1.Simultaneous Determination of Contents of 6 Components in Aerial Part of Rheum tanguticum Maxim.ex Balf by HPLC Method

Objective To develop an HPLC method to determine the contents of chrysophanol, emodin, physcion, aloe-emodin, sennoside A, and sennoside B simultaneously in the aerial part of Rheum tanguticum Maxim.ex Balf. Methods The analysis was achieved with an Agilent ZORBAX SB-C18 analytic column (250 mm × 4.6 mm, 5 μm) by gradient elution of methanol-0.1% phosphoric acid in water gradient elution system (0 min, 70% B; 5 min, 65% B;8–16 min, 60% B; 18–23 min, 55% B; 25–30 min, 45% B; 32–39 min, 35% B; 49–57 min, 24% B; 65–72 min, 20% B). The flow rate was 1.0 mL/min; detection wavelength was 254 nm; the column temperature was room temperature; the quantification used external standard method. Results The peak areas and concentrations of chrysophanol, emodin, physcion, aloe-emodin, sennoside A, and sennoside B showed good linear relationship within a certain concentration range (r≥0.9995); the RSD of precision was 0.75%–1.17%; the RSD of repeatability was 0.99%–2.06%; the RSD of stability was 0.97%–1.76%; the average recoveries were 99.7%–100.4%. The results showed that there were differences in content between the root and aerial part of Rheum tanguticum Maxim.ex Balf. Conclusion HPLC method for simultaneous determination of contents of 6 contents of the aerial part of Rheum tanguticum Maxim.ex Balf can be used as the references for quality control.

Objective To evaluate the efficiency of injection of colloidal chromic phosphate 32 P cavity in rats with rheumatoid arthritis.Methods Collagen induced arthritis (CIA) was produced in Sprague Dawley rats.After one week immune,the right post ankle was given colloidal chromic phosphate 32 P intra articular injection.Severity of arthritis was observed by the left to right diameter of the right post ankle joint,the volume of right post paw and the arthritic index.Results The group treated with colloidal chromic phosphate 32 P intra articular injection developed milder arthritis compared with control CIA group after 6 weeks.The left to right diameter of the right post ankle joint was (3 96?0 05) mm of the colloidal chromic phosphate 32 P intra articular treated group compared with (4 11?0 08) mm of the control CIA group ( P

Objective To investigate the expression of T cell immunoglobulin domain and mucin domain (TIM)-3 and its ligand Galectin-9 in the peripheral blood of initial systemic lupus erythematosus (SLE)patients,and explore their effects on SLE.Methods The percentages of CD4+TIM-3+,CD8+TIM-3+cells from 33 SLE patients and 26 normal controls were detected by flow cytometry,and the Galectin-9 gene expression of PBMCs was determined by real-time PCR The SLE Disease Activity Index(SLEDAI),C3 level and lymphocyte count were evaluated.Mann-Whitney U test was used for independent samples analysis and Spearmen's test was used for correlation analysis.Results The percentages of CD4+TIM-3+ and CD8+TIM-3+ cells were markedly increased in SLE group than those of the control group(P<0.01).In particular,the CD4+TIM-3+,CD8+TIM-3+ level Was positively correlated with SLEDAI (r=0.517,P<0.01;r=0.400,P<0.05);but negatively correlated with C3(r=0.487,P<0.05;r=0.395,P<0.05).The Galectin-9 mRNA in SLE PBMCs was higher than that of the controls(P<0.05).Conclusion TIM-3-Galectin-9 pathway may be involved in T cell immune regulation of SLE,and is related to disease activity.
5.Monte Carlo simulation and experimental investigation of 125I interseed dose attenuation

Objective To investigate the interseed dose attenuation for multiple 125I seeds.Methods Monte Carlo simulation was done by Geant 4 to investigate the dose distribution of single seed and multiple seeds.The results were compared with the dose calculation method from TG43-U1 and examined by experiments.Results The difference of single seed dose distribution between Monte Carlo method and line source model was ± 3%.The difference between Monte Carlo method and experiment result was ± 5%.The interseed dose attenuation of multiple seeds at the interesting points was 3.8% to 13.2% and the average interseed dose attenuation was 7.2%.The difference of experiment result and Monte Carlo result was less than 6%.Conclusions The interseed attenuation is about 7% and the maximum value may be larger than 13% for multiple seeds.The interseed dose attenuation may be larger in tissue.So it is not accurate to calculate the dose distribution by using TG43-U1.
6.Evaluation on reliability and validity of Chinese version of childhood trauma questionnaire

BACKGROUND: At present, the internationally public recognition of childhood trauma questionnaire is the authorized version of Bernstein,American psychologist, in 1998.OBJECTIVE: To set up the scale of Chinese version of childhood trauma questionnaire with 28 items and analyze its reliability and validity.DESIGN: Community investigation was designed.SETTING: Mental Health Institute, Xiangya Second Hospital, Central South University.PARTICIPANTS: Totally 441 students from 8 classes were randomized from a countryside middle school of a city in Henan in October 2004.METHODS: A total of 441 students were measured with childhood trauma questionnaire and 93 of them were re-measured 2 months later. Childhood trauma questionnaire included 28 items and divided into 5 subscales,named emotional abuse, physical abuse, sexual abuse, emotional neglect and physical neglect. Five grades were adopted in each item, named 1 score: never; 2 scores: occasionally; 3 scores: sometimes; 4 scores: often;and 5 scores: always. Every subscale was varied from 5 to 25 scores, and the total results were in the range from 25 to 125 scores. It was to analyze the internal identical property, reliability of re-measuring, average correlation coefficient among items and correlation coefficient between total score and every subscale. And the analysis was carried on validation factors. MAIN OUTCOME MEASURES: Homogeneity reliability, re-measuring reliability and validity, absolute fit index, relative fit index and parsimony index of childhood trauma questionnaire were involved.RESULTS: Totally 441 pieces of questionnaire were distributed on the spot and 435 pieces of questionnaire with integral and regular answers were collected. Totally 93 pieces of questionnaire were delivered for the second evaluation 2 months later and 93 pieces with regular answers were collected. All of those were used for the evaluation of re-measuring reliability of such measuring table. ① Cronbach α coefficient was 0.64 in Chinese version of childhood trauma questionnaire of and re-measuring reliability was 0.75. Cronbach αcoefficient of every subscale was varied from 0. 16 to 0.65 and re-measuring reliability was in the range from 0.27 to 0.73. The correlation coefficient among items was varied from -0.20 to 0. 44; ②The correlation coefficient between total score and every subscale was varied from 0. 36 to 0.68 and the correlation coefficient among subscales was from-0.01 to 0.39; ③ Indexes of validation factor analysis:The load coefficient of physical neglect was varied from 0.09 to 0. 64 and that in 2 items was less than 0.20. Multiple correlation coefficient was varied from-0. 20 to 0. 82, X2/df was 2.48 and root mean square error of appr0ximation(RMSEA) was 0.06. Added indexes: IFI (0. 76), CFI (0.75) and TLI (0.72) .Parsimony index: PNFI (0.58) and PCFI (0. 67).CONCLUSION: Chinese version of childhood trauma questionnaire provides better reliability and validity. According to validation analysis, except physical neglect, every index tallies with psychometric standards. The results of analysis on normative pathways are satisfactory, which explains that the subscales of such measuring model provide good matching property and conception validity.
7.Experience of Clinical Pharmacists Participating in the Treatment of One Case of Brucellosis

Objective: To explore the approaches and ideas of clinical pharmacists participating in drug therapy of patients with brucellosis. Methods: Clinical pharmacists participated in the clinical consultation for one patient with brucellosis. Clinical pharmacists analyzed comprehensively by asking medical history, occupation and contact history in details, adjusting the treatment scheme and performing medical education etc. Results: The consultation comments and suggestions proposed by clinical pharmacists were recognized by clinics, which played an important role in assisting doctors in the rational drug use and significantly improved the medical treatment. Conclusion: Clinical pharmacists should participate in the individualized medication and help clinicians optimize drug therapy, which can improve the safety and efficacy of medication.
8.The clinical study on the risk of permanent hair dye use for connective tissue disease with positive antinuclear antibodies

Objective To investigate the risk of permanent hair dye use for connective tissue disease ( CTD ) with positive antinuclear antibodies ( ANA ).Methods We conducted a population-based case-control study to analyze 440 CTD patients from the department of our hospital.Four hundreds and forty patients' relatives or friends who were free from any CTD were selected as controls.Patients and controls were matched in sex and age.A face-to-face interview was adopted to finish questionnaires about subjects' demographic data and the use of permanent hair dye.Data about the duration,frequency,times of usage and allergy related to hair dye were collected.Medical history was also reviewed,such as the diagnosis,disease course,primary symptoms,serum tests and organ involvements.Statistical analysis was performed by SPSS 17.0 software using x2 test and Logistic regression.Results The association between the use of permanent hair dye and CTD did not reach statistical significance (OR=1.282,95%CI 0.966-1.700,P=0.085 ),when the relationship of hair dye use with systemic lupus erythematosus (SLE) was tested,the same result (OR=1.092,95%CI 0.795-1.500,P=0.587 ) could be revealed.Conclusion This preliminary study has shown that the use of permanent hair dye couldn't induce the occurrence of CTD.
9.Quality of life of 151 patients with systemic lupus erythematosus

Objective To determine health related quality of life (QOL) of patients with systemic lupus erythematosus (SLE) and to identify the factors affecting QOL. Methods One hundred and fifty-one consecutive patients with SLE were separated into different groups according to the type or stage of disease. Health status were measured by 1 health transition (HT) and 8 composite scales of the medical outcomes study short form-36 (SF-36). The mean scores for each subscale of the SF-36 for patients with SLE in China were compared to those of a health population in China and of foreign SLE patients.The effect of patients′age,sex,education level,disease duration,disease activity (assessed by SLEDAI score),and organ damage(assessed by SLICC/ACR DI) and treatment were analyzed. ANOVA, rank sum test and correlation analysis were applied for statistical analysis. Results Patients with SLE had significantly lower HT and mean scores in each subscale of the SF-36 compared to those of a health population in China,while the scores in such subscales as physical functioning (PF), bodily pain (BP), vitality (VT), role-emotional (RE) and mental health (MH) were higher than those of foreign SLE patients. The mean scores of those patients with lupus nephritis and center neural lupus or in active state were lower than those not or those in steady-going. The SLEDAI score and SLICC/ACR DI were the most important elements that affected the QOL in SLE patients and showed obviously negative correlated with one′s scores in SF-36. The glucocorticoid dose of patients was closely negatively correlated with one′s scores. The latter were also associated with age, education level and disease duration. Conclusion The health status of patients with SLE is mainly associated with disease activity,damage and therapy. The SF-36 is a valid instrument for measuring the health related QOL in SLE.
