Objective To explore the impact of prenatal exposure to lipopolysaccharide on reninangiotensin system of offspring rats Methods Six pregnant SD rats were randomly divided into 2 groups. The rats in the lipopolysaccharide (LPS) group were treated with LPS 0. 79 mg/kg (I. P. ) on the 8th, 10th and 12th day of gestation, and rats in the control group were treated with saline at the same time points. The blood pressure of offspring rats was measured by the tail cuff method. Protein expression of Angiotensin Ⅱ (Ang Ⅱ ) in thoracic aorta vessel was determined by immunohistochemisry. Protein expressions of Ang Ⅱtype 1 and type 2 receptor in thoracic aorta vessel were detected by Western blot. Results Blood pressure of 12-week-old offspring rats of LPS group was significantly higher than that of 12-week-old offspring rats of control group( P < 0. 01 ). The protein expression of Ang Ⅱ and Ang Ⅱ type 1 receptor in thoracic aorta vessel were significantly higher while protein expression of Ang Ⅱ type 2 receptor was lower in 15-week-old offspring rats of LPS group than in control group, resulting in a significant increase in the ratio of Ang Ⅱtype 1 receptor/Ang Ⅱ type 2 receptor in the aorta at 15-week-old of offspring rats than in 15-week-old offspring rats of control group( P < 0. 01 ). Conclusion Prenatal lipopolysaccharide exposure results vascular renin-angiotensin system dysfunction, which my play an important role on the pathogenesis of hypertension development in offspring rats.

The aim of this study was to investigate the homing characteristics of bone marrow cells in leukemia mice after allogenic bone marrow transplantation with different conditioning regimens on the basis of a leukemia mouse model. Allogenic bone marrow transplantation was performed after three different kinds of conditioning regimen, including nonmyeloablative conditioning regimen (5 Gy (60)Co γ ray total body irradiation, A group), radiotherapeutic myeloablative conditioning regimen (9 Gy (60)Co γ ray total body irradiation, B group) and chemotherapeutic myeloablative conditioning regimen (large dose chemotherapy, C group). In the recipient mice, the nucleated cell number in peripheral blood, bone marrow and spleen was counted, the percentage of positive cells capable of connecting with FITC labeled anti-mouse H-2K(b) antibody was detected by flow cytometry and the homing ratio in bone marrow and spleen was calculated at 24, 48, 72, 96 h after bone marrow transplantation. The results showed that donor myeloid cells displayed homing and then mobilization (going out of home) in group A; homing, mobilization, and rehoming in group B and C, and there was a little delay of homing in the spleen in group C. In bone marrow, the homing efficiency of A group was the highest in early period and the lowest [(0.90 ± 0.09)%] in the fourth day with the mobilization of myeloid cells (P < 0.05), and the homing efficiency of B and C groups was lower in the early period and the highest [(2.17 ± 0.26)%, B group] in the fourth day with the rehoming of myeloid cells (P < 0.05). In spleen, the homing efficiency was similar to that in bone marrow and there still was a little delay in C group. It is concluded that the homing ratio is high in the early period and decrease obviously in 72 h after bone marrow of leukemia mice treated with nonmyeloablative conditioning regimen. The homing ratio is low in the early period and increases obviously in 72 h after bone marrow of leukemia mice treated with radio-or chemotherapeutic myeloablative conditioning regimens. The homing ratio does not obviously change between the early period and 72 h after bone marrow of leukemia mice treated with chemotherapeutic myeloablative conditioning regimen, and lies between group A and B.

This study was aimed to analyze the correlation of JAK2V617F mutation burden with clinical features in patients with polycythemia vera (PV) and essential thrombocythemia (ET), The JAK2V617F mutation ratios in 47 PV samples and 43 ET samples were detected by real-time PCR. The correlation of mutation allele ratio in PV and ET samples with clinical features (hemoglobin, hematocrit, white blood cell count and platelet count) was analyzed. The results showed that the JAK2V617F mutation burden was higher in PV (0.441 +/- 0.270) than that in ET (0.209 +/- 0.192). The JAK2V617F mutation burden was positively correlated with levels of hemoglobin (PV: R = 0.518, p < 0.001; ET: R = 0.528, p = 0.005), hematocrit (PV: R = 0.510, p < 0.001; ET: R = 0.524, p = 0.005) and leukocyte (PV: R = 0.584, p = < 0.001; ET: R = 0.471, p = 0.013) in PV and ET samples. The higher JAK2V617F mutation burden was negatively correlated with levels of platelet count in PV samples (R = -0.354, p = 0.020), but there was no correlation between the JAK2V617F mutation burden and platelet count in ET samples (R = 0.233, p = 0.242). It is concluded that the higher JAK2V617F mutation burden is related with higher hemoglobin, hematocrit and leukocyte count in both PV and ET samples. The higher JAK2V617F mutation burden is correlated with lower platelet count in PV samples, but there is no correlation between JAK2V617F mutation burden and platelet count in ET samples.

OBJECTIVETo investigate the short-term therapeutic effectiveness of body gamma-knife in patients with advanced pancreatic carcinoma.
METHODSForty-eight patients with advanced pancreatic carcinoma were treated by body gamma-knife therapy. The dosage distribution and other radiotherapeutic plans were established on the basis of the carcinoma position, clinical target volume, and patient health condition. The isodose curve was 50%-60% and covered about 95% of the target volume. The single dose was 350-450cGy. The radiation was performed once every one or two days for 10-12 times.
RESULTSThere were 33 patients with back pain. 63.6% of the patients got completely controlled, 30.3% pain remitted, and 6.1% ineffective after 2 to 18 months of therapy. The analgesic effective rate was 93.9%. Among 28 patients with obstructive jaundice, 21 patients (75.0%) recovered. Among 42 patients who received CT, tumor disappeared in 5 patients (11.9%), tumor size decreased in 30 patients (71.4%), remained unchanged in 5 patients (11.9%), and enlarged in 2 patients (4.8%). The 6-month, 12-month, and 18-month overall survival rates were 77.1%, 37.5%, and 10.4%, respectively. The whole process was tolerable for all patients and no severe side-effect was observed.
CONCLUSIONSBody gamma-knife can achieve a high local control rate and survival rate. Its short-term therapeutic effectiveness is satisfactory. Body gamma-knife is a safe and reliable treatment option for patients with locally advanced pancreatic carcinoma.

·AIM: To investigate the effect of sodium hyaluronate eye drops on ocular inflammatory response and dry eye after phacoemulsification combined with intraocular lenses (IOL) implantation for age-related cataract. ·METHODS: The clinical data of age-related cataract patients treated with phacoemulsification combined with IOL implantation in 200 cases (200 eyes) of our hospital from June 2016 to July 2017 were retrospectively analyzed. According to the different postoperative application of eye drops they were divided into observation group and control group (100 cases each). The observation group was given sodium hyaluronate eye drops and the control group was given tobramycin dexamethasone eye drops. The results of BUT,FL,proteins concentration in aqueous and ocular surface disease index (OSDI) score were compared before and at the first day,the seventh day and the fourteenth day after the operation. And the incidence of conjunctival hyperemia and ciliary hyperemia was compared between 1d after surgery and 14d after surgery. ·RESULTS: There was no significant difference in OSDI scores between the two groups before and at 1d after surgery (P>0.05). The OSDI score of the two groups was higher than that before the operation on the first, the seventh and the fourteenth day after surgery (P<0.05). The OSDI score of the two groups decreased gradually after reaching the highest level on the first day after surgery (P<0.05). The OSDI scores of the observation group on the 7 and 14d after surgery were lower than those in the control group (P<0. 05). There was no significant difference in the BUT between the two groups before and at 1d after surgery (P>0.05). The levels of BUT in the two groups were lower than those before operation on the first, the seventh and the fourteenth day after surgery (P < 0. 05). BUT levels in both groups increased gradually after reaching the lowest level on the first day (P<0.05), the levels of BUT in the observation group were higher than those in the control group on the 7 and 14d after operation (P< 0. 05). There was no significant difference in FL level between the two groups before and the first day after operation (P>0.05). The levels of FL in the two groups were different from those before operation (P<0.05) on the first, the seventh and the fourteenth day after operation (P<0.05). FL levels in the two groups decreased gradually after reaching the highest level on the first day after surgery (P<0.05), the levels of FL in the observation group on the 7th day and the 14th day after operation were lower than those in the control group (P < 0. 05). There was no significant difference in the incidence of conjunctival hyperemia and ciliary congestion between the two groups on the first day after operation (P>0.05); the incidence of conjunctival hyperemia and ciliary hyperemia in the two groups on the fourteenth day after surgery was lower than that on the first day after surgery (P<0.05). However, the incidence of conjunctival hyperemia and ciliary congestion on the fourteenth day after operation was significantly lower in the control group than in the observation group (P<0.05). Postoperatively 1,7,14d, aqueous humor protein concentration in the two groups of patients compared with preoperative had difference (P< 0. 05), and two groups of patients with aqueous humor at postoperatively 1d after reaching the highest protein concentration decreased ( P < 0. 05 ), aqueous humor protein concentrations of the control group 7,14d after operation were lower than the observation group (P<0.05). ·CONCLUSION: The effect of sodium hyaluronate eye drops on improving dry eye after age-related cataract phacoemulsification combined with IOL implantation is greater than that of tobramycin dexamethasone eye drops, but it is less effective for eye inflammation than tobramycin dexamethasone eye drops. After comprehensive consideration, for the patients with light inflammation, small incision phacoemulsification combined with IOL implant, sodium hyaluronate eye drops is a good selection.

Chinese Journal of Analytical Chemistry 2018;46(3):332-341
In this work, two chiral chloride probes were used to differentiate landiolol hydrochloride by mass spectrometry. Two chiral chloride probe reagents, N-(p-Tosyl)-L-phenylalaninyl chloride (TSPC) and (-)-Camphanic acid chloride, were chosen to react with landiolol hydrochloride and its stereoisomers to form covalent bonding derivatives, which enlarged the difference of stereo structure between landiolol and its stereoisomers. Result of tandem mass spectrometry showed that fragment from derivative products prefers to losing water to form fragment ions m/z 793 and m/z 672. The relative abundance of ions m/z 793 and m/z 672 was quite different in each isomer. The fragment ions m/z 603 from (-)-Camphanic acid chloride derivative products showed distinction relative abundance because of the different stability of each stereoisomers, which gave rise to the enlarged difference of stereo structure between landiolol and its stereoisomers. By comparing the different relative abundance ratio of analyte and each stereoisomer in MS/MS spectra, we could realize recognization landiolol hydrochloride and its stereoisomers. Accurate masses of precursor and fragment ions were confirmed on an IT-TOF mass spectrometer. This method by using ion-trap mass spectrometry could rapidly and simply differentiate landiolol hydrochloride and its stereoisomers. This work could also contribute to differentiation and discrimination of landiolol hydrochloride and its stereoisomers.

OBJECTIVETo observe the effect of Chinese herbal medicine for supplementing Shen and strengthening the bone on distal femoral trabecular ultrastructure of male rats subjected to simulated weightlessness.
METHODSFifteen male SD rats were randomized equally into 3 groups, including a control group (group A, in which the rats were allowed free movement) and two simulated weightlessness (via tail suspension) groups (group B and C). The rats in group C were treated with the commercially available Chinese herbal preparation, and those in the other two groups received normal saline at the same dose, for a treatment course of 28 days. Observation of trabeculae was performed with scanning electron microscopy.
RESULTSIn group B, the trabeculae of the femur became thinner, fragile, discontinuous with reduced quantity as compared with those in group A. The rats in group C had greater number of the trabeculae than those in group B. Resorption surface decreased and the collagenous fiber were much more regular in group B.
CONCLUSIONThe Chinese herbal medicine may produce beneficial effect on bone microstructure of rats subjected to simulated weightlessness.
