1.Study on Quality Standard of Apricot Kernel Cough Mixture

Objective To study the quality control method of apricot kernel cough mixture.Methods Chinese ephedra,liquorice and honeysuckle in apricot kernel cough mixture were qualitative examined by thin-layered chromatography(TLC),and the content of ephedrine hydrochloride was measured by high performance liquid chromatography(HPLC).Results The spots in the TLC of Chinese ephedra,liquorice and honeysuckl were in the same color with those in the chormatograms of control articles.For ephedrine hydrochloride,linear range was 0.020 0~0.200 0 ?g and the average recovery was 99.22%.Conclusion The quality control is simple,accurate and highly sensitive.The methods can be used for quality control of apricot kernel cough mixture.
2.Mesohepatectomy combined with bile duct reconstruction for the treatment of hepatic cancer: a report of one case

Comprehensive treatment dominated by surgery is the mainstay in the treatment of hepatic cancer,and hepatectomy is still the most effective treatment method.Bile duct reconstruction after hepatectomy is still the difficult point for the treatment of hepatic cancer complicated by bile duct invasion.A 45-year-old patient with hepatic cancer and gallstone was admitted to the Affiliated Hospital of Guiyang Medical College,magnetic resonance cholangiopancreatography and enhanced computed tomography indicated that intrahepatic duct was dilated and tumor had invaded both left and right hepatic ducts.Cholecystectomy,mesohepatectomy,duct to duct anastomosis of left hepatic duct and common hepatic duct,duct to duct anastomosis of right hepatic duct and cystic duct were performed during the operation.The patient was cured 2 weeks after surgery.
3.Drug Resistance of Enterococcus in Bile of Patients with Cholelithiasis

OBJECTIVE To analyze the resistance feature and the distribution of Enterococcus species isolated from bile specimens.METHODS Totally 715 samples of bile in intra-operation were cultured,and the drug resistances were tested.RESULTS From them 511 strains in 469 cases were detected out(65.6%),among whith 156 were Enterococcus strains and 159 were strains of Escherichia coli.The mixed infection with two kinds of germs was found in 35 cases.The results of drug susceptibility showed the resistant rates of Enterococcus faecalis to penicillin and high concentration gentamicin were lower than those of E.faecium.None of E.faecalis,and E.faecium resisted to vancomycin.CONCLUSIONS The detective rate of the Enterococcus is high in bile samples.The results reveal a different resistance to common antibiotics,but a high sensistivity to vancomycin and teicoplanin.The result of antibiotic susceptibility testing guides rational use of antibacterial agents.
4.Glioma cells promote expression of cancer-related genes in human bone marrow-derived mesenchymal stromal cells in vitro

Objective We investigated the expression profile of cancer related genes in hMSCs co-cultured with U251 glioma cells, to evaluate the risk of malignant transformation of hMSCs in glioma environment. Methods hMSCs were co-cultured with U251 glioma cells for 5 days and the expression profile of cancer-related genes were investigated by using microarray assay, followed by Real-time quantitative RT-PCR and Western blot. Results Of the 440 cancer-re?lated genes covered by Oligo GEArray Human Cancer Microarray OHS-802, SPINT2, TK1, STC1, MMP1, CCND1, SORT1, SEPT6, CDC20, SHB, CDK5, RELA, XRCC4, KIT, CTPS, CAPNS1 and ETV6 were significantly upregulated (>3-fold) whereas none was downregulated in hMSCs co-cultured with U251 glioma cells. The upregulation of oncogenes KIT, CAPNS1, TK1, MMP1, CCND1, CDC20, RELA and STC1 in co-cultured hMSCs were confirmed by Real-time quan? titative RT-PCR. The upregulation of protein expression of oncogenes KIT, MMP1, CCND1 and RELA were detected by Western blot. Conclusion The present study demonstrates that co-culture of hMSCs with human glioma cells leads to up?regulation of some important oncogenes in hMSCs, indicating the tumorigenic potential of hMSCs in glioma environment.
5.Influence of expressing IrrE from Deinococcus radiodurans on osmotic stress tolerance of succinate-producing Escherichia coli.

Hyper-osmotic stress is one of the key factors that decrease the efficiency of biological succinic acid production. To increase the osmotic stress tolerance of succinate-producing Escherichia coli, we studied the influence of IrrE, an exogenous global regulator, on cell osmotic stress resistance. Fermentation results showed that cell growth and succinic acid production by the recombinant increased under different Na+ concentrations. Meanwhile, the maximum dry cell mass, glucose consumption and succinic acid concentration increased 15.6%, 22% and 23%, respectively, when fermented in a 5-L bioreactor. Expressing IrrE improved cell resistance to hyper-osmotic stress. Further comparison of intracellular osmoprotectants (trehalose and glycerol) concentrations showed that trehalose and glycerol concentrations in the recombinant increased. This suggested that introduction of IrrE could enhance intracellular osmoprotectants accumulation which conferred cell with improved resistance to osmotic stress.
