1.Progress in research on acceptance strategy coping with chronic pain

Objective Chronic pain has brought great suffering to patients,as well as a heavy burden to the society.More attention has been paid on the efficiency of psychological intervention as supplementary means.Among the treatments,acceptance strategy shows its superiority and gradually enters people's field of vision.However,researches in this area are still very scarce.This study intends to review relevant research to provide reference to clinical work and research.Methods Theacceptance andchronic pain as the keywords were searched from the American Psychological Association (APA),China National Knowledge Infrastructure (CNKI),Wanfang data from 1994 to 2014 in English and Chinese articles,and the articles were chosen according to the criteria:① chronic pain and its current situation;②acceptance strategy or acceptance-based therapy;③intervention to chronic pain based on acceptance strategy;④to explore the intervention mechanism of aueptance strategy.Results Eventually,7 Chinese articles and 42 English articles were adopted.Conclusion Acceptance strategy shows some advantages in clinical pain improvement:it can significantly improve the psychological and social function of patients,but it is not always efficient in reducing the pain intensity.There are still some limitations in the present study.It is necessary to develop an acceptance-based therapy with Chinese culture to treat the domestic patients.
2.Roles and Applications of Information Technology in the Researches on TCM Clinical Practice Guidelines

TCM clinical practice guidelines are not only the norms to guide medical work, but also the basis for medical management and insurance. Researches on TCM clinical practice guidelines are profoundly significant to promote standardization, modernization and internationalization of TCM. At present, researches on TCM clinical practice guidelines at home and abroad have just started. Based on successful practice of previous researches, adopting information technology to establish computer-aided TCM clinical practice guidelines is practical to promote the compilation, revision, application and generalization. This article aimed to discuss roles of researches on information technology in TCM clinical practice guidelines.
3.Research and Construction of Transformation Platform of TCM Literature Electronic Publication Based on App Store

Digital construction and network service are the inevitable trend of the development of TCM literature modernization, and the model based on App Store has become the trend of electronic publications. This article analyzed the current status and existing main problems of the platform of TCM literature electronic publication. In view of the above problems, combined with the development of TCM literature electronic publication, this article proposed a transformation platform based on App Store, and further elaborated the function, design and technology of the platform.
4.Analysis of Researches on TCM Literature Digitization

5.Comparison of the effects of phenylephrine and norepinephrine in patients with restrictive transfusion

Objective To compare the effects of phenylephrine and norepinephrine in patients with restrictive transfusion.Methods 65 patients undergoing laparoscopic resection of rectal cancer were selected.According to the random number table method,the patients were randomly divided into 32 cases of observation group (phenylephrine composite restriction infusion) and 33 cases of control group(norepinephrine combined restrictive transfusion).The operation and hemodynamics of the two groups at different time points were compared.At T0,T1,T2,the central venous blood and arterial blood were collected,the central venous blood saturation (ScvO2),arterial blood lactate (Lac),central venous oxygen partial pressure (PcvO2),PaO2,Hb concentrations,venous oxygen content (CcvO2),arterial oxygen content(CaO2),arteriovenous oxygen content difference (Ca-cvO2) and oxygen uptake rate (ERO2) were compared between the two groups.Results The ASA levels (Ⅰ / Ⅱ grade) of the observation group and the control group were 11 cases/21 cases and 13 cases/20 cases,respectively,the difference was not statistically significant(x2 =5.298,P =0.052).The BMI of the observation group and the control group were (20.61 ± 1.65) kg/m2,(21.57 ± 1.42) kg/m2,respectively,the difference was not statistically significant (t =0.652,P =1.980).The fluid replacement of the observation group and the control group were (1 162 ± 113) mL,(1171 ± 104) mL,respectively,the difference was not statistically significant (t =1.734,P =0.065).The blood loss of the observation group and the control group were (276 ± 41) mL,(294 ± 42) mL,respectively,the difference was not statistically significant (t =0.083,P =1.023).The urine volume of the observation group and the control group were (524 ± 113) mL,(532 ± 109) mL,respectively,the difference was not statistically significant(t =1.834,P =0.053).The operation time of the observation group and the control group were (192 ± 24) min,(197 ± 21) min,respectively,the difference was not statistically significant(t =1.367,P =0.073).The MAP at T1-T2 of the two groups increased,the CVP of the two groups was significantly higher at T1,the HR of the two groups was significantly higher at T2.The MAP,CVP,HR between the two groups had no significant differences (all P > 0.05).The levels of ScvO2,PcvO2 and PaO2 at T1 were significantly higher than those at T0,on the contrary,the levels of ScvO2,PcvO2 and PaO2 at T2 were significantly lower than those at T0,and the Hb in the observation group was significantly lower at T1-T2,the Hb in the control group was significantly lower at T,-T2,which in the observation group was lower than the control group,the differences were statistically significant(all P < 0.05).There was no statistically significant difference in Lac between the two groups (P > 0.05).There were statistically significant differences in CcvO2,CaO2,Ca-cvO2 and ERO2 between the two groups at different time points (all P < 0.05).Conclusion Phenylephrine is more effective than norepinephrine in limiting infusion in elderly patients,and it has more obvious effects on tissue oxygen metabolism.
6.AC092127.1-miR-451a-AE binding protein 2 Signaling Facilitates Malignant Properties of Breast Cancer

Purpose:
The purpose of the current study was to explore the functions and potential mechanism of miR-451a in breast cancer (BC).
Methods:
Quantitative reverse transcription real-time polymerase chain reaction was used to analyze the expression of miR-451a in human normal mammary cells (MCF-10A) and BC cells. Colony formation assay, terminal-deoxynucleoitidyl transferase mediated nick end labeling assay and transwell assays were conducted to validate the effect of miR-451a on proliferation, apoptosis, migration and invasion of BC cells, respectively. RNA pull-down, RNA immunoprecipitation and luciferase reporter assays were applied to investigate the upstream and downstream mechanisms of miR-451a in BC cells.
Results:
MiR-451a was expressed at a low level in BC cells. Overexpression of miR-451a repressed BC cells proliferation, migration and invasion. Moreover, long non-coding RNA AC092127.1 acted as a sponge of miR-451a to enhance the expression level of AE binding protein 2 (AEBP2) that was demonstrated to be the target gene of miR-451a in BC cells. Finally, rescue experiments validated that miR-451a and AEBP2 involved in AC092127.1-mediated BC cell growth, migration and invasion.
Conclusion
In a word, AC092127.1/miR-451a/AEBP2 axis contributes to BC cell growth, migration and invasion. Our results may help to find novel potential targets for BC treatment.

8.Effects of hyperthermia on the expression of E6 and E7 genes of human papillomavirus type 6 and 11

Objective To investigate the effect of hyperthermia on the expression of E6 and E7 genes of human papillomavirus (HPV) type 6 and 11 in HPV-infected human skin. Methods Tissue samples were obtained from the lesions of condyloma accuminatum (CA) in 6 patients after informed consent. Each sample was divided into 4 parts: one was embedded and directly stored at -80 ℃; the other 3 parts were placed in culture medium and the surface of the samples was irradiated for 30 minutes with a thermotherapy apparatus at 37℃, 42 ℃, 45 ℃, respectively, then the samples were taken out and stored at -80 ℃. RNA was extracted from the specimens, real time quantitative PCR (qPCR) was performed to detect the expression of E6 and E7 genes of HPV-6 and -11. Results Of the 6 patients, 2 were infected with HPV-6 and -11 respectively, 4 with both HPV-6 and HPV-11. The expression of E6 and E7 mRNA decreased with the increase in irradiation temperature. The relative mRNA expression levels at 37 ℃, 42 ℃ and 45 ℃ were 1.00 ± 0.00, 0.61 ± 0.17, 0.27 ± 0.15, respectively, for HPV-6 E6 gene, 1.00 ± 0.00, 0.56 ± 0.21, 0.16 ± 0.11 respectively, for HPV-6 E7 gene, 1.00 ± 0.00, 0.60 ± 0.22, 0.16 ± 0.08, respectively, for HPV-I1 E6 gene, 1.00 ± 0.00, 0.55 ± 0.15, 0.24 ± 0.06, respectively, for HPV-11 E7 gene; statistical difference was noted among them between the specimens irradiated at different temperature (all P < 0.01). Conclusion Hyperthermia can remarkably suppress the expression of HPV-6/I 1 E6 and E7 genes, which may be a possible mechanism under the regression of warts induced by local hyperthermia.
9.Effect of PNCG on the Expression of CLA+T Cell Derived Cytokines in Atopic Dermatitis

Objective To investigate the influence of polysaccharide and nucleoside extract from bacille Calmett-Gueiin (PNCG) on the frequency of peripheral cutaneous lymphocyte-associated antigen positive (CLA~+) T lymphocyte derived lymphokines in atopic dermatitis (AD) and to explore their associations with the disease severity. Methods A randomized,double blind and placebo cross-over control method was used to treat the AD patients. Before and after the treatment,flow cytometry was used to measure the frequencies of IL-4,IL-5, IFN-γ,TNFα positive CLA~+T cells. Disease severity was evaluated by atopic dermatitis area and severity index score (ADASIS). Results After the PNCG treatment,the frequency of IL-5~+ CLA~+T cell decreased significantly,while IFN-γ increased,compared with the placebo con-trol group. PNCG could improve the ADASIS. Conclusion PNCG may treat AD through restoring the immune balance of CLA~+T cells.
10.Effects of B7-1 on the cytoskeleton rearrangement in podocytes induced by angiotensin Ⅱ

Objective To observe the effect of costimulatory molecule B7-1 on cytoskeleton rearrangement in mouse podocytes induced by angiotensin Ⅱ (Ang Ⅱ),and to study the underlying molecular mechanism of B7-1 in the pathological changes of podocytes.Methods All cultivation of conditionally immortalized mouse podocytes (MPC) in vitro were divided into the following groups:normal control group,CTLA-4 group,Ang Ⅱ group (10-6 mmol/L 12 h,24 h;10-8 mmol/L 12 h,24 h) and CTLA-4 with Ang Ⅱ group.Transfect B7-1 RNA interference fragment (siRNA) to the mature podocytes,and then restimulated by Ang Ⅱ (10-6 mmol/L 12 h),the change of podocyte cytoskeleton after Ang Ⅱ stimulation were observed.The expression of B7-1 in each group was assayed by flow cytometry and Western blotting.The nephrin and p-nephrin protein levels in the four groups were also analyzed by Western blotting.At the same time,the podocyte cytoskeleton distribution as indicated by F-actin was observed by fluorescence microscopy.Results Flow cytometry and Western blotting showed that B7-1 was not expressed in the normal control group.Ang Ⅱ showed a concentration and time dependent induction of B7-1 expression in mouse podocytes (P < 0.05).Western blotting indicated that Ang Ⅱ induced B7-1 protein expression (P < 0.05).Expression of nephrin and p-nephrin was significantly down-regulated by Ang Ⅱ (P < 0.05).Compared with the normal control group,the expression of podocyte protein nephrin and p-nephrin in Ang Ⅱ stimulation group was significantly reduced (P < 0.05).Using FITC phalloidin fluorescence staining showed that CTLA-4+Ang Ⅱ stimulation group cytoskeleton rearrangement was improved significantly and F-actin recombinant score (mCFS) decreased compared with Ang Ⅱ group (P < 0.05),suggesting that Ang Ⅱ led to the disorder of the podocytes cytoskeleton and the destruction of the cytoskeleton of podocytes by Ang Ⅱ could be improved after B7-1 blocking.Compared with the Ang Ⅱ stimulation,transfection of B7-1 siRNA + Ang Ⅱ stimulation group improved F-actin cytoskeletal rearrangement,and mCFS also decreased significantly (P < 0.05),suggesting that transfection of B7-1 siRNA might improve the damage of Ang Ⅱ on podocytes cytoskeleton.Conclusions B7-1 participates in the process of cytoskeleton reconstruction and plays an important role in the pathological changes of podocytes.
