1.Effect of conditioned medium with concanavalin A-activated spleen cells supernatant on cultured rabbit corneal endothelial cells

Objective To evaluate the effect of conditioned medium made up of ConA-activated spleen cells supernatant on cultured rabbit corneal endothelial cells (RCECs).Methods RCECs were primarily cultured in vitro by revealing descemet membrane and endothelium combining tissues masses.Culture medium was RPMI 1640 with 10% FBS and varied concentrations of conA-activated spleen cells supernatant.Experimental groups were divided by different concentrations of ConA-conditioned supernatant:5%,10%,15%,and 20%.In the control group only RPMI1640 and 10% FBS were used.Methyl thiazolyl tetrazolium colorimetry analysis was used to evaluate the proliferation of RCECs cellular population.The cultured cells were identified by immunohistochemical staining for neuronal specific enolase (NSE).Results The RCECs were cultured successfully.RCECs presented positive expression in cytoplasm with NSE immunohistochemical assay.There were significant differences in the experimental groups compared with the control group (P

Objective To compare the effects of biofeedback (BF) training on clinical symptoms , psychological status and quality of life in different subtypes of patients with functional defecation disorders (FDD) .Methods According to Rome Ⅲ criteria ,50 FDD patients were divided into non coordinated defecation (F3a) and the lack of promoting defecation (F3b) two subtypes .The patients of these two subtypes received BF training three times every week ,five to ten times each course ,two to three times training at home were required during and after treatment .The changes ,of clinical symptoms scores ,self‐rating anxiety scale (SAS) ,self‐rating depressive scale (SDS) and the patient assessment of constipation quality of life question naire (PAC‐QOL ) were compared between before and after BF training in the patients of two subtypes .The difference between two subtypes were also analyzed as well .Paired t‐test or Wilcoxon signed‐rank test was performed for comparison between before and after treatment ,and group t‐test or Wilcoxon rank sum test was for comparison between groups .Results Among 50 FDD patients , there were 13 cases of F3a type and 37 cases of F3b type .After BF training ,the clinical symptoms scores of two subtypes patients were both lower than those before BF training (5 .62 ± 3 .91 vs 8 .77 ± 3 .59 , 5 .89 ± 3 .67 vs 9 .35 ± 3 .22 ,t = 3 .264 and 6 .272 ,both P< 0 .01) .There were no statistically significant differences in the scores of clinical symptoms between two subtypes at before and after BF training (t =- 0 .545 and - 0 .230 ,both P > 0 .05) .After BF training ,the SAS scores of two subtypes patients were both lower than those before BF training (30 .85 ± 6 .67 vs 42 .46 ± 8 .37 ,30 .65 ± 7 .51 vs 38 .59 ± 8 .38 , t= 4 .536 and 6 .402 ,both P< 0 .01) ,and the scores of SDS were both lower than those before BF training (42 .85 ± 8 .30 vs 53 .92 ± 7 .98 ,43 .95 ± 12 .17 vs 55 .39 ± 10 .83 ,t = 4 .788 and 6 .830 ,both P< 0 .01) . There were no statistically significant differences in the scores of SAS and SDS between two subtypes at before and after BF training (t= 1 .431 ,0 .084 ,- 0 .447 and - 0 .301 ,all P> 0 .05) .After BF training ,the PAC‐QOL scores of two subtypes [0 .54 (0 .15 ,0 .88) 、0 .98 (0 .51 ,1 .34)] were both lower than those before BF training [2 .08(1 .18 ,2 .34) 、1 .86(1 .34 ,2 .29)] ,Z= - 2 .903 、- 4 .825 ,both P < 0 .01) .There were no statistically significant differences in the PAC‐QOL scores between two subtypes at before and after BF training (Z= 0 .409 ,1 .891 ,both P > 0 .05) .Conclusions BF training is both effective on two subtypes of FDD ,which can improve clinical symptoms ,psychological status and quality of life ,and with There is no difference in efficacy between the two subtypes .
3.Effects of Shen-wu Capsule on learning and memory ability and brain β-amyloid content in APP transgenic mice

ObjectiveTo investigate the effects of Shen-wu capsule on learning and memory ability and its mechanism in APP transgenic mice. MethodsThe APP 695V717I transgenic mice were randomly divided into model group, Shen-wu low dose group (0.4g/kg·d) and high dose group (1.2g/kg·d). Normal control adopted the same age and background C57BL/6J mice. The animals were administered intragastrically by the drug or water from 4 month old to 10 month old. Morris water maze and object recognition test were performed to measure the learning and memory ability. The content of β-amyloid (Aβ) in the brain cortex homogenate was detected with RIA,and amyloid plaques were measured with Congo red staining. ResultsIn the Morris water maze test, swimming time and swimming distance of model group were prolonged distinctly(P<0.01). Shen-wu high dose group obviously shortened swimming time(P<0.05). In the object recognition test, the relative time to the new objection in model group was obviously shorter than that in the control group(P<0.05). The relative time to the new objection for Shen-wu high dose group was obviously longer than the model group(P<0.05). The content of soluble Aβ in model group was higher than that of the control group(P<0.05). Shen-wu group decreased the soluble Aβ distinctly(P<0.01). The amyloid plaques were increased in the brain of model mice(P<0.01). Each group of Shen-wu decreased amyloid plaques significantly(P<0.01).ConclusionShen-wu Capsule ameliorated the learning and memory function disorder and decreased Aβ formation in the brain of APP transgenic model mice.
4.Application of Electrical Stimulation in Regeneration of Peripheral Nerve(review)

Injury of peripheral nerve is a so common complication in trauma, that it is important to improve regeneration to recover the function of peripheral nerve. This paper has reviewed the pathology after injury of peripheral nerve, condition of successful regeneration of peripheral nerve, experiment and clinical study of electrical stimulation promoting regeneration of peripheral nerve and the possible mechanism. The effect of electrical stimulation on promoting regeneration of peripheral nerve has been gradually accredited. However, further study is still needed to discover the many unknown field.

8.Research progress in the study of brain microdialysis in glioma.

Glioma is the most common form of brain cancer. Despite recent advances in the treatment of solid tumors, there are few effective treatments for malignant gliomas due to its infiltrative nature. It has important significance to improve the treatment of glioma through in-depth understanding the intracerebral metabolic characteristics and pharmacokinetics of chemotherapeutics. Brain microdialysis (B-MD), an effective method to monitor central nervous system anticancer drug disposition, conditions of drugs through the blood-brain barrier, basic pathophysiologic metabolism, bioactive compounds and the changes of neurotransmitter in brain, provides the unique opportunity to allow the simultaneous determination of unbound concentrations of drugs in several tissues, and directly measure gliomas biochemistry continuously. B-MD has been able to monitor the change of brain drugs, metabolites and neurotransmitters, dynamic analysis of the drug concentration and pharmacological effect after administration, pharmacodynamic interaction between drugs, receptor mechanism of drug transport, as well as feedback information of internal environment. B-MD is expected to provide reference for clinical individual chemotherapy of glioma, but also provide powerful tools for the evaluation of new anticancer drugs in vivo. In this review, a comprehensive overview of B-MD for studies on glioma is elucidated with special emphasis on its application to neurochemistry and pharmacokinetic studies.

9.Research of the dynamic expression and effects of Nrf2 in delayed encephalophathy in rats after acute carbon monoxide poisoning

Objective To study the effects of nuclear factor E2-related factor 2 (Nrf2) in delayed encephalopathy in rats after acute carbon monoxide poisoning. Methods One hundred and sixty-eight rats were randomly divided into three groups: forty-eight rats in the blank control group (BC group), sixty rats in the air control group (AC group) as well as in the carbon monoxide poisoning group (CO group). The DEACMP model was established by improved intraperitoneal injections. The animals were valuated at 1st ,3rd, 7th, 14th, 21th, 28th day after acute carbon monoxide poisoning. Tunnel method was used to test the pyramidal cell apoptosis in the hippocampal CA1 area. The expression of Nrf2 was tested by immunohistochemical method and Western Blot method. Result In the CO group, the apoptosis index (AI) started to increase from first day and achieved it's peak at the 7th day (20.20 ± 1.78), then began to decrease slowly. The apoptosis index was still higher than that in the other groups at 28th day. And the apoptosis index of the CO group was markedly higher than the other two groups at each time point. The Nrf2 protein started to increase from 1st day in the CO group , reached its peak at 3rd day (8.20 ± 1.08), reduced later, maintained at a high level at 28th day, and expressed significantly higher than other groups at each time point. Conclusions The Nrf2 has a linear correlation with apoptosis , and plays a dual role in DEACMP because it rapidly increased in first three days to against apoptosis. But it continuously has been excessive expressed from 7th to 28th day in promoting the apoptosis of hippocampus pyramidal cells and may be a positive factor in DEACMP.
