1.The Effect of Theophylline on Eosinophil Apoptosis

Objective To vie the effect of theophylline on eosinophil (Eos) apoptosis. Methods EDTA-anticoagulated venous blood was obtained from 15 healthy volunteers and pre-incubated with saline or theophylline (10 -4 M) or dexamethasone and rhIL-5(10 -8 M),respectively.The mRNA of bcl-2 and bax were determined by reverse transcriptase polymerase chain reaction(RT-PCR),and their cytoplasm proteins were determined by flo cytometry. Results Analysis revealed that the bax protein express intracellular without any stimulator,but not bcl-2 in protein and mRNA levels.And culturing Eos in the presence of 10 -8 M IL-5 for 24 hours up-regulating bcl-2 mRNA ( P
2.Immunohistochemical study of the synaptophysin in hirschsprung’s disease

Objective:To study the neuromuscular junction of the colon in patients with Hirschsprung’s disease by immunohistochemical methods.Methods:The distribution of the neuromuscular junction in the muscle layers of bowel from 10 normal controls and 20 Hirschsprung’s disease patients was studied by synaptophysin immunohistochemistry.Antimouse synaptophysin serum was used for visualization of neuromuscular junction.Results:In the colon of the control cases,synaptophysin-immunoreactivity was widely distributed in the circular muscle layer.In the dilated colon with Hirschsprung’s disease cases,synaptophysin-immunoreactivity was moderately distributed within normal ganglion cell and scarcely in constricted segment within degenerated ganglion cell.In contrast to the high degree of synaptophysin-immunoreactivity in the controls and dilated colon,the synaptophysin-immunoreactivity in the constricted segment were nil in the circular muscle layer.Conclutions:Dense synaptophysin-immunoreactivity nerve endings were prominent in the circular muscle layer in the control and there was almost no synaptophysin imm-unoreactivity activity in the circular muscle layer in the constricted segment.The circular muscle layer from the constricted segment is confirmed to be a region with poor innervation including extrinsic and intrinsic nerve. Based on the Cannon’s Law of Denervation,the muscles that are supplied by these nerves shorten and tighten,resulting in muscle spasm and stiffness.The constricted segment which was denervated,is incapable of transmitting normal propulsive peristaltic waves and therefore acts as a physiologic obstruction.
3.The protective effect of sufentanil postconditioning on myocardial ischemia-reperfusion injury in rats

Objective To evaluate the effect of sufentanil postconditioning on myocardial ischemia-reperfusion injury in rats, and to investigate the possible mechanism. Methods Thirty male Sprague-Dawley ratswere randomly divided into 5 groups (n = 6 each): sham group (group S), I/R group (group I/R), low dose of sufentanil postconditioning group (group SL), high dose of sufentanil postconditioning group (group SH) andsufentanil postconditioning plus wortmannin group (group WM). Different drugs were injected before reperfusion: normal saline (NS) and sufentanil 1 μg/kg in group SL, NS and sufentanil 3 μg /kg in group SH, wortmannin 15 μg/kg andsufentanil 1 μg/kgin group WM, and NS in group S and I/R. At the end of reperfusion, artery blood was collected for assessment of plasma cTnI concentration; And the heart was harvested for determination of infarct size (IS) and area at risk (AAR). Results Compared with group S, cTnI concentration was increasedin the rest groups (P< 0.01); Compared with group I/R, cTnI concentration was decreased in group SL, SH and WM, while IS/AAR reduced in group SL and SH (P < 0.01); Compared with group SL, cTnI concentration and IS/AAR were decreased in group SH while increased in group WM (P <0.01). Conclusion Sufentanil postconditioning could attenuate myocardial ischemia-reperfusion injury in rats, and the mechanism involved PI3K related pathway.
4.Survey on Two-week Prevalence and Ethical Analysis in Rural Preschool Leftover Children in Shandong Province

Objective:To understand the status of two -week prevalence and the influencing factors in rural preschool leftover children of Shandong , so as to provide evidence for putting forward effective health intervention means .Methods:Using the method of stratified -cluster-random sampling and elect 646 cases of preschool lefto-ver children from 2 counties of Shandong .We surveyed with self -made questionnaire .Results:The two-week prevalence of the rural preschool leftover children is high;the influencing factors of the two -week prevalence with the rural preschool leftover children including whether children are only -child, the type of children guardianship , whether children piddle and whether left -behind children can avoid dangerous .Conclusion: The two -week prevalence present situation of rural preschool left -behind children is low , and its problems of health and ethics are clear.Therefore, effective measures should be carried out to improve the health level of rural leftover children .
5.Eukaryotic expression and identification of human pSecTag2B-CD226 fusion protein

Objective: To construct and express the eukaryotic expression vector of human pSecTag2B-CD226(PTA1).Methods: The gene fragment encoding extracellular region of human CD226 was cloned into the eukaryotic expression vector pSecTag2B.After sequencing,the vector was transfected into COS-7 cells,and the expressed molecule was purified by affinity chromatography.Finally,the product was characterized by ELISA.Results: hCD226-6His was successfully expressed.After purification,the concentration of hCD226-6His was 50?g/ml.Conclusion: Human CD226-6His fusion protein involving the extracellular region of CD226cDNA has been successfully expressed and purified,which helps prepare the ground for further functional studies of this molecule.
6.The effect of intrauterine injection of rat fetal hepatocytes on rat homograft rejection reaction

Aim To investigate the effect of intrauterine injection of rat fetal hepatocytes on homograft rejection reaction. Methods The skin graft from a male LOU/CN rat was transplanted to a female recipient CHN rat of 7 to 9 weeks after parturition, and then survival time of the graft was observed. Simultaneously, homograft rejection reaction was examined by mixture lymphocyte culture. Results As compared with control group, survival time of transplanted skin graft was obviously prolonged. Mixure lymphocyte culture demonstrated that homograft rejection reaction was inhibited markedly. Conclusion Intrauterine injection of rat fetal hepatocytes wuld obviously inhibite homograft rejection reaction, thus prolonging survival time of the graft.
7.Molecular cloning of mouse CD226(PTA1) and its isoforms

Objective:To clone mouse CD226(platelet and T cell activation antigen1,PTA1).Methods:Specific primers were designed and sythesized according to the EST sequence from GenBank,which had 51% homology to human CD226 on the amino acid level.And then,the cDNA of mouse CD226 was cloned from the thymus of 4 week old BALB/c mouse by using RACE technique.Results:The length of mouse CD226 cDNA is 2 223 bp,with the open reading frame(ORF) of 1 002 bp,encoding 333 amino acids,which is 3 amino acids shorter than its counterpart in human.The mouse CD226 belongs to IgSF,and shares 53% homology with human PTA1 on amino acid level.Besides,three isoforms of mouse PTA1 were also cloned at the mean time.Conclusion:The molecular cloning of mouse PTA1 lays the foundation for the in vivo studies on the biological function of this molecule,as well as the studies of this molecule in gene knockout mouse and transgenic mouse.
8.Pedicle flap of nasal septum-basis nasi and temporal muscucofascial flap to repair nasoseptal perforation.

OBJECTIVE:
To explore curative effect with pedicle flap of nasal septum-basis nasi and temporal muscucofascial flap to repair nasal septal perforation.
METHOD:
Dissecting mucoperichondrium and mucoperioseptum around the perforation and taking dowm and out xia-ward to the floor of nasal cavity to make a inferior extremity pedicle flap. Then,the flap was tumbled and sutured onto raw surface of contralateral side through perforation. Reapplicating autoallergic temporal musculofascial flap to repair another side perforation.
RESULT:
Repairing perforation Sin twelve cases were sucessfully healed in endoscope.
CONCLUSION
The pedicle flap of nasal septum-basis nasi and temporal muscucofascial flap is easy to acquire and no rejection. The flap has good blood supplying, high survival rate and provides adequate transplantating materail to repair comparatively large perforation.

9.Identification of the Peptides that Bind to PTA1 mAb(LeoA1) by Phage Displayed Library

Aim To screen peptides binding specifically to anti-human PTA1mAbs from a random twelve-peptide phage-disp-layed library. Methods Series of PTA1mAbs(LeoA1、 1B11、 C9、 2D1、 2E9、 2G8、 2H2 and E8)were purified using protein-A affinity column. PTA1mAbs which could bind PTA1-Fc fusion protein binding to its ligand were confirmed by flow cytometry, and then used as target to screen phage library. After three rounds of affinity screening, the peptide sequences of positive phage clones were determined and analyzed. Results LeoA1 could block PTA1-Fc fusion protein binding to its ligand. 13 phages which could bind specifically to LeoA1 were isolated from phage library and further confirmed by ELISA. Conserved motifs were found among the sequences of the peptides. Conclusion It was shown that the conserved motifs were candidafe regions binding to PTA1 ligand,which is important to identify functional epitopes for seeking ligand of PTA1 and further investigation of biological function of PTA1.
10.Transumbilical laparotomy: a cosmetic and minimally invasive operative route in pediatric patients

Objective:The feasibility of a minimally invasive cosmetic transbilical laparotomy was studied in fifty-seven pediatric patients.The advantages of this technique will be discussed.Methods:The circumferential incision was made following the ring fold of the right umblilicus,which was less than 2700.The peritoneum was opened in the midline via rectus abdominis.Intra-peritoneal lesions were detected and explored.The surgical procedures were able to be carried out either inside or outside of the peritoneum.After closure of peritoneum,the umbilical incision was closed subcutaneously with an absorbable suture.Results:From January 2001 to January 2005,58 procedures via transumbilical route have been done in 57 pediatric patients.None of those cases required conversion to a traditional abdominal incision.The duration of surgery was from 20 to 170 minutes.All patients were discharged five days postoperatively and follow-up was conducted from 3 months to 4 years.No complications were observed in all but one patient.This patient had a leaking anastamosis.No visible scars were present and the cosmetic results were excellent.All patients had shorter hospital stay and lower operating cost.Conclusion:Transumbilical approach is a feasible,safe,economical,minimally invasive procedure with excellent cosmetic results which appears to be superior to traditional procedures.It suggests that the transumbilical route should be widely used clinically and might be applicable for elder children and possibly would be of benefit in the treatment of other conditions.
