1.Antioxidant Compounds and Radical Scavenging Property of Fructus Xanthii

Objective To isolate and purify the antioxidant compounds from fructus xanthii and to study their antioxidant activity.Methods Compounds were isolated by repeated silica gel,macroreticular resin Diaion HP-20 and Sephadex LH-20 column chromatography and their antioxidant property was determined by scavenging activity to DPPH and ?OH.Results Five antioxidant compounds were isolated from the ethyl acetate extracts of Xanthium sibiricum fruits and their structures were identified as 5,7,3',4'-tetramethoxyisoflavone,myricetin-3'-methyl ether,falcalindiol,1,8-heptadecadiene-4,6-diyne-3,10-diol and 3,4-dihydroxybenzoic acid.Among them,3,4-dihydroxybenzoic acid had the highest radical scavenging activity.Conclusion All above 5 compounds have high antioxidant activities,and antioxidant activity of 3,4-dihydroxybenzoic acid is the highest.
2.Application of Accelerated Solvent Extraction(ASE) for the Extraction of Compounds in Fructus Xanthii

Objective To establish a method for extracting compounds in Fructus Xanthii by accelerated solvent extraction(ASE),and to simultaneously determine the compounds by GC-MS.Methods The extraction was performed by ASE in which the conditions were optimized by orthogonal test,then the compounds in different extracts with different polarity were determined by GC-MS.Results The optimized conditions of ASE for compounds in Xanthium sibiricum fruits were as follows: petroleum ether,ethyl acetate and methanol as extracting solvent in turn in 33 mL extracting tube and the volume of solvent being 65 mL;6 g sample,9 MPa of extracting pressure,90 ℃of extracting temperature,5 min of extracting time,two cycles and one time for each extraction.The GC-MS conditions were as follows: 1 ?L extract was analysed by GC-MS(Shimadzu Model QP-2010) instrument fitted with a flame ionisation detector,separation was achieved on a DB-5ms,DB-17ms and DB-WAX column(J&W Scientific,30 m,0.25 ?m id film thickness,1.4 ?m,USA) respectively,the column temperature was maintained at 60 ℃for 3 min and increased to 250 ℃,the scanning range was from m/z 40 to m/z 700(scanning interval: 0.5 s),and the compounds were identified by searching NIST and Wiley registry of mass spectral data.Conclusion There are 61 compounds found in the petroleum ether extract and in which 48 are identified,22 in the ethyl acetate extract and 18 identified,as well as 34 in the methanol extract and 29 identified with the methods of optimized ASE combined with GC-MS.
3.Regulatory effect of hydrogen sulfide on adiponectin secretion in fatty liver adipocytes

Objective To observe whether hydrogen sulfide modulates adipocytes to secrete adiponectin.Methods Thirty-two male SD rats were randomly divided into normal diet group (n =8),high-fat diet group (n =8),high-fat + cysteine group (n =8),and high-fat + propargylglycine (cystathionine-γ-lyase inhibitor) group (n =8).The fatty liver animal model was established using the high fat diet; meanwhile,L-cysteine was applied to the animals to produce endogenous hydrogen sulfide and propargylglycine to inhibit the production of endogenous hydrogen sulfide.Fatty degeneration of the hver of animals and plasma within the adiponectin concentration was observed after 8 weeks.The omental adipose tissues were excluded and digested using collagenase Ⅰ to isolate fat cells.Sodium hydrosulfide and propargylglycine was applied to stimulate the cells in vitro for 7 days before the detection of the cell supernatant adiponectin content.Results Liver steatosis was established after highfat diet for eight weeks.Steatosis was significantly extenuated after the application of L-cysteine and aggravated by propargylglycine.Plasma hydrogen sulfide and adiponectin levels in fat diet group animals were (21.13 ± 7.06) mmol/L and (3.16 ± 1.15) mg/L respectively,which significantly decreased when compared with the normal diet group with the levels of hydrogen sulfide (29.13 ± 13.06) mmol/L and adiponectin (8.98 ± 2.84) mg/L (P=0.0229,P=0.0062).Hydrogen sulfide [(35.47 ±9.04) mmol/L] and adiponectin [(6.54 ± 1.38) mg/L] levels in the high fat + cysteine group significantly increased (P =0.0032,P =0.0131).However,hydrogen sulfide [(16.65 ±8.79) mmol/L] and adiponectin [(2.50±0.91) mg/L] in high fat + propargylglycine group were significantly decreased (P =0.0191,P =0.0021).Hydrogen sulfide was not significantly differem in the supematam of adipocyte isolated from normal diet group [(26.77 ± 12.65) mmol/L] and from high-fat diet group [(28.76 ±9.09) mmol/L] (P =0.0927),but that in the sodium hydrosulfide and propargylglycine ceils supematants significandy either increased or decreased (P =0.0000,P =0.0000).Adiponectin in the supematants of high-fat diet group and normal diet group adipocyte showed no significant difference [(4.21 ± 1.61) mg/L vs (4.49 ± 1.09) mg/L,P =0.1076)].The levels of adiponectin in sodium hydrosulfide and propargylglycine cell supernatants increased and decreased respectively than that in higher fat diet group cells [(5.77 ±0.86) mg/L vs (4.21 ±1.61) mg/L,P =0.0094; (3.01 ±1.26) mg/L vs (4.21 ± 1.61) mg/L,P =0.0108].Conclusion Hydrogen sulfide promotes adipocytes to secrete adiponectin.
4.Observations on the Efficacy of Vastus Medialis Oblique Enhancement plus Electroacupuncture Intervention in Treating Patellar Chondromalacia

ObjectiveTo investigate the clinical efficacy of vastus medialisoblique enhancement plus electroacupuncture intervention in treating patellar chondromalacia.MethodFifty-two patients meeting the inclusion criteria were randomly allocated to a treatment group of 32 cases and a control group of 20 cases. The treatmentgroup received vastus medialis oblique enhancement plus electroacupuncture intervention and the control group, electroacupuncture intervention alone. Both groups were treated once daily, 10 times as a course, for four consecutive courses. The Visual Analogue Scale (VAS) score was counted in the two groups before and after treatment. The clinical therapeutic effects were evaluated in the two groups.ResultThe VAS score improved significantly in both groups of patients after treatment compared with before(P<0.05). It improved significantly more in the treatment group than in the control group (P<0.05). The cure rate and the total efficacy rate were 37.5% and 96.9%, respectively, in the treatment group, which were higher than a cure rate of 20.0% and a total efficacy rate of 75.0% in the control group, respectively. ConclusionVastus medialis oblique enhancement plus electroacupuncture intervention can improve the clinical symptom of pain more markedly and heighten the clinical therapeutic effect in patients with patellar chondromalacia. Its effect is better than that of electroacupuncture intervention alone.
5.The detection of Rotavirus and Norwalkvirus in stool and cerebrospinal fluid of children with infantile convulsions associated with mild gastroenteritis

Objective To explore the common pathogen of infantile convulsions associated with mild gastroenteritis, and to study the differences between the seizures caused by the two kinds of virus.Methods RT-PCR was used to detect Rotavirus (RV) and Norwalkvirus (NoV) in stool and cerebrospinal fluid of 30 cases with infantile convulsions associated with mild gastroenteritis. The differences between the frequency of seizures caused by two kinds of virus were analyzed by statistical methods (two-sample t-test).Results 17/30 (56. 7%) were RV-positive in stool and 3/17 (17. 7%) in cerebrospinal fluid; 6/30 (25.0%) were NoV-positive in stool and 1/6 (16. 7%) in cerebrospinal fluid. The seizure frequency with NoV infection was (4. 33 ± 1.75) times, and RV infection patients was (2. 53 ± 1.12) times (P < 0. 01).The seizure frequency of CSF virus-positive children was (4. 75 ± 1.71) times compared to (2. 63 ± 1.21)times in virus-negative children (P < 0.01). Conclusion The common pathogens causing infantile convulsions associated with mild gastroenteritis were RV and NoV. The degree of NoV infection affecting the central nervous system may be greater than RV. The presence of the virus in cerebrospinal fluid may lead to higher incidence of seizures,but their exact roles related to the occurrence of seizures remain to be further studied.
6.Clinical Application of Myeloperoxidase in Diagnosis and Prognosis of Non-ST Elevation Acute Cornary Syndromes

Objective To evaluate the diagnostic value of myeloperoxidase (MPO)in diagnosis of non-ST elevation acute cor-onary syndromes (NSTE ACS).Methods The MPO was determined in 61 unstable angina pectoris (UAP)patients and 54 non-ST-elevation myocardial infarction (NSTE MI)patients,then analyzed the data with receiver operationg characteristic curve (ROC curve)statistical software.The major adverse cardiac events (MACE)were obseved in 106 NSTE ACS patients through 1 year of follow-up.Results The MPO values of NSTE MI group and UAP group were 672±418 ng/ml and 459± 328 ng/ml,the difference was statistically significant (t=3.928,P <0.001).The cTnI values were 558±261 pg/ml and 405 ±278 pg/ml of respectively.The difference was statistically significant (t = 3.089,P = 0.003).The area under the ROC curve of MPO was 0.726 when the optimal cutoff value was 382 ng/ml in the diagnosis of NSTE MI.The sensitivity and specificity was 92.6% and 39.8%.The area under the ROC curve of cTnI was 0.799 when the optimal cutoff value was 484 ng/ml in the diagnosis of NSTE MI.The sensitivity and specificity was 65.5% and 92.3%.High MPO maintained a strong association with the risk of major adverse cardiac events through 1 year of follow-up.Conclussion MPO is ahigh sensitivity marker of the NSTE MI and of clinical value for prognosis of NSTE ACS.
7.Combined measurement of H-FABP and MPO in the prognosis of non-ST elevation acute cornary syndromes

Objective To evaluate the prognosis value of heart‐type fatty‐acid binding protein(H‐FABP)and myeloperoxidase (MPO) in non‐ST elevation acute coronary syndromes(NSTE ACS) .Methods 181 NSTE ACS patients were divided into 4 groups according to the level of H‐FABP and MPO ,and the baselines of 4 groups such as gender ,age ,hypertension ,smoking ,body mass in‐dex ,diabetes mellitus ,hyperlipidemia were compared .The incidences of adverse cardiac events in 4 groups were compared after a 2 years′follow‐up .Results By multivariate COX regression adjustment for other risk factors ,the relative risk(RR) of H‐FABP for adverse cardiac events was 2 .023(95% CI:1 .029 -3 .987 ,P=0 .002) ,and MPO was (95% CI:2 .196 -5 .325 ,P< 0 .05) .The 2 years′follow‐up showed the incidence of adverse cardiac events in NSTE ACS patients with higher H‐FABP and MPO levels was higher than the NSTE ACS patients with one or two indicators of normal levels .Conclusion The combined application of H‐FABP and MPO could has clinical significance for the prognosis of patients with NSTE ACS .

Calcium oxalate (CaOx) stone is the main type, and its formation is closely related to the metabolism of oxalic acid and calcium. Gut Microbiome is normal microflora which settled in the human intestinal tract and plays an important role in regulating a variety of metabolism in the body. In the past, Oxalobacter formigenes in gut was a protective factor for the formation of CaOx stones. Recently, it has been found that the bacteria regulating oxalate metabolism were not limited to Oxalobacter formigenes. Gut Microbiome of CaOx stones formers is different from healthy people. It regulates the metabolism of oxalic acid in the body through the gut-kidney axis and affect the formation of CaOx stone. The purpose of this study is to describe the characteristics of intestinal flora in patients with CaOx stones, and to summarize its potential function in the formation of CaOx stones and its possible clinical application in the future.
9.Relationship between resting heart rate and carotid artery structure in young hypertensive patients

Objective To investigate the relationship between resting heart rate (RHR)and carotid artery structure in young hypertensive patients.Methods A total of 663 primary hypertensive patients aged between 18 and 45 (38.01 ± 5.78) were chosen from the First Affiliated Hospital of Xinjiang Medical University from January,2009 to January,2012.Patients under this study were divided into three groups according to their RHRs:group A (RHR<70 bpm,n=163),group B(70 bpm≤RHR<80 bpm,n=268),group C (RHR≥80 bpm,n=232).Data on bilateral carotid artery intima-media thickness,common carotid artery diameter were detected by carotid ultra-sonography.Indexes of carotid artery were analyzed by both variance and bi-variate correlation analysis.Results 1)Differences in the blood pressure among three groups showed statistical significance (P<0.01),with diastolic pressure in group A higher than in group B (P<0.05),while both systolic and diastolic pressure in group C higher than in group B (P<0.01).2)In group A,the right carotid artery diameter was higher than in group C(P<0.05) and the left carotid artery diameter higher than in group B(P< 0.05) with the left carotid artery diameter higher than in group C (P<0.05).3) Significant positive correlations were found between RHR and blood pressure (r =0.112,0.118) and negative correlation between RHR and left carotid artery diameter (r=-0.89).However,correlation between RHR and right carotid artery diameter was not significant (r=-0.075,P=0.068).Conclusion When blood pressure had an increase,we found that the carotid artery diameter was decreasing along with the increasing RHR among the young hypertensive patients.
10.Laminin may be a predictor of recurrence after liver transplantation for HBV -related hepatocellular carcinoma patients

Objective To explore the prognostic role of laminin (LN ) as a tumor biological marker in predicting the recurrence of hepatocellular carcinoma (HCC) related with HBV infection after liver transplantation (LT ) .Methods Tissue samples from 251 HBV-related HCC patients undergoing LT were immunohistochemically stained with anti-LN antibody .The relevant prognostic factors were analyzed using Spearman's rank test , Kaplan-Meier method , log-rank test and multivariate step-wise Cox regression analysis .Results The expressions of LN in tumor tissues were significantly positively correlated with tumor number (P=0 .00) ,microsatellite (P=0 .02) ,venous invasion (P=0 .048) ,pTNM tumor stage (P=0 .00) ,pre-LT serum α-fetoprotein (AFP) level (P=0 .00) ,HBV DNA level (P= 0 .02) ,HBeAg level (P= 0 .02) and tumor recurrence (P= 0 .00) respectively .Significant differences existed in 1/3/5-year overall survival or tumor recurrence-free survival rate post-LT among LN different expression (-,+ ,≥ + + ) in HBV-related HCC patients (P< 0 .05 ) . Multivariate analysis indicated that LN was a significantly independent predictor in predicting poor tumor recurrence-free survival post-LT (P=0 .01) .Conclusions LN may be a feasible marker in predicting HCC recurrence post-LT for HBV-related HCC patients .
