1.Clinical efficacy of the treatment of inverted nipple with retractor

Objective To observe the surgical effect of nipple retractor to inverted nipples.Methods Between January 2001 and January 2014,this nipple retractor to inverted nipples was used to correct 604 inverted nipples of 322 patients.The follow-up period ranged from 6 to 24 months.The clinical results and complications were evaluated.Results Excellent nipple morphology and patient satisfaction were achieved in 211 cases (93.8 %),basically normal nipple morphology and patient satisfaction were achieved in 12 cases (5.3%),and recurrent nipple inversion occurred in three cases (1.3%).The latter three patients had experienced failed open correction for their inverted nipples,and some degree of nipple inversion occurred after removal of the frame.In this study,28 patients (12.4 %) experienced successful pregnancy and breast-feeding.No loss of nipple sensation occurred in 225 cases.Nipple erection was observed after stimulation in 215 cases (95.6%).Conclusions The described technique is minimally invasive,simple,safe,and reliable.Satisfying aesthetic results can be achieved without destroying breast feeding function and nipple sensation.
2.Finite element analysis of the prosthodontic design for maxillary first molar mesio-lingual cusp defect with subfissue

Objective:To study the prosthodontic design for the treatment of maxillary first molar mesio-lingual cusp defect with subfis-sue.Methods:A finite element model of maxillary first model,including mesio-lingual cusp defect with subfissue,periodontal support-ing tissue and a section of the maxilla was established by cone beam CT.Different prosthodontic designs for respective restoration and simulating different bite force were adopted,the Von Mises stress,maximum compressive stress and the crack distribution,J integral and the root displacement were analysed.Results:In the metal pile restoration model,the stress,JINT value and the equivalent stress and displacement of root were the smallest.With the increase of crack depth of the vertical and oblique fissue,the equivalent levels of stress (equivalent stress of J integral),the stress experienced by the dentin along with the peak of the maximum principal,increased signifi-cantly.Under 4 kinds of force direction and 3 repair methods,the equivalent stress peak value,J integral dentin stress,maximum prin-cipal stress peak value and dentin stress peak value of the cracks was 200 N,90°>200 N,45°>600 N,0°>200 N.Conclusion:High elastic modulus and all ceramic crown may lead to minimal equivalent stress,J integral,dentin equivalent stress and maximum principal stress.When subfissure is repaired,teeth fracture remains possible and any possible propagation may exist.The effect of later-al force on the crack and the dentin is greater than that of vertical force for crack expansion and tooth fracture.

5.Chronic granulomatous disease caused by compound heterozygous mutation in NCF2 gene: case report and literature review

Objective:To explore the relationship between the type of mutation and clinical features, prognosis, and clinical characteristics of chronic granulomatous disease (CGD) caused by compound heterozygous mutations in the NCF2 gene in children. Methods:The clinical data of 1 case of neonatal CGD caused by compound heterozygous mutations of NCF2 gene at Tianjin Children′s Hospital in August 2019 was analyzed, and domestic and international literatures were searched to summarize the clinical characteristics, gene mutation type and prognosis of CGD caused by NCF2 mutation. Results:The diagnosis of CGD was confirmed by the presence of compound heterozygous mutations c. 196_197insA (p.Arg66Glnfs23X) and c. 1180T>G (p.Tyr394Asp) in the NCF2 gene, accompanied with the clinical manifestations of fever, cough, multiple clumps and nodules in the chest CT at 25 days after birth, and the neutrophil respiratory burst test stimulation index(SI) 23.This new mutation was not reported in the Human Genetic Mutation Database.The child had a residual portion of nicotinamide adenine dinucleotide phosphate (NADPH) oxidase activity and was followed up until the age of 9 months with an antifungal drug without recurrent infection.A total of 101 cases of CGD patients with NCF2 gene mutation were reported in domestic and international databases.Totally, 33 cases had SI results, with 22 cases below 3, 11 cases above 3, and 8 cases of missense mutations. Conclusions:c. 196_197insA and c. 1180T>G are new mutations in NCF2 gene that can lead to CGD.CGD patients containing missense mutations in the NCF2 gene may have more residual NADPH oxidase activity.
6.Diagnosis of Alport Syndrome by Immunohistochemical Staining of Type Ⅳ Collagen ? Chains on Paraffin-Embedded Renal Sections

Objective To investigate the diagnostic technique of Alport syndrome(AS)by immunohistochemical staining of type Ⅳ collagen ? chains on paraffin-embedded renal sections.Methods Renal biopsies were obtained from 2 patients with autosomal recessive form of AS,2 female patients and 2 male patients with X-linked dominant form of AS and 2 patients with hematuria(1male and 1 female).AS was diagnosed according to symptoms,family history,pathology,immunofluorescence staining of type Ⅳ collagen ? chains on renal and skin biopsies and gene analysis.Normal portions of nephrectomized kidneys from 2 patients with renal tumor were used as controls.Type Ⅳ collagen ? chains were stained by two-step immunohistochemistry staining method on paraffin-embedded renal sections.Three antigen retrieval methods including autoclave heating,pepsin digestion and proteinase were investigated to find the best antigen retrieval method for type Ⅳ collagen ? chains.The findings were compared with those examined by immunofluorescence staining on fresh frozen sections.Results By immunohistochemistry staining,type Ⅳ collagen ?3 and ?5 chains showed continuous linear pattern along glomerular basement membrane on sections from the controls and the hematuria patients,intermittent linear pattern for X-linked dominant female AS patients,negative for X-linked dominant male AS patient.For patients with autosomal recessive AS,the staining of type Ⅳ collagen ?3 and ?5 chains were negative on glomerular basement membrane,but ?5 chain was positive on glomerular capsules and partial tubular basement membrane.The results were the same as those examined by immunofluorescence staining.Conclusion AS can be diagnosed by immunohistochemistry staining of type Ⅳ collagen on paraffin-embedded renal sections,which is a new technique for diagnosis of AS in China.
7.Gastric cancer related miR-148 a targets gastrin receptor CCKBR

Objective To investigate the regulation role of gastric cancer related miR-148a on gastrin receptor CCKBR expression, and find the correct binding sites of miR-148a in CCKBR 3’UTR.Methods The potential binding sites of miR-148a in the CCKBR 3’UTR were predicted with the bioinformatic tools;The miR-148a expressing plasmid was constructed by PCR, and miR-148a expression was verified by Northern Blot;The luciferase report plasmids containing the wild type and mutated binding sites of CCKBR 3’ UTR were constructed, and were used to study the regulation mechanism and identify the binding sites of miR-148a by luciferase activity analysis; The regulation effect of miR-148a on CCKBR protein expression was checked by Western Blot.Results Three potential binding sites of miR-148a in the CCKBR 3’ UTR were found; The miR-148a expressing plasmid was constructed successfully, and highly expressed miR-148a after transfected to gastric cancer cells;The inhibitory effect of miR-148a on CCKBR protein expression was checked by Western Blot.Over-expression of miR-148a inhibited CCKBR expression by directly binding to the binding site in CCKBR 3’UTR 423bp.Conclusion CCKBR is a target of miR-148a, and its expression is inhibited by the binding of miR-148a on its 3’ UTR, indicating that miR-148a may participates in the progression of gastric cancer by regulating CCKBR expression.
8.Effect of arsenic trioxide on telomerase and telomerase reverse transcriptase in KM3 cell line.

To explore the effects of arsenic trioxide on multiple myeloma (MM) cell line KM(3) and its possible mechanism, cell viability was counted by trypan-blue exclusion, apoptosis was detected by morphology and DNA ladder; cell cycle was assayed by flow cytometry (FCM), telomerase activity was determined by semi-quantitative telomeric repeat amplification protocol (TRAP)-reverse transcription polymerase chain reaction (RT-PCR)-enzyme linked immunosorbent assay (ELISA), while the expression of hTERT mRNA in transcriptional level was measured by using RT-PCR. The results showed that arsenic trioxide inhibited the growth and viability of KM(3) cell and induced apoptosis; cell cycle was arrested in G(2) phase; arsenic trioxide could inhibit telomerase activity, which consisted with the downtrend of hTERT mRNA expression. In conclusion, down-regulation of telomerase activity and hTERT may play an important role in the apoptosis of MM cell line KM(3) induced by arsenic trioxide.

9.Establishment of chronic heart failure rat model of Xin-qi deficiency complicated blood stasis and edema syndrome and judgment of diagnosis information integration.

OBJECTIVETo establish and evaluate chronic heart failure (CHF) rat model of Xin-qi deficiency complicated blood stasis and edema syndrome (XQD-BSES).
METHODSTotally 40 SD rats were randomly divided into the normal control group (Control), the propylthiouracil (PTU) group, the adriamycin (ADR), and the ADR + PTU group. Normal saline was used as equivalent solvent of each group. Rats in the Control group were intragastrically and intraperitoneally injected with normal saline. Rats in the PTU group were intragastrically injected with PTU suspension and intraperitoneally injected with normal saline. Rats in the ADR group were intragastrically injected with ADR solution and intraperitoneally injected with normal saline. And rats in the ADR + PTU group were intragastrically injected with PTU suspension and intraperitoneally injected with ADR solution. The dose of PTU was 0.2% of daily forage weight, once daily. The dose of ADR was 3.5 mg/kg, once per week. The modeling lasted for 6 weeks. Left ventricular ejection fraction (LVEF), left ventricular fraction shortening (LVFS), brain natriuretic peptide (BNP), heart rate (HR), respiratory rate (RR), urine output, ear temperature, exhaustive swimming test (EST), Tri-iodothyronine (T3), tetra-iodothyronine(T4), thyroid stimulating hormone (TSH) as well as heart, lung, liver weight indices and their pathological sections were integrated and compared.
RESULTSCompared with the Control group, LVEF, LVFS, BNP, HR, RR, heart, lung, liver weight indices, urine output, ear temperature, EST, and T3, T4, and TSH changed significantly in the ADR group, the PTU group, and the ADR + PTU group with statistical significance (P < 0.05), and pathological changes of heart failure occurred in pathological sections of heart, lung, and liver. Compared with the ADR group, LVEF, LVFS, BNP, and lung, liver weight indices, urine output, ear temperature, T3, T4, and TSH changed significantly in the ADR + PTU group with statistical significance (P < 0.05), and pathological changes of heart failure were more serious in pathological sections of heart, lung, and liver. Compared with the PTU group, LVEF, LVFS, BNP, HR, RR, urine output, EST, T4, heart and lung weight indices changed significantly in the ADR + PTU group with statistical significance (P < 0.05), and pathological changes of heart failure were quite serious in pathological sections of heart, lung, and liver.
CONCLUSIONADR + PTU was an appropriate method to establish CHF rat model of XQD-BSES.

10.Simultaneous determination of four components in Hedyotis diffusa oral solution by reversed-phase HPLC.

OBJECTIVEA new method for simultaneous determination of four methyl-3, 4-dihydroxybenzoate ( I ), P-coumaric acid (II), ferulaic acid (III) and E-6-O-P-coumaroyl scandoside methyl ester (IV) in Hedyotis diffusa oral solution by reversed-phase HPLC was developed.
METHODThe separation was performed on a Diamonsil C18 column (4.6 mm x 250 mm, 5 microm) with gradient elution. A-acetonitrile, B-methonal-water-glacial acetic acid (5: 95: 0.25), 0-20 min, 1% -16% A; 2042 min, 16% A; 42-46 min, 16%-20%A; 46-65 min, 20% A. The UV detection was set at 265 nm; flow rate 1.0 mL x min(-1).
RESULTThere was good linearity between the peak area and concentration at the ranges of 2.1-105 (r = 0.999 8), 3.5-175 (r = 0.999 8), 1.72-86 (r = 0.999 9), 4.0-200 mg x L(-1) (r = 1.000 0) for I, II, III and IV respectively. The average recoveries of I, II, III and IV were 99.9%, 97.9%, 98.6% and 98.1%.
CONCLUSIONThe method is rapid, simple and accurate, and it can be used for the evaluation of H. diffusa oral solution.
