Oligodeoxynucleotide containing unmethylated cytosine phosphate-guanosine motif(CpG ODN) may induce high expression of CD80, CD86, CD83, HLA I and HLAⅡ molecules on dendritic cells(DC) and stimulate DC to produce high level of IL-6, IL-12, TNF-? and IFN-?. CpG ODN is demonstrated in vivo to be a very potent adjuvant for Th1 cells, regulating Th0 cells to develop toward Th1 cells. Its role for DC is characteristics of CpG ODN sequence specificity and species specificity. CpG ODN is, at present, considered as a pathogen associated molecular pattern which binds its specific receptor,Toll-like receptor 9,then functions through TLR/IL-1R signaling pathway. It may represent a new therapeutic drug for broad applications in infectious disease, autoimmune disease, allergy and cancer therapy.

AIM: To study the effect of oligonucleotides containing unmethylated CpG motif (CpG ODN) on mouse bone marrow-derived dendritic cells (BMDCs) in anti-HcaF cytotoxicity. METHODS: BMDCs were stimulated by CpG ODN in combination with tumor antigen (TAg). The expression of CD80 on BMDC surface was analyzed by FCAS. Level of IL-12 (p70) in supernatants of BMDC culture was detected by ELISA. The proliferation of T cells was examined by MTT assay. Cytotoxicity of CTL induced by CpG ODN combining with TAg was detected by MTT assay. RESULTS: CpG ODN combining or not combining with TAg up-regulated the expression of CD80 on BMDC surface and stimulated BMDCs to produce a high level of IL-12. CpG ODN-activated BMDC promoted the proliferation of T cells. CTL induced by CpG ODN in combination with TAg appeared strong specific cytotoxicity on Hca-F cells. CONCLUSION: CpG ODN may effectively induce the functional maturation of mouse BMDC in vitro. CpG ODN in combination with TAg can enhance the anti-HcaF cytotoxicity of CTL. [

Objective To explore the relationship between subjective well-being (SWB) and behavioral inhibition/activation system (BIS/BAS) of older adults.Methods Using cluster sampling method and convenient random sampling method respectively for older adults in the nursing home and living home,we got 185 older subjects; then using BIS/BAS scale and SWB scale to test 185 older adults by oral questionnaire interviews face to face.Results (1) The SWB average score was 27.39±10.96,the BIS average score was 11.29±4.30,the BAS average score was 32.96±9.13.The SWB of older adults had a significant difference between older adults in the nursing (29.81±9.91) and living home (25.09± 11.47) (t=-2.69,P＜0.01).The negative experience of SWB had significant differences between older adults in the nursing (4.25±3.37) and living home (4.25±3.37) (t=3.58,P＜0.01),and also between males (4.25±3.37) and females (5.91±3.83) (t=-1.96,P＜0.05).The BAS fun seeking had a significant gender difference (t=2.05,P＜0.05),which was higher in males (12.60±3.48) than in females (11.40±3.67).(2) BAS was positively correlated with SWB (r=0.40,P＜0.01),and BIS was negatively correlated with SWB (r=-0.40,P＜0.01).(3) The BAS drive could forecast the 19％ variance of SWB.Conclusion BIS/BAS are correlated with SWB for older adults and the BAS drive is one of factors contributing to the SWB of older adults.

Objective: To investigate the relationship between depressive symptoms and frailty among middle-aged and elderly populations, so as to provide insights into the early identification and prevention of frailty. Methods: Based on the 2018 database of China Health and Retirement Longitudinal Study (CHARLS), demographic information, lifestyle and self-rated health status were collected from people aged 50 years and older. Depression symptoms were evaluated using the Short Version of Center for Epidemiological Studies Depression Scales (CES-D-10), and the frailty status was evaluated using frailty index. The relationship between depressive symptoms and frailty among middle-aged and elderly populations were using a multivariable logistic regression model, and the dose-response relationship was analyzed using a restricted cubic spline model. Results: A total of 5 172 individuals were enrolled, including 2 495 males (48.24%) and 2 677 females (51.76%), with a median age of 63.00 (interquartile range, 12.00) years, a median CES-D-10 score of 7.00 (interquartile range, 9.00) scores. There were 1 560 participants (30.16%) detected with mild depressive symptoms, 360 participants (6.96%) detected with moderate to severe depressive symptoms. Multivariable logistic regression analysis showed that after adjusting for gender, age, educational level, marital status, smoking, alcohol consumption and self-rated health status, participants who had mild depressive symptoms (OR=4.226, 95%CI: 3.625-4.928) and moderate to severe depressive symptoms (OR=10.737, 95%CI: 8.259-13.958) had a higher risk of frailty. The restricted cubic spline model showed a nonlinear relationship between depressive symptoms and frailty among middle-aged and elderly populations. When the CES-D-10 scores were greater than 7, the risk of frailty increased with higher CES-D-10 scores. Conclusion The degree of depressive symptoms in middle-aged and elderly people is associated with frailty, and there is a nonlinear relationship between the two.

Objective To study the therapeutic effect of Danshen injection combined with conventional therapy on children with recurrent asthmatic bronchitis. Methods A prospective study was conducted. Fifty-five children with asthmatic bronchitis admitted in People's Hospital of Henan Province from January 2012 to July 2014 were enrolled and randomly divided into a integrative Chinese and western medicine group(30 cases)and a conventional therapy group(25 cases);in the same period,18 healthy children having passed physical examinations were chosen as the healthy control group. In the conventional therapy group,rest,oxygen inspiration,anti-infection, fluid supplement and aerosol inhalation,etc. were given,while in the integrative Chinese and western medicine group,based on the similar conventional therapies,additionally Danshen injection 0.4 mL · kg-1 · d-1 was given,once per day for a total of 14 days. The wheezing remission time and the pulmonary rale extinction time were observed in the two patient groups,and the fractional exhaled nitric oxide(FeNO)content was investigated in the three groups. Results The wheezing remission time and the pulmonary rale extinction time of the integrative Chinese and western medicine group were obviously shortened compared with those in the conventional therapy group〔the wheezing remission time(days):2.3±0.8 vs. 5.2±1.1,the pulmonary rale extinction time(days):6.8±1.5 vs. 8.4±1.1,both P<0.01〕. The contents of FeNO in the integrative Chinese and western medicine group and the conventional therapy group before treatment were significantly higher than those in the healthy control group(×10-9:30.3±9.5,30.4±8.2 vs. 13.6±3.4,both P<0.01),but after treatment the FeNO levels of both patient groups were markedly lowered,and the degree of change in integrative Chinese and western medicine group was more significant(×10-9:18.6±4.7 vs. 24.0±6.8,P<0.01). Conclusion Danshen injection combined with conventional therapy can obviously down-regulate the FeNO content,alleviate the airway inflammation,effectively relieve the children recurrence of asthmatic bronchitis.

Objective To study the expression of CD147 and collagen Ⅳ protein in gastric carcinoma.Methods Immunohistochemical assay was used to detect the expression of CD147 and collagen Ⅳ protein in 119 cases of gastric carcinoma, 20 cases of normal gastric tissues, and the clinical data was analyzed.Results In 119 cases of gastric carcinoma, the positive expression of CD147 protein was 83.2 %(99/119), the positive expression of collagen Ⅳ protein was 31.9 %(38/119). The expression of CD147 and collagen Ⅳprotein had relationship with metastasis of lymph nodes,depth of invasion and clinical phases (P ＜0.05). There was negative significant correlation between the expression of CD147 and collagen Ⅳ protein in gastric carcinoma (P＜0.01). Conclusion The expressions of CD147 and collagen Ⅳ protein may be used as one of the marks to study gastric carcinoma.

Objective To explore the impact of executive function intervention on the executive functioning of elderly patients with cerebral infarction,and to seek the best way to improve executive function in such patients.Methods Eighty elderly patients with cerebral infarction were randomly divided into an intervention group and a control group.A series of executive function interventions were administered to those in the intervention group with no intervention in the control group.The two groups were given the same conventional medication.The Behavioral Assessment of the Dysexecutive Syndrome (BADS) was taken before the intervention,then 3 months and 6 months after recruitment.Results In the intervention group the average score and the total standard score had increased significantly after 3 months except for the action program test and the modified six elements test.Both indicators were still significantly increased from the baseline afar 6 months except for the modified six elements test.The average score and the total standard score had increased significantly between 3 months and 6 months except on the key search test.Both the time determination test and the total standard score had statistically significant average differences.The control group showed no significant differences.After 3 months,the average scores and the total test standard scores in the intervention group were signifieantiy higher than those in control group except on the action program test.The rules conversion card test,the time determination test and the total standard score difference all showed statistically significant differences between the groups.After 6 months the intervention group's scores remained significantly higher than those of the control group except on the action program test.Conclusions Executive function interventions are effective in delaying the progress and improving the prognosis of executive dysfunction in elderly patients with cerebral infarction,and the therapy has persistent effects.
