechanism of their relieving pain.Methods:Fifty-four male Sprague-Dawley rats were randomly divided into a sham-operation group, a monoiodoacetate group and an exosome group, each of 18. The knee cavities of the left hind limbs of all of the rats except those in the sham-operation group were injected with 50μl of monoiodoacetate to establish an arthritis pain model. The sham-operation group received only 50μl of saline solution as controls. Two weeks after the modelling, the knee joint cavities of the exosome group were injected with 50μl of exosomes, while the other two groups were injected with 50μl of normal saline. The rats′ mechanical and thermal pain thresholds were measured 1 day before the modeling, 7 and 14 days after the monoiodoacetate injection, as well as 7, 14 and 28 days after the exosome injection. Western blotting was used to detect the expression of ATF-3 and GAP-43 in the rats′ DRG, while hematoxylin and eosin staining was used to detect any cartilage repair.Results:Compared with the monoiodoacetate group, the latency of the mechanical and thermal pain thresholds had increased significantly in the exosome group 7 days after the exosome injection. The difference remained significant until the 28th day after the injection. The expression of ATF-3 protein decreased significantly and that of the GAP-43 protein increased significantly. Significant differences were observed in the average Osteoarthritis Research Society International (OARSI) knee cartilage score.Conclusions:Exosomes can alleviate the pain induced by monoiodoacetate adjuvant. The analgesic mechanism may be related to reducing nerve injury and promoting nerve and cartilage repair, with the nerve repair earlier than cartilage repair.
5.Cultivation of college students' innovative and entrepreneurial thinking and ability based on Synthetic Biology and iGEM.

Synthetic Biology is one of the most promising fields of modern Biology and a frontier interdisciplinary subject in the 21st century. With the rapid development of synthetic biology, the International Genetically Engineered Machine (iGEM) competition has emerged. The iGEM competition, based on the subject foundation of Synthetic Biology, intends to solve the biological problems in our daily life by applying modern biological technology. In recent years, with the continuous increase of participating teams, the iGEM competition has received extensive attention and achieved great progress. On the basis of the development of Synthetic Biology, we analyzed the 2018-2020 award-winning projects of the iGEM competition and illustrated the role and significance of the iGEM competition in cultivating college students' innovative thinking and ability with the participation experience of the iGEM team of Southwest Jiaotong University as an example.
