Objective To screen the target genes that contribute to cisplatin resistance in ovarian cancer treatment. Methods Gene expression and methylation profiles of ovarian cancer cells that were sensitive or resistant to cisplatin with accession number GSE15709 were downloaded from GEO database. Differential expressed and methylated genes were identi?fied through associating packages in R. DAVID database to screen the enriched GO terms and pathways of the different ex?pressed genes between A2780 and A2780/DDP. Gene Set Enrichment Analysis (GSEA) of different gene was performed against DAVID database. Genes that exhibited difference in both expression and methylation profiles between the two types of ovarian cancer cells as well as genes that present contradictory profile between expression and methylation were verified via qRT-PCR. Results We found 416 different expressed genes and 281 methylated genes between the two types of ovari?an cancer cells respectively. These differential genes were rich in pathways of cell cycle, DNA replication, nucleus division , p53 signaling , and negative regulation of protein modification process etc. Four genes demonstrated contradictory profile be?tween expression and methylation in the two types of ovarian cancer cells and were verified by qRT-PCR. Conclusion Combination of bioinformatics and molecular biology is useful in the identification of target genes that contribute to resis?tance of cisplatin in ovarian cancer treatment and further reveal molecular mechanism behind it.

Objective To investigate effects of celecoxib on the expression of cyclooxygenase-2 (COX-2), apoptotic protease activation factor-1 (Apaf-1) and function of mobility in rat model of severe craniocerebral trauma. Methods For?ty-eight adult male Wistar rats were randomly divided by random number table into four groups. Normal group was given no manipulation. Sham group was given scalp incision and sutured. The severe closed craniocerebral injury model was estab?lished via Foda method in rats of injury group. Treatment group was given intraperitoneal injection of celecoxib [ 250 mg/(kg·6 h)] on the basis of injury group. The intraperitoneal injection of same volume of normal saline was given in the other three groups. Samples were taken altogether after 72 hours. Changes of COX-2 and Apaf-1 were detected by immunohistochemis?try and Western blot assay. Ten days after the restoration, six rats were taken from each group for assessing neurological im?pairment scale (NSS). Results The expression levels of COX-2 and Apaf-1 were significantly higher in injury group than those of other groups. The expression levels of COX-2 and Apaf-1 were significantly lower in treatment group than those of injury group but the levels were significantly higher than those of sham group and normal group (P < 0.05). NSS scores showed that rats in treatment group improved mobility compared with that of injury group (P<0.05), but there was difference compared with Sham group and control group (P<0.05). Conclusion Celecoxib, with its specific inhibitoty effect on pro?tein COX-2, can effectively reduce inflammatory reactions lower the expression of Apaf-1 and reduce apoptosis of neurons, improving the prognosis of dysfunction of mobility after craniocerebral injury.

Objective To study the effect of celecoxib on learning and memory function,cyclooxygenase(COX-2) and the apoptotic protease-activating factor-1(Apaf-1) protein expression after traumatic brain injury in rat.Methods A total of 72 adult male Wistar rats were equally and randomly divided into the normal control group,sham operation group,trauma group and Celecoxib treatment group.Postoperative 72 h-reperfusion was performed for taking brain specimens.The immunohistochemical method and Western blot were used to respectively detect COX-2 and Apaf-1 protein expression change;the Morris water maze test was adopted to detect the learning and memory function on preoperative 5 d and at postoperative 72 h.Results The COX-2 and Apaf-1 protein expression in the trauma group was significantly higher than that in other groups (P＜0.05),and the protein expression in the treatment group and trauma group was decreased,but still higher than that in the sham operation group and normal group(P＜ 0.05);in the Morris water maze test,the prolongation of escape latency time in the trauma group was maximal among 4 groups (P ＜0.05),but the treatment group had a shorter time compared with the trauma group (P＜0.05).Conclusion Craniocerebral trauma can cause different degrees of learning and memory dysfunction,and COX-2 inhibitor celecoxib can downregulate the expression of COX-2 and Apaf-1 protein,inhibit inflammation reaction and cellular apoptosis,and improve the learning and memory dysfunction after traumatic brain injury.

Objective To develop a real-time electronic nursing records quality control system based on No.1 Military Medical Project to solve the existing problems in range,representativeness of detection results,information feedback,information storage and etc.Methods An integrated platform was implemented for information acquisition,integration and display on the bases of No.1 Military Medical Project as well as the interfaces with information management systems for electronic nursing records,clinical laboratory examination,medical examination,imaging and etc,which had the functions of auto monitoring of timeliness and logic of electronic nursing records,manual quality control of the connotation as well as the feedback,statistics and analysis on the problems.Results The system behaved well in stability,compatibility,and could be used for real-time and dynamic control of electronic nursing records,timely information feedback and statistical analysis.Conclusion The system contributes to the quality control of electronic nursing records,and is of great significance for its enhancement.

Objective To investigate the outcome of damage control sugery in patients with acute superior mesenteric occlusion (ASMO).Methods Seventeen patients with acute superior mesenteric occlusion from June 2008 to May 2012 were reviewed retrospectively.Diagnosis was confirmed by CT and/or DSA.Patients were divided into two groups:seven patients in damage control group (shortening surgical time,bowel exteriorization,temporary abdominal closure) ; ten patients in conventional surgical treatment group.The treatment outcomes of the two groups were compared.The survival of all patients was followed up for 6 months to 5 years at outpatient clinic.Results The overall mortality rate was (5/17) and surgical complications rate was (8/17).Operation time was (97 ±42) min and (236 ± 137) min(t =-4.72,P ＜0.01,P ＜ 0.05) respectively in damage control group and conventional surgical treatment group.Intestine resection length was (114 ± 94) cm and (229 ± 93) cm (t =-2.49,P =0.03,P ＜ 0.05) respectively.Complications rate was (1/7) and (7/10) (Fisher's dxact test P =0.05,P ≤0.05) respectively; mortality was (1/7) and (4/10) (Fisher's dxact test P =0.34,P ＞ 0.05) respectively.Conclusions Although damage control does not significantly reduce mortality,but the rational use of damage control can effectively reduce operation time,shorten intestine resection length and decrease surgical complications.

AIM: To establish the quality standard for Baixuan Xitare Tablets (Alone, Herba Euphorbiae Humifusae, Fructus Chebulae, etc.) METHODS: Herba Euphorbiae Humifusae and Scammonia resin were identified by TLC. The content of aloin was determined by HPLC. RESULTS: Herba Euphorbiae Humifusae and Scammonia resin could be identified by TLC. Aloin showed a good linear relation in a range of 0.9360?g～ 2.184?g, r=0.9995(n=5). The average recovery was 98.54% and RSD was 0.67%(n=9), respectively. CONCLUSION: The method is simple, accurate and specific and can be used for the quality control of Baixuan Xitare Tablets.

Oral and maxillofacial surgery is one kind of special subjects involving multiple clinical disciplines.The postgraduate training is different from other professional postgraduate training model,which emphasizes the cultivation of students,should be first medical students,then dentist.In new situation,we advance the experiences in exploring how to cultivate high quality postgraduate of oral and maxillofacial surgery.

Objective To analyse the long-term result and prognosis of 18 FDG PET/CT positioning three - dimensional conformal radiotherapy ( 3 DCRT ) for stage Ⅲ non - small cell lung cancer. Methods Sixty-four cases with stage Ⅲ non-small cell lung cancer (clinical stage Ⅲa- Ⅲb ) were randomly divided into two groups: PET/CT positioning three-dimensional conformal radiotherapy group (PET/CT group) and the conventional CT positioning three-dimensional conformal radiotherapy group (conventional CT group). In the PET/CT group, the target volume and critical organs were sketched according to PET/CT after fusion of the PET and the CT images; the treatment plan was worked out, then conventional fractionated 3DCRT ( total dosage around 40 Gy) followed by field-shrinked radiotherapy to a total dose of 65 Gy or sowas performed ;in the conventional CT group, the target volume and critical organs were sketched according to CT and 3DCRT were performed to the same total dose; All cases were treated with the TP scheme (paclitaxel 175 mg/m2,d1 ,cisplatin 40 mg,d2-4) adjuvant chemotherapy for 6 cycles after the radiotherapy. Results The followup rate was 100%. The number of patients who completed the 1-,2-and 5-year follow-up were 40,20 and 11 respectively ;The number of patients of the PET/CT group and conventional CT group were 23 and 17,11 and 9,7 and 4 respectively. Target volumes of 13 cases in the PET/CT group were changed. The complete remission and partial remission rates of the two groups were 13% 、66% and 19% 、53% (x2 = 0. 33, P =0. 564), respectively. The 1-,2-and 3-year local control rates of the PET/CT group and conventional CT group were84 % 、66% 、53 % an d72% 、59% 、44% ( x2 = 2.36, P = 0. 124 ) respectively. The1 -, 2-and 3-year survival rates were 72% 、34% 、22% and 53% 、28% 、13% (x2 =2. 46,P =0. 117) respectively. The level-1 and level-2 lungs' and trachea's late radiation injury of the PET/CT group and the conventional CT group were 28% and 53% ( x2 = 4. 14, P = 0. 042 ), respectively. The hilar and mediastinal lymph node recurrence rates of the PET/CT group were lower than those of the conventional CT group, were 3% ,25%(P = 0. 026) and 6%, 28% ( P = 0. 042 ), respectively. The main reason for treatment failure was distant metastasis both in the PET/CT group and conventional CT group,56% and 47% (x2 = 0. 56,P = 0. 453 ),respectively. Conclusions PET/CT, as a method of sketching the target of stage Ⅲ non-small cell lung cancer, can improve the radiation treatment plan, reduce the recurrence rate of hilar and mediastinal lymph nodes, meanwhile it can not improve the long-term survival rate; Distant metastasis was the main reason of failure.
