1.Response surface methodology applied to the optimization for supercritical CO2 extraction of the lipids from tempeh

Objective To optimize the supercritical carbon dioxide(SC-CO2)extraction of lipids from tempeh(TE-C)and further improve the lipid classes ratio. Methods The experimental parameters of SC-CO2 extraction including extraction temperature, pressure,and moisture content of tempeh were optimized using a Box-Behnken design combined with response surface methodology (RSM),according to the weighted extraction ratio of TE-C and lipid classes after the experimental results of single factors. Detailed chemical compositions of TE-C obtained by optimum conditions of SC-CO2 extraction were analyzed by high performance liquid chroma?tography with an evaporative light-scattering detector(HPLC-ELSD)and high performance liquid chromatography-atmospheric pres?sure chemical ionization mass spectrometry(HPLC-APCI-MS). Results TE-C was composed of three lipid classes:fatty acids(Ⅰ), diacylglycerols(Ⅱ)and triacylglycerols(Ⅲ). The optimum SC-CO2 extraction conditions of TE-C were 50℃extraction temperature, 25 MPa pressure,1.99%moisture content of tempeh and 1.5 hour extraction time. Conclusion The optimum value of RSM for SC-CO2 extraction was(5.97±0.15)g/100 g.
2.Studies on chemical constituents of Ervatamia hainanensis.

OBJECTIVETo study the chemical constituents of Ervatamia hainanensis.
METHODThe compounds were separated and purified by column chromatography with silica gel, and identified by IR, MS, NMR and 2D-NMR.
RESULTFive compounds were identified as I (isolariciresinol 9-O-beta-D-glucopyranoside), II (cycloartenol), III (beta-amyrin acetate), IV (beta-sitosterol), V (daucosterol), respectively.
CONCLUSIONAll the compounds were isolated from this plant for the first time.

3.Expression of connexin 36 in central nervous system and its role in epileptic seizure.

OBJECTIVEThis review discusses the experimental and clinical studies those show the expression of connexin 36 in the central nervous system and the possible role of connexin 36 in epileptic seizure.
DATA SOURCESAll articles used in this review were mainly searched from PubMed published in English from 1996 to 2012.
STUDY SELECTIONOriginal articles and reviews were selected if they were related to the expression of connexin 36 in the central nervous system and its role in epilepsy.
RESULTSThe distribution of connexin 36 is developmentally regulated, cell-specific and region-specific. Connexin 36 is involved in some neuronal functions and epileptic synchronization. Changes in the connexin 36 gene and protein were accompanied by seizures. Selective gap junction blockers have exerted anticonvulsant actions in a variety of experiments examined in both humans and experimental animals.
CONCLUSIONSConnexin 36 plays an important role in both physiological and pathological conditions in the central nervous system. A better understanding of the role of connexin 36 in seizure activity may contribute to the development of new therapeutic approaches to treating epilepsy.

4.Effect of the chelator Zn-DTPA on the excretion of lead in lead intoxication mice detected with ICP-MS.

To study the lead excretion effect of the chelator Zn-DTPA on the lead intoxication mice, inductively coupled plasma mass spectrometry (ICP-MS) was applied to detect the lead content of biological samples. The acute lead intoxication mice model was established by injecting lead acetate intraperitoneally with the dose of 1 mg. Zn-DTPA was administered intraperitoneally to mice once daily for five consecutive days 4 h after intoxication. Control group, model group, combination of Zn-DTPA and Ca-DTPA group were evaluated at the same time. The urine was collected every day. The mice were sacrificed in batches in the 2rd, 4th, 6th day. Biological samples including urine, whole blood, femur and brain were prepared and nitrated. Lead concentration was detected by ICP-MS. The result showed that Zn-DTPA could increase lead content in urine markedly and reduce lead content in blood, femur and brain.

5.The changes and impact factors of carotid-femoral and carotid-radial pulse wave velocity in patients with essential hypertension.

OBJECTIVETo analyze the changes of carotid-femoral and carotid-radial pulse wave velocity in patients with essential hypertension, and effects of age and levels blood pressure levels on these changes.
METHODSAutomatic pulse wave velocity (PWV) measuring system was applied to examine carotid-femoral PWV (CFPWV) and carotid-radial PWV (CRPWV) as the parameters reflecting central elastic large arterial and peripheral muscular medium-sized arterial elasticity respectively. 517 hypertensive patients aged 17-82 years (272 males and 245 females, mean age 52.0 +/- 13.0 years) and 118 healthy subjects aged 19-82 years (52 males and 66 females, mean age 54.2 +/- 13.8 years) were recruited in the study.
RESULTSCFPWV increased with age in both groups (P < 0.001), whereas CRPWV did not change significantly. CFPWV and CRPWV both increased with blood pressure. A stepwise multiple regression analysis demonstrated that age and systolic blood pressure were positively related to CFPWV independently, while diastolic blood pressure was significantly associated with CRPWV.
CONCLUSIONSAge and components of blood pressure exert different effects on elasticity of central elastic large artery and peripheral muscular medium-sized artery. Examining the change of large arterial elasticity has important clinical value in hypertensive patients.

6.Transmembrane transport behavior of in vitro HepG2 cells of ananas and its effect on lipids and glucose distribution.

Pineapple (Ananas comosus) leaves contain mainly phenolic components with antioxidant and hypolipidemic effects. One of the principle components is p-coumaric acid. In this study, the transport behavior of p-coumaric acid, was observed after the administration of pineapple leaf phenols in vitro. Simultaneously, the effect of the phenols on glucose, total cholesterol and triglycerides transportation and metabolism in HepG2 cells was also observed. The results showed that the phenols had good transport characteristics. 5 min after the administration, p-coumaric acid of the phenols could be detected, and the content of p-coumaric acid reached the peak concentration after 60 min of the administration. p-coumaric acid of phenols have time-and dose-dependent manner. While promoting glucose transporter (GLUT4) and low density lipoprotein receptor (LDLR) expression, the phenols decreased intracellular lipid content. This reduction of intracellular lipid content was highly correlated with the promotion of lipoprotein lipase (LPL) and hepatic triglyceride lipase (HTGL) expression, while the reduction of intracellular glucose levels was correlated with glycogen synthesis in the cells.

7.Difference of gene expression between the central and the peripheral epithelia of the bovine lens.

BACKGROUNDEquatorial lens epithelial cells proliferate and differentiate into fiber cells throughout life, while central lens epithelial cells proliferate little and do not form fiber cells. This study aimed to investigate the differences in gene expression between the central and the peripheral epithelial cells of the bovine lens.
METHODSLens epithelia were dissected into central ( RESULTSBy microarray analysis, 67 transcripts were at least two-fold lower and 269 at least two-fold higher in pLEC compared with that in cLEC. Thirty-four protein spots, including 20 in cLEC and 14 in pLEC, were identified by two dimensional electrophoresis and mass spectrometry. Of these 34 protein products, 28 were represented by probe sets on the microarray. Nine transcripts changed in the same direction and four transcripts in the opposite direction to their protein products. Immunoanalyses revealed that three (mitogen-activated protein kinase 1 (MAPK1), nidogen (NID), small nuclear ribonucleoprotein N (SNRPN)) out of four transcripts with opposite change between 2-DE and microarray assay showed the same changes as the results of 2-DE gel analyses. The genes differently expressed between cLEC and pLEC mainly include those related to the MAPK, transforming growth factor beta (TGFbeta) signaling and glycolysis pathways. CONCLUSIONThe results suggested that there were distinctly different genome activities, including a specific group of pathways, between central and peripheral lens epithelial cells.

8.A study on the risk factors of cardiovascular between hypertensive patients and normal subjects in the same family.

OBJECTIVEThrough studying the difference between the hypertensive patients and those with normal blood pressure who all from the same hypertensive pedigree, we tried to find the factors which would decrease the risk of hypertension.
METHODSHypertensive patients, when coming to the cardiovascular clinic of Xuan Wu Hospital from 2003 to 2004, were defined as the members of the hypertensive pedigrees through inquiry. 84 families including 368 persons, with 192 syblings were involved. Metabolic syndrome could be defined by the criterion of adult treatment panel III.
RESULTSWhen compared with normal blood pressure persons, clinical examinations of the hypertensive patients had an higher levels of triglyceride, total cholesterol, glucose and body mass index (BMI). The hypertensive group showed higher ratios among male patients, smokers, alcohol drinkers, having irritable personality and high-salt-intake. Through logistic regression, overweight, smoking and irritable temperament showed positive relations with hypertension. The rate of metabolic syndrome was higher in hypertension group.
CONCLUSIONIt is important that either persons with normal blood pressure or hypertension should control their body weights (BMI) since the rate of metabolic syndrome in hypertension group was high.

9.Genomic diversity of the Avian leukosis virus subgroup J gp85 gene in different organs of an infected chicken.

The genomic diversity of Avian leukosis virus subgroup J (ALV-J) was investigated in an experimentally infected chicken. ALV-J variants in tissues from four different organs of the same bird were re-isolated in DF-1 cells, and their gp85 gene was amplified and cloned. Ten clones from each organ were sequenced and compared with the original inoculum strain, NX0101. The minimum homology of each organ ranged from 96.7 to 97.6%, and the lowest homology between organs was only 94.9%, which was much lower than the 99.1% homology of inoculum NX0101, indicating high diversity of ALV-J, even within the same bird. The gp85 mutations from the left kidney, which contained tumors, and the right kidney, which was tumor-free, had higher non-synonymous to synonymous mutation ratios than those in the tumor-bearing liver and lungs. Additionally, the mutational sites of gp85 gene in the kidney were similar, and they differed from those in the liver and lung, implying that organ- or tissue-specific selective pressure had a greater influence on the evolution of ALV-J diversity. These results suggest that more ALV-J clones from different organs and tissues should be sequenced and compared to better understand viral evolution and molecular epidemiology in the field.

10.Seroprevalence and Risk Factors of Fascioliasis in Yaks, Bos grunniens, from Three Counties of Gansu Province, China.

The aim of this study was to determine the seroprevalence and risk factors of fascioliasis in yaks, Bos grunniens, from 3 counties of Gansu Province in China. A total of 1,584 serum samples, including 974 samples from white yaks from Tianzhu, 464 from black yaks from Maqu, and 146 from black yaks from Luqu County, were collected and analyzed using ELISA to detect IgG antibodies against Fasciola hepatica. The overall F. hepatica seroprevalence was 28.7% (454/1,584), with 29.2% in white yaks (284/974) and 27.9% in black yaks (170/610). The seroprevalence of F. hepatica in yaks from Tianzhu, Luqu, and Maqu was 29.2%, 22.6%, and 29.5%, respectively. Female yaks (30.9%) had higher F. hepatica seroprevalence than male yaks (23.4%). Also, F. hepatica seroprevalence varied by different age group from 24.1% to 33.8%. Further, the seroprevalence ranged from 21.8% to 39.1% over different seasons. Interestingly, the season and age of yaks were associated with F. hepatica infection in yaks in the investigated areas. These findings provided a basis for further studies on this disease in yaks from 3 counties of Gansu Province in northwestern China, which may ultimately support the development of effective control strategies of fascioliasis in these areas.
