1.The characteristics of invasion and metastasis in malignant melanoma and apoptosis

Impaired ability to undergo apoptosis in response to a wide range of external stimuli acquires melanomas a selective advantage for progression and metastasis as well as their notorious resistance to therapy. Changes at genomic, transcriptional and post-translational levels of G-proteins and protein kinases (Ras) , their transcription factor effectors (c-jun,ATF2, stat3 and NF-?B) and upregulator of antiapoptosis gene (Bcl-2、Bcl-X_L、survivin、ML-IAP)affects apoptosis-induced of TNF, Fas and TRAIL receptors, which play important roles in acquiring melanoma's resistance to apoptosis. The complexity of the molecular variants involved in signal transduction along apoptotic pathways suggests that the cell may have a variety of possibilities for regulating apoptosis and generating apoptosis deficiency.
2.The regulation of ?-melanocyte-stimulating hormone to melanogenesis of skin

Cutaneous dyschromia is a common problem of plastic and cosmetic surgery. There are not good methods to resolve this problem in present. The mechanisms of skin pigmentation process are not well understood. It is now recognized the correlation between melanogenesis and alpha-melanocyte-stimulating hormone. Alpha-msh enhances activity and quantity of tyrosinase as well as stimulates melanogenesis, specifically to stimulate eumelanin synthesis by activating the melanocortin-1 receptor (MC-1R) on melanocytes. Alpha-msh stimulates melanocyte prolixferation, formation of dendricity, and transfer of melanosomes to keratinocytes, It also protects melanocytes from the damaging effects by immunomodulation. Also, alpha-msh interact with other factors to affect pigmentation indirectly. Agouti signaling protein(ASIP) is a endogeneous antagonist of alpha-msh can induces pheomelanin synthesis by competing with alpha-msh for binding to the MC-1R.Follwing better understood the mechanisms of skin pigmented process, it will provide effective methods to resolve acquired human cutaneous hyperpigmentation and hypopigmentation.

7.The practice of situational teaching in the course of “Introduction to Medical Humanities” from the perspective of narrative medicine

The situational teaching of the course of introduction to medical humanities has some problems, such as low participation, poor communication, and lack of humanities thought. By reading text and creating text, narrative medicine can make students deeply participate in the scene, provide an intro-spective opportunity of being close to the patients, and make students better redefine the story of the other so as to be able to help patients to take a positive view of the disease. Therefore, by using the method of reading text and creating text of narrative medicine, this paper has studied the “4+3” situational teaching model reform of the course of “introduction to medical humanities”, analyzed the present situation and achievements and pointed out that the teaching should pay attention to the key elements, which is “to build a correct value view of medical humanistic, strengthen the emotional education of humanistic care, as well as culture humanistic knowledge and culture humanistic ability”, and it has also put forward some sugges-tions on improving the teaching level of“Introduction to medical humanities”in the future.

10.Developmental toxicity of 2,4-dinitrochlorobenzene in zebrafish embryos

Objective:
To investigate the developmental toxicity of 2,4-dinitrochlorobenzene in zebrafish embryos.
Methods:
AB wild-type male and female zebrafish were selected to mate and spawn, then the eggs were cultured with Holt buffer solution. Six dose groups ( 0.4, 0.8, 1.0, 1.2, 1.6, 2.0 μg/mL ), a solvent control group and a cosolvent control group, were set up with 20 embryos each. Malformations and death of embryos were observed at 48, 72 and 96 hpf ( hours post fertilization ), the mortality and 50% lethal concentration ( LC50 ) were also calculated.
Results:
At 48, 72 and 96 hpf, the LC50 of 2,4-dinitrochlorobenzene on zebrafish embryos were 1.668, 1.043 and 0.895 μg/mL, respectively, with a downward trend. After 72 hpf, when the concentration reached 2.0 μg/mL, all the zebrafish died. In the range of 0.4-2.0 μg/mL, the mortality of zebrafish at 48, 72 and 96 hpf increased with the increase of 2,4-dinitrochlorobenzene concentration ( all P<0.05 ); the malformation rate of zebrafish embryos at 48 hpf increased with the increase of 2,4-dinitrochlorobenzene concentration ( P<0.05 ). Zebrafish embryos exposed to 2,4-dinitrochlorobenzene led to yolk sac edema, pericardial edema and spinal curvature.
Conclusion
2,4-dinitrochlorobenzene can affect the development of zebrafish embryos, which will lead to lethal and teratogenic effects.
