1.Changes of T lymphocyte subsets in mice immunized with recombinant Bb-Em Ⅱ/3-Em14-3-3 vaccine of Echinococcus multilocularis

Objective In order to investigate the changes of T lymphocytes subsets in mice immunized with recombinant Bb-Em Ⅱ/3-Em14-3-3.vaccine of Echinococcus multilocularis (Em) and challenged with Em protoscoleces.Methods BALB/c mice were immunized with recombinant Bb-Em Ⅱ/3-Em14-3-3 vaccine by subcutaneous injection,intramuscular injection,nasal mucosa inoculation and oral administration,Bifidobacterium (Bb) and PBS were used as controls.After 12 weeks of immunization,all the mice were challenged with 50 protoscoleces of Em by intraperitoneal injection.Eighteen weeks later,mice were killed to measure alveolar hydatid weight and spleens were taken to separate spenocytes,in which the percentages of CD4+ and CD8+ T cell subsets were determined by flow cytometry.Results Alveolar hydatid weight was (0.77 ± 0.52),(0.87 ± 0.60),(2.17 ± 0.50),(3.06 ± 0.15) g in subcutaneous injection,intramuscular injection,nasal mucosa inoculation and oral administration groups,respectively,which was obviously lower than that in PBS control group [(3.54 ± 0.32) g,P < 0.05 or < 0.01].The level of CD49 T cell subset was (28.2 ± 2.5)%,(25.0 ± 2.7)%,(24.0 ± 1.3)%,(23.0 ± 1.8)% in subcutaneous injection,intramuscular injection,nasal mucosa inoculation and oral administration groups,respectively,which was significantly higher than that in PBS control group [(16.1 ± 2.2)%,all P < 0.01].The level of CD8+ T cell subset in the immunization groups was slightly elevated,but there was no statistical significance between groups (F =1.36,P >0.05).The level of CD4 + T cell subset in subcutaneous injection group was higher than that in intramuscular injection,nasal mucosa inoculation and oral administration groups (all P < 0.05).Conclusion CD4+ T cell subset may play an important role in the protection of mice induced by recombinant Bb-Em Ⅱ/3-Em14-3-3 vaccine and the subcutaneous injection route appears to be a better way for immunization.
2.Epidemiological characteristics of Mycoplasma pneumoniae infection among children in Guilin area during 2015

Objective To understand the infection situation and epidemiological characteristics of Mycoplasma pneumonia (MP) among children with MP infections in Guilin area .Methods The IgM antibody of MP was determined in 1 704 pediatric outpatients and inpatients with respiratory tract infections in our hospital during 2015 by using the MP IgM antibody test reagent kit(colloidal Gold method) .The detection situation was investigated and analyzed .Results The total MP positive rate among children patients with respiratory tract infections in Guilin was 18 .19% ,the female was higher than male with statistical difference (22 .48% vs . 15 .37% ,P<0 .05) .The MP positive rates in children 0 - <1 years old ,1 - <3 years old ,3 - <6 years old and 6 -14 years old were 6 .25% ,18 .59% ,25 .56% and 32 .40% respectively ,and the differences were statistically significant (P<0 .05) .The MP posi‐tive rates in spring ,summer ,autumn and winter were 21 .56% ,15 .29% ,16 .52% and 18 .48% respectively ,and the differences were not statistically significant(P>0 .05) .Conclusion MP is one of the most common pathogens leading to respiratory tract infection in children ,its infection shows gender and age differences .Summer and autumn are mainly epidemic seasons .Children aged 6―14 years old are susceptible to be infected by MP .So it is necessary to strengthen the prevention of MP infection .
3.Development of the electrohydraulic lithotripter

This paper presents the components and the principles of an internal electrohydraulic lithotripter.Cooperating with the cystoscope,the lithotripter introduces the high-pressure discharge pulse into the bladder through the microelectrode.Then the little,easily-moved stone is targeted,and the re-discharge of the lithotripter 5mm away from the stone results in the shock wave to perform lithotritization.
4.Results analysis of urine cellular components tested by Urit-1500 urine analyzer

Objective To explore the clinical accuracy of urine cellular components of RBC,WBC,etc.detected by the Urit-1500 urine analyzer.Methods 1 085 urine samples in this hospital from June to October 2013 were selected and detected by the Urit-1500 urine analyzer and the microscope detection respectively.The results of urine cellular components detected by the urine analy-zer were analyzed.Results In 1 085 urine samples,the coincidence rate of two detection methods in detecting RBC and WBC was 93.8%(1 018/1 085)and 94.0%(1 020/1 085)respectively.In 295 RBC positive urine samples tested by the urine analyzer,the number of negative results tested by microscopy was 49,accounting for 16.6%.In 790 RBC negative urine samples tested by the u-rine analyzer,the number of negative results tested by microscopy was 18,accounting for 2.3%.In 197 WBC positive urine samples tested by urine analyzer,the number of negative results tested by microscopy was 28,accounting for 14.2%.In 888 WBC negative urine samples by urine analyzer,the number of negative results tested by microscopy was 37,accounting for 4.2%.Conclusion De-tecting urine RBC and WBC by using the Urit-1500 urine analyzer should combined with the microscopy and other test indexes and even clinical data to conduct the comprehensive analysis for reducing the misdiagnosis and missed diagnosis as far as possible.
5.Drug Sensitivity and Identification of 393 Strains of Candida

OBJECTIVE To get the message of drug sensitivity and characteristic of Candida infecting patients in our hospital. METHODS The CHROMagar candida color medium was applied to isolate and identify Candida strains.Drug sensitivity was analyzed by the K-B slip diffusion method. RESULTS Totally 393 strains of Candida in our hospital were isolated in the three years,including Candida albicans;C.tropicalis;C.glabrata;(C.krusei);Cryptococcus neoformans and the others;and their infection rate was 63.10%,12.47%,9.41%,8.40%,1.53%,and 5.09%,respectively.Among all strains,156(39.69%) strains were isolated from sputum,(66(16.79%))from stool,45(11.45%) from urine,38(9.67%)from leucorrhea,and 34(8.65%)from throat swabs.Sensitivity results to nine antifungals showed that 5-flucytosine,fluconazole,clotrimazole,amphotericin B,and nystatin had higher drug sensitivity to these fungi. CONCLUSIONS Isolated rate of Candida is higher from clinical samples.Drug sensitivity of different Candida to various antibiotics is different.Culturing and drug sensitivity of Candida should be put on important place and antibiotics should be used reasonably based on drug sensitive test to reduce drug resistance.
6.Distribution and Drug Resistance of Pathogens in Respiratory Tract in Children

OBJECTIVE To investigate pathogenic bacteria isolated from children with respiratory tract infection(RTI) and bacterial resistance to commonly used antimicrobial agents in Guilin area,so as to instruct the rational use of antibiotics.METHODS Routine methods were used to isolate and identify bacteria species and samples of sputum or throat swabs were collected from children with respiratory infection in inpatient departments.The drug susceptibility tests were performed by Kirby-Bauer method.RESULTS Total 733 strains of bacteria were isolated.The Haemophilus influenzae(HI) was the most common(165 strains),followed by Escherichia coli(136 strains) and Streptococcus pyogenes(51 strains).The resistance of HI to trimetoprim-sulfa methoxazole and ampicillin was 59.39% and 36.36%,respectively.Cephalosporins and quinolones were more sensitive to HI.Imipenem,quinolones and amikacin were more sensitive to E.coli as yet.The resistant rate of Streptococcus to penicillin-G,erythromycin,clindamycin and trimetoprim-sulfa methoxazole was 56.79-69.63%.CONCLUSIONS Gram-negative bacilli are the main pathogenic bacteria in childrens respiratory tract infection.The drug resistance is conspicuous.Antibiotics for treating RTI should be selected based on pathogenic bacteria variance and the drug susceptibility test.
7.Epidemiological characteristics of rotavirus infection in infants with diarrhea in Guilin area

Objective To investigate the epidemiologic features of infant rotavirus(RV) diarrhea in Guilin area .Methods The detection of group A rotavirus were performed on 1 300 patients from Guilin 181st hospital by using immune colloidal gold method , and the epidemiological analysis were done according to the detection results .Results In the 1 300 patients ,132 were found rotavir‐us positive ,the total positive rate was 10 .15% .The positive rate of >1-2 years old group was the highest(23 .08% ) ,compared with other age groups(P<0 .05) .In November ,December and January ,the positive rates were 24 .14% ,20 .00% ,23 .53% ,respec‐tively .Compared with the positive rate in other months ,there was statistically significant difference(P<0 .05) .Conclusion Infants of 6 months to less than 2 years old in Guilin have higher detection rate of RV infection ,and the infection peak appears in autumn and w inter .
8.The analysis of curative effect of the modified McBride's procedure on correcting mild-moderate hallux val-gus

Objective To explore the curative effect of clinical application of modified McBride's procedure on correcting mild-moderate hallux valgus.Methods We had retrospectively assessed 32 patients(52 feet)treated with the procedure of modified McBride's procedure.All patients were followed up,the follow -up period from 6 months to 6 years(3.1 years on average).There were 2 male(4 feet)patients and 30 female(48 feet)patients in this group.The average age at the time of surgery was 41.6 years old(from 21 to 59 years).Results According to the forefoot score of American Orthopedics Foot and Ankle Society(AOFAS),29 feet(55.8%)were excellent,17 feet (32.7%)were good,And the rate of excellent and good was 88.5%.The average correction of HVA and IMA was 13.68°and 3.24°respectively compared with the preoperative cases.Conclusion This procedure can not only effec-tively reduce the increased hallux valgus angle,but also narrow the angle between the 1st and 2nd metatarsal,relocate the sesamoid system,and effectively relieve patients'pain.This approach is of minor side effects to bone and joint structure,and of rapid recovery.It is a preferential choice to treat mild-to-moderate hallux valgus deformity.
9.Clinical application of detection of procalcitonin and high sensitivity C-reactive protein in the early diagnosis of bloodstream infection

Objective To investigate the clinical value of detection of procalcitonin(PCT ) and high sensitivity C-reactive protein (hs-CRP) in early diagnosis of bloodstream infection(BSI) .Methods The serum levels of PCT and hs-CRP of 58 BSI patients(test group) and 58 non BSI patients(control group) were detected .The early diagnosis value of PCT and hs-CRP was evaluated by ROC curve .Results There were significant difference between the serum PCT and hs-CRP levels of test group and control group(P<0 .05) .The ROC AUC of PCT and hs-CRP were 0 .902(95% CI:0 .850-0 .955) and 0 .706(95% CI:0 .611-0 .801) ,respectively . With 2 .24 ng/mL being the diagnostic cut-off value ,the diagnostic sensitivity ,specificity ,positive predictive value ,and negative pre-dictive value of PCT were 77 .6% ,91 .4% ,90 .0% ,and 80 .3% ,respectively .With 64 .83 mg/L being the diagnostic cut-off value ,the di-agnostic sensitivity ,specificity ,positive predictive value ,and negative predictive value of hs-CRP were 74 .1% ,62 .1% ,54 .4% ,and 59 .5% , respectively .Conclusion Detection of serum PCT and hs-CRP has important clinical value in early diagnosis of BSI .
10.Genome-wide analysis of DNA 5-hydroxymethylcytosine in uremia patients using hMeDIP-chip

Objective To investigate the role of the 5-hydroxymethylcytosine (5-hmC) DNA modification in the onset of systemic lupus erythemosus (SLE),we compared tihe levels 5-hmC between SLE patients and normal controls.Methods With informed consent,whole blood was obtained from patients,and genomic DNA was extracted.Using hMeDIP-seq analysis and validation by quantitative real-time quantitative polymerase chain reaction (RT-PCR),we identified the differentially hydroxymethylated regions that were associated with SLE.Results There were 1 701 genes with significantly different 5-hmC levels at the promoter region in the SLE patients compared with the normal controls.The CpG islands of 3 826 genes showed significant difference at 5-hmC levels in SLE patients compared with the normal controls.Out of the differentially hydroxymethylated genes,three were selected for validation,including TREX1,CDKN1A,and CDKN1B.The hydroxymethylation levels of these three genes were confirmed by quantitative RT-PCR.Conclusion Our studies indicate that there are significant alterations of 5-hmC in SLE patients;these differentially hydroxymethylated genes may contribute to the pathogenesis of SLE.Such novel findings show the significance of 5-hmC as a potential biomarker or promising target for epigenetic-based SLE therapies.
