1.Effects of Adiponectin on Cell Activity and Expression of T-cadherin in Human RPE Cells Cultured with High D-glucose

Objective To investigate the effects of adiponectin on cell activity and expression of T-cadherin in human retinal pigment epithelial(hRPE) cells cultured with high glucose.Methods ①The hRPE cells were divided into three groups:normal glucose group(5.5mmol/L glucose),high glucose group (33mmol/L glucose),high glucose plus adiponectin group (33mmol/L glucose+2.5ug/ml adiponectin).48h later,the cell activity was observed by MTT assay.②After the above groups were cultured respectively for 24h,48h,72h again,the expression of T-cadherin mRNA was observed by RT-PCR. Results ①Cell activity in high glucose group significantly decreased. Cell activity adiponectin group was higher than that in the high glucose group.②With the extension of time ,the expression of T-cadherin decreased in high glucose group,but increased in adiponectin group. Conclusion Adiponectin may be protect the hRPE cells by T-cadherin.
2.Expression of c-met stimulated by high glucose in human renal tubular epithelial cells and its implication.

The expression of c-met stimulated by high glucose in human renal tubular epithelial cells and the role of HGF/c-met system in diabetic nephropathy were examined. The proximal tubular epithelial cells were cultured in vitro under different conditions. MTT was used for the detection of cellular proliferation and RT-PCR was employed for measurement of c-met mRNA level. Our results showed that under different conditions, there were no significant differences in the proliferation of proximal tubular epithelial cells 12 h and 24 h after the culuture (P>0.05). The proliferation of proximal tubular epithelial cells showed a significant change 96 h after the culture and the cellular proliferation induced by hepatocyte growth factor (HGF) was very active (P<0.05). Moreover, no significant difference in the expression of c-met mRNA was found 12 h after the culture under different conditions (P>0.05), while 24 and 96 h after the culture, a persistent and significantly higher expression of c-met mRNA was found in HGF-induced proliferation. It is concluded that addition of exogenous HGF could inhibit the apoptosis induced by high-level glucose, promote the proliferation of proximal tubular epithelial cells, and induce the expression of c-met. Our study suggests that local up-regulation of HGF/c-met system plays an important role in the repair of renal damage in diabetic nephropathy.
3.The analysis of cisplatin resistance in ovarian cancer treatment

Objective To screen the target genes that contribute to cisplatin resistance in ovarian cancer treatment. Methods Gene expression and methylation profiles of ovarian cancer cells that were sensitive or resistant to cisplatin with accession number GSE15709 were downloaded from GEO database. Differential expressed and methylated genes were identi?fied through associating packages in R. DAVID database to screen the enriched GO terms and pathways of the different ex?pressed genes between A2780 and A2780/DDP. Gene Set Enrichment Analysis (GSEA) of different gene was performed against DAVID database. Genes that exhibited difference in both expression and methylation profiles between the two types of ovarian cancer cells as well as genes that present contradictory profile between expression and methylation were verified via qRT-PCR. Results We found 416 different expressed genes and 281 methylated genes between the two types of ovari?an cancer cells respectively. These differential genes were rich in pathways of cell cycle, DNA replication, nucleus division , p53 signaling , and negative regulation of protein modification process etc. Four genes demonstrated contradictory profile be?tween expression and methylation in the two types of ovarian cancer cells and were verified by qRT-PCR. Conclusion Combination of bioinformatics and molecular biology is useful in the identification of target genes that contribute to resis?tance of cisplatin in ovarian cancer treatment and further reveal molecular mechanism behind it.
4.Experimental study of nifedipine inhibitory effect on calcium oxalate crystallization in rats

Objective To determine the effect of nifedipine on the formation of nephrolithiasis in rat models and its mechanism of action. Methods A total of 60 male Sprague-Dawley rats (body weight, 200-250g) were randomly divided into 6 groups, ie, control group, only nifedipine group, stone forming group, stone forming plus 3, 6, 10 mg?kg-1?d-1 nifedipine group. Each group is 10 rats respectively. Hyperoxaluria and calcium oxalate crystals were produced in rats by ethylene glycol in drinking water. Four weeks later, all rats were sacrificed and the calcium oxalate crystallization in kidney, the renal free radical level, the renal cell apoptosis indexes and the blood and urine biochemical indexes were detected. 5.Effects of ERC61-Extract from Rhizoma Curcumae on Hematopoietic Functions in Mice Treated by Chemotherapy

OBJECTIVE:To explore the effects of ERC61,an extract from Rhizoma Curcumae on hematopoietic functions of mice pretreated by cyclophosphamide(CTX) and the proper regime of the administration of ERC61.METHODS:The experimental mice were treated with CTX(100mg? kg-1? d-1) by ip for 3 consecutive days to establish leucopenia model.ERC61 was ip administered to the prevention group,the treatment group,and the combined group(prevention+ treatment).The numbers of blood leucocytes and bone marrow cell were counted by conventional methods;spleen index was measured as well.RESULTS:ERC61 enhanced the count of leucocytes and nucleated cells in bone marrow and improved the level of spleen index.CONCLUSION:ERC61 is able to improve the disorders of hematopoietic functions induced by CTX,and the combination therapy of prevention and treatment is the optimum one in efficacy.
6.Determination of Curdione in Nano-micelles Loaded with Curdione by UV-Vis Spectrophotometry

OBJECTIVE:To establish a UV-Vis spectrophotometric method for content determination of Curdione in Nano-micelles loaded with Curdione.METHODS:Curdione was colored by sulfuric acid-vanillin reagent.The absorbance was measured by visible-spectrophotometry at a wavelength of 500nm.RESULTS:The content of Curdione in the range of 0.050~ 0.400mg? mL-1 was in accordance with the Lambert-Beer law(r=0.999 3).The average recovery was 99.14%,and the RSD was 1.23%(n=9).CONCLUSION:This method is simple,accurate and suitable for the content determination of curdione in Nano-micelles loaded with Curdione.

Objective The recurrence and metastasis of malignant tumor could affect the survival of patients. Early detection of cancer cells in peripheral blood and effective therapy promptly are of clinical importance. The present study was to investigate the carcinoembryonic antigen (CEA) mRNA expression and serum CEA level in blood from patients with colorectal cancer and to evaluate its clinical significance. Methods Peripheral blood samples were collected from 35 eligible patients with colorectal cancer and 9 post operative patients. CEA mRNA was detected by reverse transcriptase polymerase chain reaction (RT PCR) and serum CEA was detected by time resolved fluorometric methods. Results The positive rate of CEA mRNA in blood samples of 35 patients with colorectal cancer was 45.7%, and 2.7% in healthy volunteers. Two of the nine post operative patients (22.2%) showed CEA mRNA positive. The positive rate increases with the pathological staging and showed no significant relation with the malignant extent of cell differentiation. Serum CEA mRNA positive rates in patients with CEA positivity (72.2%) were significantly higher than those in patients with negative CEA mRNA expression (42.3%).Conclusion The expression of CEA mRNA in peripheral blood cells correlates with the pathological staging of colorectal cancer and it could be of potential use to monitor the micrometastases of tumor. Long term follow up is needed to evaluate its clinical significance.
9.Characteristics and outcome of pediatric in-hospital cardiopulmonary resuscitation

Objective To investigate the present situation and outcome of cardiopulmonary resuscitation (CPR) for pediatric in-hospital cardiopulmonary arrest and to analyze the prognostic risk factors.Methods Data were collected from October 2008 till October 2011 using Ustein style.Patients older than 28 days who received CPR were evaluated.Returning of spontaneous circulation(ROSC) more than 24 hours was the primary outcome.Neurological outcome was assessed by pediatric cerebral performance categories half year after discharge.Results Of the 36 patients,15 (41.7%) achieved sustained ROSC.Seven (19.4%) patients survived to discharge.Single factor analysis indicated that the beginning heart rhythm,defibrillation and original disease were significantly different between the two groups(P <0.05).The beginning heart rote of the patient in ROSC > 24 h group was mostly sinus bradycardia.Patients who need defibrillation had bad prognosis.Patients with heart disease had a lower rate of ROSC > 24 h.At half year follow-up study,4 patients had 1 or 2 score,1 patient had 4 and 1 had 5 score in the pediatric cerebral performance categories scales.Condusion The successful rate of CPR in our hospital was the same as that in developed country.The beginning heart rhythm,defibrillation and original disease were associated with the outcome.Most of the patients who survived to discharge had a good neurological outcome.
10.Effects of Adenosine and the Selective Adenosine Receptor Agonist on the Expression of Interleukin-18 in Patient with Unstable Angina

Objective To observe the effects of adenosine and adenosine receptor agonist on the expression of inter?leukin-18 (IL-18) in patients with unstable angina (UAP), and the mechanism of adenosine receptor agonists thereof. Meth?ods Fifteen UAP and 15 healthy volunteers were included in this study. The effects of adenosine and the selective adenos?ine receptor agonists on the expression of IL-18 were measured by enzyme-linked immunosorbent assay (ELISA). Results The lower concentration of adenosine (1-100μmol/L) increased the expression of IL-18 in UAP group;whereas the higher concentration of adenosine (1 mmol/L) inhibited the expression of IL-18. The adenosine A1 receptor agonist and A3 receptor agonist increased the expression of IL-18, while the adenosine A2a receptor agonist inhibited the expression of IL-18. There was no significant effect for A2b receptor agonist on the expression of IL-18. Conclusion The lower concentrations of ade?nosine can enhance the expression of IL-18 through adenosine A1 and A3 receptors in UAP. The higher concentrations of ade?nosine can inhibit the expression of IL-18 through adenosine A2a receptors. Adenosine can promote IL-18 expression in lower concentrations.
