1.Study on the serum interleukin and T lymphocyte subsets levels in patients with gallstones before and after treatment

Objective To observe the changes of serum interleukin and T lymphocyte subsets levels in patients with gallstones before and after treatment. Methods 72 patients with gallstones were treated as research subjects and 30 cases with normal physical examination as the control group,the serum interleukin and T lymphocyte subsets levels of observation group were detected before and after surgery. Results The preoperative serum IL-6, IL-8 and TNF-α levels of observation group were(61.5 ±4. 8) ng/L, (246. 2 ±15.3)ng/L and(1.79 ±0.72)mg/L respectively which were significantly higher than postoperative and control group,the difference was statistically significant (P < 0. 05 ) , while there was no significant difference between postoperative and normal control group ( P > 0.05); The preoperative CD3+ , CD4+ and CD8+ T cells ratio of observation group were (0. 53 ± 0. 06) % , (0. 28 ±0.05)% and(0.21±0.04)% respectively which were significantly higher than postoperative and control group,the difference was statistically significant(P < 0.05) , while there was no significant difference between postoperative and normal control group(P>0.05). Conclusion Patients with gallstone had obvious abnormal serum IL-6, IL-8, TNF-α,CD3+ ,CD4+ and CD8+ T cells levels,surgery could not only eliminate inflammation,but also help the recovery of immune function in patients.
2.Study the minimally invasive surgery for treatment of cholecystolithiasis together with common bile duct stones

Objective To study the effect of minimally invasive surgery for treatment of cholecystolithiasis together with common bile duet stones.Methods 245 patients suffered from common bile duct stones were admitted and received the therapeutical procedures.All patients were randomly divided into two groups.138 patients in the treatment group treated with minimally invasive surgery;107 patients in the control group treated with the traditional way of laparotomy.Results Compared with the control group,the treatment group showed high cure rate of 97.83%(P>0.05),low complication rate of 5.19%,short hospital stays of(7.5±2.1)d,less cost of(8 134±1 676)CNY (x2=18.34,t=8.75,6.93,all P<0.01).Conclusion Minimally invasive surgery had more advantages and wide application for both doctors and patients.
3.Comparison of efficacy between the surgical treatment and plaster external fixation for treatment of unstable distal radius fractures.

OBJECTIVETo compare the differences of the efficacy between the conservative treatment and surgical treatment for unstable distal radius fractures with surgical indications.
METHODSFrom March 2008 to July 2011, the patients with unstable distal radius fractures were devided into the surgical treatment group and the conservative treatment group in a non randomized manner. In the surgical group,there were 16 males and 32 females with an average age of 50.26 years old ranging from 22 to 76 years; in the conservative treatment group there were 10 males and 27 females with an average age of 51.12 years old ranging from 42 to 79 years. The patients of two groups were almost all with falling damage trauma,which accounting for 92.94% (79/85), all the patients were closed injury, 41 cases were in the left side, and 44 cases in the right. Fracture in accordance with AO classification: C1 in 12 cases, C2 in 32 cases, C3 in 38 cases, B3 in 3 cases. Patients in the surgical treatment group were treated with volar locking compression plate (LCP) fixation and patients in the conservative were treated with manual reduction and plaster external fixation. The active wrist range of motion, pain assessment, subjective evaluation, Gartland and Werley score, Batra radiology score and complications were observed in the last follow-up.
RESULTSForty-three patients in the surgical treatment group were followed up for an average time of 8.03 months (4 to 15 months); 33 paitents in the conservative group were followed up for an average time of 6.54 months (2 to 9 months). The difference of the wrist joint active dorsiflexion and supination rotation range in the two groups were statistically significant (t = 2.212, P < 0.05; t = 2.392, P < 0.05); the difference of the proportion of patients with ulnar wrist pain in the two groups was statistically significant (chi2 = 5.71, P < 0.05); the difference of patients' subjective evaluation score of two groups was statistically significant (Z = 2.13, P < 0.05); the difference of Gartland and Werley score between the two groups was statistically significant (Z = 2.36, P < 0.05); the difference of Batra radiology score in the two groups was statistically significant (Z = 2.58, P < 0.05); the differences in the X-ray measurement of the proportion of distal radial shortening > or = 5 mm, the radial height (shortening) was statistically significant (for the correction chi2 = 7.57, P < 0.05; t = 2.016, P < 0.05); the differences of malunion rate, ulnar impaction syndrome, the incidence rate of ulnar styloid fracture nonunion of two groups was statistically significant (adjusted chi2 = 10.05, P < 0.05; chi2 = 8.39, P < 0.05; chi2 = 5.51, P < 0.05).
CONCLUSIONFor patients with surgical indications for unstable distal radius fractures, every indicator of the outcome after surgery in the treatment is better than that of the manipulative reduction and plaster fixation.

4.Recombinant porcine interferon-gamma expressed in CHO cells and its antiviral activity.

The aim of this study was to produce recombinant porcine interferon gamma (rPoIFN-γ) by Chinese hamster ovarian (CHO) cells expression system and to analyze its antiviral activity. Firstly, we constructed the recombinant eukaryotic expression plasmid pcDNA3.1-PoIFN-γ and transfected into suspension cultured CHO cells for secretory expression of rPoIFN-γ. The rPoIFN-γ was purified by affinity chromatography and identified with SDS-PAGE and Western blotting. Subsequently, the cytotoxicity of rPoIFN-γ was analyzed by CCK-8 test, and the antiviral activity of rPoIFN-γ was evaluated using standard procedures in VSV/PK-15 (virus/cell) test system. Finally the anti-Seneca virus A (SVA) of rPoIFN-γ activity and the induction of interferon-stimulated genes (ISGs) and cytokines were also analyzed. The results showed that rPoIFN-γ could successfully expressed in the supernatant of CHO cells. CCK-8 assays indicated that rPoIFN-γ did not show cytotoxicity on IBRS-2 cells. The biological activity of rPoIFN-γ was 5.59×107 U/mg in VSV/PK-15 system. Moreover, rPoIFN-γ could induced the expression of ISGs and cytokines, and significantly inhibited the replication of SVA. In conclusion, the high activity of rPoIFN-γ was successfully prepared by CHO cells expression system, which showed strong antiviral activity on SVA. This study may facilitate the investigation of rPoIFN-γ function and the development of novel genetically engineered antiviral drugs.
