1.Effect of 5-lipoxygenase and 12-lipoxygenase on proliferation and apoptosis of pancreatic cancer cell

Objective To examine the expression of lipoxygenases (LOXs)on human pancreatic carcinoma and their effects on proliferation and apoptosis of human pancreatic carcinoma in vitro.Methods Expression of 5- LOX and 12-LOX in the tissue of human pancreatic carcinoma and pancreatic cancer cell line (SW1990) was detected by immunohistochemical staining and RT-PCR.To examine the effects of polyunsaturated fatty acids on pancreatic cancer cell line,the proliferation rate of SW1990 cells treated with arachidouic acid (AA),docosahexaenoic acid (DHA) and eicosapentenoic acid (EPA) was analyzed by MTT and the incorporation of bromodeoxyuridine (BrdU) methods.An annexinV/propidium iodide (PI) assay with flow cytometry and in situ terminal deoxynucleotidyl transferase-mediated dUTP-digoxigenin end labeling (TUNEL) assay were used to identify potential induction of apoptosis by the unsaturated fatty acids.The effects of 5-LOX inhibitor (MK-886) and 12-LOX inhibitor (baicalein) on the proliferation of SW1990 cells were determined by above methods.Results Both 5-LOX and 12-LOX were over expressed in the tissue of human pancreatic carcinoma and the pancreatic cancer cell line.AA had a stimulatory effect on the growth of SW1990 cells while EPA and DHA had an inhibitory effect in a dose-dependent manner.The apeptosis of SW1990 cells co-cultured with DHA or EPA lasting for 24 hours was increased markedly.Furthermore, both MK-886 and baicalein in a dose-dependent-manner inhibited the growth of SW1990 cells ard induced a significant increase of apoptotic cells rate.Conclusions Expression of lipoxygenase is up-regulated in human pancreatic cancer. Polyunsaturated fatty acids have regulatory effects on the growth of pancreatic carcinoma in vitro.AA stimulates proliferation of pancreatic cancer cell line,while DHA and EPA suppress proliferation,simultaneously inducing apoptosis of pancreatic cancer cell line.Blocking the functions of lipoxygenase with its inhibitor exerts a negative regulatory effect on the growth of pancreatic carcinoma in vitro.Lipoxygenase might be a novel therapeutic target for the pancreatic cancer.

Objective:To evaluate the clinical efficacy of benazepril in combination with valsartan in treating chronic kidney patients with proteinuria and hypertension.Method:75 chronic kidney patients were divided into benazepril group(n=28)and valsartan group(n=26)and combination treatment group(n=21).The changes of 24 hours urinary protein excretion and blood pressure and serum BUN and SCr were observed before the treatment and 4 and 8 weeks after it. Objective Target-controlIed-infusion(TCI) system makes administration of intravenous anesthetics simpler, more accurate and adjustable. Bispectral index (BIS) is generally accepted as a measurement of depth of sedation and anesthesia. The aim of this study was to evaluate the usefulness of BIS as a feedback control in propofol anesthesia with TCI for patients undergoing laparoscopic cholecystectomy. Methods Forty ASA Ⅰ - Ⅱ patients (12 male, 28 female) aged between 20-70 yr were randomly allocated to one of two groups of twenty each: feedback TCI group and TCI group. In feedback TCI group, BIS value of 50 was set as the control variable. A bolus of 3 ?g?kg-1 fentanyl was given Ⅳ 3 min before induction. The target concentration of propofol was set at 3 ?g?ml-1 throughout anesthesia. Real time BIS, the highest and lowest systolic and diastolic pressure during operation, doses of adjunctive drugs, time for emergence and orientation, awareness during operation and the standardized unit dose of propofol were recorded and compared between groups. The term epigenetics refers to a number of biochemical modifications of chromatin that, without altering the primary sequence of DNA, play a role in genomic regulation and in control of particular gene expression. These modifications involve several kinds, such as DNA methylation, histone code modifications and chromatin remodeling. It is accepted that these modifications is as common in solid tumors as it is in hematologic malignancies such as myelodysplastic syndromes(MDS). So many specialists suggest that the study of this area is very valuable. The current clinical information regarding different forms of epigenetic therapy in patients with MDS were focused in this review.

? Choroidal neovascularization is the primary pathogenesis of neovascularage - related macular degeneration ( nAMD ) , and the role of vascular endothelial growth factor ( VEGF ) in neovascularization has been widely recognized. Currently, drugs target different targets of VEGF have been widely used in the treatment of nAMD. As a subtype of VEGF, placental growth factor ( PlGF) has synergistic effects with VEGF-A on promoting angiogenesis, stimulating the migration of endothelial cell proliferation and mediating immune inflammatory response. There is no expression of PlGF in mature blood vessels so PlGF hashigh specificity. ln this paper, the role of PlGF in the pathogenesis and treatment of nAMD is reviewed.
9.Effect of Enhanced Physical Therapy with Integrated Traditional and Western Medicine on Muscular Weakness after Selective Posterior Rhizotomy

Objective To explore the effect of enhanced physical therapy with integrated traditional and western medicine on muscularweakness after selective posterior rhizotomy. Methods 44 children with muscular weakness after selective posterior rhizotomy were dividedinto the treatment group (n=28) and the control group (n=16). Exercise therapy, physical therapy, electroacupuncture and Chinese massagewere applied to the treatment group, while exercise therapy was applied to the control group only. The muscle strength of the leg of childrenwere compared right after the surgery and 2 weeks after the surgery. Results The muscle strength of the leg of children in treatment group recoveredbetter than the control group 2 weeks after the surgery (P<0.05), and it was almost at the same level with that before surgery (P<0.05). Conclusion The enhanced physical therapy with integrated traditional and western medicine method could rapidly recover the musclestrength of the leg of children after SPR.
10.Correlation of metabolic syndrome with recurrence of urolithiasis

Objective To investigate the relationship between metabolic syndrome (MS) and recurrence of urolithiasis.Methods A retrospective analysis was performed in urinary stone patients from March 2008 to February 2012.Patients were divided into MS group and non-MS group according to the diagnose criteria of metabolic syndrome (2007 version) by the joint committee for developing Chinese guidelines on prevention and treatment of dyslipidemia in adults.The patients were followed up for 24-72months (median 47 months) since operation.The difference of stone recurrence was compared between the 2 groups.Results Two hundred and eighteen patients with urinary stone disease were enrolled.Of them,52 patients were diagnosed with MS.Stone recurrence occurred in 29 patients (55.8%) of MS group,whereas 66 patients (39.8%) of non-MS group suffered stone recurrence.It demonstrated the median recurrence free survival of group MS and non-MS was 36 months and 59 months by Kaplan-Meier analysis,respectively (Log-rank test,P =0.019).Multivariate Cox regression analysis results revealed that MS was significantly associated with stone recurrence (HR 1.817,95% CI 1.105-2.988,P =0.011),however,the gender (P =0.336),age (P =0.246) and recurrence urolithiasis at first visit (P =0.051) were not associated with stone recurrence.Conclusions MS is an independent risk factor for urinary stone recurrence.It is suggested that the treatment of MS may have a important role in prevention of stone recurrence in MS patients with urolithiasis.
