BACKGROUND:Aconitum carmichaeli Debx (ACD) is the tuberous root of Aconium carmicgaekum, used as cardiotonic to restore yang for the treatment of colapse and shock, to warm the kidney and reinforceyang, and to expel cold and promote the flow ofyang-qi. Studies have found that ACD has obviously cardiotonic, antihypertensive, vasodilatory, analgesic, anti-inflammatory and toxic effects. OBJESTIVE: To observe the vasodilatory effects of a water decoction of ACD on rabbit’s aorta rings and its mechanism. METHODS:Rabbits aorta arteries were isolated, pre-contracted with noradrenaline (10-6 mol/L) and their responses to different concentrations of ACD (0.5, 1.0, 2.0, 4.0, 8.0 g/L) were investigated. The effects of removal of vascular endothelium and different signaling pathway inhibitors (Nω-nitro-L-arginine: 1×10-4 mol/L, methylene blue: 1×10-5 mol/L, indomethacin: 1×10-5 mol/L, propranolol: 1×10-5 mol/L) on ACD-induced vasodilation were also assessed. RESULTS AND CONCLUSION:ACD could not change the resting tension of rabbit aortic rings, but ACD treatment resulted in an obvious relaxation in narodrenaline-precontracted aortic rings and the relaxant effect was dose-dependent. The vasodilatory effect of ACD was significantly reduced by removal of endothelium, 1×10-4 mol/L Nω-nitro-L-arginine and 1×10-5 mol/L methylene blue but not reduced by indomethacin and propranolol. In addition, 4 g/L water decoction of ACD did not decrease the dose-response curves of artery rings to narodrenaline or KCl in the absence of endothelial cels. ACD can relax isolated rabbit’s aorta, which may be related to endothelium-released nitric oxide, but has no significant relevance with receptor-operated and voltage-dependent calcium channels.

AIM: To study the influence of PD098059 on the rat platelet aggregation rate and the phosphorylation of ERK1/2 induced by the different agonists, and to observe the effects of phosphorylation of ERK1/2 on the platelet aggregation. METHODS: The maximal aggregation rate (MAR) was measured by nephelometry. The inhibitory rate of PD098059 and the appearing time of MAR were also observed. ERK1/2 phosphorylation was detected by Western blot. RESULTS: The phosphorylation of ERK1/2 was detected during aggregation induced by thrombin and ADP. PD098059 inhibited the MAR and phosphorylation of ERK1/2. Effects of PD098059 were different on the aggregation induced by thrombin and ADP. CONCLUSIONS: The phosphorylation of ERK1/2 is one of the cellular signal transduction mechanisms of platelets aggregation. Phosphorylation of ERK1/2 plays different roles during the platelet aggregation induced by thrombin and ADP. [

Objective To evaluate the expression of bcl-2, p53, Ki-67 in malignant lymphoma (ML)and the relationship between the expression of bcl-2, Ki-67, p53 and prognosis for the patients who were given autologous hematopoietic stem cell transplantation (AHSCT). Methods The expression of bcl-2,Ki-67, p53 were measured by immunohistochemistry (IHC) on paraffin-embedded slices in 33 cases of ML who received AHSCT. Survival analysis was done by the Kaplan-Meier method and Log-Rank test.Multivariate analysis was carried out using COX proportional hazard model. Results For patients with ML who received AHSCT, the 3-year disease free survival (DFS) of bcl-2(+) group was 35.71%, while that bcl-2(-) group was 88.89 %. There was significant difference of DFS between the two groups. Meanwhile, for these patients, the 3-year DFS of Ki-67(+) and Ki-67(-) were 43.75 % and 85.71%, respectively (P <0.05).Multivariate analysis showed that the expression of bcl-2 and Ki-67 were the independent prognostic factors. Conclusion The expression of bcl-2 and Ki-67 were closely related with relapse after AHSCT in patients with ML. They were useful molecular makers for predicting the prognosis of patients with ML after AHSCT.

BACKGROUND: Micro-arc oxidation, also be known as micro-plasma oxidation or anodic spark deposition, is a new technology that can in situ grow ceramic coating directly on the surface of non-ferrous metals. And the ceramic coating has such characteristics as high corrosion resistance, good wear resistance, etc. It is advantageous to the osseointegration between implant and bone that the ceramic coating is able to produce porous, rough oxide film on the surface layer of the implant, which improves the biological reaction of the bone interface so as to affect the number of bone-forming cells, ype, products of cell, and the expression of the product.OBJECTIVE: To observe the osseointegration between bone and implant, and to analyze induced action of metal trace elements to bone-formation's marker (ALP). METHODS: A computer-based online search was conducted in PUMMED and Chinese Journal Full-text Database with the key words of "micro-arc oxidation, synosteosis, titanium, magnesium" in both English and Chinese between 1995 and 2009. Additionally, hand-made retrieval was performed for articles about micro-arc oxidation and microelement. Among 96 articles, 34 references were excluded due to unrelated and duplicated articles, and 28 ones were excluded due to long-term publication, 34 ones were included in the final analysis.RESULTS AND CONCLUSION: It is believed that the surface modification methods on microelement and micro-arc oxidation ceramic coating on Ti-based implant will become the mainstream process to improve the rate of implant-bone integration. However, the clinical application still needs to be further studied.

Objective This study was designed to investigate the expression changes of osteopro-tegerin (OPG), tartrate-resistant acid phosphatase (TRAP) and vascular endothelial growth factor (VEGF) mRNA in collagen induced arthritis(CIA) rats. Methods After CIA was induced in Sprague-Dawley rats, the experimental animals were treated with PBS or rAAV-EGFP or rAAV-hOPG (100 μl/day) intra-articular injection for 10 days. Messenger RNAs (mRNAs) were obtained from CIA synovium 40days after first immun-ization. Reverse transcriptase-polymerase chain reactions (RT-PCR) were carried out to detect the mRNA encoding OPG, TRAP, VEGF and β-actin, which acted as inner control. The genes detected clearly by RT-PCR were quantified using real-time PCR. Results The expression of all genes was confirmed by specific single bands in RT-PCR. Real-time PCR showed that the expression levels of TRAP and VEGF were increased, whereas those of OPG mRNA were decreased in CIA group compared with normal controls. The intra-articular gene transduction markedly increased the gene copies of OPG by 128.21% (P<0.01). The expression change of OPG in synovium also caused the decrease of the expression levels of TRAP and VEGF by 58.79% (P<0.01)and 17.85% (P>0.05) respectively, however, the expression change of VEGF was not statistically significant. Conclusion OPG gene mediated by rAAV can be successfully tranfered to knee joint synovium in vivo. The results of this study suggest that gene transfer using rAAV-OPG may be a feasible and effective therapeutic approach to treat or prevent joint destruction in inflammatory arthritis.

Objective Using an in vivo adeno-associated virus(AAV)-mediated gene transfer technique, this study was designed to evaluate the protective effects of human osteoprotegerin(OPG)transgene against joint destruction in collagen induced arthritis(CIA)model.MethodsAfter CIA was established in the Sprague-Dawley rats, the experimental animals were treated with PBS or rAAV-EGFP or rAAV-hOPG (100μl/d)intra-articular injection 25 days after arthritis induction for 10 days.Paraffin-embedded joints were then analyzed histologically. The joint destruction was evaluated by Larsen Score. The protein expression of OPG,IL-1,MMP-3 was identified by enzyme-linked immunosorbent assay(ELISA).Results Suecessful trans-gene expression was confirmed by the detection of OPG by ELISA and positive fluorescence of the frozen joint section. Image analysis revealed that the expression of OPG significantly protected against joint destruction by 30% compared with the CIA group. Conclusion OPG gene transfer mediated by rAAV effectively protects against bone destruction induced by CIA model. Those data suggest that gene transferring using rAAV-OPG may be a feasible and effective therapeutic approach to treat or prevent joint destruction in inflammatory arthritis.

Objective To investigate procedure and therapeutic effect of resection and reconstruction for axis tumors through the sub mandible approach. Methods Between December 2004 to June 2010,17 patients with axis neoplasm underwent tumor resection and antero-posterior reconstruction through the combined the sub mandible-inner sternocleidomastoid muscle (SMIS) approach and posterior approach. Tumor lesions involved C2 in 11 cases, C2-3 in 4, C2-4 in 2. Eight cases involved vertebral body, and 9 involved both vertebral body and element. Fourteen primary lesions including 4 giant cell tumors, 4 plasmocytomas, 2 chordomas, 2 eosinophilic granulomas, 1 hemangiopericytomas and 1 lymphoma, and 3 metastatic lesions were involved in this study. Three types of reconstruction in upper cervical spine including titanium mesh plus vertically placed titanium plate, titanium mesh plus obliquely placed titanium plate and trimmed titanium mesh alone, were adopted after anterior tumor resection, and then posterior tumor resection and reconstruction were performed. Results All patients experienced pain relief and neurological improvement after surgery. Except for one incidence of screw pull-out which was corrected by a revision surgery, solid fusion was achieved in all patients. A follow-up period of 6 months to 6 years was available for this study. One patient died of cerebral infarction 9 months postoperative. Two patients with chordoma relapsed 13 months and18 months postoperative, respectively, of whom one died of high plegia and respiratory failure, and the other was alive with disease. Two patients with metastasis died of multiple remote metastases 12 months and 18 months postoperative, respectively. Conclusion Through the SMIS apporach, a satisfactory exposure can be obtained for axis tumor resection and reconstruction. Anterior reconstruction of upper cervical spine after tumor resection can be achieved with internal fixation system of cervical spine, which can improve intraopera-tive safety. The combined anterior reconstruction and posterior occipito-cervcial fixation can provide immediate stability, and benefit maintaining stability of upper cervical spine.
