Objective To investigate the association of polymorphisms in dopamine D3 receptor (DRD3) gene exon 1 Ser9Gly with childhood tourette syndrome and efficacy of aripiprazole in Chinese Han population.Methods Polymorphisms of Ser9Gly in DRD3 gene was genotyped by using Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP) in 158 tourette syndrome patients (study group) and 187 healthy controls (control group).Patients in study group were assessed with Yale global tic severity scale(YGTSS) at baseline and at the end of the 8th week of treatment to evaluate the efficacy of aripiprazole.Results (1) There were no significant differences in genotypic frequency and allelic frequency of Ser9Gly in DRD3 gene between study group and control group (P＞0.05).(2) Significant difference was found in frequency of Ser9Gly in DRD3 gene between effective and ineffective groups (genotype:Ser/Ser 40 vs 17;Ser/Gly 52 vs 28;Gly/Gly 7 vs 14;alle:Ser 132 vs 62;Gly 66 vs 56)(P＜0.05).(3) There were no significant differences in distribution of genotypes and alleles of Ser9Gly among three-type of tourette syndrome (P＞0.05).Conclusion The polymorphisms of Ser9Gly in DRD3 gene are associated with the efficacy of aripiprazole.

Objective To explore the influence of olanzapine combined with risperidone on the serum level of homocysteine (Hcy) in patients with schizophrenia.Methods Participants included sixty patients diagnosed with schizophrenia,patients were divided into two groups by means of random digital method.The group A was given olanzapine treatment,and the B group was given olanzapine combined with risperidone therapy.After two months follow-up,the curative effect and the level of Hcy were observed.Results Before treatment,Hcy level [(24.4 ± 6.1)μmol/L,(25.8 ± 7.7)μmol/L] and HH number (22 cases,23 cases) in the two groups had no obvious differences (t =1.11,x2 =1.34,all P ＞ 0.05) ;2 months after treatment,Hcy level (11.6 ± 8.6) μmol/L and HH number(16 cases) in group B were significantly lower than those in the group A [(16.3 ± 3.8) μmol/L,8 cases],the differences were statistically significant (t =4.65,x2 =4.32,all P ＜ 0.05).Conclusion Olanzapine combined with risperidone in treatment of schizophrenia have good curative effect,it can improve the symptoms of patients,decrease serum Hcy level and HH number.

Aim A method of coculture of brain capillary endothelial cells (BCECs) and astrocytes of rats was used to evaluate nanoparticle' s blood-brain barrier (BBB) transcytosis and toxicity at the endothelial tight junction. Methods A lipophilic fluorescent probe, 6-coumarin, was incorporated in poly(ethyleneglycol)-poly (lactide) nanoparticle using double emulsion/solvent evaporation method. BCECs and astrocytes were firstly isolated from brain of newborn rats and characterized by their morphology and immunocytochemistry staining, separately. Subsequently, a coculture model with BCECs on the top of micro-porous membrane of cell culture insert and astrocytes on the bottom side was established. The permeability of 14C-labeled sucrose and nanoparticle were determined, separately. Results The meanweight-based diameter of 6-coumarin loaded nanoparticles was ( 102.4 ± 6.8) nm, with zeta potential of ( - 16.81 ± 1.05) mV. BCECs were positive for factor Ⅷ staining and glial fibrillary acidic protein was tight junction between BCECs in the coculture model could be visualized by both scanning electron microscopy and transmission electron microscopy. The unchanged paracellular transport of sucrose proved vivo situation for examination of the permeability of nanoparticle and toxicity evaluation.

Objective To investigate the effects of mobility training on mobility and the mRNA levels of both synaptophysin and growth associated protein 43 (GAP-43) in the region around an infarction in rats with acute cerebral infarction. Methods Models of cerebral infarction were created in 100 rats through middle cerebral artery occlusion. They were then randomly divided into training and control groups. The motor skill of the rats was examined using a beam walking test. The mRNA levels of both synaptophysin and GAP-43 in the region around the infarction were observed at the 1st, 3rd, 7th, 14th and 28th days after model-creation using a semi-quantitive reverse transcrip-tion polymerase chain reaction. Results The rats' mobility scores increased with training, and significant differ-ences were observed between the average scores of the two groups at the 3rd, 7th and 14th days. The scores were higher in the training group. The mRNA levels of both synaptophysin and GAP-43 in the region around the infarction increased significantly from the 1st to the 3rd and 7th days. Synaptophysin mRNA levels were significantly higher in the trained group at each time point, but the levels of GAP-43 mRNA were significantly higher in the trained group only on the 3rd and 7th days. Conclusions Motor skill and the mRNA levels of synaptophysin and GAP-43 in the region around an infarction can be increased by motor skill training, at least in rats with model acute cerebral infarc-tion.

Objective To understand the cross-resistance of Culex pipiens pallens to common pesticides,so as to provide the evidence for improving the application of chemical pesticides. Methods The IV instar larvae of DDVP-resistant,propoxur-resistant and cypermethrin-resistant strains as well as the sensitive strain of Culex pipiens pallens were collected to detect the re-sistance to DDVP,propoxur and cypermethrin based on the WHO bioassay method. Results The resistance coefficients of DDVP-resistant strain to DDVP,propoxur and cypermethrin were 14.47,8.96 and 207.27 respectively. The resistance coeffi-cients of propoxur-resistant strain to DDVP,propoxur and cypermethrin were 3.27,6.93 and 8.65 respectively. The resistance coefficients of cypermethrin-resistant strain to DDVP,propoxur and cypermethrin were 2.93,1.61 and 501.11 respectively. Con-clusion The resistance and cross-resistance could be generated during the long-term application of a single kind of chemical pesticide,and we should pay more attention to the varieties and dosages of them.
