1.Variation of blood-platelet parameters in patients with cerebral infarction

BACKGROUND: The parameters of blood platelet(B-PLT) can reflect its thrombo-function, but the variation of blood-platelet parameters in patients with thrombotic disease is controversial and not unanimous.OBJECTIVE: To observe the changing rule of the B-PLT parameters in patients with cerebral infarction(CI), and investigate whether blood cell counter can be used in the evaluation of the variation of B-PLT parameters in CI patients.DESIGN: A case-control and experimental study based on patients and healthy people.SETTING: Department of laboratory and internal medicine in a university.PARTICIPANTS: During the January and March 2002, 37 CI patients including 28 males and 9 females aged 45 - 87 years were selected from Department of Neurology, Union Hospital Affiliated to Tongji Medical College,Huazhong University of Science and Technology. Among 37 healthy people,20 males and 17 females aged 40 - 60 years were selected as control group.METHODS: Platelet distribution width (PDW), mean platelet volume (MPV), platelet-large cell ratio(P-LCR) and the changing quantity of B-PLT of 37 CI patients were analyzed with blood cell counter, meanwhile,the healthy people were analyzed as control subjects.MAIN OUTCOME MEASURES: Changes of B-PLT parameters in two groups respectively.RESULTS: A positive relation between PDW and P-LCR was observed and between P-LCR and MPV as well( r = 0. 99, 0.92, P < 0. 001 ). A negative correlation between B-PLT quantity and MPV was observed and also between B-PLTquantity and P-LCR(r= -0.58, -0.59, P <0.05) .In CI patients, PDW, MPV and P-LCR were(14.51 ± 2. 88)%, (10.95 ± 1.48) fl and(34.24 ± 11.23)% respectively, which were higher than those of healthy people [ (12. 86 ± 2.02)%, (10. 19 ± 1.29) fl, (28.47 ± 9.41 )% ]respectively, and the difference was significant( P =0. 019 31, 0. 021 77 and 0.026 35), but there was no significant difference between the two groups in B-PLT quantity.CONCLUSION: The blood platelets' parameters in CI patients, which reflect the function of the B-PLT, are significantly higher than those in healthy people. It shows that the enhanced function of platelet is a risk factor of cerebral infarction, the use of blood cell counter in analysis can observe the variation of the platelets' function rapidly and accurately. It can be a useful method of the therapy and prevention in CI patients.
2.Research progress of biomechanics and finite element analysis of lumbar interspinous devices

BACKGROUND:The lumbar interspinous device, as a kind of non-fusion technology has been extensively applied in the clinic and exerts superiority in biomechanics compared with the traditional fusion technology. With the development of prosthesis design and impanation technology, it reveals a better application prospect. OBJECTIVE:To review the biomechanics and finite element analysis of lumbar interspinous devices. METHODS:The first author retrieved the databases of CNKI, WanFang, PubMed and SpringerLink using the keywords of“lumbar spine, interspinous devices, biomechanics, finite element analysis”in Chinese and English, respectively. Researches related to the biomechanics and finite element analysis of lumbar interspinous devices were included and repeated researches were excluded. A total of 44 literatures were enrol ed for review, including 8 Chinese and 36 English literatures. RESULTS AND CONCLUSION:(1) Biomechanical y, several interpinous devices, which are commonly used in the clinic, can increase the stability of the implanted segment in sagittal alignment by limiting the range of flexion-extension, with no significant change in lateral bending and axial rotation. (2) Few studies analyze the influence of the implant size and placement on the implanted segment and on the adjacent segments. (3) Through the stress nephogram, three-dimension finite element analysis can intuitively analyze the changes of the stress distribution in the intervertebral disc, isthmus and facet joints before and after implantation. Both biomechanical studies and finite element analysis indicate that interspinous devices can share the load of the disc and facet joints, and at the same time, make no effect on the range of motion and stress of the adjacent segment. (4) In conclusion, the short-term biomechanical advantage of the interspinous devices is obvious, but further studies are needed. The finite element analysis can simulate different body physical environment, and can analyze mechanical distribution changes after implantation, which is an effective way to evaluate the mechanical mechanism of the interspinous devices.
3.Purification of an anti-HBsAg scFv and measurement of its affinity constant

AIM: To purify and refold the inclusion body of a human anti-HBsAg scFv with a 6?His tag, and to determine the affinity constant of the purified recombinant product.METHODS: Solubilizing in buffers containing urea or guanidine hydrochloride (GuHCl), the inclusion body was purified by IMAC, and then refolded by dialysis against urea or GuHCl, at the same time, Ni 2+ charged chelate column was utilized for in situ refolding. The affinity constant of the refolded scFv, polished by immune-affinity chromatography, was determined by non-competitive ELISA. RESULTS: The refolded scFv with highest specific bioactivity was produced by dialysis against GuHCl. Under this condition, the recovery of target protein reached (61.08?1 45)%. The affinity constant of the polished scFv was confirmed to be(2.30?0.32) ?10 7 L/mol. CONCLUSION: The inclusion body studied in this paper can be refolded efficiently under optimal dialysis condition in vitro . The antigen-binding property of this recombinant scFv is not affected by the purification tag fused to the N terminal of the protein.
4.Meta-analysis on interspinous dynamic stabilization system Wallis versus Coflex for lumbar degenerative disease

BACKGROUND: It is still controversial that interspinous dynamic stabilization system Wallis and Coflex which one can provide better clinical effects for lumbar degenerative disease.OBJECTIVE: To systematically assess the clinical effectiveness and safety of Wallis and Coflex for lumbar degenerative disease.METHODS: According to the computer-based online search of PubMed, Embase, Medline, Cochrane Library, CBM,CNKI, Wanfang Database, and VIP, articles published before August 1st, 2016 were searched. Articles about Wallis comparing with Coflex for lumbar degenerative disease were included; the quality score of methodology was assessed by MINORS. Research data abstracted and synthesized by Review Manager 5.3 were used for meta-analysis.RESULTS AND CONCLUSION: (1) Six studies were included, and all studies were designed for non-randomized controlled trial. (2) There were no significant statistical differences in Japanese Orthopedic Association, Oswestry disability index, visual analogue scale score, Prolo functional score, segmental lordosis angle, and segment movement degree. Incidence of adverse events was significantlue scale less in the Wallis group than in the Conflex group (P < 0.05).(3) There was no significant difference in clinical efficacy between Wallis and Coflex in the early and mid-term follow-up.We can conclude that Wallis may provide better clinical safety than Coflex.
5.A study on the relationship between the Wnt receptor signaling pathway and severe retinopathy of prematurity

Objective To study the relationship of Wnt receptor signaling pathway and severe retinopathy of prematurity (ROP).Method From January 2011 to June 2015,preterm infants with severe ROP admitted to the NICU of our hospital were enrolled prospectively.Preterm infants with similar gestational age,gender,and age (in days) admitted to our hospital during the same period were selected as the control group.FZD4,LRP5,and ND gene mutations in Wnt receptor signaling pathway were examined.Result A total of 61 Chinese preterm infants were screened for these three candidate genes of Wnt receptor signaling pathway,32 in ROP group and 29 in control group.ND and FZD4 gene mutations were not found among all cases.Eight types of LRP5 mutations were found in 26 cases of ROP group,including 7 cases of Exon18 missense mutation [c.3989C > T;p.Ala1330Val (rs3736228)],5 cases of Exon8 synonymous mutation (c.1647T > C;p.Phe549Phe),5 cases of Exon6 intronic mutation [c.1412 + 8G > A (rs4988319)],3 cases of Exon2 missense mutation [c.266A > G;p.Gln89Arg (rs41494349)],2 cases of Exon21 intronic mutation [c.4349-17C > T (rs372086596)],2 cases of Exon19 synonymous mutation (c.4089C > T;p.Asp 1363 Asp),one case of Exon9 synonymous mutation (c.1 932G > A;p.Glu644Glu),and one case of Exon16 missense mutation (c.3580C >T;p.Arg1194Cys).Three types of LRP5 mutations were found in 6 cases of the control group,including 4 cases of Exon8 synonymous mutation,one case of Exon19 synonymous mutation,and one case of Exon9 synonymous mutation.The positive rates of Exonl8 missense mutation and Exon6 intronic mutation in severe ROP group were significantly higher than the control group (P < 0.05).Conclusion LRP5 gene mutations in Wnt receptor signaling pathway may be associated with the occurrence of severe ROP.

7.Investigation of proper treatment for giant omphalocele with liver protrusion in neonates

Objective:To investigate the proper choice and improve the survival rate in neonates suffering from giant omphalocele (GO)with liver protrusion by analyzing the clinical characteristics, different treatments and outcomes.Methods:Neonates with GO admitted in Department of Neonatal Surgery in Wuhan Children′s Hospital were retrospected from January 2009 to May 2019.The treatment methods include: (1) repaired with similar silo-bag by staged operation; (2) one-stage repair with patch or not; (3) delayed repair by conservative treatment for some time (from 2 to 8 weeks) firstly.Data included gestational age, gender, birth weight, average diameter of omphalocele, contents of omphalocele, associated malformation, treatment method, duration of ventilator use and clinical outcome.These neonates were divided into 2 groups according to the contents of protrusion: group with liver protrusion and group with liver and other organs protrusion.Clinical data were summarized and compared between different groups.Results:Sixteen cases were collected, including 9 males and 7 females.The average age was (1.25±0.45) d (1-2 days), the average birth weight was (2.48±0.37) kg, and pregnant week was (36.23±1.17) weeks.The average diameter of the omphalocele was (9.88±3.30) cm, ranging from 5 cm to 15 cm.Seven cases were belonged to the group with liver protrusion, cases undergone one-stage repair, delayed repair were 6 cases and 1 case, respectively.There were 9 cases in group with liver and other organs protrusion, and the protruded organs included liver, intestine, colon or spleen.Among them, cases needed one-stage repair, delayed repair and silo-bag repair were 3 cases, 3 cases, 3 cases, respectively.Cases needed respiratory machine were 3 cases (42.8%) and 8 cases (88.89%) in group with liver protrusion and group with liver and other organs protrusion, respectively.Neonates had a longer time of needing respiratory machine in group with liver protrusion, comparing with the neonates in group with liver and other organs protrusion[(30.67±19.0) h vs.(106.25±69.36) h, P=0.021], and the risk rate to use respiratory machine was 5.143(95% CI: 0.727-36.368). There were no dead cases in group with liver protrusion.Three cases were dead in group with liver and other organs protrusion.All the patients were followed from 3 months to 4 years, and hernia in abdominal wall was found in 5 cases, including 1 case in group with liver protrusion and 4 cases in group with liver and other organs protrusion.If the dead cases had been excluded, the rate of hernia was 66.7% (4/6 cases) in group with liver and other organs protrusion. Conclusions:Individual treatment should be emphasized in neonates suffering from GO with liver protrusion.Neonates with single liver protrusion have a better prognosis than those with liver and other organs protrusion.One-stage repair is recommended in the former, and the delayed repair is highly recommended in the latter.The accessory liver should be considered when GO neonates presenting liver protrusion.
8.Effects of S-3307 on the yield and main ingredients of Alisma plantago-aquatica.

UNLABELLEDTo study the effect of S-3307 on the yield and main ingredients of Alisma plantago-aquatica.
METHODThe contents of 24-acetyl alisol A and the 23-acetyl alisol B in tuber were determined by HPLC.
RESULTSThe contents of 24-acetyl alisol A and the 23-acetyl alisol B as well as yield were significantly increased in all groups applied with different concentrations of S-3307 comparing with control group. The optimal concentration of S-3307 was 80 mg x kg(-1). The residues of S-3307 was detected under 0.316 8 mg x kg(-1) (detecting limit).
CONCLUSIONThe optimal concentration of S-3307 is 80 mg x kg(-1), it reached the best result when applied 36 d after seedling.

9.Effects of S-3307 on photosynthesis of Ligusticum chuanxiong.

OBJECTIVETo study the effects of S-3307 spraying time and density on photosynthetic characteristic of Ligusticum chuanxiong.
METHODThe photosynthetic characteristic of L. chuanxiong under different S-3307 spraying time and density was studied by plot cultivation experiment.
RESULTThe content of chlorophyll a and chlorophyll b in leaf increased when the spraying density was 20, 40, 80 mg x L(-1), while the net photosynthetic rate was the maximum. When the spraying density was 160 mg x L(-1), the content of chlorophyll a and chlorophyll as well as net photosynthetic rate were not increased.
CONCLUSIONS-3307 spraying can raise the photosynthetic capacity of L. chuanxiong and promote the form of assimilation products.

The chemical constituents of 95% ethanol extract of Melastoma dodecandrum were isolated and purified by chromatography on silica gel, Sephadex LH-20, and HPLC, to obtain thirteen compounds eventually. On the basis of their physico-chemical properties and spectroscopic data, these compounds were identified as quercetin (1), quercetin-3-O-β-D-glucopyranoside (2), quercetin-3-O-(6"-O-p-coumaroyl) -β-D-glucopyranoside (3), kaempferol (4), kaempferol-3-O-β-D-glucopyranoside (5), kaempferol-3-O- [2",6"-di-O-(E)-coumaroyl]-β-D-glucopyra-noside (6), luteolin (7), luteolin-7-O-(6"-p-coumaroyl) -β-D-glucopyranoside (8), apigenin (9), apigenin-7-(6"-acetyl-glucopyranoside) (10) , naringenin (11), isovitexin (12), and epicatechin-[8,7-e] -4β-(4-hydroxyphenyl)-3,4-dyhydroxyl-2(3H)-pyranone (13). Eight compounds(3,5,6,8-11 and 13) were obtained from M. dodecandrum for the first time.
