2.Establishment of HPLC fingerprints of Qingfeiyucuo Pills and determination of four constituents

AIM To establish the HPLC fingerprints of Qingfeiyucuo Pills (an agent for the management of acne,containing Scutellariae Radix,Eriobotryae Folium,Paeoniae Radix Rubra,etc.) and to determine the contents of four constituents.METHODS The analysis of methanol extract of Qingfeiyucuo Pills was carried out on a 35 ℃ thermostatic Agilent HC-C1s column (4.6 mm ×250 mm,5 μm),with the mobile phase comprising of acetonitrile-0.4% phosphoric acid flowing at 0.8 mL/min in a gradient elution manner,and the detection wavelength was set at 230 nm.RESULTS Eleven common peaks with the similarities of more than 0.9 were observed in the HPLC fingerprints of ten batches of samples.Paeoniflorin,hesperidin,baicalin and salvianolic acid B showed good linear relationships within the ranges of 0.08-0.25 μg,0.07-0.22 μg,0.2-0.60 μg and 0.06-0.18 μg,whose average recoveries were 98.7%,96.9%,97.4% and 98.2% with the RSDs of 0.98%,1.09%,0.82% and 1.66%,respectively.CONCLUSION This sensitive and specific method can be used for the quality control of Qingfeiyucuo Pills.
3.The development and changes of pressure of putrefactive gas in cadaveric enterocelia in spring and its forensic application.

OBJECTIVE:
To study the development and changes of pressure of putrefactive gas (PPG) in cadaveric enterocelia in spring, and to explore its application in estimation of postmortem interval (EPI).
METHODS:
57 goats were divided into 2 groups according to means of death, on land or in water. Celiac PPG were observed timely and systematically.
RESULTS:
The development of PPG in cadaveric enterocelia, which can be divided into raising phase, peak phase, and declining phase was observed, and a model to estimate postmortern interval by changes of PPG was founded.
CONCLUSION
Measuring PPG in cadaveric enterocelia could be used in forensic EPI.

4.Application of associated symptom and sign-based acupoint selection in acupuncture-moxibustion treatment.

Based on traditional acupuncture-moxibustion treatment ideas, with differentiation of channels and collaterals as main part and feature, the important role of associated symptom and sign-based acupoint selection in acupuncture-moxibustion treatment is explained from angles of philosophy and medicine. Combined with clinical experience of acupuncture and moxibustion, the category of associated symptom and sign-based acupoint selection is explained in detail to make sure the accuracy of acupuncture-moxibustion differentiation. It could show uniqueness and advantage of theory and clinic in acupuncture-moxibustion and provide theoretical references in making acupuncture-moxibustion prescription to improve effectiveness of clinical treatment.

5.Effect of Ang II on PDGF receptor expression in vascular smooth muscle cell.

AIMTo study the effect of angiotensin II (Ang II) on platelet-derived growth factor(PDGF) receptor expression in vascular smooth muscle cell(VSMC).
METHODSRestenosis model was established by balloon injury in rat aorta. The morphologic change and level of Ang II were measured at 14th day after operation. The expression of PDGF-beta receptor was detected by Western blot. The cultured VSMC pretreated with or without losartan were treated with Ang II.
RESULTSCompared with sham group, the sections of injured aorta showed marked intimal thickening with large numbers of VSMCs proliferation throughout intima and media, the level of Ang II obviously increased by 78.8% (P < 0.05), the expression of PDGF-beta receptor significantly increased by 83.9% (P < 0.05) at 14th day after operation. The expression of PDGF-beta receptor in cultured VSMC treated with Ang II was higher than that of control group (P < 0.01). The effect of Ang II was inhibited remarkably by pretreatment with losartan.
CONCLUSIONAng II can stimulate PDGF receptor expression in VSMC, it may be an important mechanism of Ang II-induced VSMC proliferation.

6.Influence of the number of removed axillary lymph nodes on the prognosis of node-negative primary breast cancer.

OBJECTIVETo analyze the relationship between the number of removed axillary lymph nodes and prognosis of axillary node-negative breast cancer.
METHODSThe clinicopathological data of 655 patients with breast cancer were analyzed retrospectively. The disease-free survival curves were generated according to the number of removed axillary lymph nodes using Kaplan-Meier plots. The correlation between the co-variables and rate of breast cancer-related events was analyzed using Cox model.
RESULTSThe overall five year-disease free survival rate of the 655 cases was 94.4%. The rate of patients with lymph node number ≤ 12 was 90.3%, and that of lymph node number > 12 was 96.5%, with a statistically significant difference (P = 0.009). Significantly less breast cancer-related events were observed in patients with lymph node number > 12 (15/426, 3.5%) than that in patients with lymph node number ≤ 12 (22/229, 9.6%) (P = 0.009).
CONCLUSIONSWhen axillary node dissection is indicated, dissection of lymph nodes >12 leads to much less breast cancer-related events than that in patients with dissected lymph node ≤ 12. The more lymph nodes are dissected, the more accurate prognosis can be estimated.

7.Treatment of spondylolysis and spondylolisthesis with posterior STB thoracolumbar transpedicular screw-plate spine fixation system.

OBJECTIVETo observe the clinical effect of a self-designed posterior STB thoracolumbar transpedicular screw-plate system in the treatment of spondylolysis and spondylolisthesis.
METHODSThis STB screw-plate system is developed with titanium alloy (TC4, Ti6Al4V). During December, 1999 and January, 2001, this system was applied in 51 cases, including lumbar spondylolysis and spondylolisthesis (36 cases), degenerative lumbar instability (15 cases). The patients were aged 22 - 78 years, mean age: 47.5 - years; among them there were 14 male cases, 37 female cases; There were 15 cases degenerative spondylolisthesis with grade I-II slip, 36 cases of spondylolysis and spondylolisthesis, including 34 cases with grade I-II slips and 2 cases with grade III slips; single level of 33 cases, two levels 3 cases. Decompression was performed for the 43 cases with interbody fusion using iliac crest or bilateral-lateral fusion using demineralized bone matrix and 8 cases were fixed with demineralized bone matrix fusion without decompression.
RESULTSAll the operations heeded about 60 - 120 minutes, and during the operations bleeding amounts were 200 - 500 ml. The cases completely recovered to work or normal action after 3 months of surgeries. 46 of all cases were achieved to satisfactorily reduction and clinical effect, 5 cases of grade II-III remained incomplete reduction (grade I slip).
CONCLUSIONThis STB system has advantages of biomechanical stability and reduction capability and its effect in clinical application is undoubtedly positive.

The cardiovascular circulation feedback control treatment instrument (CFCTI) is an automatic feedback control treatment system, which has the function of monitoring, alarming, trouble self-diagnosis and testing on the line in the closed loop. The instrument is designed based on the successful clinical experiences and the data are inputted into the computer in real-time through a pressure sensor and A/D card. User interface window is set up for the doctor's choosing different medicine. The orders are outputted to control the dose of medicine through the transfusion system. The response to medicine is updated continually. CFCTI can avoid the man-made errors and the long interval of sampling. Its reliability and accuracy in rescuing the critical patients are much higher than the traditional methods.

10.Determination of bis (p-fluorobenzyl) trisulfide and bis (p-fluoro-benzyl) disulfide in lungs of rat by HPLC.

OBJECTIVETo establish an HPLC method for analysis of bis(p-fluorobenzyl) trisulfide(BFTS) and bis(p-fluorobenzyl)disulfide(BFDS) in the lungs of rat.
METHODS5.0 ml extract solvent (n-hexane: isopropyl alcohol=95:5, v/v) and 20 microl of 11.50 microg/ml dibenzyl disulfide (internal standard) were added to 0.2 g lung sample followed by homogenization. After centrifugation, 4.0 ml of supernatant was separated and vaporized to dryness, and the residue was reconstituted in mobile phase for HPLC analysis. The HPLC analysis was performed on an SB C18 column using acetonitrile and water (65:35, v/v) as mobile phase with a flow rate of 1.0 ml/min with UV detection at 220 nm.
RESULTThe calibration curves for BFTS and BFDS in sample were linear over the concentration ranges of 0.04712-14.78 microg/g(r=0.999) and 0.04831-23.96 microg/g(r=0.999), respectively. The limits of quantification were 0.04712 microg/g and 0.04831 microg/g for BFTS and BFDS, respectively. The assay recoveries for BFTS and BFDS ranged from 95.71%-107.2% and 90.00%-110.5%, respectively. The precisions were obtained with RSD of <10%. The developed method was successfully applied to study the content of BFTS and BFDS in the lungs of rats after intravenous injection of 12.5 mg/kg BFTS.
CONCLUSIONThe method developed is simple, selective, repeatable and accurate, which can be applied to study the tissue distribution of BFTS and BFDS.
