1.Determination of Content and Related Substances of Tadalafil Tablets by HPLC

Objective:To establish a determination method for the content and related substances of tadalafil tablets. Methods:An HPLC method was adopted. The determination was performed on a ZORBAX Eclipse XDB C8 (250 mm × 4. 6 mm,5μm) column with the mobile phase consisting of 0.1% trifluoroacetic acid-acetonitrile(65∶35)at a flow rate of 1.0 ml· min-1,the column temperature was controlled at 35℃, the detection wavelength was 285 nm and the injection volume was 20 μl. Results:The related substances could be completely separated from the maln component under the selected chromatographic conditions. A good linear relationship was found be-tween the peak area and the concentration of tadalafil within the range of 20. 13-201. 30μg·ml-1(r=0. 999 3) with the average recover-y of 99. 5% (RSD=1. 1%, n= 9). The limit of detection was 0. 6 ng and the limit of quantification was 2 ng. Conclusion:The method is specific and sensitive, and can be used in the determination of content and related substances of tadalafil tablets.

3.The efficacy and safety of induction therapy with autogenous mesenchymal stem cells perfusion before operation in cadaveric kidney transplantation

Objective To evaluate the efficacy and safety of mesenchymal stem cells(MSCs)in preventing early acute rejection after renal transplantation.Methods Eighty-eight primary cadaveric renal allograft recipients in our department were randomized into two groups treated with bone marrow MSCs (BMSCs group,n =43) or not (control group,n =45).Main immunosuppressive therapy regimen consisted of steroids,tacrolimus or cyclosporine and mycophenolate mofetil in all recipients.Estimated glomerular filtration rate (eGFR) of transplant kidney,incidence of acute reaction (AR),graft survival and incidence of adverse events were recorded within 24 months.Results In BMSCs group,the incidence of AR was 4.7 % and 9.3 % at 3rd month and 6th month respectively,significantly lower than 20.0 % and 26.7 % (P<0.05) in the control group.The eGFR at day 7,14and 30 post-transplantation was significantly higher in the BMSCs group than in the control group (P<0.01,P<0.01,P<0.05 respectively).The incidence of adverse events in the BMSCs group and the control group was 44.2 % (19/43) and 66.7 % (30/45,P < 0.05) respectively and the rate of infection was 37.2 % (16/43) and 33.3 % ( 15/46,P > 0.05) respectively within 24 months.Conclusion Induction therapy with autogenous BMSCs appeared to be more effective in the prevention of AR following cadaveric kidney transplantation and was associated with better clinical outcomes as far as early renal graft function without compromising patient safety.
4.Analysis of antibodies of poliviruses in persistent populations in Beijing, 2012.

OBJECTIVETo analyze the polio immunity level of persistent population in Beijing, 2012.
METHODSA total of 1 676 subjects residing more than 6 months in Beijing were selected by stratified random cluster sampling design in 2012. Demographic characteristics, history of oral poliovirus vaccine (OPV) immunization were investigated by questionnaire. All 5 ml blood sample were collected for testing of polio neutralizing antibody using the method of microcell neutralization. The positive rate and the geometric mean titer (GMT) of polio neutralizing antibody type I, II and III were analyzed in different groups.
RESULTSThe positive rate of type I, II and III were 98.2% (1 645/1 676), 98.1% (1 644/1 676), 97.6% (1 635/1 676); The GMT were 1:130.2, 1: 113.4 and 1: 79.7. Three types of positive rates in<15 years group (99.7% (664/666), 99.8% (665/666), 99.5% (663/666)) were higher than those of ≥ 15 years group (97.1% (981/1 010), 96.9% (979/1 010), 96.2% (972/1 010)), the differences were significant (all the values of P < 0.01); The GMT in<15 years group (1:325.9, 1:250.5, 1:190.7) were higher than that of ≥ 15 years group (1: 71.1, 1: 67.2, 1: 44.8), the difference was significant (all the values of P < 0.01). The positive rate (99.0%-100%) and GMT (1: 128.8-1: 300.7) in vaccination information confirmed population were higher. The highest positive rate (all were 100%) and GMT(1: 409.7-1: 636.7) were observed in children who vaccinated three times.
CONCLUSIONThe polio antibody of healthy population was at a high level in Beijing in 2012; Especially the age groups of < 15 years which were covered by vaccines.Immunization barrier had been formed firmly to interrupt the transmission of wild poliovirus and vaccine-derived poliovirus.
