2.Effects of artesunate on improving airway remodeling in asthmatic young rats

Objective To investigate the effect of artesunate on airway remodeling induced by ovalbumin in asthmatic young rats and its mechanism.Methods SD rats were randomly divided into normal group,model group(ovalbumin activation),positive control group(on the basis of model group,injected with 0.2 mg·kg-1 dexamethasone),low-dose group(on the basis of model group,injected with 10 mg·kg-1 artesunate),high-dose group(on the basis of model group,injected with 20 mg·kg-1 artesunate)and agonist group(on the basis of model group,injected with 20 mg·kg-1 artesunate and 100 mg·kg-1 nigerin sodium salt),with 10 rats in each group.Airway remodeling related indexes were detected after treatment.Western blot and real-time fluorescence quantitative polymerase chain reaction were used to detect Nod-like receptor protein 3(NLRP3)pathway-related protein and mRNA expression;enzyme-linked immunosorbent assay were used to detect inflammatory factors expression level,and the classification and count of inflammatory cells in alveolar lavage fluid were detected by Wright's-Giemsa Staining.Results The mRNA expression levels of NLRP3 in normal group,model group,positive control group,high-dose group and agonist group were 1.00±0.10,2.36±0.26,1.08±0.11,1.33±0.12,2.14±0.26,respectively;cysteine aspartate proteinase 1(caspase-1)mRNA expression levels were 1.00±0.13,1.92±0.22,1.22±0.12,1.44±0.10,1.82±0.14,respectively;interleukin-17(IL-17)inflammatory factor expression quantity were(22.41±1.15),(56.74±6.54),(28.72±2.75),(32.69±3.73),(58.40±4.46)pg·mL-1;neutrophil count were(4.04±0.32)×106,(12.70±1.05)×106,(4.53±0.30)×106,(4.67±0.18)× 106,(10.19±0.58)× 106 cell·mL-1.Compared the model group with the normal group,the positive group,the high dose group compared with the model group,the agonist group compared with the high dose group,the differences of the above indicators were statistically significant(all P<0.05).Conclusion Artesunate can significantly improve airway remodeling in ovalbumin induced asthmatic pups,which may be achieved by inhibiting the NLRP3/caspase-1/interleukin 1 β(IL-1β)inflammatory pathway.
3.The prevalence characteristic and prevention strategy of Japanese B Encephalitis in Henan province.

OBJECTIVETo analyze the prevalence trend, prevalence characteristics and influence factors of Japanese B Encephalitis (JE) in Henan province.
METHODSThe data that of 64 401 JE patients in Henan from 1980 to 2008 were statistically analyzed by SPSS12.0 and EXCEL2003 software. Luoshan, Xinan, Xihua, Deng county and Hua county were chosen as monitoring sites. The mosquito specimens were gathered with the artificial hour method and the mosquito curtain method, the mosquito density was calculated one time each ten day period from May to July. At the same time, 30-40 newborn pig blood samples were gathered each ten-day period and the pig serum JE IgG antibody was detected by ELISA method.
RESULTSThe Cumulative incidence of JE was 64 401 cases in Henan province from 1998 to 2008, the range of incidence rate was from 0.34/100 000 (315/93 599 969) to 6.72/100 000 (5246/78 076 567); The average incidence of JE was 4.39/100 000 (3530/80 381 469) from 1980 to 1994; The average incidence of JE was 0.86/100 000 (811/94 217 549) from 1995 to 2008; In 2008, the incidence rate reached the lowest point for 0.34/100 000 (315/93 599 969); The incidence occurred mainly in July-September, accounting for 89.40% of the total cases (57 572/64 401); the patients were concentrated mainly in 5 cities, which were Xinyang, Nanyang, Zhumadian, Zhoukou, Luoyang, accounting for 81.02% (52 175/64 401). The 0 - 14 years old age group was the dominant group (79.01%, 50 884/64 401). In Luoyang city, incidence of >/= 15 years old group was significantly increased (57.83%, 2120/3666), the constitution of JE incidence were significantly different between 0 - 14 years old group and >/= 15 years old age group (chi(2) = 2705.32, P < 0.05) in Henan province and Luoyang city. The different density of the mosquitoes and the different positive-times for 50% of the antibodies of JE in piglets on the monitor sites showed the intensity of JE disease.
CONCLUSIONThe incidence of JE showed a decreasing trend, seasonal, regional characteristics and age distribution difference in Henan province. The monitoring of host animal pig JE antibody level and the medium mosquito density may forecast the JE prevalence tendency. To control the incidence in the younger groups in Henan province, older age group in Luoyang city and high-incidence areas, it is important to strengthen the monitoring and forecasting measures to prevent JE in Henan province.

Objective To explore a kind of simple and high efficient approach to differentiate human embryonic stem cells(hESCs)into neural stem cells(NSCs).Methods hESCs were cultured in bacterial culture dish filled with serum free medium to gain embryoid bodies.Then the mature embryoid bodies grew in the special medium including B27 and noggin by adherent culture to differentiate into NSCs. Results The hESCs kept floating in the bacterial culture dish and growing all the time and then formed mature embryoid bodies 7 to 10 days later.The embryoid bodies could be differentiated into highly pure (96.4%)nestin positive cells.And these cells were differentiated into all kinds of neural cells if cultured further.Conclusions This kind of method is less time-consuming,cheaper,and more efficient than those of the results in literatures reported.It affords very good source of seed cells for cell transplantation therapy in the future.
6.The effects of safflower injection on lipid peroxidation level and expression of heme oxygenase-1 of the rat liver with hypoxia and hypercapnia.

AIMTo investigate the changes of lipid peroxidation level and expression of heme oxygenase-1 of the rat liver with chronic hypoxia and hypercapnia, and the effects of Safflower injection (a compond of Chinese Traditional medicine).
METHODSThirty male SD rats weighing 180 approximately 220 g were divided into three groups (n=10): control group (N group), chronic hypoxia and hypercapnia for four weeks group(F group), and Safflower injection group (H group). SOD and MDA in liver tissue were measured by spectrophotometric method. And methods Immunohistochemical assay was used to detect the distribution of HO-1 protein. Pathological changes in liver tissues were observed in HE staining section. The mRNA expressions of HO-1 in liver were detected by semi-quantitative RT-PCR.
RESULTSThe activity of SOD of the liver in F group were significantly lower than those in N group, and the content of MDA were significantly higher. The activity of SOD of the liver in H group were significantly higher than those in F group, and the content of MDA were significantly lower. In F group there were multiple dispersed immunoreactivity cells in liver. And compared to those in F group, the immunoreactivity cells were significantly decreased in H group. HE staining revealed that there were many hepatocytes with obvious adipose degeneration. Hepatic pathological damage in H group was slighter than that in F group. The expression of HO-1 mRNA of the liver in F group were significantly higher than those in N group (P < 0.01), and those in H group were significantly lower than those in F group (P < 0.01) .
CONCLUSIONChronic hypoxia and hypercapnia increases the level of oxidative stress. Safflower injection have a protective effect, maybe because of the accommodation of the expression of HO-1 of the liver and the elimination of free radicals.

Objective To analyze the epidemiological characteristics and risk factors of Japanese encephalitis (JE) in Henan province.Methods Epidemiological characteristics and related factors of JE cases in Henan province from 2006 to 2010 were analyzed with descriptive epidemiological method and JE IgM antibodies of the serum or cerebrospinal fluid of the cases were detected by ELISA.Results 3099 cases were reported in Henan province from 2006 to 2010.The incidence fluctuated from 0.39/100 000 to 1.08/100 000,and the incidence rate was decreasing.Patients were concentrated mainly in Xinyang,Nanyang and Luoyang cities,which accounted for 60.12% of the total.The peak season was in July-September,accounted for 93.26% of all the cases.Most cases were in 0-14 year old (83.61%) in the whole province.However,in Luoyang city,number of cases in the ≥ 15 year old group,had an obvious increase (57.63%).Compared with the provincial data,significant difference was found between the two ≥ 15 year old groups (x2=330.0341,P<0.05)statistically.20.23% of the JE cases were vaccinated but 75.54% of them did not complete the whole course.79.77% of the cases were not sure if they had received the vaccination.42.98% of the JE cases were confirmed through laboratory tests but other 45.95% of them were clinically diagnosed,with the rest 11.07% were suspected cases.Conclusion The distribution of JE cases showed seasonal,regional characteristics and crowd differences in Henan province.Basic immunization and the program on strengthening immunization of JE should be further standardized.The laboratory diagnosis rate of JE cases should be further improved.
9.Expression and significance of PTEN/PI3K/AKT/mTOR signaling pathway in extranodal NK/T cell lymphoma

Objective To investgate the pathogenesis of PTEN/PI3K/AKT/mTOR signaling pathways in nasal type extranodal NK/T cell lymphoma(ENKTCL-N).Methods The expression of PTEN,p-AKT,p-4EBP1 and p-mTOR in 51 patients with ENKTCL-N and 20 patients with reactive proliferative lymph nodes was detected by immunohistochemistry EnVision.The clinical data were collected and the clinical features,curative effect and prognosis were analyzed retrospectively.Survival analysis was performed by Kaplan-Meier method.Results The positive rate of PTEN in ENKTCL-N patients was lower than that in patients with reactive hyperplastic lymph nodes,and the positive rates of p-AKT and p-4EBP1 were higher than those in patients with reactive hyperplastic lymph nodes,and the difference was statistically significant(P<0.05);The positive rate of p-mTOR in ENKTCL-N patients was higher than that in patients with reactive hyperplastic lymph nodes,but the difference was not statistically significant (P>0.05).The survival rate of PTEN-positive patients was higher than that of PTEN-negative patients,and the difference was statistically significant(P<0.05).Conclusion PTEN/PI3K/AKT/mTOR signaling pathway is abnormally active in ENKTCL-N,which may be related to its pathogenesis;PTEN-positive patients may have better prognosis.
10.Effect of Sinisan on ultra structure of hippo-campus in the intervene rats by post traumatic stress disorder

Objective To study the effect of Sinisan on the ultra structure of hippo-campus to the intervention in rats with post-traumatic stress disorder(PTSD).Methods SD rats were divided equally into 5 groups,each group had ten rats:blank control group,model group,negative control group,positive control group and experimental group.The blank control group did not copy the model,the normal feeding.Model group,repetitive post-traumatic stress disorder model was induced by current stimulation,but not treated.Negative control group,equal volume of 0.9% NaC1.Positive control group (paroxetine hydrochloride 0.42 mg · mL-1) and experimentalgroup (Sinisan,containing crude drug 0.24 g · mL-1).It given to drugs 1 h before the model establishment.The rats were administered with 10 mL · kg-1,once a day,for a total of 7 d.In each group,rats was cardiac perfusion and the hippo-campus tissue was collected.The changes of ultra structure of CA1 and CA3 area in hippo-campus with rats were observed by transmission electron microscope.Results In the blank control group,the CA1 and CA3 neurons in the hippocampus were rich in cytoplasmic organelles,and the mitochondria were round or long,the structure of the mitochondrial cristae was clear,the rough endoplasmic reticulum was like a cord like distribution,and the ribosome was abundant.Compared with the blank control group,the organelle of CA1 and CA3 area in hippo-campus of the model group were significantly damaged,the cytoplasm was open,the mitochondria were swollen,the mitochondrial membrane and cristae disappeared,and the rough endoplasmic reticulum was dilated.The results showed that the damage of the organelles in the hippocampal neurons of rats was induced by the electric shock.The model group was similar to the negative control group,which indicated that there was no significant effect on the stress of rats after intragastric administration.The intracellular organelles in the CA1 and CA3 neurons in the hippocampus of the positive control group and the experimental group were significantly recovered,and the changes in the two groups were similar.These results suggest that both paroxetine hydrochloride and Sinisan can significantly improve the structure of CA1 and CA3 neurons in the hippocampus of PTSD rats.Conclusion Snisan as traditional Chinese medicine compound can significantly improve the hippocampus of rats with PTSD CA1 and CA3 neurons.
