1.Investigation of the polymorphism of HPA-1-17 gene and the platelet transfusion refractoriness in Li population on Hainan island,in China

Objective To investigate the allele polymorphism and characteristic of HPA-1-17 genotyping of Li ethnic on Hainan island,and to evaluate the HPA mismatched ratio between donor and recipient in randomly chosen cases of blood transfusion,in order to investigate the platelet transfusion refractoriness in Li population.Methods Genotyping tests of A-1—17 allele of the blood sample from 180 Lis were conducted with PCR-SSP.Results A total of 180 blood samples were genotyped successfully in HPA genetic system,among which 0.9972 were HPA-2a,0.0028 were HPA-2b,0.4889 were HPA-3a,0.5111 were HPA-3b,0.9667 were HPA-5a,0.0333were HPA-5b,0.9972 were HPA-6a,00028 were HPA-6b,0.4250 were HPA-15a,0.05750 were HPA-15b,and they all showed polymorphism. But polymorphism was not detected out in other allelic genes,such as HPA-1,-4,-7—14,-16,and -17. The bb homozygous were detected in two alleles,among which 0.2834 was HPA-3 and 0.3667 was HPA-15,but it wasn't detected in the others systems. In the randomly chosen cases of blood transfusion,the mismatch rate of HPA-3 ,HPA-15 and HPA-5 was 37.49%,36.93% and 6.23%,respectively.Conclusion This research revealed the distribution of HPA gene in Li population on Hainan island,and found the allelic frequency and thier characteristics. According to our findings,HPA-2,HPA-3,HPA-5 and HPA-15 genes between donor and recipient should be matched in order to avoid platelet transfusion refractoriness.
2.Recent progress in the study of reduction-sensitive drug carriers.

With rapid and efficient drug release, few side effects and excellent biodegradable properties, the reduction-sensitive carriers is not only the new hot point in the field of pharmaceutical research, but also the most promising intelligent drug carrier on clinical application. This paper reviews the latest research of reduction-sensitive drug and gene carriers, including the mechanisms of drug release and the synthesis of the reduction-sensitive conjugates, reduction-sensitive nano polymer micelles, nano vesicles, nano hollow microspheres, nano liposomes, as well as the characteristics and advantages of various kinds of carrier system. It will provide a theoretical basis for its further application.
3.Establishment of Thromboelastography reference interval for healthy adults in Hainan

【Objective】 To establish the reference interval of Thromboelastography(TEG)of healthy adult in Hainan, compare with the interval provided by the manufacturers, and analyze the influencing factors. 【Methods】 A total of 308 healthy adult volunteers were included according to the inclusion and exclusion criteria. The TEG was analyzed based on adequate quality control, and the reference interval of each parameter was calculated with 95% CI. 【Results】 The reference interval of these volunteers were R 4.27~9.20 min, K 1.60~3.83 min, Angle 44.22°~67.78°, MA 47.82~64.17 mm, CI -5.63~1.12 and G 4.58~9.03. 14.94% (46/308) of these volunteers had at least one index exceeded the reference interval provided by manufacturers. A total of 74 healthy volunteers were diagnosed with coagulation disorder, with the specificity at 79.22%. Significant differences of R, K, Angle, MA, CI, G were observed between males and females (P<0.05). Hypercoagulability was not associated with the age. 【Conclusion】 This study established a reference interval of TEG of healthy adults in Hainan, which provided reference for related clinical and basic science study.
4.Allelic polymorphism of 13 rare blood group systems in Li Ethnic Group in Hainan

【Objective】 To detect alleles of rare blood group systems of red blood cells (RBC) of Duffy, Diego, Kidd, Dombrock, MNS, Lutheran, Kell, Colton, Scianna, YT, Knops, Indian and Vel of Li Ethnic Group in Hainan, so as to investigate the distribution characteristics of rare blood group alleles and provide references for safe blood transfusion. 【Methods】 The genotypes of 13 rare blood group systems in 300 Li people in Hainan were analyzed by PCR-SSP, and the gene frequency was analyzed . 【Results】 The allele frequencies of Duffy, Diego, Kidd, Dombrock, MNS, and Lutheran blood group systems of 300 Li people were as follows: Duffy: Fya=0.958 3, Fyb=0.041 7; Diego: Dia=0.066 7, Dib=0.933 3; Kidd: Jka=0.450 0, Jkb=0.550 0; Dombrock: Doa=0.101 7, Dob=0.898 3; MNS: M=0.818 3, N=0.181 7, S=0.028 3, s=0.971 7, Mur+ =0.576 7; Lutheran: Aua=0.835 0, Aub=0.165 0. The antigen genes of Kell, Colton, Scianna, Yt, Knops, Indian, Vel blood group systems were not polymorphic, and the genetypes were kk, CoaCoa, Sc1Sc1, YtaYta, KnaKna, InbInb and Vel+, respectively. 【Conclusion】 The allele frequencies of Duffy, Diego, Kidd, Dombrock, MNS and Lutheran blood group systems of Li ethnic group in Hainan were polymorphic, while the allele frequencies of Kell, Scianna, Yt, Knops, Colton, Indian and Vel blood group systems were monomorphic.
5.Correlation betweenthromboelastography and routine coagulation tests in malignant tumor patients

【Objective】 To investigate the correlation between preoperative thromboelastography (TEG) parameters and routine coagulation tests of malignant tumor patients. 【Methods】 A total of 79 patients with malignant tumors, receiving preoperative TEG tests and routine coagulation tests between January 2019 and June 2020, were included. The correlation and regression between coagulation indexes and TEG parameters were analyzed. 【Results】 In patients with malignant tumors, the coagulation time(K) was negatively correlated with Plt and fibrinogen (FIB), while Angle, Ma, and G were respectively positively correlated with Plt and FIB. 【Conclusion】 There is a correlation between preoperative TEG parameters and routine coagulation tests in patients with malignant tumors. There is complementarity but not reciprocal substitutability between these two tests.
