1.Progress in reguIation of phosphoIipase D mediated by Wnt/β-catenin and NF-κB signaIing pathways and its reIationships with tumorigenesis

Over the last few years,elevated phospholipase D(PLD)expression and activity found to be mediated by Wnt/ β-catenin and NF-κB signaling axis sharing the same downstream genes have been involved in tumorigenesis,controlling cancer cell invasion and metastasis. New studies have revealed that Wnt signaling/ NF-κB signaling can induce PLD upregulation and increased enzymatic activ-ity. PLD/ PLD-generated phosphatidic acid( PA),as a critical regulator,might positively modulate β-catenin-dependent T-cell factor and NF-κB transcriptional activity via a targeted motif and a positive feed-back loop to reinforce pathway output including cyclin D1 ,c-myc,survivin,vascular endothelial growth factor and cyclooxygenase-2,while PLD/ PA inhibitors can reverse all the effect mentioned above and achieve significantly better anticancer effects. The PLD/ PA signaling pathway can be a novel therapeutic target for the treatment of cancer.
2.Accuracy of continuous noninvasive hemoglobin monitoring in patients undergoing cesarean section

Objective To evaluate the accuracy of continuous noninvasive hemoglobin ( Hb ) monitoring in the patients undergoing cesarean section. Methods A total of 200 patients, at 36-42 weeks of gestation, aged 19-40 yr, with body mass index of 20.5-35.1 kg∕m2 , of ASA physical statusⅠorⅡ, undergoing elective cesarean section from June 2014 to October 2014 in our hospital, were enrolled. A sensor was positioned at patient′s finger and connected to the Masimo Radical?7 Pulse CO?Oximeter, a continuous noninvasive Hb measurement device. Noninvasive Hb obtained with Pulse CO?oximeter ( SpHb) was recorded. Before skin incision ( T0 ) , after delivery of the placenta ( T1 ) , after suturing the uterus ( T2 ) and at the end of operation ( T3 ) , blood samples from the radical artery were collected for determination of total Hb ( tHb) , and SpHb was also recorded. The agreement between two methods was assessed using Bland?Altman analysis. Results At T0-T3, tHb was 111±9, 103±8, 94±8 and (89±7) g∕L, respectively, and SpHb was 124 ± 9, 120 ± 12, 108 ± 9 and ( 103 ± 8 ) g∕L, respectively. Bland?Altman analysis showed that at T0-T3 , the mean difference between SpHb and tHb was 13.5, 17.1, 14.1 and 13.9 g∕L, respectively, and 95% confidence interval was 13.1-13.9, 16.5-17.7, 13.6-14.6 and 13.4-14.4 g∕L, respectively. The limit of agreement was 8.4-18.6, 9.1-25.1, 7.8-20.4 and 7.4-20.4 g∕L at T0-T3 , respectively, and the interchangeable limits of the two methods ranged between 3.5-23.5, 7.1-27.1, 4.1-24.1 and 3.9-23.9 g∕L at T0-T3 , respectively. The repeatability coefficient of tHb and SpHb was 16.5 and 15.8 g∕L, respectively. The relative error of SpHb was (4.6±1.0)%, (5.3±1.4)%, (4.9±1.2)% and (4.8±1.2)% at T0-T3, respectively. Conclusion Continuous noninvasive Hb monitoring provides good accuracy in the patients undergoing cesarean section.
3.Expression level and clinical features of STAT3 in breast cancer

Objective To detect the expression of STAT3 in breast cancer and adjacent paracancerous tissues ,then analyze the correlationship between its expression level and some other clinical features of breast cancer,and to evaluate its significance in prognosis of patients with breast cancer. Methods 130 breast cancer patients were enrolled in the Jiangmen Central Hospital ,and 61 cases of adjacent normal tissues. The expression of STAT3 was detected by IHC. We compared the expression of STAT3 between the two groups and analyzed the relationship between STAT3 and other clinical features. Results The expression of STAT3 in breast cancer group was much higher than the adjacent paracancerous tissues. STAT3 expression showed obvious correlationship with clinical stage ,tumor size ,lymph node metastasis ,the expression of ER and PR ,but no statistical correla-tion with age ,histological grade and Her-2 expression. Survival analysis showed that patients with high STAT3 expression level had a significantly poor prognosis. The Cox regression analysis showed that PR and STAT3 expres-sion were independent prognostic factors for patients with breast cancer. Conclusion These results may implicate that STAT3 will promote the development of breast cancer and higher expression of STAT3 indicates the poor survival rate.

Objective To study the clinicopathological significance of the expression of glutameta decarboxylase 65(GDA65) and protein kinase C(PKC) in the central cancer tissues, cancer edge tissues, paracancerous liver tissue and non-cancer liver tissues. Methods The expression of GDA65 and PKC were detected by immunohistochemical method in 10% neutral formalin- fixed and routinely paraffin-embedded sections in 37 hepatic cancer specimen. Results The positive rate and the score of GDA65 and PKC in the cancer tissues were significantly higher than that in the paracancer tissues or non-cancer liver tissues, but the PKC expression was no difference between the central cancer tissues and the cancer edge tissues . The expression of GDA 65 was related to the pathological types, differentiated degrees, liver cirrhosis or metastasis of hepatocarcinomas. No correlation was found between the expression of PKC and the clinicopathological features of hepatocarcinomas. Conclusions The expression of GDA65 and PKC might be closely related to the carcinogenesis of hepatocarcinoma, they might be important biological markers of hepatocarcinoma.
7.Non-motor Symptoms of Parkinson's disease Associated with Quality of Life

Objective To investigate the status about non-motor symptoms (NMS) in Parkinson's disease (PD) patients and the effect on quality of life. Methods 50 PD patients and 50 normal controls were investigated with the NMS questionnaire and the Healthy Survey short form (SF-36), and analysed with Spearman correlation. Results 98% of PD patients presented NMS, and recent memory loss, restless legs,constipation were the most frequent. All the NMS were more frequent than in the controls (P<0.05) except sexual desire, sexual difficulties and nausea or vomiting. All the dimensions of SF-36 were significantly different between the groups (P<0.05). The symptoms of constipation,decline in recent memory, decreased attention, restless legs and sleep disorders correlated with the scores of SF-36 (P<0.05). Conclusion NMS is common in PD patients, and impacts their quality of life.
8.Effect of caffeine on hyperoxic lung injury of premature rats and its relationship with p38 motigen ̄acti ̄vated protein kinase signal pathway

Objective To study the effect of caffeine on hyperoxic lung injury of premature rats and its relationship with p38 motigen-activated protein kinase( MAPK) signal pathway. Methods Sixty Wistar premature rats were divided into 4 groups(n=15) according to the random number table:air + normal sa-line group(A+N group),air + caffeine group( A+C group),hyperoxia + normal saline group( H+N group),and hyperoxia + caffeine group(H+C group). Among them,H+N group and H+C group were continually exposed to hyperoxia ( oxygen concentration was 60% ~70%). For A + C group and H + C group,the premature rats were injected with caffeine of 29 mg/(kg·d) into their peritoneal cavities every day after birth. For A+N group and H+N group,the premature rats were injected with normal saline of the same volume into their peritoneal cavities. In each group,the lung tissues of 5 premature rats were randomly select-ed on the third, seventh and fourteenth day respectively. The pathological changes of lung tissue, radiated alveolar count(RAC) and collagen content in lung tissue were observed under a light microscope. The wet/dry ratio ( W/D) was measured. Two-step immunohistochemistry was used to detect the distribution of p38MAPK in lung tissue. The content of phosphorylated p38MAPK( p-p38MAPK) protein was detected by western blot. Results Compared with the air groups,the lung tissues of premature rats in high oxygen expo-sure groups showed different degree of inflammatory changes on the third,seventh,and fourteenth day. The changes were more obvious with the prolonged exposure to hyperoxia. Pulmonary fibrosis was visible on the fourteenth day,which was improved after caffeine intervention. The RAC value of premature rats in hyperoxia exposure groups was significantly lower than that in air-exposure groups(P<0. 05),and the W/D ratio and collagen content in lung tissue increased significantly (P<0. 05),which were improved after caffeine inter-vention(P<0. 05). The results of two-step immunohistochemistry showed that the number of p-p38MAPK positive cells in the lung tissue of premature rats in hyperoxia exposure groups increased and widely distribu-ted, but decreased after caffeine intervention. The results of western blot showed that the content of p-p38MAPK protein in lung tissue of premature rats in hyperoxia exposure groups was significantly higher than that of air groups(P<0. 05),but it decreased after caffeine intervention(P<0. 05). Conclusion Hy-peroxia can promote the formation of pulmonary fibrosis by activating p38MAPK signal pathway. Caffeine can interdict the expression of p38MAPK to alleviate the fibrosis of lung tissue exposed to hyperoxia and thus protects the lung tissue.
9.Clinical Characteristics and Impact of Diabetes Mellitus on Outcomes in Patients with Nonvalvular Atrial Fibrillation.

PURPOSE: Studies have shown that diabetes mellitus (DM) is a risk factor for cardiovascular disease, including atrial fibrillation (AF); however, the clinical characteristics and prognostic impact of DM in patients with nonvalvular AF have not been well understood in China. MATERIALS AND METHODS: Included were 1644 consecutive patients with nonvalvular AF. Endpoints included all-cause mortality, cardiovascular mortality, stroke, major bleeding, and combined endpoint events (CEE) during a 1-year follow-up. RESULTS: The prevalence of DM was 16.8% in nonvalvular AF patients. Compared with non-diabetic AF patients, diabetic AF patients were older and tended to coexist with other cardiovascular diseases. Most patients with DM (93.5%) were eligible for anticoagulation, as determined by CHADS2 scores. However, only 11.2% of patients received anticoagulation. During a 1-year follow-up, the all-cause mortality and CEE rate in the DM group were significantly higher than those of the non-DM group, while the incidence of stroke was comparable. After multivariate adjustments, DM was still an independent risk factor for 1-year all-cause mortality [hazard ratio (HR)=1.558; 95% confidence interval (CI) 1.126-2.156; p=0.007], cardiovascular mortality (HR=1.615; 95% CI 1.052-2.479; p=0.028), and CEE (HR=1.523; 95% CI 1.098-2.112; p=0.012), yet not for stroke (HR=1.119; 95% CI 0.724-1.728; p=0.614). CONCLUSION: DM is a common morbidity coexisting with nonvalvular AF and is associated with an increased risk of 1-year all-cause mortality, cardiovascular mortality, and CEE. However, no increased risk of stroke was found during a 1-year follow-up in patients with AF and DM.

10.Evaluation the application of intra-operative cell salvage in cesarean section based on multicenter data

Objective:To investigate the safety, efficacy and application indication of intra-operative cell salvage (IOCS) in cesarean section.Methods:A total of 1 265 pregnant women who received IOCS blood transfusion during cesarean section in 11 tertiary A hospitals from August 2016 to January 2019 were collected and divided into <1 500 ml group (796 cases) and ≥1 500 ml group (469 cases) according to the amount of blood loss during cesarean section. The general clinical data, ultrasonic imaging data, perinatal and puerperium indicators were analyzed retrospectively. The risk factors of intraoperative blood loss ≥1 500 mL using IOCS transfusion were analyzed by logistic multivariate regression.Results:(1) A total of 848 001 ml of blood was recovered and a total of 418 649 ml of blood was transfused in 1 265 pregnant women who received IOCS transfusions, which was equivalent to 23 258 U red blood cell suspension, greatly saving medical resources. The intraoperative blood loss in <1 500 ml group and ≥1 500 ml group was 800 ml (300-1 453 ml) and 2 335 ml (1 500-20 000 ml), respectively. No amniotic fluid embolism, severe adverse reactions, shock and death occurred in the two groups. (3) Multivariate regression analysis showed that age ≥35 years ( OR=1.5, 95% CI: 1.1-1.9), prenatal hemoglobin level <110 g/L ( OR=1.7, 95% CI: 1.3-2.2), history of uterine surgery ( OR=1.8, 95% CI: 1.3-2.6), placenta previa ( OR=1.9, 95% CI: 1.1-3.1), placenta accreta ( OR=2.6, 95% CI: 1.8-3.9), blood pool in the placenta ( OR=1.6, 95% CI: 1.1-2.3), abnormal posterior placenta muscle wall ( OR=1.8, 95% CI: 1.2-2.6), placenta projecting to the anterior uterine wall ( OR=3.0, 95% CI: 1.3-7.0) were risk factors for blood loss ≥1 500 ml in obstetric transfusion using IOCS technique, with statistical significance (all P<0.05). Conclusion:IOCS is safe and effective in cesarean section, which could save the medical resources and reduces medical expenses, however, it is necessary to strictly master the application indication.
